Globisporangium recalcitrans is an important phytopathogenic oomycete that causes soybean root rot. To develop a loop-mediated isothermal amplification (LAMP)-based system for rapid and specific detection of G. recalcitrans , four LAMP primers and two loop primers were designed with the Ribosomal DNA transcribed spacer 2 sequence (ITS2) as the target gene. The specificity and sensitivity of these primers were validated, and the system was also successfully applied to detect G. recalcitrans in soybean tissues after artificial inoculation and natural infection. The nucleic acid amplification reaction was performed under isothermal conditions at 63 °C for 60 min. Specificity was compared with those for 71 strains of G. recalcitrans , other Globisporangium spp. The minimum detection limit of the system was 100 fg·μL −1 for detecting fungal genomic DNA. This LAMP-based system provides a technique for specific detection of G. recalcitrans and rapid diagnosis of the disease it caused.

Soybean root rot, caused by Fusarium neocosmosporiellum , poses a significant threat to soybean productivity. Although chemical fungicides are widely used, their environmental risk necessitate sustainable alternatives. This study proposes an integrated control strategy combining the biocontrol bacterium Pseudomonas aeruginosa E1 - 23 with prochloraz. Initial screening of seven candidate bacterial strains identified E1 - 23 as the most potent antagonist, exhibiting 64.1% inhibition against F. neocosmosporiellum in dual-culture assays. Concurrent fungicide evaluation revealed prochloraz as the optimal agent (EC 50 = 0.1646 μg/mL). Compatibility analysis demonstrated E1 - 23's robust tolerance to prochloraz concentrations at ≤ 50 μg/mL. In vitro combination of prochloraz (0.25 μg/mL) and E1 - 23 (1 × 10 6 CFU/mL) achieved 91.86% pathogen suppression, surpassing individual treatments by > 10% and theoretical additive effects. Greenhouse trials confirmed enhanced efficacy, with integrated application increasing disease control efficacy by 34.35%—significantly outperforming standalone bacterial (18.96%) or fungicide (20.52%) controls. The qRT-PCR analysis revealed that the combination application of E1 - 23 and prochloraz up-regulated the expression of some defense genes of soybean plants 5 days post-inoculation (dpi), but the induced plant disease resistance was low. This study highlights the potential of E1 - 23-prochloraz as an eco-friendly approach to mitigate soybean root rot while reducing fungicide dependency.

Background and aims Root rot is a catastrophic disease of perennial medicinal plants which could be caused by various soil-borne fungal pathogens. However, the understanding of the composition and diversity of root rot pathogenic fungi, their successions and potential drivers remains limited. Methods Sanqi ginseng ( Panax notoginseng ) was selected as a model plant and the rhizosphere soils of root rot-infected Sanqi ginseng at different growth years were collected. Phenolic acid content, pathogen abundance, and fungal microbiota were examined by using the high-performance liquid chromatography (HPLC), real-time PCR and Miseq sequencing. Results Pathogenic agents dominated in the rhizospheric fungal microbiota of rotten Sanqi ginseng and mainly comprised of Ilyonectria , Plectosphaerella , Clonostachys , Gibberella , Pestalotiopsis , Fusarium , and Chalara , with Ilyonectria and Fusarium being most abundant. The composition of pathogenic microbiota, but not α-diversity indices, changed considerably with Sanqi ginseng growth. The shifts of phenolic acid profile were an important driver in changes of rhizosphere pathogenic microbiota, and same phenolic acid exerted distinct functions on differential pathogenic taxa. Particularly, the contents of p -coumaric acid, ferulic acid, and benzoic acid were positively correlated with the abundance of genus Fusarium and negatively correlated with the abundance of genus Ilyonectria . Conclusions Root rot-infected Sanqi ginseng rhizosphere harbors dynamically pathogenic microbiota driven by the shift of phenolic acids. This study provides insights into the composition, variation, and driver of root rot pathogenic microbiota; these insights are important for the development of targeted measure for the prevention and control of root rot diseases.

Atractylodes chinensishas a high medicinal value and is widely cultivated. However, root rot disease seriously affects the yield and quality ofA. chinensis. To develop green and safe pesticides, the inhibitory effect of essential oils (EOs) of three Labiatae plants on the pathogenic fungi that causes root rot disease inAtractylodes chinensiswas investigated. The results showed that theOriganum vulgareEO andThymus mongolicusEO exhibited strong inhibitory effects onFusarium oxysporum,Fusarium solani, andFusarium redolens, with 100% inhibition rate. The low MIC values of EOs and their main components against the three pathogenic fungi indicated that all of them showed strong fungicidal effects. The MIC values ofO. vulgareEO againstF. oxysporum,F. solani, andF. redolenswere 2.60 mg/mL, 3.13 mg/mL, and 1.56 mg/mL, respectively. Analyses using scanning electron microscopy (SEM) and transmission electron microscopy (TEM) showed that theO. vulgareEO severely damaged the cell wall and cell membrane of mycelial cells. TheO. vulgareEO increased cell permeability, leading to a large leakage of cell contents (DNA and proteins). In addition,O. vulgareEO inhibitedF. oxysporumby inducing ROS production and reducing the amount of intracellular GSH, leading to a large accumulation of ROS. This study showed that plant EOs have excellent fungicidal activity and can be used as novel natural and environmentally friendly pesticides for the control of root rot inA. chinensis. Keywords:Atractylodes chinensis;root rot;essential oils;Origanum vulgare;antifungal activity;mechanisms of action

Soil-borne diseases have caused substantial economic losses to agricultural production due to their rapid onset and few means of control. Here, pyraclostrobin (Pyr) microcapsules (Pyr@MCC) with temperature-responsive pesticide release capability were prepared using temperature-sensitive phase-change materials as capsule cores. Then, the Pyr@MCC was organically combined with poly(vinyl alcohol)-starch film (PVA/ST) to prepare a composite mulch film (PM/PVA/ST) with intelligent control of pesticide release rate according to external temperature change, which realized sustainable and efficient control of Phytophthora root rot (PRR) on soybean. Tensile tests showed that the PM/PVA/ST film exhibited good tensile deformation ability (134.9%) and high tensile stress (18.0 Mpa). The water vapor transmission rate test showed that the PM/PVA/ST film exhibited good barrier performance to H2O, CO2, and O2. The release test data proved that the PM/PVA/ST film could slowly release Pyr at lower temperatures (15 °C) and release Pyr rapidly at higher temperatures (35 °C), corresponding to the high temperature at which the Phytophthora root rot outbreak. In vitro antifungal and pot experiments showed that the PM/PVA/ST film had a good control efficiency on Fusarium spp. These results show that the functional pesticide microcapsules loaded onto the film have good application potential for controlling PRR on soybean. In general, this composite mulch film provided some innovative research ideas for optimizing the pesticide delivery pathway and thereby efficiently controlling soil-borne pests and diseases, reducing the frequency and amount of pesticide use and minimizing white pollution from traditional plastic mulch films.

Fusarium oxysporumis a widespread soil-borne fungal pathogen that can infect various plants, causing wilt and root rot diseases. The root rot disease ofAtractylodes macrocephalacaused byF. oxysporumis among the most serious diseases associated with continuous cropping, significantly hindering its sustainable development. In this study, we aimed to investigate the effect of exogenous application of double-stranded RNA (dsRNA) on silencing theF. oxysporum Tup1gene to reduce its virulence and to evaluate its potential application in controlling root rot disease inA. macrocephala. TheTup1gene was amplified from theF. oxysporumgenome, and different lengths ofTup1-dsRNA were designed and synthesized. The uptake of dsRNA by the fungus was verified usingTup1-dsRNA labeled with fluorescein, and in vitro dsRNA treatment experiments were conducted to assess its impact on the growth and virulence ofF. oxysporum. Additionally,Tup1-dsRNA was applied to the roots ofA. macrocephalato evaluate its effectiveness in controlling root rot disease. The experimental results showed thatF. oxysporumcould effectively uptake exogenously appliedTup1-dsRNA, significantly reducingTup1gene expression. All lengths ofTup1-dsRNA inhibited fungal growth and caused morphological changes in the fungal hyphae. Further plant experiments and Reverse Transcription Quantitative Polymerase Chain Reaction (RT-qPCR) analysis indicated thatTup1-dsRNA treatment significantly reduced the incidence of root rot disease inA. macrocephala, which was supported by the reduction in peroxidase (POD) and catalase (CAT) enzyme activities, malondialdehyde (MDA) content, and proline (Pro) levels in treated root tissues. This study demonstrated that exogenous dsRNA could reduce the virulence ofF. oxysporumby silencing theTup1gene and effectively mitigate the root rot disease it causes inA. macrocephala. The successful application ofTup1-dsRNA provided strong evidence for the potential of RNA interference (RNAi) technology in plant disease control. Future research could further optimize the design and application of dsRNA to enhance its practical value in agriculture.

Medicine plants such as Panax notoginseng (Sanqi) frequently experience replant failure owing to the accumulation of soilborne pathogens and autotoxins in continuous cropping soil. Bacillus sp. is a commonly used biocontrol agent to combat soilborne pathogens, but further research is needed to understand its impact on controlling Sanqi root rot and the underlying mechanisms. Here, we isolated and screened a Bacillus strain, B19, from Sanqi rhizosphere soil that exhibited potential for inhibiting Sanqi root rot pathogens. We then tested the strain's effect on the incidence of Sanqi root rot in continuous cropping soil, along with its motility/chemotaxis and biofilm formation ability (i.e., potential for root colonization). We also analyzed the strain's genomic characteristics, including potential secondary metabolite synthesis gene clusters. Our results showed that the strain B19 exhibited high inhibitory activity against fungal pathogens ( F. solani and F. oxysporum ) and could metabolize saponins as a sole carbon source. The application of B19 to continuous cropping soil significantly reduced the incidence of root rot in Sanqi seedlings. B19 was identified as B. velezensis through morphological characteristics, 16S rRNA and gyrA gene sequence analysis, and genome-level annotation. Notably, B19′s genome encodes many genes associated with functions such as motility/chemotaxis and biofilm formation, as well as 13 biosynthetic gene clusters that could encode secondary metabolites, including surfactin, fengycin, and siderophore. Additionally, B19 demonstrated the capability to produce volatile organic compounds that inhibit pathogenic fungi. The motility/chemotaxis and biofilm-forming ability of B19 were enhanced by saponins. Collectively, these properties could enhance B19′s biocontrol ability, thereby reducing the incidence of root rot.

Soybean root rot, primarily caused by Fusarium and Phytophthora spp., severely damages root tissues, impairs nutrient uptake, and leads to plant death, particularly in saline-alkaline soils, where high pH, salt stress, and deteriorated soil conditions reduce the stress resistance of soybean plants. Conventional chemical treatments are less effective owing to the poor vertical mobility of fungicides in alkaline soils. Stimuli-responsive controlled-release formulations can improve the bioavailability of active ingredients, reduce application frequency, and minimize environmental impact. Here, we present a sustainable, biodegradable, and environmentally benign delivery system based on zein, a plant-derived protein. Zein was modified with non-toxic succinic anhydride to improve water dispersibility, resulting in a green carrier (SZ) with reduced ecological risk. The UV-sensitive fungicide difenoconazole (Di) was encapsulated into this carrier via nanoprecipitation to construct a stimuli-responsive nano-enabled delivery system (Di@SZ). Di@SZ exhibited alkaline-triggered release, with rapid release in alkaline conditions. Compared to the commercial emulsifiable concentrate (EW), Di@SZ nanoparticles showed a 3.03-fold improvement in UV resistance. They enhanced vertical mobility of Di in soil, enabling targeted root delivery during drip irrigation in saline-alkaline soils. Pot experiments demonstrated that Di@SZ provided stronger and longer-lasting protection against soybean root rot than EW. Moreover, the system exhibited superior biosafety, with 2.77-fold lower toxicity to soil earthworms relative to EW. These findings underscore Di@SZ as a sustainable, materials-based plant protection technology that integrates renewable resources, reduces environmental impact, and enhances efficacy, offering a promising approach for eco-friendly disease management in agriculture.

Plant-beneficial microbes can be effective as biological agents for promoting development and controlling diseases in plants. However, direct inoculation of non-encapsulated plant-beneficial microbes into the soils can affect their vitality and efficacy. A novel bio-based encapsulant, sporopollenin-chitosan microspheres (SCMs), was developed to load an endophytic fungus Talaromyces neorugulosus R-209 with antagonistic activities against the root rot pathogen ( Rhizoctonia solani AG4) and plant growth-promoting functions. The results showed that T. neorugulosus R-209 encapsulated in SCMs ( Tn R-209−SCMs) could significantly enhance fungal spore germination rates and available nitrogen/phosphorus levels in the soil compared to the non-encapsulated fungus. In addition, the preliminary evidence suggests that Tn R-209−SCMs have a basic safety profile for practical applications. Inoculation with Tn R-209−SCMs could effectively promote development and enhance resistance in pigeon pea seedlings by promoting chlorophyll synthesis, improving photosynthesis, and enhancing phenolic compound accumulation. Meanwhile, T. neorugulosus R-209 was found to endogenously colonize root intercellular spaces. Moreover, co-inoculation of Tn R-209−SCMs and R. solani AG4 could reduce host defense responses compared to R. solani AG4-infected roots, as reflected by lower levels of phenolic compound accumulation and pathogenesis-/biosynthesis-related gene expression. Overall, Tn R-209−SCMs is a promising biological agent that can promote development and control root rot in plants, which also provides an innovative approach to biomaterial-supported agricultural practices.

Background and aims Overaccumulation of allelochemicals in soil from trees has a negative effect on the growth of Panax notoginseng and cause root rot. Many plant secreted phenols are water-soluble substances that can be depleted by leaching. However, comprehensive understanding of the effect of leaching of phenols in Chinese fir soil on P. notoginseng root rot disease is still lacking. Methods Soil with high level of phenols was leached with five intensities and their effect on the root rot of P. notoginseng was assessed. Then, the relationship of root rot with changes in soil chemical properties and phenol content as well as rhizospheric microorganisms was analyzed. Results With increasing leaching intensity, the levels of phenols in soils and the incidence of root rot decreased significantly. Decreased phenol content could alleviate root rot by modifying the soil microbial community. Network analysis showed that root rot incidence was positively correlated with the p-hydroxybenzoic acid-enriched pathogenic fungus Ilyonectria but negatively correlated with p-hydroxybenzoic acid (HA)-, ferulic acid (FA)- or vanillic acid (VA)-suppressed bacteria, including Sphingomonas , Lysobacter , Massilia and Burkholderia . Culture experiments confirmed that individual HA, FA and VA as well as a mixture of the five phenols with increasing concentrations could stimulate the growth and sporulation of I. destructans but suppress the growth and biofilm formation of bacteria ( Burkholderia and Lysobacter ). Moreover, Burkholderia isolates showed antagonistic activity against the growth of I. destructans . Conclusions Leaching could relieve the stimulation of pathogenic fungi by phenols and reduce the pressure on phenolic-suppressed bacteria, thereby reducing the incidence of root rot disease.

Fusarium oxysporum f.sp.radicis-lycopersici(Forl) is a destructive soil-borne phytopathogenic fungus that causes Fusarium crown and root rot (FCRR) of tomato, leading to considerable field yield losses. In this study, we explored the antifungal capability of linalool, a natural plant volatile organic component, againstForland its role in controlling FCRR symptoms in tomatoes. Our results showed thatForlmycelial growth was inhibited by the linalool treatment and that the linalool treatment damaged cell membrane integrity, enhanced reactive oxygen species levels, depleted glutathione, and reduced the activities of many antioxidant enzymes inForl. Transcriptomic and proteomic analyses demonstrated that linalool also downregulated metabolic biosynthetic pathways at the transcript and protein levels, including redox, transporter activity, and carbohydrate metabolism inForl. Moreover, linalool significantly decreased the expression of manyForlpathogenic genes, such as cell wall degrading enzymes (CWDEs) and G proteins, which is likely how aForlinfection was prevented. Importantly, exogenously applied linalool activated the salicylic acid (SA) and jasmonic acid (JA) defensive pathways to improve disease resistance and relieved the negative effects ofForlon plant growth. Taken together, we report that linalool is an effective fungicide againstForland will be a promising green chemical agent for controlling FCRR.Keywords:volatile compounds;antifungal mechanism;Fusarium oxysporum f.sp.radicis-lycopersici;Fusarium crown and root rot;transcriptomic and proteomic
