New butyrylcholinesterase inhibitor derived from mulberry twigs, a kind of agricultural byproducts

Yuyuan Zhu,Wei Xiang,Yao Shen,Yanan Jia,Yuansong Zhang,Lingshu Zeng,Jiaxin Chen,You Zhou,Xin Xue,Xianzhi Huang,Li Xu

Alzheimer's disease (AD) is one of the most common diseases in the elderly, imposing a significant financial and emotional burden on patients and their families. Inhibiting the enzyme cholinesterase is currently one of the main methods of delaying the symptoms of AD. The mulberry extract has good butyrylcholinesterase (BChE) inhibitory activity, but its main active ingredients and the mechanism of action are unknown. In this study, three potential BChE inhibitors, Kuwanon G, kuwanon H, and morusin, were isolated and identified from mulberry twigs by magnetic nanoparticles Fe 3 O 4 @BChE coupled with high-performance liquid chromatography-high-resolution mass spectrometry-solid-phase extraction-nuclear magnetic resonance (HPLC-HRMS-SPE NMR). Kuwanon G, kuwanon H, and morusin displayed potential inhibition activity, and the half maximal inhibitory concentration (IC 50 values) were 71.0 ± 2.62 μM, 10.9 ± 0.39 μM and 78.7 ± 2.61 μM, respectively, and the positive control, Galanthaminel was 60.4 ± 6.88 μM. It was found for the first time that Kuwanon H had butyrylcholinesterase inhibitory activity. The results of molecular docking revealed that Kuwanon H interacted primarily with the amino acid residues (Asp70, Ser72, Thr284, Leu286, and Ser287) of the BChE via hydrogen bonding, and interacted with Pro285 to produce hydrogen bonds via water molecular bridge bond, thereby exerting inhibitory activity. This research is critical in explaining the material basis and mechanism of mulberry twigs inhibiting or alleviating Alzheimer's disease as well as contributing to the development and utilization of mulberry twigs as an agricultural by-product.

Hierarchical tubular porous carbon derived from mulberry branches for long-life lead-carbon battery

Puqiang He,Jian Tu,Yi Yang,Hui Huang,Buming Chen,Chao Gao,Yapeng He,Zhongcheng Guo

Lead-carbon batteries (LCBs) possess remarkable technical merits over traditional lead-acid batteries, and the development of applicable carbon-based negative additives is considered as an essential mission for the further exponential advancement of LCBs. Herein, hierarchical tubular porous carbon (HTPC) with high specific surface area derived from mulberry branches was synthesized while evaluating the role of activation temperature on the structural evolution. As a result, the increase of activation temperature improves the specific surface area, reduces the graphitization degree of HTPC, whereas intrinsic tubular structure would collapse under high activation temperature. The critical function of HTPC as negative additive in LCBs is exhaustively elaborated, where the LCBs with 1.2 % HTPC deliver a discharge capacity of 165.4 mAh g −1 at 0.1 C compared with 140.5 mAh g −1 of blank sample. Especially, the cyclic life of LCBs under HRPSoC condition is tremendously extended by 3.17 times compared with the control sample. The performance improvement is attributed to the cooperation of HTPC with excellent conductivity, high specific surface, rich functional groups, and abundant tubular array structures, which facilitate the reduction of the internal resistance, increase the conversion rate of active materials, and inhibit the irreversible sulphation on negative lead. The proposed research provides a sustainable and scalable strategy to recycle the agricultural waste into outstanding carbon materials for high-performance LCBs.

Dihydromyricetin as a Dietary Supplement Enhances Hypoglycemic Efficacy: Synergistic Modulation of the Gut-Liver with Sangzhi Alkaloids

Yuwen Guan,Qiuyue Lv,Xinyu Wang,Baoping Jiang,Peigen Xiao

Chronic hyperglycemia, a hallmark of Type 2 diabetes mellitus, presents significant challenges in long-term management due to drug resistance and adverse effects. Dihydromyricetin (DHM), a natural flavanol from vine tea ( Ampelopsis grossedentata ), widely recognized as a nutraceutical, shows potential in enhancing the efficacy of pharmacological therapies. This study investigates the role of DHM in improving the hypoglycemic effects of Sangzhi Alkaloids (SZ-A), a commercially used glucosidase inhibitor. Our results demonstrate that DHM supplementation allows for a reduction in SZ-A dosage while maintaining or surpassing its glucose-lowering efficacy, thereby mitigating gastrointestinal side effects. Mechanistic studies reveal that this combination modulates the gut-liver axis, reducing ER stress, and enhancing gut microbiota diversity, including increased beneficial bacteria and non-12OH bile acids. These findings highlight DHM’s potential as an effective adjunctive nutraceutical, offering a safer and more efficient therapeutic strategy for managing hyperglycemia and related metabolic disorders.

Ramulus Mori (Sangzhi) Alkaloids Enhance Pancreatic β-Cell Maturation and Function by Targeting the CD81/Endoplasmic Reticulum Stress Pathway

Nan Wu,Wenjing Zhang,Sunyue He,Lusi Lu,Yiming Liu,Shuainan Liu,Lei Lei,Shuiya Sun,Yuchen Zhao,Chunyi Xu,Xihua Lin,Jiaqiang Zhou

Background β-cell dedifferentiation and impaired maturation are crucial contributors to β-cell dysfunction in type 2 diabetes mellitus (T2DM), yet targeted therapies remain limited. Objective To investigate the protective effects of Ramulus Mori (Sangzhi) Alkaloids (SZ-A) on β-cell function and maturation in T2DM. Methods A T2DM mouse model was induced by high-fat diet and streptozotocin, followed by SZ-A treatment. Single-cell RNA sequencing (scRNA-seq) was used to profile β-cell subpopulations, identifying Cd81 as a marker of β-cell immaturity. Cd81 -overexpressing diabetic mice, as well as β-TC-6 cells with either Cd81 overexpression or knockdown, were employed to explore the role of CD81 in SZ-A-mediated β-cell maturation. Western blotting was performed to evaluate the effects of SZ-A on endoplasmic reticulum (ER) stress. The potential interactions between SZ-A components—1-deoxynojirimycin (DNJ), fagomine (FAG), and 1,4-dideoxy-1,4-imino-D-arabinitol (DAB)—and CD81 were investigated using molecular docking. Results SZ-A treatment improved glucose metabolism in T2DM mice in a dose-dependent manner. ScRNA-seq revealed an increased proportion of Cd81 high β-cells with reduced expression of mature β-cell markers in diabetic islets, which was reversed by SZ-A. CD81 overexpression attenuated the metabolic benefits of SZ-A both in vivo and in vitro. Mechanistically, SZ-A alleviates CD81-mediated ER stress, at least in part via its active components DNJ and FAG, which may bind to CD81, suppress its expression, and thereby promote insulin secretion. Conclusion SZ-A promotes β-cell maturation and function by suppressing CD81-mediated ER stress. These findings reveal a novel therapeutic mechanism of SZ-A and identify CD81 as a potential target for T2DM intervention.

Discarded sericultural mulberry branch based triple layer composite phase change material with lignin enhanced thermal management capability

Qianqian Yu,Xiaohan Sun,Zejun Pan,Meiying Wu,Zhihui Dong,Yunyun Xia,Wan Nong,Xin Li,Zhaolin Yang,Yudong Li,Chengyu Wang

With the exhaustion of fossil fuels, prior phase change materials are characterized by such drawbacks as poor thermal conductivity, weak shape stability, and high costs. Therefore, the preparation of phase change materials with brilliant thermal-insulating properties, high thermal conductivity, and leakage-free properties has emerged as a crucial research focus. Herein, a sericultural mulberry branch-derived (SMB) composite phase change material was prepared by deep eutectic solvent pretreated SMB and vacuum-assisted impregnated paraffin wax with cupric oxide (CuO). The anisotropic porous SMB features a whole trunk structure similar to pith, xylem, and bark, which is closely tied physiological functions and provides a foundation for thermal storage and release. Triple layer porous structure of SMB as supporting material determined the excellent shape stability of composite phase change material. The addition of CuO significantly enhances the thermal conduction capacity. After 50 cycles heating, CuO/PW/DSMB retained 98 % of latent heat capacity and enthalpy change for heat absorption was measured at 91.66 J/g, which represents a decrease of 17.94 J/g, demonstrating a significant superior latent thermal effect and excellent thermal stability. Moreover, the triple layer of SMB uneven thermal transfer generate a self-regulating thermal management functional structure to achieve mutual thermal compensation. In summary, this composite phase change material, with its excellent properties of thermal insulation, high thermal conductivity, leakage-free nature, low cost, and easy availability, has broad application prospects in thermal energy storage and energy conservation.

An Eco-Friendly Slow-Release Urea Fertilizer Based on Waste Mulberry Branches for Potential Agriculture and Horticulture Applications

Yong Zhang,Xiying Liang,Xiaogang Yang,Hongyi Liu,Juming Yao

Development of a compound fertilizer for agricultural and horticultural applications remains an important challenge in the field of biomass synthesis on account of its raw material source and biodegradability. In this work, an eco-friendly slow-release urea fertilizer (SRUF) employing mulberry branch-g-poly(acrylic acid-co-acrylamide) (MB/P(AA-co-AM)) superabsorbent was prepared. The MB/P(AA-co-AM) superabsorbent was blended with sodium alginate, urea, and CaCl2 solutions to accomplish this synthesis process. The synthesis conditions of the SRUF and its application performance were examined. The results showed that under the optimal synthesis conditions the water absorbency and water absorbency rate of the SRUF reached 420.0 g/g and 60.0 (g/g)/min in deionized water, respectively. The water retention of the SRUF was 7.2 wt % after 25 d. The urea release in deionized water and soil both exhibited a typical slow release behavior. A degradation rate of 32.0 wt % was attained for the SRUF while it was buried for 90 d in soil.

Lyophilized aqueous extracts of Mori Fructus and Mori Ramulus protect Mesenchymal stem cells from •OH–treated damage: bioassay and antioxidant mechanism

Jiang Qian,Li Xican,Tian Yage,Lin Qiaoqi,Xie Hong,Lu Wenbiao,Chi Yuguang,Chen Dongfeng

Background Mori Fructus and Mori Ramulus are two traditional Chinese herbal medicines from mulberries. The present work explores their beneficial effects on •OH–treated mesenchymal stem cells (MSCs) and discusses possible mechanisms. Methods Lyophilized aqueous extracts of Mori Fructus (LAMF) and Mori Ramulus (LAMR) were prepared and analyzed using HPLC. LAMF and LAMR (along with morin) were further investigated for their effects on •OH-treated MSCs using the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl (MTT) assay. The direct antioxidation mechanisms were studied using 2-phenyl-4,4,5,5-tetramethylimidazoline-1-oxyl-3-oxide (PTIO•)-scavenging, 2,2′-azino-bis (3-ethylbenzo-thiazoline-6-sulfonic acid (ABTS+•)-scavenging and 1,1-diphenyl-2-picryl-hydrazl (DPPH•)-scavenging, as well as Cu2+-reducing and Fe3+-reducing antioxidant power. Finally, the indirect antioxidant mechanism was investigated based on the UV-vis spectra of Fe2+-chelation.Results In each LAMF and LAMR, seven phytophenols were successfully measured by HPLC, including five flavonoids (morin, rutin, astragalin, isoquercitrin and luteolin) and two non-flavonoids (chlorogenic acid and maclurin). MTT assays revealed that LAMF, LAMR and morin could effectively increase the survival of •OH-treated MSCs at 10–100 μg/mL, and could effectively scavenge PTIO• (IC 50 6609.7 ± 756.6, 4286.9 ± 84.9 and 103.4 ± 0.9 μg/mL, respectively), DPPH• (IC 50 208.7 ± 3.0, 97.3 ± 3.1 and 8.2 ± 0.7 μg/mL, respectively) and ABTS+• (IC 50 73.5 ± 5.8, 34.4 ± 0.1 and 4.2 ± 0.2 μg/mL, respectively), and reduce Cu2+ (IC 50 212.5 ± 7.0, 123.2 ± 0.9 and 14.1 ± 0.04 μg/mL, respectively) & Fe3+ (IC 50 277.0 ± 3.1, 191.9 ± 5.2 and 5.0 ± 0.2 μg/mL, respectively). In the Fe2+-chelating assay, the five flavonoids produced much stronger shoulder-peaks than the two non-flavonoids within 420–850 nm.Conclusion Mori Fructus and Mori Ramulus, can protect MSCs from •OH-induced damage. Such beneficial effects can mainly be attributed to the antioxidant action of phytophenols, which occurs via direct (ROS-scavenging) and indirect mechanism (Fe2+-chelating). The ROS-scavenging mechanism, however, include at least a H+-transfer and an electron-transfer (ET), and possibly includes a hydrogen-atom-transfer (HAT). In the Fe2+-chelating, flavonoids are more effective than non-flavonoids. This can be attributed to several adjacent planar chelating-sites between the 3-OH and 4-C = O, between the 4-C = O and 5-OH, or between the 3′-OH and 4′-OH in flavonoids. Such multiple-Fe2+-chelating reactions cause overlap in the UV-vis absorptions to deepen the complex color, enhance the peak strength, and form shoulder-peaks. By comparison, two non-flavonoids with catechol moiety produce only a weak single peak.

Efficient extraction of highly reactive lignin from waste mulberry branches

Gong Xiujin,Wang Dengfeng,Zheng Qiannan,Liu Lin,Wu Han,Li Zihan,Hong Haojie,Yao Juming

Lignin extraction and upgrade are the great importance to lignin valorization but are closely related to raw material sources, separation methods, and conditions. The advantage of this work is to firstly exploit an efficient and practical approach to extract highly reactive lignin from waste mulberry branches (MB) using green deep eutectic solvent (DES). To better understand the relationship between the extraction conditions and the properties of lignin, the influences of DES components, extraction temperature and time on the extraction efficiency, average molecular weight, chemical structure, and thermal stability of lignin were systematically investigated and analyzed. The synergistic effects of processing conditions resulted in high extraction efficiency with 62.9% of yield and 94.5% of purity. Under optimal conditions with LA/ChCl ratio of 2:1, extraction temperature of 150 °C, and time of 6 h, the obtained lignin had relatively low molecular weight ( M n = 1900 g/mol and M w = 2841 g/mol) and narrow distribution (PDI = 1.49), simultaneously had high total hydroxyl content of 9.15 mmol/g. Benefiting from high hydroxyl content and low molecular weight, the extracted lignin possessed excellent miscibility and reactivity which could participate in polymerization reaction to synthesize lignin-derived rigid polyurethane foam (LRPUF). The integration of the highly reactive lignin significantly improved the specific compressive strength and modulus of LRPUF to 6.4 and 76.0 MPa/(g/cm 3 ), which increased by 7% and 22.4% than those of petroleum-based RPUF. Therefore, this work not only provided an efficient and practical strategy for lignin extraction and upgrade, but also proposed a potential of high value-added utilization of waste mulberry branches. Non-targeted metabonomics and transcriptomics revealed the mechanism of mulberry branch extracts promoting the growth of Sanghuangporus vaninii mycelium

Huo Jinxi,Sun Yuqing,Pan Meiliang,Ma Huanyan,Lin Tianbao,Lv Zhiqiang,Li Yougui,Zhong Shi

Sanghuangprous vaninii is a wood-inhabiting fungus, and its mycelium and fruiting body show excellent medicinal values. Mulberry is one of the major hosts of S. vaninii, however, the mechanism of mulberry affecting the growth of S. vaninii has not been reported. In the present study, a mulberry-inhabiting strain of S. vaninii was selected to explore the effects of mulberry branch extracts (MBE) on the growth of the strain. Results showed that MBE could significantly promote the growth of S. vaninii mycelium at the concentration of 0.2 g/l. After 16 days of liquid culture, the dry weight of mycelium in 0.2 g/l MBE medium was higher by three times compared with that in the control. The non-targeted metabonomic analysis of the culture medium at different culture times and concentrations was conducted to find the key components in MBE that promoted the growth of S. vaninii mycelium. Under the different concentrations of MBE culture for 10 and 16 days, 22 shared differential metabolites were identified. Next, in accordance with the peak value trend of these metabolites, HPLC–MS and liquid culture validation, four components derived from MBE (i.e., scopoletin, kynurenic acid, 3,5-dihydroxybenzoic acid and 2,4-dihydroxybenzoic acid) could significantly increase the growth rate of mycelium at the concentration of 2 mg/l. Transcriptomic and qRT-PCR analyzes showed that MBE could upregulate hydrolase-related genes, such as serine–glycine–asparaginate–histidine (SGNH) hydrolase, alpha-amylase, poly-beta-hydroxybutyrate (PHB) depolymerase, glycosyl hydrolase family 61, cerato-platanin protein and Fet3, which might enhance the nutrient absorption ability of S. vaninii. Importantly, MBE could significantly increase the content of harmine, androstenedione and vesamicol, which have been reported to possess various medicinal effects. Results suggested that MBE could be an excellent additive for liquid culture of S. vaninii mycelium, and these hydrolase-related genes also provided candidate genes for improving the nutrient absorption capacity of S. vaninii.

Mulberroside A ameliorates CCl4-induced liver fibrosis in mice via inhibiting pro-inflammatory response

Baozhang Shi,Jinqiang Qian,Hongyue Miao,Shuo Zhang,Yue Hu,Puqing Liu,Liping Xu

Liver fibrosis is caused by a variety of pathogenic factors. It is mainly characterized by chronic liver damage mediated by the imbalance between extracellular matrix synthesis and degradation. If the injury factor cannot be removed for a long time, fibrosis will progress to cirrhosis or even cancer. The development of liver fibrosis is a very complex process which is related to the activation of hepatic stellate cells (HSCs), oxidative stress, and cytokines produced by immune cells. At present, screening of substances with anti-inflammatory activity from natural plant extracts has become a new research focus in the prevention and treatment of liver fibrosis. Mulberry twig is a commonly used traditional Chinese medicine. Pharmacological studies have shown that mulberry twig has anti-inflammatory and antioxidant activities. Thus, it is likely that Mulberry twig contains active substances with liver protection functions. The present study aimed to explore the effect of Mulberroside A (MulA), the main active ingredient from Mulberry twig, on acute liver injury induced by CCl 4 in mice. MulA treatment could significantly alleviate the CCl 4 -induced liver injury, as evidenced by histological analysis and Masson staining. However, we observed that MulA inhibited the expressions of collagen I and α-SMA in livers of CCl 4 -treated mice but did not directly inhibit the proliferation and activation of HSCs. Finally, we analyzed the anti-inflammatory effect of MulA and demonstrated that it could markedly inhibit the pro-inflammatory cytokines release in liver tissues and in cultured macrophages, thereby alleviating liver fibrosis. Our findings suggest MulA as a potential therapeutic candidate for liver injury and inflammatory diseases.

Rapid Screening of Nonalkaloid α-Glucosidase Inhibitors from a Mulberry Twig Extract Using Enzyme-Functionalized Magnetic Nanoparticles Coupled with UPLC-MS/MS

Fanshan Feng,Wei Xiang,Han Gao,Yanan Jia,Yuansong Zhang,Lingshu Zeng,Jiaxin Chen,Xianzhi Huang,Li Xu

Mulberry twigs are an important source of α-glucosidase inhibitors. To date, research studies on α-glucosidase in mulberry twigs have mainly focused on alkaloids such as 1-deoxynojirimycin (DNJ). Preliminary studies have shown that there may be more active nonalkaloid α-glucosidase inhibitors in mulberry twigs. In this study, we immobilized α-glucosidase on Fe3O4@SiO2 for the first time and rapidly screened four nonalkaloid α-glucosidase inhibitors (kuwanon G, kuwanon C, kuwanon H, and morusin) using ligand fishing technology with ultrahigh-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) from the mulberry twig extract of Jialing 20, the excellent artificial triploid variety of mulberry cultivated extensively in Southwest China. The half maximal inhibitory concentrations (IC50) of kuwanon H and kuwanon G were 2.82 ± 0.68 and 2.83 ± 0.31 μM, respectively, with better inhibition activity than that of DNJ (with an IC50 of 7.04 ± 0.82 μM). Meanwhile, the molecular docking results showed that the action sites of these two isopentenyl flavonoids on α-glucosidase were different from that of DNJ. In brief, this work is beneficial to discovering new α-glucosidase inhibitors from mulberry twigs quickly and accurately and provides a theoretical basis for the mulberry twig extract as a functional food or a natural hypoglycemic drug source, as well as a reference for directional breeding of mulberry, which greatly improves the exploitation and utilization value of mulberry twigs as an agricultural byproduct in the fields of agricultural production, functional food, and natural medicine.
