High-speed counter-current chromatographic separation and thermodynamic mechanism of an antioxidant from Morus alba leaves

Shuang Guo,Dongyu Gu,Chang Liu,Shanshan Tang,Yi Wang,Yi Yang

Morus alba leaves have a long history as a plant medicine and possess many biological activities. Antioxidant activity is the basis of its many biological activities. In order to explore the antioxidant effect of Morus alba leaves, antioxidant and its mechanism of scavenging free radicals have been investigated in this study. As results, rutin with the EC 50 of 27.94 ± 1.18 μg/mL and 25.86 ± 0.09 μg/mL to DPPH· and ABTS· + respectively was enriched by liquid-liquid extraction, and isolated by high-speed counter-current chromatography and identified by NMR, and was recognized as the main antioxidant in Morus alba leaves. The results of reaction thermodynamics and frontier molecular orbital theory indicated that 4′-OH in rutin was the most active group, which scavenged free radicals by hydrogen atom transfer in non-polar media and single electron transfer followed by proton transfer in polar media. Polar solvents were more helpful for rutin to exert its antioxidant effect. This study not only explored the antioxidant basis of Morus alba leaves, but also revealed its mechanism of scavenging free radicals.

Production and characterization of novel antioxidant peptides from mulberry leaf ferment using B. subtilis H4 and B. amyloliquefaciens LFB112

Zaheer Abbas,Jing Zhang,Yucui Tong,Baseer Ahmad,Junyong Wang,Haosen Zhang,Zhenzhen Li,Xubiao Wei,Rijun Zhang,Dayong Si

The objective of this study was to isolate and characterize antioxidant peptides from mulberry leaves fermented with Bacillus subtilis H4 and Bacillus amyloliquefaciens LFB112. The results indicated that fraction F4 (<1 kDa) exhibited superior DPPH activity compared to the F1 (>10 kDa), F2 (3–10 kDa), and F3 (1–3 kDa) fractions, and cytoprotective effect against lipopolysaccharides (LPS)-induced oxidative stress in RAW264.7 cells. Three novel peptides, FRFDP, RFGG, and GPPLAFGGGP, were identified in the F4 fraction of the mulberry leaf ferment, and docking results showed that these peptides could form stable carbon (covalent) and hydrogen bonds to the active sites of Keap1, thus regulating the Keap1-Nrf2 pathway by blocking the Nrf2 binding sites on Keap1. These peptides significantly upregulated the mRNA expression of Nrf2 , HO-1 , and NQO1 , providing indirect evidence of their potential to enhance cellular antioxidant defense via the possible activation of Keap1-Nrf2 pathway. Furthermore, these peptides showed good DPPH and ABTS scavenging activities, indicating their potential as antioxidant peptides. This study offers valuable insights into the integration of these novel peptides in developing mulberry leaf ferment as a functional food and their potential use as a feed additive.

Nutritional and functional components of mulberry leaves from different varieties: Evaluation of their potential as food materials

Yanfang Yu,Hongyan Li,Bing Zhang,Junwen Wang,Xuping Shi,Jinzhi Huang,Jianyuan Yang,Yanfei Zhang,Zeyuan Deng

Mulberry leaves had been used as material of medicine, drink, and functional foods in many countries, but the variety suitability for different applications is still not clear. In this study, the nutritional and phytochemical components of mulberry tender shoots from 19 varieties in China were investigated. Obvious genotypic diversity was observed in all the assessed components. The contents of crude protein (CP), total soluble sugars (TSS), crude fiber (CF), 1-deoxynojirimycin (DNJ), total phenolic (TP), and total flavonoid content (TF) were 27.63–37.36 g/100 g dry weight (DW), 58.71–150.31 mg/g DW, 9.90–13.85 g/100 g DW, 0.08–1.12 mg/g DW, 8.76–20.26 mg gallic acid equivalents (GAE)/g DW, and 21.36–56.41 mg rutin equivalents (RE)/g DW, respectively. Chlorogenic acid, rutin, isoquercitrin, and astragalin were considered to be mainly potential antioxidant compounds in mulberry leaves. According to the result of correlational analysis, principal component analysis (PCA), and hierarchical cluster analysis (HCA), the NS14 variety had high comprehensive healthy properties. GS8, D10, XS, TP2, and DHS could be appropriate materials for functional food or drink because of high content of phenolics or DNJ. Some varieties may have a potential application as protein-rich vegetables. The study suggests that some variety of mulberry could be selected and utilized rationally for their dietary properties.

Therapeutic mechanisms of mulberry leaves in type 2 diabetes based on metabolomics

Ma Quantao,Li Yaqi,Zhao Ruixue,Tang Ziyan,Li Jialin,Chen Cong,Liu Xiaoyao,Hu Yujie,Wang Ting,Zhao Baosheng

Background: Type 2 diabetes (T2D) is considered as one of the most significant metabolic syndromes worldwide, and the long-term use of the drugs already on the market for T2D often gives rise to some side effects. The mulberry leaf (ML), Morus alba L., has advantages in terms of its comprehensive therapeutic efficacy, which are characterized as multicomponent, multitarget, multipathway, and matching with the complex pathological mechanisms of diabetes. Methods: T2D rats were established by a high-fat diet combined with an intraperitoneal injection of streptozotocin; an evaluation of the hypoglycemic effects of the ML in combination with fasting blood glucose and other indicators, in addition to the utilization of metabolomics technology, was performed to analysis the metabolite changes in serum of rats. Results: MLs significantly reduced the fasting blood glucose of T2D rats, while improving the symptoms of polyphagia and polyuria. ML treatment altered the levels of various metabolites in the serum of T2D rats, which are involved in multiple metabolic pathways (amino acid metabolism, carbohydrate metabolism, and lipid metabolism), played a role in antioxidative stress and anti-inflammation, modulated immune and gluconeogenesis processes, and improved obesity as well as insulin resistance (IR).Conclusion: The ML contains a variety of chemical components, and metabolomic results have shown that MLs regulate multiple metabolic pathways to exert hypoglycemic effects, suggesting that MLs may have great promise in the development of new hypoglycemic drugs.

TGase-induced crosslinking of mulberry leaf protein particles as stabilizer of high-internal-phase Pickering emulsions: characterization and stability

Mingze Zhi,Yingshan Xie,Hongyan Li,Zeyuan Deng,Han Peng,Yanfang Yu,Bing Zhang

BACKGROUND Mulberry leaf protein (MLP) is a high-quality protein with significant nutritional value and functional properties. Enzymatic modification of proteins can enhance their functional properties by using proteases to covalently crosslink or hydrolyze proteins. This study investigates the potential of transglutaminase (TGase)-induced crosslinked MLP as an emulsifier in the formation of high-internal-phase Pickering emulsions. RESULTS Crosslinked MLP samples were prepared with TGase concentrations ranging from 0 to 25 U g −1 . High-internal-phase Pickering emulsions (80% v/v) were formed at pH 8, with a crosslinking temperature of 50 °C, a TGase concentration of 20 U g −1 and an optimal crosslinking time of 60 min. As the enzyme concentration increased, the content of exposed sulfhydryl groups progressively increased, while the total free sulfhydryl content remained relatively stable. After varying crosslinking durations, both total free and exposed sulfhydryl group contents initially increased before declining. Additionally, the content of free amino groups in MLP gradually decreased with higher enzyme dosages and longer crosslinking times. The surface hydrophobicity of crosslinked MLP increased initially, followed by a decrease, reflecting changes in the spatial structure of MLP. SDS-PAGE analysis confirmed the formation of polymer masses after TGase-catalyzed crosslinking. Under optimal crosslinking conditions, the high-internal-phase Pickering emulsion prepared with TGase-induced crosslinked MLP exhibited a relatively uniform droplet distribution. CONCLUSION TGase-induced crosslinking enhances both the emulsifying activity and stability of MLP emulsions. © 2025 Society of Chemical Industry.

Extraction optimization, characterization and antioxidant activity in vitro of polysaccharides from mulberry (Morus alba L.) leaves

Qingxia Yuan,Yufeng Xie,Wei Wang,Yuhua Yan,Hong Ye,Saqib Jabbar,Xiaoxiong Zeng

Extraction optimization, characterization and antioxidant activity in vitro of polysaccharides from mulberry leaves (MLP) were investigated in the present study. The optimal extraction conditions with an extraction yield of 10.0   ±   0.5% for MLP were determined as follows: extraction temperature 92   °C, extraction time 3.5   h and ratio (v/w, mL/g) of extraction solvent (water) to raw material 34. Two purified fractions, MLP-3a and MLP-3b with molecular weights of 80.99 and 3.64   kDa, respectively, were obtained from crude MLP by chromatography of DEAE-Cellulose 52 and Sephadex G-100. Fourier transform-infrared spectroscopy revealed that crude MLP, MLP-3a and MLP-3b were acidic polysaccharides. Furthermore, crude MLP and MLP-3a had more complicated monosaccharide compositions, while MLP-3b had a relatively higher content of uronic acid. Crude MLP, MLP-3a and MLP-3b exhibited potent Fe 2+ chelating power and scavenging activities on 1,1-diphenyl-2-picrylhydrazyl, hydroxyl, superoxide and 2,2′-azinobis-(3-ethyl-benzothiazolin-6-sulfonic acid) radicals. The results suggested that MLP could be explored as natural antioxidant.

Effects of enzymatic hydrolysis on physicochemical property and antioxidant activity of mulberry (Morus atropurpurea Roxb.) leaf protein

Chongzhen Sun,Yangwei Shan,Xin Tang,Duo Han,Xiyang Wu,Hui Wu,Marzieh Hosseininezhad

To improve the antioxidant efficiency of mulberry leaf protein (MLP), alcalase, protamex, papain, flavourzyme, neutrase, and trypsin were used to hydrolyze MLP. The yield of soluble peptides, secondary structures, molecular weight distributions, and antioxidant activities of MLP hydrolysates (MLPHs) were investigated. Results showed that the native MLP was rich in the fraction above 6.5 kDa and was mainly composed of β-sheets, while MLPHs were abundant in the fractions of 0.3–0.6 kDa and 0.6–6.5 kDa and were mainly composed of disordered coils and β-folds. Limited hydrolysis of MLP could lead to better antioxidant activity than extensive hydrolysis. After enzymatic hydrolysis, the content of total sugar and total phenol in MLP increased significantly. MLP hydrolysates prepared with neutrase, alcalase, and protamex were preferable to other enzymes. Meanwhile, an enzyme to substrate level of 1% and a hydrolysis time of 2 hr were the optimum conditions to obtain higher antioxidant hydrolysates using neutrase.

Protective Effects of Mulberry (Morus atropurpurea Roxb.) Leaf Protein Hydrolysates and Their In Vitro Gastrointestinal Digests on AAPH-Induced Oxidative Stress in Human Erythrocytes

Chongzhen Sun,Hongyan Li,Xiaodan Hui,Yurong Ma,Zhina Yin,Qingsong Chen,Cong Chen,Hui Wu,Xiyang Wu

Mulberry leaf protein hydrolysates (HMP), and their in vitro gastrointestinal digests (GHMP), have shown favorable chemical antioxidant activities. The aim of this study is to investigate the potential protective effects of HMP and GHMP against 2,2′-azobis(2-amidinopropane) dihydrochloride (AAPH)-induced oxidative stress in human erythrocytes. The inhibition rate of hemolysis, the reactive oxygen species (ROS) level, the concentration of malondialdehyde (MDA), the reduced glutathione (GSH) and oxidized glutathione (GSSH), and the enzymatic activities of total superoxide dismutase (SOD), catalase (CAT), and cellular glutathione peroxidase (GSH-Px) were evaluated as the biomarkers of oxidative status in human erythrocytes. The results showed that HMP and GHMP effectively inhibit the occurrence of erythrocyte hemolysis in the range of 0.025–1.0 mg/mL, and the inhibition rates of HMP and GHMP reached 92% and 90% at concentrations of 0.4 mg/mL and 1.0 mg/mL, respectively. HMP and GHMP reduced the AAPH-induced oxidative hemolysis damage via suppressing the generation of ROS by inhibiting the formation of MDA, maintaining the balance of GSH/GSSG, and preserving the activities of the antioxidant enzymes, including SOD, GSH-Px, and CAT. Our findings revealed that both HMP and GHMP could be used as natural antioxidants, and have the potential for further application in the development of functional foods. Keywords:mulberry leaf;digested protein hydrolysates;erythrocytes;oxidative stress;natural antioxidants

Simultaneous extraction and in situ separation of flavonoids and alkaloids from mulberry leaves using a pH-responsive deep eutectic solvent/water system

Ruili Ma,Weiguo Wu,Yikang Shu,Ran Gao,Ziyu Wang,Daoquan Tang,Yan Du,Shuai Ji

This paper reports a novel method for simultaneous extraction and in situ separation of flavonoids and alkaloids from mulberry leaves using a pH-responsive deep eutectic solvent (DES)/water system. Five novel DESs were synthesized, and isoquercitrin and deoxynojirimycin were respectively selected as the representative compounds of flavonoids and alkaloids to evaluate the extraction and separation efficiency. The optimal DES, namely [HexA (hexanoic acid)][MPD (2-methyl-2,4-pentanediol)], was identified, and the extraction and in situ separation conditions were optimized. After extraction and pH-induced phase separation, flavonoids were mainly enriched in the DES phase with alkaloids in the H 2 O phase, and the distribution ratios were 6.69:1 and 1:9 ([HexA][MPD]:H 2 O), respectively. Compared to traditional solvents, the [HexA][MPD]/H 2 O system showed higher extraction efficiency of flavonoids and alkaloids with higher purity. Moreover, [HexA][MPD] possessed excellent reusability. This study demonstrates the application of pH-responsive DES/H 2 O systems for simultaneous extraction and separation of natural products for the first time.

Minor Non-Cytotoxic Enantiomeric Constituents from the Leaves of Morus alba

Jie Xu,Yanfang Yang,Qianqian Li,Ziyi Dong,Yaoyang Guo,Zhansheng Jiang

ObjectivesThe current work aimed to isolate and identify new interesting small-molecule natural products from the leaves of a well-known medicinal plant Morus alba.MethodsThe ethanol extract of M. alba leaves was partitioned between EtOAc and water, and the EtOAc soluble part was fractionated by macroporous resin column chromatograph (CC) eluted with gradient EtOH. The 80% EtOH eluate was subject to silica gel, reversed phase C18, MCI gel and Sephadex LH-20 CC and finally purified by HPLC to afford four minor compounds 1–4, and the racemic 1 and 2 were further separated by chiral columns. Structural elucidation of these isolates was achieved via spectroscopic means especially HR-ESIMS and NMR, with the absolute configurations of new compounds assigned by ECD analyses. The cytotoxic screening was performed by MTT assay.ResultsA pair of new quinone enantiomers (1a/1b), a pair of new stilbenoid enantiomers (2a/2b), a known prenyl stilbenoid (3) and a known dihydrochalcone (4) were isolated and structurally characterized from the leaves of M. alba. They did not show significant cytotoxicity against MDA-MB231, A549, Hela and 4T1 tumor cell lines.ConclusionsThe M. alba leaves represent a rich source of new natural products, and the currently obtained constituents did not exert obvious growth inhibitory activity against the tested tumor cell lines. The bioactivities of the new isolates deserve further exploration in the future.

Investigation on Antioxidant Activity and Different Metabolites of Mulberry (Morus spp.) Leaves Depending on the Harvest Months by UPLC–Q-TOF-MS with Multivariate Tools

Zili Guo,Jiangxuan Lai,Yiwen Wu,Sheng Fang,Xianrui Liang

The changes in active components in mulberry leaves harvested in different months and their antioxidant activities were investigated. Ultra-high-performance liquid chromatography–tandem quadrupole time-of-flight mass spectrometry (UPLC–Q-TOF-MS) with multivariate statistical tools was used to investigate the chemical constituents in the extracts of mulberry leaves. The results indicated that mulberry leaves were rich in phenolic acids, flavonoids, organic acids, and fatty acid derivatives. In addition, 25 different compounds were identified in the different batches of mulberry leaves. The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was measured to evaluate the in vitro antioxidant activities of mulberry leaves. Among the four batches, batch A, harvested in December, exhibited the strongest DPPH radical-scavenging activity, while batch B, harvested in March, showed the weakest activity. This was related to the total phenolic content in the mulberry leaves of each batch. The optimal harvest time of mulberry leaves greatly influences the bioactivity and bioavailability of the plant. Keywords:mulberry leaf;chemical constituent;antioxidant;UPLC–Q-TOF-MS;free radical scavenging activity

The effects of exogenous GABA on the quality of processed mulberry leaf tea were investigated based on metabolomics

Baolin Han,Weitao Zhou,Rong Fan,Shufan Zhang,Xiaohu Ren,Hucheng Gong,Shulin Tian,Qiang Xu,Minghong Bian

Morus alba 'Seedless Dashi' is an excellent variety with large leaves, which is very suitable for making tea. GABA is an important functional component in mulberry leaf tea, but its role and influence on flavor components and metabolism of mulberry leaf tea are still unclear. In this study, metabolomics methods were used to comprehensively analyze the differences between externally added GABA mulberry leaf tea and non-added GABA mulberry leaf tea. The results showed that GABA treatment significantly increased the total amount and diversity of volatile components of mulberry leaf tea (+38.95%), especially the addition of 6 new pyrazine compounds, which gave mulberry leaf tea nutty and caramel characteristics. Compared with ordinary mulberry tea, 73 different metabolites were screened (65 up-regulated and 8 down-regulated). It was found that GABA tea promotes the accumulation of functional components by regulating amino acid and lipid metabolism pathways. This study provided a theoretical basis for the quality optimization of GABA mulberry leaf tea and confirmed that GABA has the dual role of flavor-enhancing and functional components in mulberry leaf tea.
