Application of sustainable natural resources in crop protection: Podophyllotoxin-based botanical pesticides derived from Podophyllum hexandrum for controlling crop-threatening insect pests

Ruige Yang,Xiaobo Huang,Zhiping Che,Yuanyuan Zhang,Hui Xu

Podophyllotoxin is isolated as a sustainable natural bioresource from the roots and rhizomes of Podophyllum hexandrum and Juniperus Sabina . In continuation of our program aimed at the discovery of biorenewable natural product-based pesticides, a series of 4α-acyloxy derivatives of 2'-chloro-5-bromopicropodophyllotoxin or 2',5-dibromopicropodophyllotoxin were synthesized by structural modification of podophyllotoxin, and evaluated as insecticidal and acaricidal agents against two serious agricultural pests, Mythimna separata Walker and Tetranychus cinnabarinus Boisduval. Among all derivatives, compounds 6a and 7a exhibited more promising insecticidal and acaricidal activities than podophyllotoxin, and the final mortality rates of 6a , 7a and 1 against M. separata (at 1   mg/mL) and T. cinnabarinus (at 0.5   mg/mL) were 50%/53.3%/36.7%, and 33%/41.8%/1.6%, respectively. This suggested that introduction of the acetoxy moiety at the C-4 position of 2'-chloro-5-bromopicropodophyllotoxin/2',5-dibromopicropodophyllotoxin was necessary for the insecticidal and acaricidal activities.

Preparation and Biological Evaluation of Novel Osthole-Derived N-Benzoylthioureas as Insecticide Candidates

Ruige Yang,Zhiyan Liu,Meiyue Han,Liping Cui,Yong Guo

To discover novel natural-product-based insecticide candidates, herein, a variety of osthole-derived N-benzoylthioureas were synthesized and assessed for their insecticidal activities against three insect pests. An insecticidal assay showed that most of the target osthole-derived N-benzoylthioureas displayed a more potent and broad-spectrum insecticidal effect than the parent osthole after the introduction of N-benzoylthioureas on the C-3′ position of osthole. Compound B24 displayed the most potent growth inhibitory (GI) effect on Mythimna separata Walker, with a final corrected mortality rate of 82.1% when treated with a concentration of 1 mg/mL, which was 1.64- and 1.53-fold higher in comparison to osthole and the botanical insecticide toosendanin, respectively. Compounds B22, B23, and B25 displayed a more promising aphicidal effect on Myzus persicae Sulzer, and their LD50 values were 0.015, 0.017, and 0.019 μg/larvae, respectively, superior to the commercially available insecticide rotenone (0.024 μg/larvae). Derivatives B19, B20, B23, and B25 displayed more potent larvicidal activity against Plutella xylostella Linnaeus, with LC50 values of 0.22, 0.26, 0.15, and 0.30 mg/mL, respectively, exceeding that of rotenone (0.37 mg/mL). Furthermore, both compounds B19 and B23 against P. xylostella were found to be more effective than rotenone in a control efficacy assay under greenhouse conditions. The structure–activity relationship (SAR) suggested that osthole-derived N-benzoylthioureas are more active in most cases when the R group is an electron-withdrawing group than when it is an electron-donating group, especially for halogenated groups. Additionally, the potent compounds B19 and B23 possessed good selectivity and were less toxic to non-target organisms. This study suggests that these osthole-derived N-benzoylthioureas could be further studied in depth as eco-friendly natural product pesticides in crop protection.

Identification and potential application of key insecticidal metabolites in Tilia amurensis, a low-preference host of Hyphantria cunea

Lisha Yuan,Tao Li,Yi Huang,Aoying Zhang,Shanchun Yan,Dun Jiang

Developing effective insecticidal strategies is an important means of reducing the spread and host plant damage by Hyphantria cunea . In this study, key metabolites with insecticidal activity against H. cunea were screened by targeted metabolomics in Tilia amurensis , a low-preference host plant. Subsequently, the potential of key metabolites that could be used as botanical pesticides was evaluated. The results showed that coumarin was the key insecticidal metabolite of T. amurensis and had a significant insecticidal effect and weight inhibition effect on H. cunea larvae. Coumarin treatment significantly decreased the larval nutrient content and the gene expression of rate-limiting enzymes in the glycolytic pathway and tricarboxylic acid cycle . A significantly enhanced detoxification enzyme activity (CarE and GST), antioxidant oxidase activity (SOD and CAT), non-enzymatic antioxidant levels (GSH), and total antioxidant capacity were observed in coumarin-treated larvae. Coumarin treatment resulted in a significant increase in the expression levels of detoxification enzyme genes ( CarE1 , CarE2 , CarE3 , GST2 , and GST3 ) and antioxidant oxidase genes ( SOD1 , CAT1 , and CAT2 ) in H. cunea larvae. Coumarin treatment significantly increased the levels of MDA and H 2 O 2 in larvae but did not cause pathological changes in the ultrastructure of the larval midgut. Coumarin solution sprayed directly or as a microcapsule suspension formulation with coumarin as the active ingredient had significant insecticidal activity against the H. cunea larvae. Overall, coumarin, a key anti-insect metabolite identified from T. amurensis , can significantly inhibit the growth and survival of H. cunea larvae and has the potential to be developed as a botanical pesticide.

Molecular characterization and functional analysis of daf-16-2b gene in the pine wood nematode, Bursaphelenchus xylophilus

Buyong Wang,Rongrong Wen,Ling Ma,Yan Ma,T. Hua Zhou

The Forkhead box O transcription factor DAF-16 is an important downstream transcription factor in the well-conserved insulin signalling (IIS) pathway in Caenorhabditis elegans . The nuclear localization of DAF-16 can activate a series of genes that mediate oxidative stress, heat shock, xenobiotic responses, innate immunity, metabolism and autophagy. In this study, a Bxdaf-16-2b gene encoding 510 amino acids was cloned from Bursaphelenchus xylophilus . Sequence analysis showed that the BxDAF-16-2b protein shared the highest similarity value (92%) with the DAF16-2 (accession no. ADD84716.1 ) of B. xylophilus . Using in situ hybridization, Bxdaf-16-2b was shown to be constitutively expressed in the circum-pharyngeal nerve rings and intestines of males, and in the head neurons and the ovaries of females. Quantitative RT-PCR showed that the highest gene expression was found in females, followed by juveniles, males and eggs. Bxdaf-16-2b expression was up-regulated after treatment with the botanical pesticides rotenone and matrine. RNA interference showed that the expression of Bxdaf-16-2b gene was reduced by dsRNA, and the silencing of Bxdaf-16-2b suppressed the expression of several IIS pathway genes. Bursaphelenchus xylophilus showed increased mortality at sublethal doses of rotenone and matrine when subjected to Bxdaf-16-2b gene silencing. Thus, Bxdaf-16-2b may participate in the stress response of B. xylophilus and play important roles in the immune system. This study paves the way towards design of new drug targets for non-polluting control of nematodes.

Citrus Essential Oils as Potential Insecticides Against Thrips flavus

Tian-Hao Pei,Yu-Long Niu,Bin Liu,Ming Yuan,Ai-Guo Zhang,Xiao-Feng Li,Lian-Xia Wang,Meng-Lei Xu,Yu Gao

BackgroundEssential oils (EOs) are recognized for their potent insecticidal activity and are widely considered promising agents for pest control.MethodsThe insecticidal activity of seven commercial citrus EOs against the global phytophagous pest Thrips flavus was evaluated using bioassays conducted under laboratory conditions. Moreover, the chemical composition of these EOs was analyzed using gas chromatography-mass spectrometry (GC-MS).ResultsAmong the 45 identified compounds, the major constituents were d-limonene (23.77%–95.10%), methyl jasmonate (38.76%), and linalyl acetate (34.55%). Orange flower oil, sweet orange oil, tangerine peel oil, bergamot oil, lime oil, lemon oil, and grapefruit oil exhibited good insecticidal activity against T. flavus without demonstrating significant repellent activity. Orange flower oil and sweet orange oil showed higher insecticidal toxicity, with LC50 values of 0.20 g/L and 0.37 g/L, respectively. In pot experiments, the higher control efficacy against T. flavus was found after 7 days of treatment for orange flower oil (91.14±1.27% at 720.00 g a.i.·hm−2 and 100% at 900.00 g a.i.·hm−2), sweet orange oil (91.14±5.52% at 900.00 g a.i.·hm−2), tangerine peel oil (96.20±2.19% at 720 g a.i.·hm−2 and 100% at 900.00 g a.i.·hm−2), lemon oil (96.20±2.19% at 900 g a.i.·hm−2), and lime oil (97.47±2.53% at 900.00 g a.i.·hm−2).ConclusionTaken together, orange flower oil and tangerine peel oil, which were found to contain d-limonene and methyl jasmonate, demonstrated rapid and more effective insecticidal activity compared to the other EOs tested, which makes these two EOs promising alternatives to chemical insecticides.

Non-food bioactive products for botanical pesticides: Insights into residue and dissipation kinetics of matrine-type alkaloids from Sophora alopecuroides

Bin Zhang,Jia Hao,Zheng Sun,Xiaomin Zhao,Jing Zhang,Zhiqing Ma

Sophora alopecuroides has important industrial value as the main source of matrine-type alkaloids , which have the potential to develop into global botanical pesticides . However, the unclear residue dynamics of matrine-type alkaloids severely limits its industrial development and large-scale application as botanical pesticides. Therefore, this study systematically analyzed residue behavior of matrine-type alkaloids of S. alopecuroides in crops and the environment. Firstly, the indirect competitive enzyme-linked immunosorbent assay (IC-ELISA) method for the detection of matrine was developed and verified. The IC-ELISA method showed excellent cross-reactivities to matrine analogues, and could be used directly to detect and analyze the total alkaloids from S. alopecuroides . Using the IC-ELISA method, the dissipation behaviors and terminal residues of matrine and total alkaloids in pakchoi and soil were investigated. The results showed that matrine and the total alkaloids from S. alopecuroides were both easily degradable pesticides. However, the half-lives of matrine in pakchoi and soil were 1.576 d and 1.531d, respectively, significantly lower than those of total alkaloids with values of 6.649 d and 2.767 d. The results also provide guidance for the maximum residue detection values (MRLs) and scientific application of matrine-type alkaloid pesticides. This study is of great significance to promote the industrial development and large-scale application of its matrine-type alkaloids as botanical pesticides and improve the industrial value of S. alopecuroides .

Designing Veratramine Nanopesticide with Strong Adhesion Performance and Plant Uptake for Efficient Control toward Multiple Target Pests

Jingyi Chen,Yuankang Guo,Yajie He,Meizhen Yin,Xiangge Du,Jixing Xia,Jie Shen,Min Dong,Shuo Yan

Botanical pesticides are promising alternatives for sustainable pest management due to their environmental compatibility, biodegradability, and multitarget mechanisms that help delay pest resistance. Unfortunately, most botanical pesticides face challenges such as limited water solubility and poor foliar adhesion, thereby limiting their large-scale field application. Herein, the star polycation (SPc) nanocarrier and X-100 surfactant were introduced to successfully develop an efficient nanodelivery platform for veratramine (VAM). The SPc could spontaneously assemble with VAM via a hydrogen bond and van der Waals force, and the integration of Triton X-100 stabilized the nanoscale size (84 nm) to prepare a VAM nanopesticide. Interestingly, this optimized system reduced the contact angle and increased the retention of VAM on plant leaves while remarkably facilitating its systemic transport through roots and leaves. Compared to commercial VAM, the VAM nanopesticide showed stronger contact and stomach toxicity against multiple pest species such as aphids, thrips, and mites, which exhibited fast-acting properties in the field with high control efficacy even at 7 d after the spraying. Subsequently, the RNA-seq and biological experiments demonstrated that the application of VAM nanopesticide influenced various pathways in aphids, inducing starch and sucrose metabolism, protein digestion and absorption, etc., with inhibitory effects on the biosynthesis of trehalose and protein. Importantly, the application of the VAM nanopesticide did not adversely influence the predatory lady beetles or cowpea seedlings, confirming its safety in actual applications. Overall, our work addressed the formulation instability and environmental loss of VAM while ensuring its ecological safety, which offered a scalable nanoplatform to optimize botanical pesticides for enhanced efficacy and minimized ecological risks.

Natural products-based: Synthesis and antifungal activity evaluation of novel L-pyroglutamic acid analogues

Ai Likun,Fu Shiqi,Li Yong,Zuo Mei,Huang Wen,Huang Jian,Jin Zhichao,Chen Yang

Botanical pesticides are one of the sources of third-generation pesticides, which have received much attention at home and abroad in recent years due to their degradable and pollution-free advantages in nature. This article explored a concise approach toward synthesizing a series of novel L-pyroglutamic acid analogues from L-hydroxyproline. Furthermore, bioassay studies of these sulfonyl ester derivatives against Pyricularia oryzae, Fusarium graminearum, Alternaria brassicae, Valsa mali, and Alternaria alternariae showed moderate antifungal activity. For instance, C08a and C08l provide potential lead agents for controlling Fusarium graminearum because of their inhibitory activity.
