In the present study, a simple and efficient approach based on the online extraction-high performance liquid chromatography coupled with ABTS antioxidant assay (OLE-HPLC-ABTS) was established to quickly and directly analyze the antioxidants in S. baumii. Through this system, the HPLC mobile phase via a guard column packed with a S. baumii sample was used for online extraction (OLE). The separation was performed on an Agilent Poroshell EC-C18 column with a gradient elution using 0.1% formic acid (A) and 0.1% formic acid–acetonitrile (B) as mobile phase systems and detected at a wavelength of 254 nm. Then, the separated compounds were reacted with the antioxidant solution (ABTS), and the response was recorded at a wavelength of 400 nm. The developed analytical method was successfully applied to S. baumii samples, and eight antioxidants were identified. The established system integrated the online extraction, separation and online antioxidant detection, which is rapid, efficient, and suitable for the rapid screening of antioxidant compounds from solid sample mixtures.
Sanghuangporus species are precious edible and medicinal fungi that are traditionally used in Asian countries. Triterpenoids, the major bioactive constituents of Sanghuangporus spp. are currently receiving a lot of attention from the scientific community due to their potent pharmacological activities. However, very few reports have been conducted about the possibility of adjusting the fermentation conditions to increase the triterpenoids yield from certain fungi such as Sanghuangporus baumii . In this study, we examined the effects of four different extracts from mulberry shavings on the production of triterpenoids and traced the major components from the effective extracts by GC-MS to provide a reference for further exploration of the triterpenoids synthetic mechanism. It was found that unsaturated fatty acids significantly promoted the production of triterpenoids. The results suggested a direct correlation between the degree of unsaturation of fatty acid and the triterpenoids yield. The best stimulatory additive was linolenic acid . It increased the triterpenoids yield to 573 mg/L, which was 2.53 times higher than the control. This study not only showed that addition of mulberry shavings extract was an efficient fermentation protocol to improve the triterpenoids yield of S. baumii , but it also highlighted the importance of fatty acid content in the production of triterpenoids.

There have been many studies on the activities and polysaccharide production of Sanghuangporus vaninii . However, few studies have looked at triterpene production from S. vaninii using liquid-state fermentation. A method for enhancing the production of triterpenes by in situ extractive fermentation (ISEF) was studied. Eight solvents were investigated as extractants for triterpene production in the ISEF system. The results showed that using vegetable oil as an extractant significantly increased the yield of total triterpenes and biomass of S. vaninii YC-1, reaching 18.98 ± 0.71 and 44.67 ± 2.21 g/L, respectively. In 5 L fermenter experiments, the added vegetable oil improved the dissolved oxygen condition of the fermentation broth and promoted the growth of S. vaninii YC-1. Furthermore, adding vegetable oil increased the expression of fatty acid synthesis-related genes such as FAD2 and SCD , thereby increasing the synthesis of unsaturated fatty acids in the cell membrane of S. vaninii YC-1. Therefore, the cell membrane permeability of S. vaninii YC-1 increased by 19%. Our results indicated that vegetable oil increased the permeability of S. vaninii YC-1 cell membranes to promote the production of total triterpenes. The use of vegetable oil as an extractant was thus effective in increasing the yield of triterpenes in the ISEF system.
