Interference-free Determination of Indole-3-Acetic Acid in Two Real Systems Using Second-order Calibration Method Coupled with Excitation-emission Matrix Fluorescence

Xiu-Fang YAN,Hai-Long WU,Xiang-Dong QING,Yan-Mei SUN,Ru-Qin YU

In this work, a simple and practicable method that combines excitation-emission matrix (EEMs) fluorescence with a second-order calibration method based on parallel factor analysis-alternative least-squares (PARAFAC-ALS) algorithm was developed for the direct interference-free determination of indole-3-acetic acid (IAA) in two real systems, coconut water (CW) and coconut milk (CM). Although the excitation and emission profiles of IAA heavily overlapped with that of unknown interferents in the complex real systems, the actual contents and satisfactory recoveries were still obtained successfully. The contents of IAA in CW and CM were 10.8 ± 0.3 and 4.9 ± 0.2 μg mL−1, respectively, which were consistent with those reported by LC-MS/MS assays in the reference material. The average spike recoveries of IAA in the validation set based on CW and CM were 102.1 ± 3.2 and 98.0 ± 1.9%, respectively. In addition, routine experiments were performed for establishing the validity of the assay to internationally accepted criteria.

Comparative analysis of cold-pressed and hot-pressed coconut oil extraction: Implications for quality and antioxidant capacity

Xingyang Guo,He Huang,Xiaoming Jiang,Chao Chang,Pan Gao,Wu Zhong,Chuanrong Hu,Dongping He,Jiaojiao Yin

Coconut oil, a natural oil widely utilized in the food, cosmetics, and pharmaceutical sectors, is significantly influenced by varieties and processing techniques. This study compared cold-pressed and hot-pressed extraction methods for coconut oil from four coconut varieties, evaluating their physicochemical properties, fatty acid composition, and antioxidant capacity. The results revealed that cold-pressed coconut oil exhibited superior control over parameters such as acid value, peroxide value, polycyclic aromatic hydrocarbons (PAHs), and 3-monochloropropane-1,2-diol (3-MCPD) esters, along with enhanced antioxidant capacity. Lauric acid, the predominant fatty acid in coconut oil, ranged from 47.68% to 50.08% in these oils. A significant negative correlation was observed between lauric acid and peroxide value/MCPD/PAHs, while a positive association existed between phenolic compounds and antioxidant capacity. Principal component analysis identified cold-pressed coconut oil from the W4 coconut variety as exhibiting the highest quality, characterized by lower acid value, MCPD and PAHs levels, higher lauric acid content, and potent antioxidant capacity. This study highlights the advantages of cold-pressed coconut oil, particularly from the W4 coconut variety, for superior quality across various parameters, offering valuable insights for the production and processing of coconut oil. These findings contribute to the advancement and application of high-quality coconut oil products in the industry.

Improving the quality of matured coconut (Cocos nucifera Linn.) water by low alcoholic fermentation with Saccharomyces cerevisiae: antioxidant and volatile profiles

Zhang Guanfei,Chen Wenxue,Chen Weijun,Chen Haiming

Matured coconut water (MCW) is a by-product in the coconut milk industry that is usually discarded due to its unpleasant flavor. In this study, low-alcohol coconut water (LACW) was fermented with Saccharomyces cerevisiae to improve the quality of MCW. Volatile components and nonvolatile flavor-related elements were estimated to compare the qualities of the MCW and LACW. Besides measuring the kinetic changes, the levels of fructose, glucose, sucrose and ethanol contents were also determined. The results of the organic acid assays showed that tartaric, pyruvic and succinic acids were the primary organic acids present in LACW and increased significantly with fermentation. The resulting volatile composition assay indicated that esters, alcohols and fatty acids were significantly influenced by fermentation and yeast strains. Moreover, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2′-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), cupric ion reducing antioxidant capacity and ferric reducing antioxidant power values increased significantly throughout the process, correlating with the enhancement of total phenolic content.

Enhancement of hydrogen bonds between proteins and polyphenols through magnetic field treatment: Structure, interfacial properties, and emulsifying properties

Yang Chen,Qiaozhu Wei,Yile Chen,Aiguo Feng,Weimin Zhang

In food systems, proteins and polyphenols typically coexist in a non-covalent manner. However, the inherent rigid structure of proteins may hinder the binding sites of polyphenols, thereby limiting the strength of their interaction. In the study, magnetic field (MF) treatment was used to enhance non-covalent interactions between coconut globulin (CG) and tannic acid (TA) to improve protein flexibility, enhancing their functional properties without causing oxidation of polyphenols. Based on protein structure results, the interaction between CG and TA caused protein structure to unfold, exposing hydrophobic groups. Treatment with a MF, particularly at 3 mT, further promoted protein unfolding, as evidenced by a decrease in α-helix structure and an increase in coil random. These structural transformations led to the exposure of the internal binding site bound to TA and strengthening the CG-TA interaction (polyphenol binding degree increased from 62.3 to 68.2%). The characterization of molecular forces indicated that MF treatment strengthened hydrogen bonding-dominated non-covalent interactions between CG and TA, leading to improved molecular flexibility of the protein. Specifically, at a MF treatment at 3 mT, CG-TA colloidal particles with small size and high surface hydrophobicity exhibited optimal interfacial activity and wettability (as evidenced by a three-phase contact angle of 89.0°). Consequently, CG-TA-stabilized high internal phase Pickering emulsions (HIPPEs) with uniform droplets and dense gel networks at 3 mT. Furthermore, the utilization of HIPPEs in 3D printing resulted in consistent geometric shapes, uniform surface textures, and distinct printed layers, demonstrating superior printing stability. As a result, MF treatment at 3 mT was identified as the most favorable. This research provides novel insights into how proteins and polyphenols interact, thereby enabling natural proteins to be utilized in a variety of food applications.

Analysis of Sugars and Fatty Acids during Haustorium Development and Seedling Growth of Coconut

Jing Li,Yin Min Htwe,Yong Wang,Yaodong Yang,Yi Wu,Dongxia Li,Abdul Kareem,Rencai Wang

Haustorium absorbs energy from the endosperms (liquid and solid) and plays a key role in nutrient mobilization during coconut germination and seedling growth. This study was conducted to shed light on possible interactions between haustorium and endosperms during this process. Germination rates and morphological changes of coconut cultivars Hainan Tall (HT) and Red Dwarf (RD), as well as the profiles of sugars and fatty acids in endosperms and haustorium were also analyzed. Results showed that liquid and solid endosperm inside the coconut exhausts successively, while haustorium occupies the nut gradually. Haustorium had significant positive correlation with seedling, while it had significant negative correlation with liquid and solid endosperms. Sugars in liquid endosperm continuously decreased, whereas in solid endosperm and haustorium accumulated at early stages and decreased later along with the aggressive seedling growth. Fatty acids in solid endosperm shared a very similar trend with sugars. In conclusion, haustorium development and seedling growth mainly depend on the nutrients stored in the liquid endosperm at early stages and then depend on the solid endosperm, in which fatty acids degrade rapidly to sugar so as to provide enough energy. These findings could be applicable in coconut germination and seedling propagation. Core Ideas Relative cool season (25–29°C) is more suitable for coconut germination and local variety (HT) is more adaptable. Development mainly depends on stored nutrients in liquid endosperm at early stages and then on solid endosperm. Fatty acids in solid endosperm degrade rapidly to sugars since liquid endosperm almost exhausted. Conversion of fatty acids to sugars helps provide enough energy for rapidly growing haustorium and seedling.

From coconut petiole residues to fluorescent carbon dots via a green hydrothermal method for Fe3+ detection

Gao Shiyu,Wang Xi,Xu Nan,Lian Hailan,Xu Li,Zhang Wenqing,Xu Changyan

Abstract Incinerating agricultural residues to recover energy is a popular method for reducing solid waste pollution and resource waste caused by the huge amount of coconut petiole residues. However, this process not only has the risk of secondary air pollution, but also the added value of comprehensive utilization is not high. In this paper, coconut petiole residues were used as eco-friendly and economical raw materials to synthesize carbon dots (CCDs) via a simple hydrothermal method. Furthermore, given the important reference significance of Fe 3+ concentration in assessing the quality of water environment and human body health, the obtained CCDs were utilized for Fe 3+ detection. The prepared spherical CCDs with an average diameter of 1.39 nm and a carbon skeleton dominated by benzene ring structure have surface functional groups that endow them with good water solubility. The CCDs with a quantum yield of 1.39% emit strong blue fluorescence under ultraviolet radiation (365 nm), and show favorable fluorescence stability with excitation-dependent and concentration-dependent emission behaviors. Based on the synergetic effect of static and dynamic quenching, the CCDs exhibit favorable selectivity and detection sensitivity toward Fe 3+ with a linear range of 0.005 to 0.2 mM and a detection limit of 2.3 μM, and also show the feasibility of Fe 3+ detection in real water samples. This work is in favor of the high value-added conversion of coconut petiole residues. In addition, the development and use of bio-based CCDs as fluorescence sensors may be beneficial to monitoring and protecting public health and the environment. Graphic abstract

Removal and mechanism investigation of polycyclic aromatic hydrocarbons in coconut oils by ionic liquids

Gege Zhao,Haoyu Xu,Hai Zhang,Hong Zhang,Yanlan Bi,Xuebing Xu

Crude coconut oil often contains high levels of polycyclic aromatic hydrocarbons (PAHs), which are commonly carcinogenic. The present work demonstrated that PAHs in coconut oil were significantly removed by ionic liquids (ILs). The relationship between the removal efficiency and the structures of ILs was investigated, using 12 different types ILs. Among these, 1-butyl-3-methylimidazolium tosylate ([Bmim][TOS]) showed the highest efficiency in removing benzo(a)pyrene (BaP). The optimal process conditions for the removal of PAHs using [Bmim][TOS] were explored, and the removal efficiency for benzo(a)anthracene (BaA), chrysene (CHR), benzo(b)fluoranthene (BbF), and BaP under the optimal conditions were 42.7 ± 1.0, 33.3 ± 1.4, 88.8 ± 0.9, and 73.8 ± 1.0%, respectively. Importantly, [Bmim][TOS] could be recycled up to five times without a significant decrease in PAHs removal. The quantum chemistry calculations and spectrum analyses were conducted to study the interactions between ILs and PAHs, using BaP as a model compound. The ability of [Bmim][TOS] to remove PAHs originate its high hydrogen bond accepting ability and the collaborative hydrogen bond interactions between the PAHs and ILs.

Metabolomics Analysis of the Deterioration Mechanism and Storage Time Limit of Tender Coconut Water during Storage

Yunwu Zhang,Wenxue Chen,Haiming Chen,Qiuping Zhong,Yonghuan Yun,Weijun Chen

Tender coconut water tastes sweet and is enjoyed by consumers, but its commercial development is restricted by an extremely short shelf life, which cannot be explained by existing research. UPLC-MS/MS-based metabolomics methods were used to identify and statistically analyze metabolites in coconut water under refrigerated storage. A multivariate statistical analysis method was used to analyze the UPLC-MS/MS datasets from 35 tender coconut water samples stored for 0–6 weeks. In addition, we identified other differentially expressed metabolites by selectingp-values and fold changes. Hierarchical cluster analysis and association analysis were performed with the differentially expressed metabolites. Metabolic pathways were analyzed using the KEGG database and the MetPA module of MetaboAnalyst. A total of 72 differentially expressed metabolites were identified in all groups. The OPLS-DA score chart showed that all samples were well grouped. Thirty-one metabolic pathways were enriched in the week 0–1 samples. The results showed that after a tender coconut is peeled, the maximum storage time at 4 °C is 1 week. Analysis of metabolic pathways related to coconut water storage using the KEGG and MetPA databases revealed that amino acid metabolism is one of the main causes of coconut water quality deterioration.

Effects of sophorolipids and coconut wax incorporation on the physical, structural, and antibacterial properties of cellulose nanofibers-based Pickering emulsion for cherry tomato preservation

Guangshun Li,Tong Wang,Qing Wei,Zhigang Jin,Huipei Han,Huixia Zhu,Xiaojing Ma

A novel acidic sophorolipid-enhanced cellulose nanofiber-based coconut wax Pickering emulsion (ASL-CCPE) using cellulose nanofibers (CNF) as a stabilizer, lactonic sophorolipids (LSL) as an antimicrobial agent, and acidic sophorolipids (ASL) as a co-stabilizer were developed. Through optimization of the oil phase type, oil-to-water ratio, and ASL content, the emulsion exhibited shear-thinning behavior and elastic gel characteristics, making it suitable for coating applications. After 182 days of storage at room temperature, the emulsion's droplet size remained uniform, with no significant flocculation observed. Compared to CCPE, ASL-CCPE exhibited reduced surface tension and formed a more uniform, dense, and washable coating. Cherry tomatoes coated with ASL-CDPE remained fresh for at least 12 days at room temperature. Furthermore, ASL-CCPE treatment resulted in a 5.10 % reduction in weight loss and a 45.83 % increase in vitamin C content even after 15 days of storage. Additionally, the inclusion of LSL significantly inhibited microbial growth, thus reducing decay. This green, safe, and effective emulsion offers a promising, environmentally friendly approach for fruit and vegetable preservation.

Eco-friendly production of leather-like material from bacterial cellulose and waste resources

Fan Yang,Zexuan Zhang,Jiugang Yuan,Jin Xu,Qiao Ji,Yutong Bai

In recent years, resource optimization has been recognized as a top priority among many challenges. Studies have indicated that valuable products can be created by combining natural or discarded resources with cutting-edge biosynthesis techniques. This approach is considered foundational for establishing a “zero-waste” socio-economic structure. This research focuses on optimizing resources and minimizing environmental impacts, especially within the manufacturing and fashion sectors. An innovative leather-like material named BC-BioLeath (Bacterial Cellulose-Based Bio Leather) has been introduced. BC was produced by the fermentation of coconut water, a byproduct of the food industry. Traditional leather-like processing, including tanning, was applied to BC using food waste such as oxidized edible oil and coffee grounds, along with timber industry by-products like vegetable tannins. Studies involving SEM observation, mechanical, style, and dyeing performance testing revealed that the product, BC-BioLeath, exhibited superior mechanical properties. For instance, BC crosslinked with oxidized edible oil showed a tensile strength of 82.14 MPa, surpassing traditional cowhide used in shoe uppers by 2.08 times. When plasticized with glycerol, the tensile strength reached 56.19 MPa, with breaking elongation of 36%. Beyond its mechanical prowess, the product also achieved economic and environmental benefits through waste repurposing, contributing to a natural energy cycle. BC-BioLeath was identified as a more humane and eco-friendly leather alternative. This method showcased not just efficient waste repurposing but also introduced a novel, microbe-focused sustainable production approach. Such a model offered a genuine circular economy solution for high-polluting sectors, such as the leather fashion industry. A cost analysis showed that BC-BioLeath's production cost of $97.92/m 2 (excluding labor and operating costs: $7.52/m 2 ) was within the commercial leather range of $39.7 to $220.8/m 2 . This indicated its sustainability as a competitive alternative.

Conjugation of coconut protein with ferulic acid promotes interfacial adsorption and mitigates lipid oxidation in O/W emulsion

Qianqian Zhu,Jianfei Pei,Weijun Chen,Haiming Chen,Wenxue Chen

The formation of small droplets in emulsion systems promotes lipid oxidation, affecting their shelf life. In this study, ferulic acid was covalently grafted onto coconut protein (CPI) through alkaline treatment method. The structural characteristics, interfacial properties, emulsifying properties, and oxidative stability of emulsions prepared by coconut protein-ferulic acid covalent complexes (CPF) were investigated. The results showed that covalent grafting of ferulic acid could change the physicochemical properties of coconut protein, including altering protein secondary and tertiary structures, enhancing the absolute value of zeta potential and antioxidant activity [2, 2-diphenyl-1-picrylhydrazyl (DPPH • ) and 2, 2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS •+ )]. Meanwhile, the interfacial behavior of CPI and CPF was characterized by dissipative quartz crystal microbalance and dilatational rheology analysis. Compared to CPI, CPF could diffuse to the oil-water interface quickly (from 0.084 ± 0.002 to 0.154 ± 0.003 mN/m/s 1/2 ), and formed a thick interfacial layer dominated by elasticity, which enhanced the centrifugal stability of CPF-stabilized emulsions and effectively inhibited lipid oxidation within a 15-day storage period. Our results clearly demonstrated that coconut protein-ferulic acid conjugated complexes could improve emulsion stability and mitigate lipid oxidation, owing to the strong electrostatic repulsion interaction and high diffusion rate at interface. These findings may provide comprehensive insights for the further design and development of protein-polyphenol covalent complexes in the food industry.

Bio-based composite phase change material utilizing wood fiber and coconut oil for thermal management in building envelopes

Aizheng Li,Yingying Yang,Yan Ren,Yuan Wan,Weidong Wu,Hua Zhang

Bio-based materials are attracting substantial attention for their eco-friendly attributes within the construction field, where they facilitate energy saving and emissions reduction. However, the performance of it, such as thermal properties, needs to be improved. Fully utilizing bio-based materials, we developed a novel composite phase change material (PCM), A-WF/CO/h-BN, with high thermal storage properties designed for application in building envelopes. It incorporates coconut oil (CO) as the PCM featuring an optimal phase change temperature . Hexagonal boron nitride (h-BN) was chosen to enhance the thermal conductivity . Acetylated wood fiber (A-WF) was employed as the supporting material. The prepared A-WF/CO/h-BN sample exhibits excellent and rapid heat storage and exothermic capabilities. In the experiment of thermal performance evaluation, the cold end of the A-WF/CO/h-BN sample is cooler than that of the A-WF when both hot ends are heated. Results indicate that the proposed A-WF/CO/h-BN sample can effectively delay the heat transfer and inhibit the temperature rise and fall. For these superior properties, when the A-WF/CO/h-BN sample is applied in building envelopes, it can minimize the heat transfer from the envelope to the interior and inhibit indoor temperature fluctuation. Thus, it can reduce building energy consumption and carbon emissions while achieving indoor thermal comfort.
