Construction of multi-component CuO/ZIF-8@CNTs modified electrode for sensitive electrochemical detection of neohesperidin dihydrochalcone in liquor samples

Xiaolun Peng,Ruijie Xu,Liang Zhang,Xingyao Li,Junhui Xu,Dongyun Huang,Yazhen Wang

A sensitive electrochemical sensor was proposed for neohesperidin dihydrochalcone (NHDC) detection using copper oxide/ zinc imidazole framework 8@ carbon nanotubes modified glassy carbon electrode (CuO/ZIF-8@CNTs GCE). The multi-component CuO/ZIF-8@CNTs GCE was prepared by two-step drop-casting method and characterized by Field-emission scanning electron microscopy (FE-SEM). The effective combination of CuO and ZIF-8@CNTs enhanced the electron transfer and catalytic activity of the electrode and greatly strengthened the oxidation current of NHDC, which was about 5.6-fold that of the CuO GCE and 2.3-fold that of the ZIF-8@CNTs GCE. Inspired by this, the CuO/ZIF-8@CNTs GCE was used for NHDC detection and some crucial parameters were optimized. The reaction mechanism at CuO/ZIF-8@CNTs GCE was confirmed to involve two-step two-electron process by cyclic voltammetry (CV) measurements. Finally, the sensor achieved NHDC detection ranging from 0.03 to 10 μM using differential pulse voltammetry (DPV) technique, with a detection limit of 16.0 nM. The satisfactory recoveries of 92.9–107.8 % in liquor samples also demonstrated its practicability in actual samples, matching well with that of high-performance liquid chromatography (HPLC). This work not only designs a simple detection method for NHDC but also opens up a paradigm for leveraging multi-component MOF composite electrode materials for food safety monitoring.

Ratiometric fluorescence sensing based on rare-earth upconversion nanoparticles for the rapid identification of antioxidant capacity

Shuwen Li,Zhiguo Wang,Siyi Chen,Jialin Gu,Yanping Ma,Jiangang Jiang,Rui Zhang,Dan Zhang,Yiping Wang,He Wang

Based on the internal filtration effect between upconversion nanoparticles and square acid-FeIII(SA-FeIII), this method was developed for the rapid identification of total antioxidant capacity of different fruits without complex modification on the surface of upconversion nanoparticles. The near-infrared light excitation of rare-earth upconversion can effectively avoid food fluorescence and the photobleaching effect; thus, the components to be detected in the samples can be monitored real time without a complex extraction process. Due to the instability of the antioxidants at room temperature, the short time required for this reaction (6 min) will facilitate the study of the true antioxidant power of the samples. This method is simple and responds well to the main antioxidants of fruits and vegetables with a detection limit of 0.1 μmol L−1, such as ascorbic acid, polyphenols, and anthocyanin, and it shows good linearity in the concentration range of 0–1.0 mmol L−1.

Methylation, Hydroxylation, Glycosylation and Acylation Affect the Transport of Wine Anthocyanins in Caco-2 Cells

Yang Liu,Jiali Lin,Tiantian Cheng,Yangjie Liu,Fuliang Han

Anthocyanins are substances with multiple physiological activities widely present in red wine, but the influence of structure (methylation, hydroxylation, acylation, glycosylation) on the transport remains ill-defined. In the present study, Caco-2 monolayers were used as an in vitro model of the absorptive intestinal epithelium to transport different types of anthocyanin samples. Results showed that both methylation and acetylation promote the level of transport. Monoglycoside standard exhibited higher transport amount and rate compared to diglycoside standard while the transport level of the monoglycoside mixture was unexpectedly lower than that of the diglycoside mixture. Caco-2 monolayers appeared to be more capable of transporting the single standard than the mixed standard. Meanwhile, the transport of anthocyanins in Caco-2 cell model showed time- and concentration-dependent trends. Anthocyanin treatment had a greater effect on sodium-dependent glucose transporter 1 (SGLT1) mRNA expression than glucose transporter 2 (GLUT2), and significantly down-regulated the protein expression of SGLT1. Although the low bioavailability of anthocyanins requires much more research, further evidence of the role of structure is provided.Keywords:wine;anthocyanins;Caco-2 cell monolayer;absorption;SGLT1;GLUT2

Nanoparticles loaded with phlorizin fabricate a fortified yogurt with antioxidant potential

Jing Zhang,Yali Song,Xia Hu,Zhen Zeng,Jie Hu,Tingting Zeng,Fang Geng,Di Wu

Phlorizin (PHL) has various biological activities, especially powerful antioxidant effects, but its water solubility limits its application. In this study, zein-encapsulated PHL nanoparticles (zein-PHL nanoparticles), as well as zein and gum arabic (GA) co-encapsulated PHL nanoparticles (zein/GA-PHL nanoparticles) were prepared by the antisolvent precipitation method. On this basis, encapsulated PHL nanoparticles were added to yogurt to make fortified yogurt. Fourier transform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) experiments showed that the main driving forces for nanoparticle formation were electrostatic interactions and hydrogen bonding between compounds. Zein-PHL nanoparticles were more sensitive to temperature and salt ions than zein/GA-PHL nanoparticles. The addition of GA can improve the stability of the nanoparticles. In addition, the nanoparticles encapsulated with PHL were applied to yogurt. It was found that the nanoparticles improved the antioxidant capacity and water-holding capacity of yogurt, which provides a reference for the development of functional yogurt containing flavonoid compounds.

Glucosylated nanoparticles for the oral delivery of antibiotics to the proximal small intestine protect mice from gut dysbiosis

Zhang Guorong,Wang Qin,Tao Wanyin,Jiang Wei,Elinav Eran,Wang Yucai,Zhu Shu

Orally delivered antibiotics can reach the caecum and colon, and induce gut dysbiosis. Here we show that the encapsulation of antibiotics in orally administered positively charged polymeric nanoparticles with a glucosylated surface enhances absorption by the proximal small intestine through specific interactions of glucose and the abundantly expressed sodium-dependent glucose transporter 1. This improves bioavailability of the antibiotics, and limits their exposure to flora in the large intestine and their accumulation in caecal and faecal contents. Compared with the standard administration of the same antibiotics, the oral administration of nanoparticle-encapsulated ampicillin, chloramphenicol or vancomycin in mice with bacterial infections in the lungs effectively eliminated the infections, decreased adverse effects on the intestinal microbiota by protecting the animals from dysbiosis-associated metabolic syndromes and from opportunistic pathogen infections, and reduced the accumulation of known antibiotic-resistance genes in commensal bacteria. Glucosylated nanocarriers may be suitable for the oral delivery of other drugs causing gut dysbiosis.

Uptake and transport of antibiotic kasugamycin in castor bean (Ricinus communis L.) seedlings

Zhang Hongzhen,Zhang Chenghua,Xiang Xiaolong,Zhang Qilun,Zhao Wei,Wei Guoyu,Hu Anlong

Kasugamycin (KSM), an aminoglycoside antibiotic, has been widely used for the management of plant diseases, especially for the control of rice blast in Asia. However, its uptake mechanism and transport in plants are still obscure. The castor bean (Ricinus communis L.) seeding, a model plant for phloem transport, was used to study the mechanism of uptake and transport of KSM. Results showed that cotyledon-applied KSM could transport into the phloem and distributed in root and shoot of plant. The temperature, concentration, and pH had significant effects on the uptake of KSM, indicating that the uptake of KSM was mediated by an active carrier system. Compared with the control, competitive inhibitors of sugar transporters D-glucose, D-chiro-inositol, and phloridzin inhibited 71.03%, 67.95%, and 61.73% uptake of KSM, respectively. Energy inhibitor dinitrophenol (DNP) and carbonyl cyanide chlorophenylhydrazone (CCCP) also affected the uptake of KSM, and the inhibition rates were 34.23% and 48.06%. All the results showed that the uptake of KSM was mediated by a sugar transporter, and it could transport from shoot to root in plants via the phloem. The study preliminary elucidated the plant–microbe interactions in the context of the transport of microbial secondary metabolites in plants. It has certain significance for scientific application of antibiotics and biological control of plant diseases and provides theoretical basis for the development of bidirectional transport pesticides.

O-Fluorobenzoic Acid-Mediated Construction of Porous Graphitic Carbon Nitride with Nitrogen Defects for Multicolor Electrochemiluminescence Imaging Sensing

Fang Li,Yachao Xi,Jianming Jiang,Hao Peng,Bing Li,Jianbo He,Jiangnan Shu,Hua Cui

Graphitic carbon nitride (g-CN) is an attractive electrochemiluminescence (ECL) luminophore. However, g-CN with wavelength-tunable ECL emission is still limited, which limits its application in multicolor ECL sensing and imaging analysis. In this study, porous g-CN (PCN) with nitrogen defects was synthesized through the condensation of melamine by using o-fluorobenzoic acid (o-FBA) as an effective regulation reagent. A series of PCNs, including PCN-5%, PCN-10%, and PCN-30%, were obtained by changing the mass ratio of o-FBA and melamine. The porous structure and tunable chemical composition change of the PCNs were carefully characterized. The nitrogen defects and porous structure of the synthesized PCNs can enlarge the specific surface area, facilitate electron transfer, and generate various surface states with gradually changed energy bands, leading to wavelength-tunable multicolor ECL emissions. Accordingly, g-CN, PCN-5%, PCN-10%, and PCN-30% can generate navy blue, turquoise blue, turquoise green, and olive green ECL emissions, respectively, with the peak ECL wavelength varied from 465 to 550 nm. Then, a multicolor ECL sensing array was proposed for the discrimination of polyphenols based on the prepared g-CN and PCNs by using a smartphone as a portable detector for the first time. Five polyphenol substances including vitamin P, resveratrol, phloretin, phlorizin, and caffeic acid were discriminated by using principal component analysis and hierarchical cluster analysis. The present work provides a simple strategy to adjust the ECL wavelength of g-CN and presents a simple way to fabricate multicolor ECL sensing array, which has great application potential for multiplexed analysis and multicolor ECL imaging sensing.

1-Methylcyclopropene (1-MCP), storage time, and shelf life and temperature affect phenolic compounds and antioxidant activity of ‘Jonagold’ apple

Yurong Ma,Qingfeng Ban,Jingying Shi,Tiantian Dong,Cai-Zhong Jiang,Qingguo Wang

The effects of 1-methylcyclopropene (1-MCP) treatment, storage time, and shelf life and temperature on phenolic concentrations and antioxidant activity in apple peel and flesh of ‘Jonagold’ apple ( Malus domestica Borkh) have been investigated. Hyperoside and chlorogenic acid were the most abundant phenolic compounds in the peel and flesh, respectively. The flesh had lower concentrations of individual phenolics, total phenolics, flavonoids and lower antioxidant activity (determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay) than the peel. In addition, the profiles of phenolics as well as the changing patterns of individual phenolics also differed between the peel and the flesh. After long-time storage (180 d), the greatest decrease of concentrations was the concentrations of hyperoside (27 %) in the peel and total phenolics (35 %) in the flesh. Also, during the 90–180 d storage period, hyperoside and rutin in the peel, chlorogenic acid in the flesh, and flavonoids and antioxidant activity decreased in both peel and flesh tissues. Individual phenolics exhibited different stabilities during the early stages after harvest. 1-MCP reduced the reduction in the concentrations of hyperoside in the peel, chlorogenic acid in the flesh, and total phenolics, flavonoids and antioxidant activity in both the peel and flesh tissues during storage and shelf life. However, for quercetin and rutin, the protective effect of 1-MCP was only observed during the shelf life period. Overall, phenolic compounds responded variously to storage, shelf life, shelf temperature and 1-MCP treatment.

Interaction mechanism of okra (Abelmoschus esculentus L.) seed protein and flavonoids: Fluorescent and 3D-QSAR studies

Chengyun He,Lu Bai,Daqun Liu,Benguo Liu

The binding capacity of 10 flavonoids with okra seed protein (OSP) was studied by fluorescence spectroscopy. The structure of flavonoids had an obvious impact on binding performance. The binding ability of flavanone was lower than that of flavone, isoflavone and dihydrochalcone. The binding capacity of flavonoid glycoside was superior to that of the corresponding flavonoid aglycone. The binding ability was positively correlated with the number of phenolic hydroxyl groups on the B ring. The steric field and electrostatic field model constructed by 3D-QSAR method could well explain the above interaction behavior. Thermodynamic analysis suggested that the quenching mechanism of OSP caused by flavonoids was static quenching, and the binding-site number was 1. In addition, hydrogen bonding and van der Waals force dominated this interaction. The 3D and synchronous fluorescence spectra showed that there was no significant change in the polarity of the environment around tryptophan and tyrosine residues during binding.

Magnetic imprinted nanoparticles with synergistic tailoring of covalent and non-covalent interactions for purification and detection of procyanidin B2

Zhang Haipin,Song Huijia,Tian Xuemeng,Wang Yue,Hao Yi,Wang Wenting,Gao Ruixia,Yang Wan,Ke YuShen,Tang Yuhai

A synergistic imprinting strategy of covalent and non-covalent interactions is proposed to prepare magnetic molecularly imprinted polymers (DI-MMIPs) for highly selective separation of procyanidin B2 (PC) from grape seed samples. Dopamine and 3-amino-phenylboronic acid as cooperative functional monomers endow the imprinted sites with synergistic tailoring. Benefiting from the synergistic effect, the DI-MMIPs exhibit enhanced imprinting performance with high adsorption capacity (27.71 mg g −1 ), fast kinetic equilibrium time (within 30 min), outstanding selectivity (IF = 5.8, SC > 3.2), and satisfactory regeneration ability. In addition, the DI-MMIPs possess good magnetism, uniform morphology with typical core-shell structure, and stable crystallization. Furthermore, the established DI-MMIPs coupled with HPLC-UV (~ 280 nm) method has a wide linearity range of 0.05–200 μg mL −1 with correlation coefficient of 0.9997, high recoveries (> 93.1%) with RSDs from 2.9 to 5.5%, and low LOD (0.0008 μg mL −1 ). Consequently, this work provides an effective and easily tailored way to fabricate magnetic imprinted nanomaterials with both rapid recognition rate and high selectivity and thus holds great promise to realize the extraction and detection of PC from real samples. Laccase Catalyzed Oxidative Polymerization of Phloridzin: Polymer Characterization, Antioxidant Capacity and α-Glucosidase Inhibitory Activity

Haoyu Zhou,Changzhong Liu,Sheng Geng

Phloridzin is a naturally occurring dihydrochalcone with various therapeutic properties. However, its low aqueous solubility and poor enzyme inhibitory capacity have limited its application in functional foods and medicines. Inspections of the properties of natural polymeric flavonoids suggest that these limitations could be mitigated by the polymerization of phloridzin, although to date, no relevant studies have been conducted. Here, oxidative polymerization was used to prepare polymeric phloridzin using laccase as the catalyst, and its structure, antioxidant capacity and α-glucosidase inhibitory activity were characterized. The results showed that laccase catalyzed polymerization via oxidative generation of phenolic radicals in the B ring of phloridzin to achieve the polymerization. The 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging capacity of the polymer (IC50, 0.48 mg/mL) was inferior to that of phloridzin (IC50, 0.20 mg/mL), but the α-glucosidase inhibitory activity of the polymer (IC50, 0.12 mg/mL) was significantly higher than that of phloridzin (IC50, 0.21 mg/mL). These effects could be attributed to the reduction of available phenolic groups and binding of the polymer to the enzyme, respectively.

Ameliorative effect of polydatin on hyperglycemia and renal injury in streptozotocin-induced diabetic rats

Lijuan Wang,Liji Huang,Nan Li,Junjun Miao,Wei Liu,Jiangyi Yu

To investigate the effect of polydatin on glucose transporter, blood glucose homeostasis and renal injury in streptozotocin (STZ)-induced diabetic rats. The in vitro inhibitory effect of polydatin on sodium-glucose cotransporter-1 (SGLT1) and 2 (SGLT2) was determined using HEK293 cells. The inhibitory effect of polydatin on GLUT1 and GLUT4 was evaluated using 3T3-L1 adipocytes. Streptozotocin-induced diabetic rats were used for this study. Fasting blood glucose (FBG), glycosylated hemoglobin (HbA1c), urea nitrogen, serum creatinine and urinary protein were determined using biochemical analyzer. Histopathological examination was performed on renal tissue. Serum levels of interleukin 1β (IL-1β), tumor necrosis factor α (TNF-α), monocyte chemoattractant protein 1 (MCP-1) and C-reactive protein (CRP) were also determined. Polydatin significantly inhibited SGLT1/2 and exhibited high selectivity for both GLUT1 and GLUT4. It significantly and dose-dependently decreased hyperglycemia, enhanced urine glucose excretion in the diabetic rats. The polydatin treatment significantly ameliorated symptoms of DN such as polyuria, polydipsia and hyperphagia. The hypoglycemic effect of polydatin was maintained throughout the treatment period. In addition, the levels of IL-1β, TNF-α, MCP-1 and CRP were significantly reduced in treated group. Treatment with polydatin significantly ameliorated most of the structural and morphological changes induced by STZ. Moreover, the levels of urinary protein, serum creatinine and urea nitrogen were significantly reduced after treatment with polydatin.  As a potential dual inhibitor of SGLT1/2, polydatin has high selectivity for GLUT1 and GLUT4. Its long-term administration delays the development of DN, protects renal function and ameliorates renal tissue injury.
