Polydopamine assisted immobilisation of copper(II) on titanium for antibacterial applications

T. He,W. Zhu,X. Wang,P. Yu,S. Wang,G. Tan,C. Ning

Copper(II) were immobilised onto medical titanium surface by polydopamine (PDA) chelating, to build intrinsic antibacterial implant materials for inhibition of implant infection. A scanning electrical microscope showed that the modified titanium surface morphology was a porous nanostructure. An energy dispersive spectrometer showed the existence of Cu, and X-ray photoelectron spectroscopy results further confirmed that the immobilised copper element was in the form of Cu(II). Bacteria plate colony counting results showed that modified titanium surface had decent antibacterial property, matching the ion release outcomes. It could be conclude that the PDA assisted Cu(II) immobilisation was an efficient and effective method for antibacterial modification of titanium.

Nickel nitride-mediated nitric oxide generation for combating implant-associated infections

Chao Wei,Haifeng Zhang,Ji Tan,Kui Xue,Zhiyu Hou,Rongxiang Ding,Xianming Zhang,Xuanyong Liu

NiTi alloys are widely used in the fabrication of biomedical implants, and it is essential to endow them with selective antibacterial properties to avoid implant-associated infection. In this study, nickel nitride (Ni 3 N), as a promising new inorganic nitric oxide (NO) donor, was in situ constructed on the surface of NiTi alloys through a straightforward two-step plasma immersion ion implantation (PIII) strategy. Nickel sites were first exposed by physical bombardment with argon or metal plasma, followed by reaction with nitrogen plasma to form Ni 3 N. Ni 3 N can disrupt the transmembrane proton electrochemical gradient by capturing protons to produce ammonia species (NH x ), resulting in the accumulation of reactive oxygen species (ROS) as a result of bacterial metabolic disruption. On the other hand, the generated NH x can further react with ROS to produce NO and reactive nitrogen species (RNS). As a result, the Ni 3 N films exhibited more than 99 % antibacterial efficiency against both gram-negative and gram-positive bacteria. They also demonstrated effective antibacterial properties in a rat subcutaneous implant model. In addition, the modified NiTi alloys exhibit good corrosion resistance and cytocompatibility. This study provides a green, safe, and inexpensive method for constructing metal nitride films on implant surfaces and offers new ideas for the clinical design and fabrication of novel antibacterial implants.

Durable antibacterial coatings on Kirschner wires via redox-assisted co-deposition

Hongjie Ding,Dasai Ban,Cong Wang,Huali Nie,Jiaxing Huang

The Kirschner wire (K-wire) is a versatile and effective external fixator in orthopedic surgery. However, a recognized challenge associated with the use of K-wires is the potential for bacterial migration and subsequent infection as the wire passes through the skin and penetrates the bone. In this study, we developed a straightforward method for depositing antibacterial ions on K-wires by redox-assisted co-deposition. This method offers several advantages over conventional coating techniques , including uniformity, high coverage, strong adhesion , and scalability. As a result, K-wires treated with this method have a rapid and long-lasting antimicrobial effect. Both in vitro and in vivo experiments have demonstrated that these antimicrobial K-wires have an excellent bacterial elimination . The results of this study provide new opportunities for the application of robust and multifunctional antibacterial coating materials on implantable devices with great potential in the fields of orthopedics, transplantation and surgery.

Graphdiyne-modified TiO2 nanofibers with osteoinductive and enhanced photocatalytic antibacterial activities to prevent implant infection

Wang Rui,Shi Miusi,Xu Feiyan,Qiu Yun,Zhang Peng,Shen Kailun,Zhao Qin,Yu Jiaguo,Zhang Yufeng

Titanium implants have been widely used in bone tissue engineering for decades. However, orthopedic implant-associated infections increase the risk of implant failure and even lead to amputation in severe cases. Although TiO 2 has photocatalytic activity to produce reactive oxygen species (ROS), the recombination of generated electrons and holes limits its antibacterial ability. Here, we describe a graphdiyne (GDY) composite TiO 2 nanofiber that combats implant infections through enhanced photocatalysis and prolonged antibacterial ability. In addition, GDY-modified TiO 2 nanofibers exert superior biocompatibility and osteoinductive abilities for cell adhesion and differentiation, thus contributing to the bone tissue regeneration process in drug-resistant bacteria-induced implant infection.

Nucleotide coordinated polymers, a ROS-based immunomodulatory antimicrobial, doubly kill Pseudomonas aeruginosa biofilms of implant infections

Jinghuang Chen,Xianqing Tang,Qihan Sun,Xin Ji,Xingbo Wang,Zhendong Liu,Xu Zhang,Haijiao Xu,Fan Yang,Jian Sun,Xiurong Yang

Pseudomonas aeruginosa causes high morbidity and mortality in nosocomial infections, and newly approved antibiotics have been declining for decades. A green and universal deprotonation-driven strategy is used to screen the guanylic acid-metal ion coordination polymer nanoparticles (GMC), instead of the failure of binding occurs when specific metal ion participation. We find that the precise pH-dependent oxidase-like activity of GMC-2 orchestrates a duple symphony of immune modulation for Pseudomonas aeruginosa biofilm infections. Specifically, GMC-2-mediated reactive oxygen species (ROS) regulation triggers mitochondrial dysfunction and releases damage-associated molecular patterns, engaging pattern recognition receptors and resulting in endogenous innate immune activation. Meanwhile, GMC-2-triggered ROS generation in a mildly acidic biofilm environment destroys the biofilm, exposing exogenous pathogen-associated molecular patterns. GMC-2 cannot cause resistance for Pseudomonas aeruginosa compared with conventional antibiotics. In an infected implant mouse model, Pseudomonas aeruginosa biofilms were effectively eliminated by GMC-2-mediated triggering of innate and adaptive immunity. These findings provide a universal approach for facilitating the binding of biomolecules with metal ions and highlight the precise ROS-regulating platform plays a critical role in initiating endogenous and exogenous immune activation targeted for bacterial biofilm infection.

Mengni Fan,Jingxing Si,Xiaogang Xu,Linfu Chen,Junpeng Chen,Chao Yang,Jingwu Zhu,Lihuang Wu,Jiang Tian,Xiaoyi Chen,Xiaozhou Mou,Xiaojun Cai

Development of multifunctional antibacterial agent with long-lasting antibacterial activity and biofilm ablation performance is significant for the effective treatment of bacterial infections. Here, by utilizing the electrostatic interaction between sulfonated chitosan (SCS) and Ag + and chitosan (CS), and the sodium borohydride reduction method, a versatile antibacterial agent (AgNPs@CS/SCS) capable of generating silver nanoparticles (AgNPs) in-situ and long-acting slow-release Ag + was developed. AgNPs@CS/SCS has a good physiological stability and can long-acting slow-release of Ag + due to the pH-dependent Ag + release behavior of AgNPs. Noteworthy, AgNPs@CS/SCS can exert both excellent short- and long-term antibacterial and biofilm ablation activity. Importantly, it also exhibits superior antibacterial activity in the treatment of implant infections, accompanied by good biocompatibility. Together, this study suggest that AgNPs@CS/CSC is indeed a versatile antibacterial agent, and is expected to provide an effective treatment modality for implant infections in the clinic settings.

Cascade catalysis nanozyme for interfacial functionalization in combating implant infections associated with diabetes via sonodynamic therapy and adaptive immune activation

Kun Xu,Yanan Zou,Chuanchuan Lin,Liangshuai Zhang,Meijun Tan,Meng Li,Jing Wu,Xuan Li,Ye He,Peng Liu,Ke Li,Kaiyong Cai

Innovative solutions are required for the intervention of implant associated infections (IAIs), especially for bone defect patients with chronic inflammatory diseases like diabetes mellitus (DM). The complex immune microenvironment of infections renders implants with direct antibacterial ability inadequate for the prolonged against of bacterial infections. Herein, a synergistic treatment strategy was presented that combined sonodynamic therapy (SDT) with adaptive immune modulation to treat IAIs in diabetes patients. A multifunctional coating was created on the surface of titanium (Ti) implants, consisting of manganese dioxide nanoflakes (MnO 2 NFs) with cascade catalytic enzyme activity and a responsive degradable hydrogel containing a sonosensitizer. The reactive oxygen species (ROS) generated by glucose-hydrogen peroxide (H 2 O 2 ) cascade catalysis and ultrasound (US) activation sonosensitizer helped kill bacteria and release bacterial antigens. Meanwhile, Mn 2+ facilitated dendritic cells (DCs) maturation, enhancing antigen presentation to activate both cellular and humoral adaptive immunity against bacterial infections. This approach effectively eliminated bacteria in established diabetic IAIs model and activated systemic antibacterial immunity, providing long-term antibacterial protection. This study presents a non-antibiotic immunotherapeutic strategy for fighting IAIs in chronic diseases.

Exchange-coupled bi-magnetic nanoparticles enhance magnetothermal/chemodynamic antibacterial therapy of poly-l-lactide scaffold

Cijun Shuai,Chong Lin,Chongxian He,Wei Tan,Shuping Peng,Wenjing Yang

Bone implant-associated bacterial infection is a common cause of transplant failure. Chemodynamic therapy (CDT) has significant antibacterial efficacy by producing reactive oxygen species (ROS). However, the ROS permeability is restricted by the natural barrier of bacterial biofilms. Herein, a hard-soft magnetic biphasic nanoparticle (CF@MF) was synthesized by in-situ growth of the soft MnFe 2 O 4 shell on the hard CoFe 2 O 4 core. Then, the CF@MF was incorporated into poly- l -lactide powders to prepare porous scaffolds by selective laser sintering. The scaffold exhibited superior magnetothermal conversion efficiency, which was attributed to the interfacial exchange coupling of CF@MF. Importantly, the scaffold effectively dispersed biofilms by magnetothermal therapy (MTT), favoring numerous ROS pre-generated by CDT to enter the interior to kill bacteria. This study provided a synergistic MTT/CDT strategy to eradicate deep bacterial infections in biomedical application.

Polysaccharide-Based Coating with Excellent Antibiofilm and Repeatable Antifouling-Bactericidal Properties for Treating Infected Hernia

Qifen Li,Leixiang Wang,Lu Yu,Chengbo Li,Xianrui Xie,Huanhuan Yan,Wenjuan Zhou,Chunhua Wang,Zhonghao Liu,Guige Hou,Yu-Qing Zhao

In recent years, the utilization of medical devices has gradually increased and implantation procedures have become common treatments. However, patients are susceptible to the risk of implant infections. This study utilized chemical grafting to immobilize polyethylenimine (QPEI) and hyaluronic acid (HA) on the surface of the mesh to improve biocompatibility while being able to achieve antifouling antimicrobial effects. From the in vitro testing, PP–PDA-Q-HA exhibited a high antibacterial ratio of 93% against S. aureus, 93% against E. coli, and 85% against C. albicans. In addition, after five rounds of antimicrobial testing, the coating continued to exhibit excellent antimicrobial properties; PP–PDA-Q-HA also inhibits the formation of bacterial biofilms. In addition, PP–PDA-Q-HA has good hemocompatibility and cytocompatibility. In vivo studies in animal implantation infection models also demonstrated the excellent antimicrobial properties of PP–PDA-Q-HA. Our study provides a promising strategy for the development of antimicrobial surface medical materials with excellent biocompatibility.

Gold nanoclusters cure implant infections by targeting biofilm

Youhuan Gong,Xueying Zhao,XiaoJie Yan,Wenfu Zheng,Huanwen Chen,Le Wang

The biofilm-mediated implant infections pose a huge threat to human health. It is urgent to explore strategies to reverse this situation. Herein, we design 3-amino-1,2,4-triazole-5-thiol (ATT)-modified gold nanoclusters (AGNCs) to realize biofilm-targeting and near-infrared (NIR)-II light-responsive antibiofilm therapy. The AGNCs can interact with the bacterial extracellular DNA through the formation of hydrogen bonds between the amine groups on the ATT and the hydroxyl groups on the DNA. The AGNCs show photothermal properties even at a low power density (0.5 W/cm 2 ) for a short-time (5 min) irradiation, making them highly effective in eradicating the biofilm with a dispersion rate up to 90 %. In vivo infected catheter implantation model demonstrates an exceptional high ability of the AGNCs to eradicate approximately 90 % of the bacteria encased within the biofilms. Moreover, the AGNCs show no detectable toxicity or systemic effects in mice. Our study suggests the great potential of the AGNCs for long-term prevention and elimination of the biofilm-mediated infections.

Biofilm-Responsive Polymeric Nanoparticles with Self-Adaptive Deep Penetration for In Vivo Photothermal Treatment of Implant Infection

Congyu Wang,Wei Zhao,Bing Cao,Zhixiong Wang,Qi Zhou,Siyu Lu,Ligong Lu,Meixiao Zhan,Xianglong Hu

Biofilm can protect bacteria from immune attacks and antibiotic inhibition, and bacterial biofilm hosted in implanted materials and medical devices is a serious threat for modern medical system. Herein, we report biofilm-responsive caged guanidine nanoparticles (CGNs) to deeply penetrate and accumulate in bacterial biofilm, and then efficient photothermal eradication of bacterial biofilm is achieved upon NIR laser irradiation via the proof-of-concept formulation of photothermal agents in CGNs. In physiological conditions and blood circulation, CGNs are negatively charged by masking the positive charge of guanidine via covalent modification with acid-cleavable moieties, exhibiting high biocompatibility and minimal hemolysis. Whereas upon blood circulation and passive accumulation at infected implant sites, CGNs are self-adaptive in acidic biofilm to release the protective caging group and expose native guanidine moieties, which can promote nanoparticle deep biofilm penetration and bacteria adhesion as well as membrane fusion. After that, remarkable photothermal effect with a high photothermal conversion efficiency of ∼40.9% can eradicate implant biofilm upon NIR laser irradiation. It can efficiently treat S. aureus biofilm-infected implant catheters in vivo via only one single treatment in a mouse model, exhibiting ∼99.6% bacteria inhibition ratio. Apart from this proof-of-concept work, current guanidine-caged biofilm responsive polymeric nanoparticles are promising general vectors to treat biofilm and resistant pathogens in medicine and daily healthcare.

Ultrasound-Activated Probiotics Vesicles Coating for Titanium Implant Infections Through Bacterial Cuproptosis-Like Death and Immunoregulation

Shuoyuan Li,Yan Yue,Wenqi Wang,Mingyue Han,Xufeng Wan,Qiaochu Li,Xiaoting Chen,Jian Cao,Yangming Zhang,Jiyao Li,Jianshu Li,Lei Cheng,Jiaojiao Yang,Duan Wang,Zongke Zhou

Implant-associated infections (IAIs) are the main cause of prosthetic implant failure. Bacterial biofilms prevent antibiotic penetration, and the unique metabolic conditions in hypoxic biofilm microenvironment may limit the efficacy of conventional antibiotic treatment. Escaping survival bacteria may not be continually eradicated, resulting in the recurrence of IAIs. Herein, a sonosensitive metal-organic framework of Cu-TCPP (tetrakis(4-carboxyphenyl) porphyrin) nanosheets and tinidazole doped probiotic-derived membrane vesicles (OMVs) with high-penetration sonodynamic therapy (SDT), bacterial metabolic state interference, and bacterial cuproptosis-like death to eradicate IAIs is proposed. The Cu-TCPP can convert O 2 to toxic 1 O 2 through SDT in the normoxic conditions, enhancing the hypoxic microenvironment and activating the antibacterial activity of tinidazole. The released Cu(II) under ultrasound can be converted to Cu(I) by exogenous poly(tannic acid) (pTA) and endogenous glutathione. The disruption of the bacterial membrane by SDT can enhance the Cu(I) transporter activity. Transcriptomics indicate that the SDT-enhanced Cu(I) overload and hypoxia-activated therapy hinder the tricarboxylic acid cycle (TCA), leading to bacterial cuproptosis-like death. Moreover, the OMVs-activated therapy can polarize macrophages to a M2-like phenotype and facilitate bone repair. The sonodynamic biofilm microenvironment modulation strategy, whereby the hypoxia-enhanced microenvironment is potentiated to synergize SDT with OMVs-activated therapy, provides an effective strategy for antibacterial and osteogenesis performance.
