Layer-by-layer coating of carboxymethyl chitosan-gelatin-alginate on cotton gauze for hemostasis and wound healing

Weishi Zheng,Chuntao Chen,Xujun Zhang,Xiaodong Wen,Yuanxiang Xiao,Lin Li,Qingbo Xu,Feiya Fu,Hongyan Diao,Xiangdong Liu

We developed a hemostatic material able to quickly stop wound bleeding by successively constructing carboxymethyl chitosan (CMC), gelatin, and alginate onto a cotton gauze surface through a simple layer-by-layer (LbL) process. Briefly, we grafted CMC onto the cotton fibers through an esterification reaction with the hydroxyl groups of the cellulose chains, and then built a gelatin layer on the CMC layer by the ionic reactions with CMC. Finally, we integrated alginate by the lamellar structure through the cross-linking reactions caused by the Ca 2+ ions permeated from the gelatin layer. Characterization analyses demonstrated that the resulting composite dressing possessed high fluid absorption, excellent biocompatibility, and hemocompatibility. A better hemostatic performance was also revealed in animal tests compared with cotton gauze, and it showed a short hemostasis time and a small amount of blood loss in the mouse liver injury model and mouse-tail amputation model. Moreover, the composite dressing also exhibited a significant promotion effect of wound healing in a mouse defect wound model. This new composite dressing, which was prepared by natural raw materials, is a promising candidate for wound dressing and demonstrated great potential in medical applications.

Superhydrophobic cotton gauze with durably antibacterial activity as skin wound dressing

Liu Gongyan,Xiang Jun,Xia Qiongfen,Li Kaijun,Lan Tianxiang,Yu Ling

Abstract Common cotton gauze was endowed with both superhydrophobic and antibacterial properties by a dip-coating method that involved sequential deposition of positively charged chitosan (CS), negatively charged gallic acid modified silver nanoparticles (GA@AgNPs) and 1H,1H,2H,2H-perfluorodecanethiol (PFDT) with low surface energy on cotton fabrics. After such surface coating, the wettability of gauze surface was converted from superhydrophilic to superhydrophobic with a water contact angle (CA) of 158 ± 2.2° and sliding angle (SA) of 5.2 ± 1.8°, exhibiting water repellency, antifouling ability as well as bacterially antiadhesive activity. Moreover, such PFDT/GA@AgNPs/CS-coated cotton fabrics showed efficiently antibacterial activities against Escherichia coli ( E. coli ) and Staphylococcus aureus ( S. aureus ), which was mainly attributed to the synergistic effect of contact-killing of CS and continuous release of Ag + . In addition, it was found that the outer PFDT deposition could act as a barrier to prevent leaching of the AgNPs during laundry, enhancing the antibacterial durability. Graphical abstract

A reusable ionic liquid-grafted antibacterial cotton gauze wound dressing

Fang Huan,Li Dongrun,Xu Longquan,Wang Yi,Fei Xu,Tian Jing,Li Yao

Abstract Rapid and convenient treatment of wounds is urgently needed in daily life. It is very essential to finish the dressing and antibacterial treatment in one step. In this study, we developed a reusable ionic liquid-grafted antibacterial cotton gauze wound dressing using 1-vinyl-3-butylimidazolium bromide, 3-aminopropyltriethoxysilane, glycidyl methacrylate and butyl acrylate. The ionic liquid was grafted onto the surface of the cotton gauze through covalent bond to obtain antibacterial properties. The successful grafting was confirmed by Fourier transform infrared spectroscopy, scanning electron microscopy and energy-dispersive X-ray spectroscopy. The biocompatibility, antibacterial ability and wound healing properties of antibacterial cotton gauze were evaluated. The antibacterial cotton gauze exhibited effective antibacterial ability against Escherichia coli ( E. coli ), Staphylococcus aureus ( S. aureus ) and Candida albicans ( C. albicans ), and they were not cytotoxic to L929 mouse fibroblasts. The reusability experiment showed that the antibacterial cotton gauze had good antibacterial activity even after three repeated antibacterial tests. In addition, the antibacterial cotton gauze effectively promoted skin wound recovery in a rat model. These results indicated that the antibacterial cotton gauze could be used in the biomedical field, especially as an antibacterial wound dressing. Graphical abstract

Synthesis of photothermal antimicrobial cotton gauze using AuNPs as photothermal transduction agents

Fengyi Cao,Changmin Wei,Gangqing Ma,Like Hou,Rencong Zhang,Lin Mei,Qi Qin

Cotton gauze has been used as a wound dressing since the 19th century, and still plays an important role in current clinical therapies. However, the antimicrobial ability of cotton gauze is limited. In this work, gold nanoparticles (AuNPs) were used as photothermal transduction agents to synthesize modified photothermal antimicrobial cotton gauze. The modified cotton gauze was synthesized by immersing and heating the clinical cotton gauze with AuNPs solution. XPS, ICP-OES, FTIR, XRD and SEM characterizations confirmed that AuNPs were successfully decorated on the surface of cotton gauzes. Besides, the mechanical properties, air and water vapour permeability performance of cotton gauze were not changed after modification. Photothermal antimicrobial experiments confirmed that AuNPs modified on the cotton gauze could convert light to heat, inducing rapid temperature increase of the cotton gauze. And the heat could kill microbial cells permeated in the modified cotton gauze, giving it the potential of being used for photothermal antimicrobial therapy.

Chitosan and polyhexamethylene guanidine dual-functionalized cotton gauze as a versatile bandage for the management of chronic wounds

Chao Yang,Guofang Liu,Junpeng Chen,Bairui Zeng,Tianxi Shen,Dongchao Qiu,Chen Huang,Lin Li,Dongfan Chen,Jiale Chen,Zhixiang Mu,Hui Deng,Xiaojun Cai

Development of versatile medical dressing with good immediate and long-lasting antibacterial, hygroscopic and moisturizing abilities is of great significance for management of chronic wounds. Cotton gauze (CG) can protect wounds and promote scabbing, but can cause wound dehydration and loss of biologically active substances, thereby greatly delays wound healing. Herein, a bi-functional CG dressing (CPCG) was developed by chemically grafting polyhexamethylene guanidine (PHMG) and physically adsorbing chitosan (CS) onto the CG surface. Due to the powerful microbicidal activity of PHMG, CPCG exhibited excellent immediate and long-lasting antibacterial activity against gram-positive and gram-negative bacteria. Moreover, the abundant hydroxyl and amino groups in CS endowed CPCG with good biocompatibility, moisture absorption, moisturizing and cell scratch healing performances. Importantly, CPCG can be easily fabricated into a bandage to conveniently manage infected full-skin wounds. Together, this study suggests that CPCG is a versatile wound dressing, having enormous application potential for management chronic wounds.

Modified cotton gauze with high hemostatic efficacy due to controllable hygroscopicity and wet tissue adhesiveness

Huiying Li,Shuichang Chen,Hongjian Huang,Yan Fang,Yunxiang Weng,Qinhui Chen,Xinqing Liu,Haiqing Liu

In circumstances of severe traumatic bleeding, urgent bleeding control is vital to the wellness and life of the wounded person. Among many hemostatic measures, gauze is widely used because of cost-effectiveness, safety and efficiency. However, it may suck a large amount of blood before bleeding stops. Thus, advanced hemostatic gauzes less blood loss and quick hemostatic rate and is greatly demanded. Herein, a novel tissue adhesive gauze (TA-gauze) is developed by coating a thin layer of polyethyleneimine/poly(acrylic acid) (PEI/PAA) on the plain cotton gauze. This TA-gauze has a reduced hydrophilicity and gets a new trait of wet tissue adhesiveness from the PEI/PAA coating layer. The TA-gauze shows much improved hemostatic performance in the rat injury models, that is, its blood loss in the rat femoral artery model is just 13% that of the cotton gauze, due to it can retard the blood movement in the gauze and at the seam of gauze/tissue through a synergistic effect of tissue adhesion and hygroscopicity. It is biocompatible and on-demand detachable. The simple preparation procedure and high hemostatic efficiency of the TA-gauze may promise a bright future for an ideal hemostatic dressing for bleeding control.

A pH-Sensitive and mitochondria-modulating intelligent multilayered dressing system for a comprehensive therapeutic strategy of diabetic wounds

Chunlin Li,Jinyuan Chen,Guoying Deng,Peilin Wang,Hao Zhang,Kai Zhu,Qitao Hu,Shiqing Feng,Qiugen Wang,Haodong Lin

Diabetic wounds management remains a significant global challenge due to impaired wounds healing, bleeding, and infection. Oxidative stress (OS), resulting from dysregulated mitochondrial reactive oxygen species (mtROS), plays a crucial role in the diabetic wounds pathogenesis. Furthermore, diabetic infected wounds can induce excessive ROS production and an acidic microenvironment due to the accumulation of bacterial metabolic products. Thus, a pH-sensitive intelligent dressing could serve as an ideal carrier for precise regulation of mtROS and promote faster diabetic wounds healing. In this study, we fabricated a pH-sensitive and mitochondria-modulating intelligent multilayered dressing system. We synthesized mesoporous silica-coated selenium particles (SS) for mtROS regulation and infection resistance. These SS were then loaded into a polyvinyl alcohol (PVA)/(N-[(2-hydroxy-3-trimethylamine) propyl] chitosan chloride, HTCC) hydrogel for pH-sensitive controlled release, blood coagulation, and antibacterial treatment. Finally, the resulting system was cross-linked with carboxymethyl cotton gauze (CG) to enhance hemostatic ability and reduce adhesiveness. Experimental results from cellular and animal models of diabetic wounds demonstrated that HTCC/PVA/SS-CG dressing promotes faster hemostasis, protects against microbial infection, and accelerates diabetic wounds healing through its mitochondria-modulating action, which offers a comprehensive treatment strategy for diabetic wounds and shows promising prospects for clinical transformation.
