Simple SummaryMyostatin is a negative regulator of muscle growth and development, and mutational inactivation of this gene increases muscle mass. Knockout of themstnbgene inPelteobagrus fulvidracodid not increase muscle mass, although our previous study of themstnagene did. Instead, it reduced the body size and growth performance ofPelteobagrus fulvidraco. Through the present study, we found that the mutantPelteobagrus fulvidracohad a reduction in the number of vertebrae, the length, and the intervertebral distance inPelteobagrus fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantPelteobagrus fulvidraco. Histological comparison of the same sites in themstnmutant and wild groups ofPelteobagrus fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutPelteobagrus fulvidracothan in wild-typePelteobagrus fulvidraco.AbstractBased on obtainingmstnbgene knockout inPelteobagrus fulvidraco, a study on the effect of themstngene on skeletal morphology and growth was performed by comparing the number and length of the vertebrae of mutant and wild-type fish in a sibling group ofP. fulvidraco, combined with the differences in cells at the level of vertebral skeletal tissue. It was found thatmstnbgene knockdown resulted in a reduction in the number of vertebrae, the length, and the intervertebral distance inP. fulvidraco, and these changes may be the underlying cause of the shorter body length in mutantP. fulvidraco. Further, histological comparison of the same sites in themstnmutant and wild groups ofP. fulvidracoalso revealed that the number and density of osteocytes were greater inmstnbknockoutP. fulvidracothan in wild-typeP. fulvidraco. Our results demonstrated that when using genome editing technology to breed new lines, the effects of knockout need to be analyzed comprehensively and may have some unexpected effects due to insufficient study of the function of certain genes. Keywords:myostatin b;genome editing;Pelteobagrus fulvidraco;body length;skeleton;vertebral centrum

The technique of anterior cervical corpectomy and fusion (ACCF) for strut grafting has become a widespread and actively applied for many cervical complaints including cervical degeneration of the intervertebral disks , cervical trauma , cancer, rheumatoid arthritis and multilevel cervical spondylotic diseases. To avoid the complications of the old techniques, the construct stability and long anterior screw-plate designs of the bone strut have been improved by using effective biomaterials. The nanostructured hydroxyapatite (HAp) incorporated with biocompatible polymer matrixes is an effective biomedical material and creating a functional properties applied for different tissue replacements such as dental, hips, knees , tendon and ligaments and tissue repair for maxillofacial reconstruction , stabilization of the jaw bone and spinal fusion . In the present investigation, we have successfully designed cylindrical nano titanium dioxide (n-TiO 2 ) interbody fusion with anterior plate fixation . The n-TiO 2 incorporated HAp/ Polyetheretherketone (PEEK) nanocomposite strut has a superior mechanical properties and larger contact area with high fusion rates as compared with the HAp/PEEK strut in the absence of n-TiO 2 nanoparticles . It is highly able to provide appropriate strength and biological activity similar to the conventional titanium cage and also mainly it has been minimizes subsidence value. The synthesized novel material of n-TiO2 incorporated HAp/PEEK nanocomposite strut is scientifically given effective outcomes for fusion and reconstruction of the ACCF.

The widespread use of veterinary antibiotics in animal husbandry, such as tilmicosin (TIL), poses potential threats to environment pollution and human health. In this study, we conducted acute TIL exposure experiment using zebrafish model to explore the toxicological effects. In the concentration range of 20–80 mg/L, TIL did not affect zebrafish body length development but induced idiopathic scoliosis (IS)-like symptoms accompanied by impaired mobility. IS predominantly occurs in adolescents, while the etiology remains unclear. Further studies showed that the combination of quercetin and luteolin effectively alleviated TIL-induced spinal curvature and motor suppression. To explore the underlying mechanism, we performed network pharmacology analysis and enriched the PI3K-Akt pathway. We validated that the expression of key target genes in the PI3K-Akt pathway were significantly activated after TIL exposure, while quercetin and luteolin treatment effectively inhibited the PI3K-Akt pathway activation. Further experiments revealed that TIL exposure decreased brain neural cell apoptosis, thereby causing an increase in neurocytes, interfering with motor ability, and promoting IS-like vertebral malformations, which were especially noticeable during movement. This study expands our understanding that TIL exposure may be a predisposing factor for IS occurrence and progression, while the combination of quercetin and luteolin combination could protect against IS-like symptom by regulating PI3K-Akt pathway. Graphical The experiments revealed that exposure to tilmicosin (TIL) induces the development of scoliosis-like deformities, which could be effectively alleviated by the quercetin and luteolin combination. Further mechanistic studies found that TIL exposure activates the PI3K-Akt pathway, resulting in reduced apoptosis of brain neurons and increased neural cells, which could be inhibited by quercetin and luteolin. As disturbed brain development is a predisposing factor for scoliosis, the TIL-induced abnormalities in brain neurodevelopment exacerbate the progression of scoliosis-like deformities and dyskinesia.

The objective of this study is to prepare a biocompatible nanohydroxyapatite/poly(methyl methacrylate) (HA/PMMA) composite bone cement, which has good mechanical property and can be used for vertebroplasty. Up to 40 wt % of nanohydroxyapatite (nano-HA) in the power, which was surface modified with poly(methylmethacrylate- co -γ-methacryloxypropyl timethoxysilane) [P(MMA- co -MPS)] copolymer, was incorporated into the composite bone cement. The content of P(MMA- co -MPS) on the surface of nano-HA (18.7%, 22.8%, and 26%) was determined through thermogravimetric analysis (TGA). The morphology of biomineralized surface of composite bone cement was observed under scanning electron microscope (SEM). The mechanical measurements of the composite cements implied that the interfacial interaction between the HA and PMMA matrix may be greatly enhanced after surface modification of HA. Biochemical assays indicated that the HA/PMMA bone cement had no cytotoxicity and induced no hemolysis. The cell adhesion and alkaline phosphatase (ALP) activity assays indicated that the biocompatibility of HA/PMMA bone cement could be promoted, demonstrating that it can be used as an ideal weight-bearing bone repair materials on clinical application. © 2015 Wiley Periodicals, Inc. With the continuous development of technology, using biological three-dimensional (3D) printing technology to prepare hydrogel stent for cartilage construction has great potential in soft tissue repair. In this study, the cartilage stent was prepared by cell-controlled assembly technology,
the gelatin sodium alginate (GSA) was selected as the basic material and the nano-hydroxyapatite (n-HAP) was undertaken as the reinforcing material of the basic hydrogel, and the cell-controlled technology was used to assemble the n-HAP hydrogel stent with different composite ratios. While
the performance of the stent material was characterized, the hydrogel material prepared for traumatic thoracolumbar fracture (TTF) was adopted for filling treatment during the percutaneous kyphoplasty (PKP). The results showed that compared with the GSA stent, addition of n-HAP can optimize
accuracy of 3D printing, increase stability and mechanical strength of stent, and tested average elastic modulus of the composite hydrogel material is 24.58 ± 1.05 MPa. It suggested that addition of n-HAP can improve biodegradability of stent while adjusting the surface roughness of
the stent. The biocompatibility test of n-HAP hydrogel stent indicated that the stent material was non-toxic and could support the adhesion and growth of mouse chondrocytes. After culture for 1 week after the 3D printing, the proportion of living cells reached more than 90%. Under the action
of the composite hydrogel stent material, the visual analogue scale (VAS) score of patient decreased dramatically (from 8.9 to 1.8), the Oswestry disability index (ODI) decreased from 83.65 before surgery to 36.89 after surgery (P < 0.05), and the anterior vertebral height (AVH)
of patient increased from 2.46 cm to 2.96 cm, which was 2.76 cm in the last follow-up test (P < 0.05). In summary, the composite hydrogel stent prepared in this study could meet the needs of cartilage tissue factories and showed good clinical effects in the treatment of TTF.

ABSTRACT The treatment of vertebral compression fractures of the spinal necessitates the use of injectable bone repair materials with the adaptive modulus of elasticity. Currently, newly developed injectable self-curing materials for minimally invasive surgery are primarily categorized into calcium phosphate and calcium silicate systems, both of which exhibit excellent biocompatibility. However, calcium phosphate bone cement lacks adequate mechanical properties and the stress does not match between bone and material. Calcium silicate cement has a long curing time, low mechanical strength, and high alkalinity. These deficiencies render them inadequate for meeting clinical requirements. Therefore, we incorporated inorganic disodium phosphate, organic polyvinyl alcohol, and solid component cerium oxide into the matrix phase of tricalcium silicate bone cement to establish a ternary self-curing hydration reinforcement system and produced an organic-inorganic composite injectable bone cement. Even though the composite bone cement has a longer setting time, it exhibited high compressive strength (61.9Mpa) and fracture strain (7.88%), demonstrating excellent mechanical properties. Furthermore, it also demonstrates good injectable properties (86%), anti-collapse properties, good biological activity, and outstanding antibacterial properties. The composite bone cement has great potential to develop a new injectable self-curing material to replace PMMA bone cement in clinical vertebral compression fractures.
