A heat shock protein protects against oxidative stress induced by lambda-cyhalothrin in the green peach aphid Myzus persicae

Bao Dong,Xi-Ya Liu,Bin Li,Mao-Ye Li,Shi-Guang Li,Su Liu

Lambda-cyhalothrin (LCT) is a pyrethroid insecticide widely used to control insect pests . Insect exposure to LCT may cause abnormal accumulation of reactive oxygen species (ROS) and result in oxidative damage. Heat shock proteins (HSPs) may help protect against oxidative stress . However, little is known about the role of HSPs in response to LCT in the green peach aphid , Myzus persicae . This insect is an important agricultural pest causing severe yield losses in crops. In this study, we characterized a cDNA sequence ( MpHsp70 ) encoding a member of the HSP70 family in M. persicae . MpHsp70 encoded a 623 amino acid protein putatively localized in the cytosol. The highest expression level of MpHsp70 occurred in fourth-instar nymphs. Treatment of M. persicae with LCT resulted in oxidative stress and significantly increased H 2 O 2 and malondialdehyde levels. This led to an elevated transcription level of MpHsp70 . Injection of H 2 O 2 into M. persicae also upregulated the MpHsp70 expression level, suggesting that MpHsp70 is responsive to ROS, particularly H 2 O 2 , induced by LCT. Recombinant MpHSP70 protein was expressed in Escherichia coli . E. coli cells overexpressing MpHSP70 exhibited significant tolerance to H 2 O 2 and the ROS generators, cumene hydroperoxide and paraquat . This indicated that MpHSP70 protects against oxidative stress. Furthermore, knockdown of MpHsp70 by RNA interference resulted in increased susceptibility in apterous adults of M. persicae to LCT. These findings indicate that MpHsp70 plays an important role in defense against LCT-induced oxidative stress and insecticide susceptibility in M. persicae .

Standard strategy for developing multicomponent nanopesticides: From gene function analysis to co-delivery nano-platform construction

Qinhong Jiang,Min Dong,Lin Che,Che Li,Xian Zhang,Meizhen Yin,Jie Shen,Shuo Yan

The continuous screening of high-efficiency RNAi targets is labor-consuming and time-costing, which constrains the fast commercialization of RNA pesticides. Thus, incorporating synergistic active ingredients via co-delivery nano-platform offers a promising solution without the need for high-efficiency RNAi targets. The current study aimed to propose a standard strategy for developing multicomponent nanopesticides. The function analysis revealed that the decapentaplegic-like ( Dpp ) participated in fatty acid biosynthesis and cuticle formation, and its RNAi resulted in molting failure and death in destructive green peach aphids. However, the morality caused by Dpp RNAi could not meet the actual demand, and thus a potential synergistic pesticide spirotetramat (SPI) was introduced to assemble with star ploycation (SPc) and ds Dpp to construct a co-delivery nano-platform. The SPc could assemble with SPI into SPI/SPc complex, and the ds Dpp could be further adhered on the surface of complex through electrostatic interaction, which formed stable spherical particles with nanoscale size (322.95 nm) and positive charge (28.22 mV). Notably, the SPI/SPc/ds Dpp complex displayed better adhesion performance on plant leaves, and thus the plant uptake of SPI/SPc/ds Dpp complex was remarkably improved in vivo and in vitro. Meanwhile, the SPI/SPc/ds Dpp complex exhibited amplified contact area on aphid cuticle, and its translocation was improved across the aphid cuticle/cell membrane. Importantly, the stomach and contact toxicity of SPI/SPc/ds Dpp complex was the strongest with the highest aphid mortalities of 94.5 %–96.6 %. The co-delivery nano-platform improved both initial action time and overall bioactivity, addressing the short life disadvantage of ds Dpp and slow-acting property of SPI. Our work proposed a standard strategy for designing multicomponent nanopesticides, which could facilitate the fast commercialization of RNA pesticides.

Multiple Insecticide Resistance and Associated Metabolic-Based Mechanisms in a Myzus Persicae (Sulzer) Population

Jinfeng Hu,Feng Chen,Jun Wang,Wenhua Rao,Lei Lin,Guocheng Fan

The green peach aphid,Myzus persicae(Sulzer) (Hemiptera: Aphididae), is an economically devastating crop pest worldwide. TheM. persicae(SEF-R) population of a cabbage field in China was tested for susceptibilities to 13 insecticides. Compared with the susceptible population (FFJ-S), extremely high and high resistance to beta-cypermethrin (324-fold) and imidacloprid (106.9-fold) was detected in SEF-R. More importantly, this is the first report of resistance in the fieldM. persicaepopulation to sulfoxaflor (32.4-fold), flupyradifurone (9.5-fold), pymetrozine (34.8-fold), spirotetramat (8.1-fold), flonicamid (5.8-fold), and broflanilide (15.8-fold) in China when compared with FFJ-S. The resistance factor decayed to a low level to sulfoxaflor and pymetrozine after 15 generations without any selection pressure. The resistance-related mutations (R81T and kdr) detected in SEF indicated target-site resistance to neonicotinoids and pyrethroids, respectively. Biochemical assays revealed the involvement of monooxygenase, carboxylesterase, superoxide dismutase, and peroxidase in a multi-insecticide resistance mechanism. The overexpression of P450s, esterases, and a UDP-glycosyltransferase might be responsible for the multi-insecticide resistance in SEF-R. The knockdown of CYP6CY3 in SEF-R increased its susceptibility to imidacloprid, thiacloprid, and thiamethoxam, which verified that P450s play vital roles in neonicotinoid metabolism. Our findings provide guidance for the rational use of insecticides to delay resistance development in GPA.Keywords:Myzus persicae;insecticides;resistance monitoring;detoxification enzymes;protective enzymes;metabolic mechanism

Molecular characterization of a catalase gene from the green peach aphid (Myzus persicae)

Mao-Ye Li,Yun Wang,Xiao Lei,Chuan-Tao Xu,Dong-Dong Wang,Su Liu,Shi-Guang Li

The green peach aphid, Myzus persicae (Sulzer), is a serious agricultural pest with a worldwide distribution. Catalase (CAT), which is encoded by the catalase ( Cat ) gene, is an extremely important antioxidant enzyme that plays a pivotal role in protecting cells against the toxic effects of hydrogen peroxide. The Cat gene has not been characterized in M. persicae ; therefore, this study describes the identification of the Cat ( MpCat ) gene from M. persicae . MpCat contains an open reading frame of 1515 bp and encodes a MpCAT protein consisting of 504 amino-acid residues. MpCAT possesses features typical of other insect catalases, including 7 conserved amino acids involved in binding heme and 15 involved in binding nicotinamide adenine dinucleotide phosphate. Phylogenetic analysis showed that MpCAT was closely related to orthologs from other aphid species. MpCat consisted of nine exons and eight introns, and the number and insertion sites of introns are consistent with those of Cat genes from Acyrthosiphon pisum (Harris) and Aphis gossypii Glover. The mRNA transcripts of MpCat were detected at all tested developmental stages, with the highest mRNA level in alate adults. The expression of MpCat was significantly upregulated when M. persicae was exposed to low and high temperatures, ultraviolet radiation, Beauveria bassiana , and permethrin. The transcription of MpCat and the activity of catalase were suppressed by RNA interference, and knockdown of MpCat significantly reduced the survival rate in M. persicae under heat stress. The results provide valuable information for further study on the physiological functions of MpCat . Research Highlights A catalase gene ( MpCat ) was characterized in Myzus persicae . Transcription of MpCat was responsive to various environmental stresses. Knockdown of MpCat increased mortality in M. persicae under heat stress.

Identification and Functional Analysis of Differentially Expressed Genes in Myzus persicae (Hemiptera: Aphididae) in Response to Trans-anethole

Ding Chao-Yang,Ma Yu-Meng,Li Bin,Wang Yun,Zhao Le,Peng Jiang-Nan,Li Mao-Ye,Liu Su,Li Shi-Guang

Plant essential oils, with high bioactivity and biodegradability, provide promising alternatives to synthetic pesticides for pest control. Trans-anethole is the major component of essential oil from star anise, Illicium verum Hook. The compound has a strong contact toxicity against the green peach aphid, Myzus persicae (Sulzer) (Hemiptera: Aphididae), which is a major insect pest of many vegetables and crops. However, little information is known about how M. persicae responds to trans-anethole at the molecular level. We conducted a comparative transcriptome analysis of M. persicae in response to a LD50 dose of trans-anethole. A total of 559 differentially expressed genes were detected in the treated individuals, with 318 genes up-regulated, and 241 genes down-regulated. Gene ontology (GO) analysis revealed that these genes were classified into different biological processes and pathways. We also found that genes encoding ATP-binding cassette (ABC) transporters, DnaJ, and cuticle proteins were dramatically up-regulated in response to trans-anethole. To study the function of these genes, we performed RNA interference (RNAi) analysis. Knockdown of an ABC transporter gene (ABCG4) and a DnaJ gene (DnaJC1) resulted in a significantly increased mortality rate in M. persicae following trans-anethole exposure, indicating the involvement of these two genes in the toxicity response to trans-anethole. The findings provide new insights into the mechanisms of M. persicae in coping with plant essential oils.

Contact Toxicity, Antifeedant Activity, and Oviposition Preference of Osthole against Agricultural Pests

Fang Dong,Xin Chen,Xingyuan Men,Zhuo Li,Yujun Kong,Yiyang Yuan,Feng Ge

Simple SummaryThe excessive utilization of chemical pesticides has resulted in the emergence of pesticide resistance, the disturbance of ecosystems, environmental degradation, and health hazards. Botanical pesticides offer a promising alternative to chemical pesticides, as they have minimal impacts on human health and the environment. Osthole, which has shown pesticidal activities, is the primary bioactive compound inCnidium monnieri(L.) Cusson. In this study, we assessed osthole for its contact toxicity, antifeedant activity, and oviposition preference against three agricultural pests: theTetranychus urticae,Myzus persicae, andBactrocera dorsalis, using laboratory bioassays. Osthole exhibited pesticidal effects against theT. urticaeandM. persicae, impeding their fecundity and development. Additionally, it demonstrated significant antifeedant activity against theT. urticaeand acted as a potent attractant for oviposition by theB. dorsalis. These findings highlight and expand the potential of osthole as a botanical pesticide for the control of these agricultural pests.AbstractOsthole, the dominant bioactive constituent in theCnidium monnieri, has shown acute pesticidal activities. However, its detailed toxicity, antifeedant, and oviposition preference effects against agricultural pests have not been fully understood, limiting its practical use. This study aimed to investigate the contact toxicity, antifeedant activity, and oviposition preference of osthole against three agricultural pests (Tetranychus urticae,Myzus persicae, andBactrocera dorsalis). Our results showed that theCnidium monnieri(L.) Cusson (CMC) has a high osthole content of 11.4 mg/g. Osthole exhibited a higher level of acute toxicity against theT. urticaeto four other coumarins found in CMC. It showed significant pesticidal activity againstT. urticaeandM. persicaefirst-instar nymphs and adults in a dose-dependent manner but not againstB. dorsalisadults. Osthole exposure reduced the fecundity and prolonged the developmental time of theT. urticaeandM. persicae. Leaf choice bioassays revealed potent antifeedant activity in theT. urticaeandM. persicae. Furthermore, the femaleB. dorsalisshowed a distinct preference for laying eggs in mango juice with 0.02 mg/mL osthole at 48 h, a preference that persisted at 96 h. These results provide valuable insights into the toxicity, repellent activity, and attractant activity of osthole, thereby providing valuable insights into its potential efficacy in pest control. Keywords:botanical pesticide;osthole;antifeedant;oviposition preference;Tetranychus urticae;Myzus persicae;Bactrocera dorsalis

Inhibition of Nicotinamidase Impacts Growth, Development, and Lifespan of Drosophila melanogaster and Myzus persicae

Hailiang Guo,Wenjin Xia,Changyang Chen,Song Yang,Dongmei Sun,Jizu Qiao,Jianya Su

Nicotinamidase (Naam), which is essential for insect NAD synthesis but absent in mammals, represents a potential pesticide target. Virtual screening identified 1-[4-(trifluoromethyl)-3-pyridinyl]-ethanone (4-TFPE) as a potent inhibitor of Myzus persicae (IC50 = 4.156 μM) and Drosophila melanogaster (IC50 = 2.855 μM) Naam. While 4-TFPE reduced pupal weight, development, and longevity in both species, it showed no lethal toxicity. Crucially, Naam expression was upregulated by nicotinamide accumulation, revealing a feedback regulation mechanism that compromises its pesticidal potential. Furthermore, Flumetnicam’s neurotoxic effects were found to be Naam-independent. These findings highlight how target compensation mechanisms can undermine pesticide development focused on single metabolic enzymes, suggesting the need for multitarget strategies.

A GDSL lipase confers resistance to piercing-sucking insects in tobacco by strengthening leaf cuticle

Na Chen,Yu-Die Xiong,Chi Zou,Yu-Wei Zhong,Hui Du,Yu-Jie Chi,Chan Zhao,Shu-Sheng Liu,Xiao-Wei Wang

SUMMARY Piercing-sucking insects, such as whiteflies and aphids, cause massive economic losses in major crops around the world. During feeding, the stylets of piercing-sucking insects navigate cuticles, cell walls, epidermal cells, and mesophyll cells; thus, these barriers are vital for the resistance of plants to insects. However, the relationship between insect stylet probing behavior and the composition and structure of these barriers remains unclear. Here, we identified a tobacco Cuticle Related Factor ( NtCRF ), which was induced significantly by whitefly infestation. Bioassays showed that NtCRF positively regulated plant resistance against whiteflies and green peach aphids. Silencing of NtCRF did not affect plant jasmonic acid (JA) and salicylic acid (SA) defenses but shortened the stylet probing time of phloem-feeders. Further studies confirmed that silencing of NtCRF resulted in significant structure destruction of the leaf cuticle and led to increased epidermal permeability. Overexpression of NtCRF in Arabidopsis also significantly enhanced the plant's resistance against whiteflies and green peach aphids. Our findings expand understanding of plant–insect interactions and provide a strategy for genetic improvement of crop resistance against piercing-sucking insects.

Comparative assays revealed distinct toxicity characterizations between pymetrozine and flonicamid

Tengfei Liu,Jianguo Niu,Yuying Gao,Xuan Liu,Jianya Su

Flonicamid is an insecticide with a unique mode of action, primarily inhibiting the feeding behavior of sap-sucking insects. However, its molecular target remains controversial. Reports indicate that both flonicamid and pymetrozine affect insect behavior and mobility. To investigate further, we compared the susceptibility of three insect species ( Myzus persicae , Nilaparvata lugens , and Drosophila melanogaster ) to flonicamid and pymetrozine under two different feeding postures. Results showed that feeding posture did not affect the survival curves or mean lifespans of the three insects. However, insect lifespans were significantly reduced under exposure to either insecticide compared to untreated controls. The effects of the two insecticides on susceptibility and lifespan under different feeding postures were markedly different: feeding posture significantly influenced the sensitivity of aphids, planthoppers, and fruit flies to pymetrozine but had minimal effects on the sensitivity of aphids and planthoppers to flonicamid. Flonicamid had only a minor impact on fruit flies' sensitivity. Survival curve analysis also revealed significant differences under pymetrozine stress, while no such differences were observed under flonicamid stress. These findings indicate that flonicamid exerts different toxicological effects on target insects (aphids and planthoppers) versus non-target insects (fruit flies), depending on feeding posture. This suggests that the toxicological mechanisms of flonicamid differ substantially between target and non-target organisms, with implications that findings in non-target insects may not directly apply to target pests. Our study provides valuable insights and directions for further research into flonicamid's mode of action, aiding in elucidating its underlying mechanisms.
