Design and characterization of 3D printed gradient scaffolds with spatial distribution of pore sizes

Fan Ye,He Yang,Chuhang Hong,Xiaopei Wu,Honglian Dai

In bone repair, scaffold structural design is essential for optimizing mechanical performance and osteogenic potential. Gradient structural scaffolds are engineered to balance mechanical qualities and permeability which can improve the material transport capacity of scaffolds. This study designed and fabricated gradient structural scaffolds with spatially distributed pore size, and investigated their mechanical and fluid dynamics properties, coupled with in vitro osteogenic experiments to understand the impact of pore size distribution. The results indicated that, compared to traditional uniform pore scaffolds, gradient scaffolds exhibited a significant increase in mechanical strength, fluid permeability, and in vitro osteogenic activity. To sum up, the gradient scaffolds demonstrated better comprehensive performance and potential for use in the structural design of bone repair scaffolds, offering an important reference value for the three-dimensional structural design of scaffolds.

Enhanced hyaline cartilage formation and continuous osteochondral regeneration via 3D-Printed heterogeneous hydrogel with multi-crosslinking inks

Zhonglian Wu,Hang Yao,Haidi Sun,Zehao Gu,Xu Hu,Jian Yang,Junli Shi,Haojun Yang,Jihang Dai,Hui Chong,Dong-An Wang,Liwei Lin,Wang Zhang

The unique gradient structure and complex composition of osteochondral tissue pose significant challenges in defect regeneration. Restoration of tissue heterogeneity while maintaining hyaline cartilage components has been a difficulty of an osteochondral tissue graft. A novel class of multi-crosslinked polysaccharide-based three-dimensional (3D) printing inks, including decellularized natural cartilage (dNC) and nano-hydroxyapatite, was designed to create a gradient scaffold with a robust interface-binding force. Herein, we report combining a dual-nozzle cross-printing technology and a gradient crosslinking method to create the scaffolds, demonstrating stable mechanical properties and heterogeneous bilayer structures. Biofunctional assessments revealed the remarkable regenerative effects of the scaffold, manifesting three orders of magnitude of mRNA upregulation during chondrogenesis and the formation of pure hyaline cartilage. Transcriptomics of the regeneration site in vivo and scaffold cell interaction tests in vitro showed that printed porous multilayer scaffolds could form the correct tissue structure for cell migration. More importantly, polysaccharides with dNC provided a hydrophilic microenvironment. The microenvironment is crucial in osteochondral regeneration because it could guide the regenerated cartilage to ensure the hyaline phenotype.

Hydroxyapatite Gradient Poly (Vinyl Alcohol)/Bacteria Cellulose Bone Scaffold via Buoyancy-Driven Gradient Method

Zhang Quanchao,Wang Huiwen,Shi Jian,Luo Honglin,Yin Chen,Wan Yizao

The repair of bone defects necessitates the establishment of sophisticated gradients. However, most strategies for gradient casting require specialized apparatus and have high equipment requirements. Therefore, there is a strong desire to develop a simple method for constructing gradient scaffolds that mimic the hierarchical structure of native bone. In this study, we prepared poly (vinyl alcohol)/bacterial cellulose (PVA/BC) scaffolds with gradient hydroxyapatite (HAp) content using the buoyancy-driven gradient (BG) method. Scanning electron microscopy characterization revealed that HAp gradient scaffold exhibited the gradient microstructure and HAp content. Furthermore, HAp gradient scaffold demonstrated enhanced adhesion, spreading, and proliferation of MC3T3-E1 cells and displayed excellent osteogenic ability. Additionally, when applied in subcutaneous implantation models in mice, HAp gradient scaffold showed superior biocompatibility compared to other scaffolds tested in vitro. These results present a straightforward approach for fabricating gradient HAp bone scaffolds that hold great potential as candidate materials for bone regeneration.

Dual gradients of bioactive components on electrospun fibers for cell migration and controlled stem cell differentiation

Longfei Li,Tong Wang,Kelly Van,Xiaodi Zhang,Xindan Zhang,Jian Song,Rui Shi,Liqun Zhang,Jiajia Xue

The construction of gradient scaffolds to mimic the in vivo microenvironment for promoting tissue regeneration is attractive, especially by incorporating biochemical cues with physical guidance to manipulate cell behavior. Here, we report a general method to generate dual concentration-gradient of two types of bioactive components on scaffolds composed of uniaxially aligned electrospun nanofibers through layer-by-layer self-assembly. Different layers of collagen (COL) and chondroitin sulfate (CS) are sequentially coated onto the specific positions of uniaxially aligned electrospun polycaprolactone (PCL) nanofibers upon electrostatic attraction. In this case, dual stepwise gradients of COL and CS are generated along the fiber alignment. In demonstrating the potential application, the dual gradients can effectively promote the migration and proliferation of bone marrow stem cells (BMSCs) and spatially control their osteogenic differentiation. This simple strategy for generating dual gradients can be easily extended to different types of gradient scaffolds for bone regeneration and related applications involving cell migration, stem cell differentiation, and tissue interface.
