Morphological, physiological and metabolomic analysis to unravel the adaptive relationship between root growth of ephemeral plants and different soil habitats

Mengwen Peng,Hao He,Meng Jiang,Zhongke Wang,Guifang Li,Li Zhuang

To gain insights into the adaptive characteristics of ephemeral plants and enrich their potential for resource exploitation, the adaptive changes in two highly dominant species ( Malcolmia scorpioides and Isatis violascens ) to soil habitats (aeolian soil, AS; grey desert soil , GS) were investigated from the aspects of root morphology, physiology, and metabolism in this study. The results revealed that changes in root morphology and enzyme activity were affected by soil habitat. Total root length (TRL), root volume (RV) and root surface area (RSA) were higher in GS than in AS. The levels of proline (Pro), glutathione (GSH), soluble sugar (SS), and lysine (Lys) were higher in GS than in AS. Untargeted LC–MS metabolomics indicates that root metabolites of both species differed among the two soil habitats. Root responses to different soil habitats mainly affected some metabolic pathways. A total of 780 metabolites were identified, common differential metabolites (DMs) in both species included amino acids , fatty acids, organic acids, carbohydrates, benzene and derivatives, and flavonoids , which were mainly involved in carbohydrate metabolism , amino acid metabolism , flavonoid biosynthesis and fatty acid metabolism , and their abundance varied among different habitats and species. Some key DMs were significantly related to root morphology and enzyme activity, and indole, malonate, quercetin , uridine , tetrahydroharmine , and gluconolactone were important metabolites associated with root growth. Therefore, the response changes in root growth and metabolite of ephemeral plants in response to soil habitats reflect their ecological adaptation, and lay a foundation for the exploitation of plant resources in various habitats.

Foliar-applied lanthanum chloride increases growth and phosphorus acquisition in phosphorus-limited adzuki bean seedlings

Lian Huida,Qin Cheng,Zhang Cong,Yan Minfei,Li Hongbing,Zhang Suiqi

Aims Lanthanum, a rare earth element, is used in agriculture and forestry to promote plant growth and productivity. We aimed to investigate its role in altering the roots and alleviating P deficiency in the adzuki bean Vigna angularis . Methods The adzuki bean variety “Jin No. 5” was pre-cultivated in sand. Fourteen-day-old seedlings were sprayed twice with 0.4 mM LaCl 3 solution. Next, we treated the seedlings with half-strength Hoagland solution containing P reduced to 1% of its original concentration (P-deficient); P reduced to 5% of the original (P-marginal); and unadjusted solution (P-sufficient). We measured plant growth, root structure, root hydraulic conductivity, P content, gas exchange, and photosynthetic pigment levels in all groups. Results LaCl 3 application significantly increased leaf P concentration, effectively improving the P uptake efficiency of roots, shoots, and leaves under severe P stress. Root morphology and acid phosphatase (APase) activity varied significantly in response to LaCl 3 under low-P stress. A greater leaf area, gas exchange ability, chlorophyll content, and carotenoid content were observed in the presence of LaCl 3 under P-marginal stress. Conclusions The application of LaCl 3 can promote growth and P acquisition in adzuki beans under P limitation by inducing changes in root morphology, APase activity, and hydraulic conductivity.

Effects of a soil collembolan on the growth and metal uptake of a hyperaccumulator: Modification of root morphology and the expression of plant defense genes

Liming Pu,Zhu Li,Mingyun Jia,Xin Ke,Hongyan Liu,Peter Christie,Longhua Wu

Soil collembolans live in close proximity to plant roots and may have a role in the phytoextraction of potentially toxic metals from contaminated soils but the underlying mechanisms remain poorly investigated. We hypothesize that soil collembolans may change the root morphology of hyperaccumulators by regulating plant physiological characteristics. Here, a pot experiment was conducted in which a cadmium (Cd) and zinc (Zn) hyperaccumulator ( Sedum plumbizincicola ) was grown with or without a collembolan ( Folsomia candida ), and plant transcriptome and hormones as well as the root characteristics of S. plumbizincicola were analyzed. F. candida promoted the growth and Cd/Zn uptake of S. plumbizincicola , the root and shoot biomass increasing by 53.3 and 34.4%, and the uptake of Cd and Zn in roots increased by 83.2 and 65.4%, respectively. Plant root morphology, total root length, root tip number and lateral root number increased significantly by 40.7, 37.2 and 33.8%, respectively, with the addition of F. candida . Transcriptome analysis reveals that the expression levels of defense-related genes in S. plumbizincicola were significantly up-regulated. In addition, the defensive plant hormones, i.e. salicylic acid in the roots, increased significantly by 338%. These results suggest that the plant in defense of the action of F. candida regulated the expression of the corresponding genes and increased the defensive plant hormones, thus modifying root morphology and plant performance. Overall, this study highlights the importance of the regulation by collembolans of plant growth and metal uptake by interaction with hyperaccumulator roots.

NOM mitigates the phytotoxicity of AgNPs by regulating rice physiology, root cell wall components and root morphology

Xitong Huang,Yong Li,Ke Chen,Haiyan Chen,Fei Wang,Xiaomin Han,Beihai Zhou,Huilun Chen,Rongfang Yuan

Natural organic matter (NOM) affects the environmental behaviors of AgNPs, which may change their phytotoxicity to plants. However, more evidence can be provided to illustrate how NOM influences AgNPs-induced phytotoxicity. In this study, using rice ( Oryza sativa ) as a model, the effects of NOM, Suwannee River humic acid (SRHA) and fulvic acid (FA), on the dissolution and phytotoxicity of AgNPs were investigated. Silver ions decreased in both AgNPs and AgNO 3 solution in the presence of NOM, and the effect of SRHA was stronger than FA. Image-XRF (iXRF) results showed that Ag mainly remained in the root rather than the shoot of rice seedling exposed to AgNPs. NOM mitigated the negative effects of AgNPs and AgNO 3 on rice with lower germination inhibition rate, less chlorophyll reduction, more relative biomass and less O 2 •− content. Moreover, NOM improved root cell viability according to FDA fluorescent dye as well as maintained the normal root morphology. Interestingly, the neutral sugars content from pectin, hemicellulose 1, hemicellulose 2 and cellulose of root cell wall in AgNPs and AgNO 3 treatments differed from the control, while it was close to the regular content in AgNPs/AgNO 3 +SRHA/FA groups, which implied that NOM regulated the changes. Besides, SRHA led to less germination and less relative biomass than FA due to different chemical characters. Thus, NOM needs to be considered when studying the phytotoxicity of AgNPs.

Herbicidal activity and biochemical characteristics of the botanical drupacine against Amaranthus retroflexus L.

Hua-long YU,Ci TIAN,Rong-yan SHEN,Han Zhao,Juan YANG,Jin-gao DONG,Li-hui ZHANG,Shu-jie MA

Botanical herbicide has been a hot topic in the research and development of novel pesticides. The herbicidal activity and biochemical characteristics of the botanical compound drupacine were studied by evaluating its effects on seed germination, seedling growth, morphological and physiological characteristics of Amaranthus retroflexus . Drupacine inhibited seed germination and seedling growth, and had a median inhibition concentration (IC 50 ) value of 38.99 mg L −1 against A . retroflexus root. The α-amylase activity and soluble sugar content in treated plants were significantly lower than that of the control. The expression of α-amylase gene was dosage-dependently inhibited compared to the untreated control. This suggested that inhibition of α-amylase activity was a mode of action on seed germination. The root hairs were significantly decreased and part of the root cap fell off after treatment with drupacine. The ultrastructure observation showed that cell damage of root tips increased with the treatment time. Drupacine also increased the relative conductivity and malondialdehyde (MDA) content. Peroxidase (POD), catalase (CAT), and superoxide dismutase (SOD) activities were significantly enhanced in the treatment compared to the control. These findings indicated that the physiological and biochemical reaction changes leading to morphological and membrane injuries were the main effects of drupacine on the inhibition of seedling growth. Drupacine can be developed as a botanical herbicide.

Dual transcriptomic and metabolomic analyses provide novel insights into the role of vitamins A and B metabolism in ectomycorrhizal symbiosis between Pinus yunnanensis and Lactarius deliciosus

Kai Su,Jing Yuan,Ran Wang,Lanlan Huang,Jian Zhan,Xueqiong Zhang,Shanping Wan,Xinhua He,Hans Lambers,Fuqiang Yu,Yanliang Wang

Ectomycorrhizal (EM) fungi play important roles in nutrient cycling and plant community establishment in forest ecosystems. Effects of EM formation on global alterations of the transcriptome and metabolome during plant-fungal interaction and the key metabolites involved in EM development are largely unknown. Here, dual RNA-Seq and untargeted metabolomic analyses were used to reveal stage-specific and core responses of Pinus yunnanensis and Lactarius deliciosus during mycorrhizal colonization. We found that L. deliciosus colonization in P. yunnanensis roots induced different transcriptional changes across three interaction stages, with a small core of genes consistently regulated at all stages. Concentrations of retinol (vitamin A) and retinoic acid increased while that of B group vitamins decreased during EM formation, which was coordinately regulated by these two plant-fungus partners, with L. deliciosus possibly playing a dominant role. Exogenous retinol altered the diameter and mantle thickness of P. yunnanensis - L. deliciosus EM tips and affected host plant growth and phosphorus acquisition . In the absence of L. deliciosus, exogenous retinol increased the root diameter and the number of root tips of P. yunnanensis . Furthermore, the concentration of auxin increased, but that of abscisic acid decreased during EM formation, and the genes involved in plant hormone signal transduction were gradually activated, and auxin and cytokinin signal transduction potentially played a positive role in this EM symbiosis. In conclusion, we propose that the interaction of P. yunnanensis and L. deliciosus alters vitamin metabolism, which may further affect plant hormone biosynthesis and signal transduction, modulating root morphology and EM traits.
