Biological control agents are beneficial microorganisms that protect plants from disease. However, the effectiveness and viability of microorganisms are often influenced by severe environmental conditions. Encapsulation of microbial cells can protect them from biotic and abiotic stresses. The objectives of this work were to encapsulate Bacillus velezensis ZF481 (ZF481) with chitosan and carrageenan to prepare macrospheres and evaluate their control effect on clubroot disease. Macrospheres with different ratios of chitosan (2 % w/w)/carrageenan (0.3 % w/v) were synthesized by an ionic gelation method, and the bacterial cells of ZF481 were loaded into macrospheres. The maximum swelling ratio (634 %) and maximum survival rate (64.72 ± 2.65 %) were obtained from macrospheres with chitosan: carrageenan ratio of 1: 1. The number of bacteria released from macrospheres into sterile phosphate buffer solution (pH 7.4) reached a release balance after 48 h. The bacteria could be slowly released from macrospheres into the soil. The survival rate of the encapsulated bacteria respectively was 73.33 % and 67.31 % when stored at 4 °C and 25 °C for 80 days, respectively. The control efficiency of ZF481 macrospheres (chitosan: carrageenan, 1: 1) on clubroot disease in pak choi under greenhouse conditions and Chinese cabbage under field conditions was significantly higher than that of ZF481 culture. Moreover, the ZF481 macrospheres also enhanced the plant growth. Collectively, chitosan and carrageenan are ideal wall materials for preparing slow-release bacterial macrospheres to control clubroot. The macrospheres retard the release of B.velezensis ZF481 in soils and significantly improve its biocontrol efficiency on clubroot in pak choi and Chinese cabbage. Thus, this study paves the way for forming formulations against the clubroot disease.

Clubroot, caused byPlasmodiophora brassicaeWoronin, poses a major threat to Chinese cabbage (Brassica rapasubsp.pekinensis) production worldwide, significantly impacting crop yield, quality, and economic value. Biological control represents a promising approach since it is non-toxic and eco-friendly, and it reduces the risk of pathogen resistance development. In this study, our objective was to screen for actinomycetes that can effectively inhibit clubroot. We screened 13 actinomycete strains, identifying 2, XDS3-6 and CD1-1, with substantial in vivo inhibitory effects, achieving infection suppression rates above 64% againstP. brassicae. Phylogenetic analysis classified XDS3-6 and CD1-1 asStreptomyces virginiaeandStreptomyces cinnamonensis, respectively. Both strains exhibited protease and glucanase production capabilities, essential for pathogenic suppression. Additionally, these strains induced host defense responses, as evidenced by increased jasmonic acid (JA) and salicylic acid (SA) accumulation and elevated activities of defense-related enzymes. Colonization studies of XDS3-6 and CD1-1 mutant strains in cabbage roots indicated sustained root colonization, with peak colony-forming units (CFUs) at 20 days post-inoculation, reaching 11.0 × 104CFU/g and 8.5 × 104CFU/g, respectively, and persisting for at least 30 days. Overall, these findings underscore the potential ofStreptomycesstrains XDS3-6 and CD1-1 as effective biocontrol agents, providing a theoretical foundation for their application in managing clubroot in Chinese cabbage.

Plant roots encounter numerous pathogenic microbes that often cause devastating diseases. One such pathogen, Plasmodiophora brassicae (Pb), causes clubroot disease and severe yield losses on cruciferous crops worldwide. Here, we report the isolation and characterization of WeiTsing (WTS), a broad-spectrum clubroot resistance gene from Arabidopsis. WTS is transcriptionally activated in the pericycle upon Pb infection to prevent pathogen colonization in the stele. Brassica napus carrying the WTS transgene displayed strong resistance to Pb. WTS encodes a small protein localized in the endoplasmic reticulum (ER), and its expression in plants induces immune responses. The cryoelectron microscopy (cryo-EM) structure of WTS revealed a previously unknown pentameric architecture with a central pore. Electrophysiology analyses demonstrated that WTS is a calcium-permeable cation-selective channel. Structure-guided mutagenesis indicated that channel activity is strictly required for triggering defenses. The findings uncover an ion channel analogous to resistosomes that triggers immune signaling in the pericycle.

Clubroot, caused by the obligate parasite Plasmodiophora brassicae , is one of the most important diseases of brassicas. The antagonistic bacterium Paenibacillus polymyxa ZF129 can suppress clubroot while its effectiveness is often unstable. To control clubroot more effectively, the macrobeads for controlled release of ZF129 were prepared using microencapsulation technology. Macrobeads with various ratios of chitosan (2 % w/w): carrageenan (0.3 % w/v) were prepared by an ionotropic gelation method and the bacteria ZF129 was loaded into macrobeads. The 1:1 chitosan: carrageenan showed the maximum swelling ratio (634 %), and the maximum survival rate (61.52 ± 1.12 %) after freeze-drying. Fourier transform infrared revealed the electrostatic interactions between chitosan and carrageenan. The macrobeads can efficiently release ZF129 strains into phosphate buffer solution and reach equilibrium in 48 h. The maximum number of bacteria cells to be released in the soil was observed after 25–30 days. The control efficacy of ZF129 macrobeads (chitosan: carrageenan, 1:1) and ZF129 culture against clubroot disease was 76.33 ± 3.65 % and 59.76 ± 4.43 % in greenhouse experiments, respectively and the control efficacy was calculated as 60.74 ± 5.00 % for ZF129 macrobeads and 40.94 ± 4.05 % for ZF129 culture under field experiments, respectively. The ZF129 macrobeads had significant growth-promoting effects on pak choi and Chinese cabbage. The encapsulation method described in this study is a prudent approach toward efficient biopesticides utilization with reduced environmental implications.
