The evolution of pest resistance to insecticidal proteins from Bacillus thuringiensis (Bt) restricts the sustainable application of transgenic Bt crops, and exploring other ecofriendly insecticides to efficiently suppress Bt-resistant populations is thus urgently needed. The plant secondary metabolite rutin possesses pest resistance effects and plays an important role in plant resistance to pests, but the effect of rutin on pink bollworms is unclear. We investigated the effects of rutin on two types (Bt-susceptible and Bt-resistant) of pink bollworms ( Pectinophora gossypiella ). Rutin had significant inhibitory effects on the growth, development, and oviposition of both Bt-susceptible and Bt-resistant strains. For both strains, rutin ingestion by larvae did not significantly affect larval survival at 7 days after treatment or egg hatching rate but significantly decreased pupation rate, eclosion rate, neonate-to-adult survival, pupal weight and eggs per female and significantly prolonged larval developmental periods, and the effect increased with increasing rutin content. The response to rutin ingestion was basically consistent between Bt-resistant and Bt-susceptible strains, indicating that the Bt-resistant strain did not exhibit cross-resistance to rutin. These results provide a reference basis for green control and resistance management of pink bollworms and may be used as a component of comprehensive management for this pest.

Carboxylesterasesare an important class of detoxification enzymes involved in insecticide resistance in insects. A subgroup ofHelicoverpa armigeraesterases, known as Clade 001, was implicated in organophosphate and pyrethroid insecticide resistance due to their overabundance in resistant strains. In this work, a novel carboxylesterasegene001DofH. armigerafrom China was cloned, which has an open reading frame of 1665 nucleotides encoding 554 amino acid residues. We used a series of fusion proteins to successfully express carboxylesterase 001D inEscherichia coli. Three different fusion proteins were generated and tested. The enzyme kinetic assay towards 1-naphthyl acetate showed all three purified fusion proteins are active with aKcat between 0.35 and 2.29 s−1, and aKm between 7.61 and 19.72 μM. The HPLC assay showed all three purified fusion proteins had low but measurable hydrolase activity towardsβ-cypermethrin and fenvalerate insecticides (specific activities ranging from 0.13 to 0.67 μM·min−1·(μM−1·protein)). The enzyme was stable up to 40 °C and at pH 6.0–11.0. The results imply that carboxylesterase 001D is involved in detoxification, and this moderate insecticide hydrolysis may suggest that overexpression of the gene to enhance insecticide sequestration is necessary to allow carboxylesterases to confer resistance to these insecticides inH. armigera. Keywords:Helicoverpaarmigera;carboxylesterase;pyrethroid;heterologous expression

Furanocoumarins are a class of chemical compounds with phototoxic properties. For herbivores, efficient detoxification of such defense compounds is the prerequisite to feed successfully on furanocoumarin-containing plants. The cotton bollworm Helicoverpa armigera is a very important polyphagous pest in agriculture, but how it copes with toxic furanocoumarins in some of its host plants is not well understood. Given that cytochrome P450s are well known for their capacity in xenobiotic metabolism, this study attempted to explore the potential roles of cytochrome P450s in furanocoumarin transformation in this pest. Our data showed that two linear structures (psoralen and xanthotoxin) could be metabolized by three recombinant CYP9A enzymes, but no detectable depletion was observed for the linear one with the 8-dimethylallyloxy substituent on the coumarin moiety (imperatorin) and the angular furanocoumarin (angelicin). Initial epoxidation of the double bond connecting C2' and C3' of the furano ring following by cleavage of the epoxidated furan ring, leading to the formation of more soluble, less reactive and nonphotosensitizing metabolites, was identified as a common mechanism of linear furanocoumarin metabolism using a quadrupole/time-of-flight (Q-TOF) mass spectrometry interfaced with a high performance liquid chromatography (HPLC) system. Our data demonstrated that multiple P450s were involved in the detoxification of linear furanocoumarins in the cotton bollworm. These findings contribute to a better understanding of the biochemical basis of adaptation to plant defense chemicals in this economically important pest. Summary CYP9A12, CYP9A14, and CYP9A17 are capable of metabolizing psoralen and xanthotoxin. Epoxidation of the C2'−C3' double bond of the furano ring is the common pathway. The three CYP9A enzymes exhibit no detectable activity against angelicin and imperatorin.

Herbivore-induced plant volatiles (HIPVs), chemicals produced by plants infested by herbivorous insects, can act as kairomones that recruit natural enemies of the pest herbivore.Agrotis segetum(Denis and Schiffermüller) is a common, important pest of seedling cotton in Xinjiang Province, China, and the braconidMicroplitis mediator(Haliday) is an important mortality factor of this pest’s larvae. In olfactometer tests, which included healthy foliage, infested foliage, or infested roots,M. mediatorpreferredA. segetum-infested cotton plants to healthy cotton plants. In GC-MS analyses of plant-emitted volatiles, we found that compounds emitted increased 14.9- and 13.3- fold after leaf infestation and root infestation, respectively, compared to healthy control plants. The volatiles were mainly p-xylene, nonanal, tetradecane, decanal, benzaldehyde, β-caryophyllene, and humulene, while linalool was only present in the leaf-infestation treatment. In addition, principal component analysis indicated that all 18 compounds were associated with the infested plants, especially β-caryophyllene, p-xylene, and decanal. Based on the above studies and previous functional evaluations of the volatile compounds, it can be demonstrated that these compounds play a crucial role in modulating the interactions betweenA. segetumandM. mediatorand regulating parasitoid behavior. It may be possible to enhance the biological control ofA. segetumbyM. mediatorthrough the application of HIPVs.Keywords:turnip moth;parasitoid wasp;herbivore-induced plant volatiles;behavioral response;biological control

As we known, inducibility is an important feature of P450 genes. Previous studies indicated that CYP6B7 could be induced and involved in fenvalerate detoxification in Helicoverpa armigera . However, the regulatory mechanism of CYP6B7 induced by fenvalerate is still unclear. In this study, CYP6B7 promoter of H. armigera was cloned and the cis -acting element of fenvalerate was identified to be located between −84 and − 55 bp of CYP6B7 promoter. Subsequently, 33 candidate transcription factors ( CYP6B7 -fenvalerate association proteins, CAPs) that may bind to the cis -acting element were screened and verified by yeast one-hybrid. Among them, the expression levels of several CAPs were significantly induced by fenvalerate. Knockdown of juvenile hormone-binding protein-like (JHBP), RNA polymerase II-associated protein 3 (RPAP3), fatty acid synthase-like (FAS) and peptidoglycan recognition protein LB-like (PGRP) resulted in significant expression inhibition of CYP6B7 , and increased sensitivity of H. armigera to fenvalerate. Co-transfection of reporter gene p (−84/−55) with pFast-CAP showed that JHBP, RPAP3 and PGRP could significantly increase the activity of CYP6B7 promoter. These results suggested that trans -acting factors JHBP, RPAP3 and PGRP may bind with cis -acting elements to regulate the expression of CYP6B7 induced by fenvalerate, and play an important role in the detoxification of H. armigera to fenvalerate.

Genetically engineered crops simultaneously produce defensive allelochemicals and Bacillus thuringiensis (Bt) toxin proteins to kill some of the world's most devastating insect pests. How the two types of toxins, when ingested sequentially or simultaneously, interact at both lethal and sublethal doses in these pests remains underexplored. Here, we examined the toxicological interactions between the Bt toxin Cry1Ac and the flavonoid allelochemical flavone in Helicoverpa armigera . Simultaneous exposure of H. armigera neonates to lethal doses (LC 25 ) of Cry1Ac and flavone caused a mortality significantly higher than that of either toxin alone and their expected additive mortality. Preexposure for 24 h to a sublethal dose (LC 10 ) of Cry1Ac followed by 6-d simultaneous exposure to the same dose of Cry1Ac plus a lethal dose (1.6 mg/g diets, LC 50 ) of flavone resulted in a mortality significantly higher than that of the LC 50 dose of flavone alone and the expected additive mortality of the LC 50 dose of flavone plus the LC 10 dose of Cry1Ac. One-day preexposure to the sublethal dose (LC 10 ) of flavone followed by 6-d simultaneous exposure to the LC 50 dose (6 ng/cm 2 ) of Cry1Ac plus the LC 10 dose of flavone yielded a mortality significantly higher than that of the LC 50 dose of Cry1Ac but similar to the expected additive mortality of the LC 50 dose of Cry1Ac plus the LC 10 dose of flavone. The results suggest that Cry1Ac induces and synergizes the toxicity of flavone against H. armigera larvae.

The cotton bollworm , Helicoverpa armigera , is a polyphagous pest threatening many economically important crops worldwide. Until recently, synthetic pyrethroids remain in wide use for controlling pest insects including the cotton bollworm. Understanding the metabolic mechanism of pyrethroids in a given pest can provide significant implication for a smart choice of insecticides, and such information is useful for the development of novel selective and safe insecticides. In this study, we used complexes of recombinant H. armigera cytochrome P450 CYP9A and NADPH-dependent cytochrome P450 reductase to investigate the capacity of three CYP9A paralogs in the transformation of seven structurally different pyrethroids by metabolism assays. The results showed that the three paralogous CYP9As were able to metabolize multiple pyrethroids. Interestingly, all the three CYP9As transformed pyrethrin-resembling pyrethroids (e.g. bioallethrin) more efficiently than the heavily modified ones (e.g. bifenthrin). These findings suggest that herbivorous insects can cope with synthetic insecticides using their physiological systems that initially evolved to survive exposure to the defensive chemicals in their host plants, adding support to the pre-adaptation hypothesis.

Campoletis chlorideae Uchida (Hymenoptera: Ichneumonidae) is an important larval endoparasitoid of the cotton bollworm Helicoverpa armigera (Hübner). A better understanding of herbivore-induced plant volatiles (HIPVs) employed by C. chlorideae to locate hosts will contribute towards the biological control of this notorious crop pest. In this study, we first investigated the electrophysiological and behavioral responses of C. chlorideae to 19 volatile compounds emitted by herbivore-infested plants. Eleven odorants induced EAG responses of C. chlorideae , while only two compounds, cis -jasmone and cis -3-hexenyl acetate, were attractive to the parasitoid in Y-tube olfactometer assays. Next, we used two- and four-choice tests in cage settings to investigate the parasitism rate of H. armigera larvae by C. chlorideae on tobacco plants treated with different doses of cis -jasmone, cis -3-hexenyl acetate, and their mixtures. Results showed that the parasitism rate of H. armigera was significantly higher on the plants treated with cis -jasmone or cis -3-hexenyl acetate, and their optimal dose for both compounds was 1 mg. However, the mixtures of cis -jasmone and cis -3-hexenyl acetate did not increase H. armigera parasitism rate by C. chlorideae when compared with control groups. Our results indicate that the single compounds cis -jasmone and cis -3-hexenyl acetate, at certain doses, enhance parasitic efficiency of C. chlorideae , but their mixtures negate this effect. These results will help us in the exploitation of C. chlorideae as biological control agent to reduce the populations of H. armigera .

Insect cuticular protein (ICP) plays an important role in insect growth and development . However, research on the role of ICP in insecticide resistance is very limited. In this study, insect cuticular protein genes LCP17 and SgAbd5 were cloned and characterized in Helicoverpa armigera based on previous transcriptome data. The functions of LCP17 and SgAbd5 genes in fenvalerate resistance were assessed by RNA interference (RNAi), and their response to fenvalerate was further detected. The results showed that LCP17 and SgAbd5 were overexpressed in the fenvalerate-resistant strain comparing with a susceptible strain. The open reading frames of LCP17 and SgAbd5 genes were 423 bp and 369 bp, encoding 141 and 123 amino acids , respectively. LCP17 and SgAbd5 genes were highly expressed in the larval stage, but less expressed in the adult and pupal stages. The expression level of LCP17 and SgAbd5 genes increased significantly after fenvalerate treatment at 24 h. When the cotton bollworms larvae were exposed to fenvalerate at LD 50 level, RNAi-mediated silencing of LCP17 and SgAbd5 genes increased the mortality from 50.68% to 68.67% and 63.89%, respectively; the mortality increased to even higher level, which was 73.61%, when these two genes were co-silenced. Moreover, silencing of these two genes caused the cuticle lamellar structure to become loose, which led to increased penetration of fenvalerate into the larvae. The results suggested that LCP17 and SgAbd5 may be involved in the resistance of cotton bollworm to fenvalerate, and LCP17 and SgAbd5 could serve as potential targets for H. armigera control.

Transcription factors play an important role in regulating the expression of detoxification genes (e.g. P450s ) that confer insecticide resistance. Our previous study identified a series of candidate transcription factors ( CYP6B7 -fenvalerate association proteins, CAPs) that may be related to fenvalerate-induced expression of CYP6B7 in a field HDTJ strain of H. armigera . Whether these CAPs can mediate the transcript of CYP6B7 induced by fenvalerate in a susceptible HDS strain of H. armigera remains unknown. Further study showed that the expression levels of multiple CAPs were significantly induced by fenvalerate in HDS strain. Knockdown of CAP19 [fatty acid synthase-like (FAS)], CAP22 [polysaccharide biosynthesis domain-containing protein 1 (PBDC1)], CAP24 [5-formyltetrahydrofolate cycloligase (5-FCL)], CAP30 [peptidoglycan recognition protein LB-like (PGRP)] and CAP33 [NADH dehydrogenase [ubiquinone] 1 alpha subcomplex subunit 11 (NDUFA11)] resulted in significant inhibition of CYP6B7 and some other P450 genes expression; meanwhile, the sensitivity of HDS strain larvae to fenvalerate was significantly increased. In addition, PBDC1, PGRP and NDUFA11, either alone or in combination, could significantly enhance the activity of CYP6B7 promoter in HDS strain, as well as the expression level of CYP6B7 gene in Sf9 cells line. These results suggested that PBDC1, PGRP and NDUFA11 may be involved in the transcript regulation of key detoxifying genes in response to fenvalerate in HDS strain of H. armigera .

BACKGROUND Carboxylesterase (CarE) is a major class of enzyme involved in the detoxification of toxic xenobiotics in various insect species. Previous work has shown that the carboxylesterase gene CarE001G found in Helicoverpa armigera is more active and can metabolize synthesized pyrethroids, such as β -cypermethrin, one of the commonly used commercial insecticides for lepidopteran pest control. In addition, CarE001G is very special as it has a very specific glycine-rich region located adjacent to its C-terminal. But whether mutations in this unique sequence can change the biochemistry and function of CarE001G are unknown. RESULTS In this study, four variants of CarE001G with different deletions in the glycine-rich region were obtained and functionally expressed in Escherichia coli . The recombinant proteins were purified and confirmed by Western blot and mass spectrometry analyses. These mutant enzymes showed high catalytic efficiency toward the model substrate α -naphthyl acetate. Inhibition study showed that β -cypermethrin had relatively strong inhibition on CarE activities. In vitro metabolism assay showed that the mutant enzymes significantly enhanced their metabolic activities toward β -cypermethrin with specific activities between 4.0 and 5.6 nmol L -1 min −1 mg −1 protein. Molecular docking analyses consistently demonstrated that deletion mutations in the glycine-rich region may facilitate the anchoring of the β -cypermethrin molecule in the active binding pocket of the mutant enzymes. CONCLUSION The data show that deletion mutations can cause qualitative change in the capacity of CarEs in the detoxification of β -cypermethrin. This indicates that deletion mutations in the glycine-rich region may have the potential to cause synthesized pyrethroid (SP) resistance in H. armigera in the future. © 2020 Society of Chemical Industry

Carboxylesterases (CarEs) usually play critical roles in the detoxification of toxic chemicals and therefore may be involved in insecticide resistance in agricultural pests. Previous work has shown that CarE 001C from Helicoverpa armigera was able to metabolize the isomers of cypermethrin and fenvalerate . In this study, seven mutants of CarE 001C with single amino acid substitution were produced and expressed in the Escherichia coli. Enzyme kinetic analysis indicated that all seven mutations dramatically reduced enzymatic activities toward the generic substrate α -naphthyl acetate, but in vitro metabolism assay showed that two of the mutations, H423I and R322L, significantly improved hydrolase activities toward fenvalerate, with their recorded specific activities being 3.5 and 5.1 nM·s −1 ·mg −1 proteins, respectively. Further, thermostability assay showed that the stability of one mutant enzyme was enhanced. This study will help us better understand the potential of CarEs in insecticide detoxification and resistance in H. armigera .
