Due to the emergence and spread of multidrug resistance in Helicobacter pylori ( H. pylori ), its eradication has become difficult. Sodium sulfite (SS), a widely used food additive for ensuring food safety and storage, has been recognized as an effective nonbactericidal agent for H. pylori eradication. However, the mechanism by which H. pylori adapts and eventually succumbs under low- or no-oxygen conditions remains unknown. In this study, we aimed to evaluate the anti- H. pylori effect of SS and investigated the multiomics mechanism by which SS kills H. pylori . The results demonstrated that SS effectively eradicated H. pylori both in vitro and in vivo . H. pylori responds to the oxygen changes regulated by SS, downregulates the HcpE gene, which is responsible for redox homeostasis in bacteria, decreases the activities of enzymes related to oxidative stress, and disrupts the outer membrane structure, increasing susceptibility to oxidative stress. Furthermore, SS downregulates the content of cytochrome C in the microaerobic respiratory chain, leading to a sharp decrease in ATP synthesis. Consequently, the accumulation of triglycerides (TGs) in bacteria due to oxidative stress supports anaerobic respiration, meeting their energy requirements. The multifaceted death of H. pylori caused by SS does not result in drug resistance. Thus, screening of the redox homeostasis of HcpE as a new target for H. pylori infection treatment could lead to the development of a novel approach for H. pylori eradication therapy.

Bacterial biofilm is notorious for causing chronic infections, whose antibiotic treatment is bringing about severe multidrug resistance and environmental contamination. Stimuli-responsive nanocarriers have become encouraging materials to combat biofilm infections with high efficiency and low side effect. Herein, a charge-switchable and pH-responsive nanocomplex is fabricated via a facile aqueous one-pot zeolitic imidazolate framework-8 (ZIF-8) encapsulation of proteinase K (PK) and photosensitizer Rose Bengal (RB), for enzymatic and photodynamic therapies (PDT) against biofilm infections. Once encountering in acidic microenvironment, the surface charge of nanocomplex can switch self-adaptively from negative to positive, hence remarkably facilitating the biofilm penetration of nanocomplex. After acid-induced decomposition of nanocomplex, the released PK degrades biofilm matrix and loosens its structure, promoting diffusion of RB inside the biofilm. Afterwards, upon visible light illumination, the RB generates highly reactive oxygen species (ROS), which can readily and efficiently kill the remained bacteria even in the biofilm core. The charge-assisted penetration makes PK and RB fully functional, resulting in a cooperative effect concerning high biofilm eradication capacity, as testified by biofilm models both in vitro and in vivo . The green synthesis and good therapeutic performance of the nanocomplex manifests its considerable potential as a nontoxic and effective platform for biofilm treatment.

Compared to the activation of acquired immunity by the immune checkpoint blockade, the activation of innate immunity via anti-phagocytosis checkpoint blockade could significantly increase the beneficiary population of immunotherapy. However, the activation of innate immunity and the occurrence of phagocytosis are only accomplished when the interaction between pro-phagocytosis signals and anti-phagocytosis signals is realized. Herein, a versatile nanoplatform (DHMR) based on mesoporous silicon nanoparticles (MSNPs) has been constructed. Two drugs, doxorubicin, a chemotherapeutic drug which could initiate tumor cells to release pro-phagocytosis signals, and RRx-001, an immunoadjuvant that could effectively implement the anti-phagocytosis checkpoint blockade, were loaded in MSNPs. Further decoration of hyaluronic acid encapsulation endows DHMR with the function of tumor targeting and long circulation. Ultimately, the DHMR system could efficiently and accurately target tumor tissue, release the drugs in the tumor microenvironment, achieve the activation of innate immunity, and finally dramatically inhibit the growth and metastasis of tumor cells.

The increasing severity of fungal infections globally and the limited availability of effective treatments necessitate novel therapeutic strategies. In this study, we developed a copper-polyphenol nanocomplexes (CPE NPs) through a one-step synthetic process by the coordination self-assembly of copper sulfate (CuSO4), protocatechuic acid (PA), and ε-Poly-L-lysine (EPL). CPE NPs demonstrated peroxidase-like activities, which could catalyze H2O2 to generate hydroxyl radicals (•OH) to effectively eradicate fungi. CPE NPs were stable in physiological neutral environment. However, since metal-phenolic coordination interaction was unstable in acidic pH, CPE NPs could be dissociated in acidic environments, such as fungal infection sites, leading to effective release of Cu2+ and EPL, thereby exerting antifungal effects. CPE NPs exhibited excellent fungicidal ability at both pH 7.4 and 6.0 against Candida albicans and Aspergillus flavus. More importantly, CPE NPs could effectively eradicate fungi in 20% Sabouraud dextrose broth (SDB) medium containing rich inorganic salts and proteins, in which dissociative Cu2+ completely lost its fungicidal ability. In summary, CPE NPs could effectively deliver Cu2+ to fungal surface for enhanced fungal eradication by generating •OH, highlighting their great potential as a promising nano-therapeutic agent for the treatment of fungal infections.

Local administration of therapeutic agents with long-term retention capabilities efficiently avoids nonspecific distribution in normal organs with an increased drug concentration in pathological tissue. Herein, we developed an injectable and degradable alginate-calcium (Ca2+) hydrogel for the local administration of corn-like Au/Ag nanorods (NRs) and doxorubicin hydrochloride (DOX·HCl). The immobilized Au/Ag NRs with strong absorbance in the near-infrared II (NIR-II) window efficiently ablated the majority of tumor cells after 1064 nm laser irradiation and triggered the release of DOX to kill residual tumor cells. As a result, injectable hydrogel-mediated NIR-II photothermal therapy (PTT) and chemotherapy efficiently inhibited tumor growth, resulting in the complete eradication of tumors in most of the treated mice. Furthermore, owing to the confinement of the Au/Ag NRs and DOX·HCl within the hydrogel, such treatment exhibited excellent biocompatibility.

There is an increasingly growing demand to balance tissue repair guidance and opportunistic infection (OI) inhibition in clinical implant surgery. Herein, we developed a nanoadjuvant for all-stage tissue repair guidance and biofilm-responsive OI eradication via in situ incorporating Cobaltiprotoporphyrin (CoPP) into Prussian blue (PB) to prepare PB-CoPP nanozymes (PCZs). Released CoPP possesses a pro-efferocytosis effect for eliminating apoptotic and progressing necrotic cells in tissue trauma, thus preventing secondary inflammation. Once OIs occur, PCZs with switchable nanocatalytic capacity can achieve bidirectional pyroptosis regulation. Once reaching the acidic biofilm microenvironment, PCZs possess peroxidase (POD)-like activity that can generate reactive oxygen species (ROS) to eradicate bacterial biofilms, especially when synergized with the photothermal effect. Furthermore, generated ROS can promote macrophage pyroptosis to secrete inflammatory cytokines and antimicrobial proteins for biofilm eradication in vivo. After eradicating the biofilm, PCZs possess catalase (CAT)-like activity in a neutral environment, which can scavenge ROS and inhibit macrophage pyroptosis, thereby improving the inflammatory microenvironment. Briefly, PCZs as nanoadjuvants feature the capability of all-stage tissue repair guidance and biofilm-responsive OI inhibition that can be routinely performed in all implant surgeries, providing a wide range of application prospects and commercial translational value.

Periodontal disease is one of the most common oral infectious diseases and closely related to many systemic diseases which is in urgent need of novel and effective drugs. In particular, the problems of deep lesion location and synchronization of antibacterial and anti-inflammatory needs to be addressed. Herein, near infrared (NIR) light triggered nanoplatform with antibacterial and anti-inflammatory dual-functions by antibacterial photodynamic therapy (aPDT), photothermal therapy (PTT) and gas therapy was realized by loading S-nitrosothiols (SNO) and ICG into mesoporous silica-coated gold nanorods . Using a single 808 nm NIR light, the excellent antibacterial efficacy can be achieved not only by aPDT-induced bacteria killing but also by PTT-triggered biofilm dispersal. Furthermore, SNO molecules would also be thermally activated to release nitric oxide (NO) gas molecules at the site of periodontal disease, exerting inflammation inhibition by suppressing the assembly of pro-inflammatory factors and NLRP3 inflammasome. Additionally, such nanoplatform could validly combat pathogenic bacterial biofilm, and modulate the inflammatory immunity, leading to favorable therapeutics on animal models with periodontal diseases. Hence, this design provides an effective solution to solve the problems of deep tissue treatment along with simultaneously antibacterial/anti-inflammatory capabilities, which provides a bright prospect for clinical treatment of periodontal diseases, even extended to other refractory infections in deep sites.
