Monoterpene emissions and their protection effects on adult Cinnamomum camphora against high temperature

Xu Chenyi,Ma Yuandan,Tian Zhengfeng,Luo Qingyun,Zheng Tiefeng,Wang Bin,Zuo Zhaojiang

Key message The four chemotypes of adult C. camphora released different monoterpenes which protected these plants against high temperature by lowering ROS levels and maintaining photosynthetic abilities. Abstract High temperature seriously impacts plant growth and development, and terpenes are believed to improve plant thermotolerance through indoor experiments using seedlings. To uncover the actual protection effects of monoterpenes on outdoor adult plants, we investigated the monoterpene emissions, reactive oxygen species (ROS) metabolism, and photosynthetic abilities in four chemotypes of adult Cinnamomum camphora that were blocked monoterpene synthesis by fosmidomycin (Fos) under high temperature in summer (the mean highest temperature at about 38.2 °C). Eucalyptol (EuL), linalool (Lnl), borneol (BeL), and camphor (CmR) chemotypes of C. camphora released an abundance of monoterpenes, with eucalyptol, linalool, endo-borneol, and camphor as the typical monoterpene, respectively. When the four chemotypes of C. camphora were treated with Fos, their monoterpene emission was inhibited remarkably, with disappearance of some monoterpenes. In that case, the content of O 2 − , H 2 O 2 , and thiobarbituric acid reactive substance (TBARS) in the four chemotypes of C. camphora significantly increased although antioxidant enzyme activities remarkably increased, and the photosynthetic abilities significantly decreased by declining the quantum production and electron transport in photosystem II and increasing the dissipation of absorbed light energy. Among the four chemotypes, monoterpenes might play the maximum roles in CmR lowering ROS levels and in EuL maintaining photosynthesis. It can be speculated that monoterpenes can protect adult C. camphora against high temperature by lowering ROS levels and maintaining photosynthetic abilities.

Effervescent salt and crown ether-assisted matrix solid-phase dispersion extraction of coumarins from Cortex fraxini

Yu-Han Hu,Qiu-Yan Wang,Li-Hong Ye,Juan Yang,Xin Dong,Hui Zheng,Jian Zhou,Jun Cao

A simple, rapid and environmentally friendly method named effervescence-assisted matrix solid-phase dispersion (EA-MSPD) was investigated for the extraction of four coumarins (esculin, esculetin, fraxin and fraxetin) from Crotex fraxini ( C. fraxini ). In this study, an effervescent tablet containing carbon dioxide sources (100 mg sodium bicarbonate and 200 mg sodium dihydrogenphosphate) and an adsorbent (25 mg benzo-15-crown-5) was prepared. The effervescent tablet was dissolved into an aqueous solution, and as a consequence of the effervescence reaction, carbon dioxide was generated, thus making the sample better a dispersion of the extraction solvent (100 mM sodium dodecyl sulfate). The variables that influenced the extraction efficiency included effervescent salts, extraction solvents and crown ether. These values were represented using ultra- high-performance liquid chromatography (UHPLC). Good linearity was exhibited by coefficients of determination all equal to 1, limits of detection ranging from 1.62 to 3.83 ng·mL −1 and limits of quantification ranging from 5.39 to 12.76 ng·mL −1 . The recoveries ranged from 91.37 to 100.29% with relative standard deviations of 0.57–4.58% under optimized conditions. In this work, the proposed EA-MSPD method was successfully applied to extract and identify four coumarins in C. fraxini . Compared with previously reported methods, the method examined herein was faster, greener and more sensitive.

Authentication of Two Different Chemical Types of Cinnamomum Camphora Leaves by Microscopic Technique with GC-MS and GC Analysis

Ying-Jiao Liu,Li-Juan Xie,Lan Xiao,Li-Min Gong,Ta-Si Liu,Nai-Hong Chen

Cinnamomum camphora Chvar. Borneol (CCCB) and Cinnamomum camphora Chvar. Cineol (CCCC) are two different chemical types of C. camphora . The clinical application of these chemical types of CC shows big differences, but sometimes they are confused because of the similarity of morphological features. To identify the two types of CC and to analyze the differences of compositions, their morphological characteristics were investigated by microscopic techniques and the chemical components were determined by GC-MS analysis and GC internal standard method. Distinguishing features of morphological characteristics were the shape, petiol, gland, and smell. Microscopic study of the transverse sections of leaves showed 13 – 15 columns xylem vessel in the leaves of CCCB, 11 – 13 columns xylem vessel, and more obvious and thicker cell walls of fiber cells in the leaves of CCCC. From this research, GC-MS analysis of essential oil of the leaves could provide mainly differential characteristics to distinguish between CCCB and CCCC. The total of 33 compounds were identified in the essential oil of CCCB leaves and the main component was d -borneol (55.11%), while d -borneol in CCCC leaves was not the main component and the amount of d -borneol might be related to the microstructure of gland. L(-)-camphor and d-borneol were quantitatively analyzed by GC with FID detector, using methyl salicylate as the internal standard. Each gram of the total essential oil contains 0.68 g of d -borneol in CCCB, and each gram of the total essential oil contains 0.150 and 0.015 g of L(-)-camphor in CCCB and CCCC, respectively, from Hunan province. Other habitats showed the same trend. This research was aimed to establish a method based on the microscopic, GC-MS, and GC techniques to identify and quantify CCCB and CCCC compositions, for providing scientific data to ensure with accuracy the origin of CCCB and CCCC plant material.

Improving effect of phytase treatment on the functional properties and in vitro digestibility of protein isolate from Cinnamomum camphora seed kernel

Xianliang Bao,Xianghui Yan,Guohua Zhang,Junxin Zhao,Zheling Zeng,Ping Yu,Deming Gong

Phytate, an anti-nutritional factor in Cinnamomum camphora seed kernel protein isolate (CPI), chelated with minerals and proteins to form complexes that reduced their bioavailability. The study aimed to evaluate the effects of phytase treatment on the structural and functional properties and in vitro digestibility of CPI. The results showed that the degree of phytate degradation was improved with the increase in phytase treatment time. Most of the original secondary structures were retained in the treated CPI samples. The removal of phytate increased the Zeta potential of CPI, and the protein isoelectric point was increased. Low phytate CPI samples showed higher thermal stability and higher Ca 2+ content. In terms of functional properties, foaming capacity was positively correlated with phytate content, whereas foam stability, emulsifying activity index and emulsion stability index were negatively correlated with phytate content. The reduction of phytate content had a positive effect on in vitro gastrointestinal digestion, and the bioavailability of CPI was significantly improved. These results have demonstrated that phytase treatment retained most of the original structure, and improved functional properties and digestibility of the protein isolate from Cinnamomum camphora seed kernel.

Analysis of endophytic bacterial community diversity and metabolic correlation in Cinnamomum camphora

Zhang Guo-fang,Huang Qiu-liang,Bi Xue-qin,Liu You-lin,Yuan Zong-sheng

The structure and diversity of microbial communities in the leaves of Cinnamomum camphora at different growth stages were studied by high-throughput sequencing. Moreover, the relationships between microbial communities and borneol content were analyzed in this paper. The results indicated that the community structure of endophytic bacteria in C. camphora exhibited temporal variations, with the microbial diversity presented as follows: T1 (low content period) > T3 (peak period) > T2 (small peak period). The population of endophytic bacteria and the ratio of primary metabolism in the leaves of C. camphora were T2 > T1 > T3, while the metabolic intensity of endophytic bacterial terpenoids and polyketides was T3 > T2 > T1, which had the same trend as borneol content in C. camphora . The metabolic ratio of terpenoids and polyketides in T3 was 7.44% higher than that in T1, while that in T2 was 4.10% higher than that in T1. The abundance and diversity of Clostridium_sensu_stricto_1 , Ochrobactrum, Escherichia-Shigella, Pseudomonas , and Massilia significantly promoted the content of terpenoids in C. camphora . Together, those results provide the first evidence that borneol content and potential metabolic intensity in leaves of C. camphora greatly depend on microbial communities composition and diversity.

Regulatory roles of indole-3-acetic acid in the physiological and biochemical responses of Cinnamomum camphora seedlings under cadmium stress

Jing Jiang,Chenyang Zhang,Meng Na,Shangqi Xu,Xiaoping Li,Shoubiao Zhou,Jihai Zhou

Soil cadmium (Cd) pollution has become increasingly severe, negatively affecting plant growth, soil safety, the ecological balance, and public health. Phytoremediation technology is an effective method for remediating soil Cd contamination because of its low cost, environmentally friendly nature, and minimal soil disturbance. However, phytoremediation efficiency is affected by factors such as the plant species, Cd concentration, and soil environment. In this study, a 180-day indoor potted experiment was conducted to explore the influences of various indole-3-acetic acid (IAA) levels on the growth, physiological and biochemical responses, Cd uptake, and soil enzyme activity of Cinnamomum (C.) camphora seedlings using 20 mg kg −1 Cd-contaminated soil with or without IAA treatments. The results showed that Cd stress significantly suppressed C. camphora seedling growth, whereas IAA application improved the basal diameter of seedlings by 22.22–133.33 %. This effect of IAA was observed primarily at low concentrations, while higher concentrations showed diminished effects. Compared to the Cd pollution treatment, adding 20 mg kg −1 IAA significantly enhanced the activities of antioxidant enzymes (SOD, POD, CAT), reduced the malondialdehyde (MDA) content, and decreased soil basal respiration (SBR). Moreover, IAA treatment promoted Cd uptake in the roots, stems, and leaves, and reduced the soil Cd content by 22.90–41.17 %. In conclusion, exogenous IAA addition effectively alleviates Cd stress by regulating the plant’s physiological and biochemical characteristics and soil enzyme activities. These findings suggest that the combined use of IAA and C. camphora represents a promising remediation method for soil heavy metal pollution.

Exploring of toxic Pb(II) removal by low-cost bio-adsorbent of camphor leaf forestry waste after camphor oil extraction

Xu Sheng,Xiao Huiru,Jiang Xinde,Liu Lingli,Cao Meng,Wang Zhenxi

Camphor leaf (CL) was widely used to extract camphor oil and thus led to abundant forestry waste. In order to reduce pollution, the waste CL was used to prepare bio-adsorbent for Pb(II) removal after alkali treatment and functional modification. The effects of solution pH, initial Pb(II) concentration, contact time and solution temperature were investigated on adsorption process to evaluate the potential application in heavy metal ions’ removal. It was found that the massive hydroxyl groups released and plenty of micro-pores formed after the alkali treatment of CL bio-adsorbent, which obviously increased the Pb(II) adsorption. And the adsorption performance promoted continually after further functional modification by ionized 1,2,3,4-butanetetracarboxylic acid (BTCA). The increase of pH was favourable for the adsorption even though the precipitation effect was deducted. Linear fitting method was more suitable to describe the adsorption process than nonlinear fitting method, including adsorption isotherms and adsorption kinetics research. The adsorption thermodynamics was better to be described by nonlinear fitting method due to its lower root mean square error ( RMSE ) value and higher R 2 value. Among which, the adsorption isotherm and adsorption kinetics were fitted well to Langmuir model and pseudo-second-order model, respectively. The adsorption thermodynamics was exothermic in nature and the process was spontaneous at low solution temperature. The adsorption mechanism was revealed as the combination of dominant chemical adsorption and assistant physical adsorption.

Near-harvest methyl jasmonate application enhances essential oil yield and citral content in Camphora officinarum Nees ex Wall

Ling Qingyan,Zhang Beihong,Xiao Zufei,Shen Yao,Jiang Junfei,Xu Zhiwei,Liu Qingqing,Xiao Changlong,Zeng Rong,Liu Yuanqiu,Jin Zhinong

Citral-type Camphora officinarum ( C. officinarum ) exhibits high economic value due to its branch and leaf essential oil enriched with citral, yet the low natural concentration of this compound necessitates urgent strategies to enhance yield and quality. Methyl jasmonate (MeJA), a recognized safe elicitor, stimulates aromatic compound accumulation in plants and promotes monoterpene synthesis in C. officinarum suspension cells. However, it remained unclear whether foliar MeJA application enhances monoterpene biosynthesis in C. officinarum . This study evaluated the effects of near-harvest foliar MeJA application at gradient concentrations 0 μmol/L (control, M0), 100 μmol/L (M100), 250 μmol/L (M250), and 500 μmol/L (M500) on growth, oil gland morphology, and essential oil composition in citral-type C. officinarum . The results demonstrated that: M100 treatment significantly increased plant height, branch biomass, leaf biomass, and total biomass compared to the control M0. M250 treatment induced structural modifications in oil cells, enlarging their diameter and increasing density, resulting in an 11.83% increase in essential oil yield per unit fresh leaf weight. Both M100 and M250 treatments enhanced citral content by 3.6% and 3.31%, respectively, while total essential oil production surged by 12.31% (M100) and 15.46% (M250). Principal component analysis (PCA) collectively demonstrated that 100–250 μmol/L MeJA (M100/M250) represented the optimal induction concentration for enhancing essential oil yield and promoting citral synthesis, whereas 500 μmol/L (M500) induced detrimental osmotic stress. These findings demonstrate that MeJA serves as a safe agricultural regulation approach for enhancing essential oil yield and citral content, offering valuable insights for other aromatic plant species.

Dextran Conjugation Improves the Structural and Functional Properties of Heat-Treated Protein Isolate from Cinnamomum camphora Seed Kernel

Xianghui Yan,Xiaofeng Gong,Zheling Zeng,Maomao Ma,Junxin Zhao,Jiaheng Xia,Meina Li,Yujing Yang,Ping Yu,Deming Gong,Dongman Wan

TheCinnamomum camphoraseed kernel (CCSK), with high contents of medium-chain oil (~59%) and protein (~19%), is an excellent source for a plant-based food ingredient. To broaden the application of the protein isolate (PI) from CCSK in the food industry, the Maillard reaction products (MRPs) were prepared by PI and dextran (DX) under mild wet-heating conditions (60 °C, 5 h), and the structural and functional properties of the PI-DX conjugates were investigated. The covalent bond between PI and DX was confirmed by the degree of grafting and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Compared with the heated PI, the PI-DX conjugates had more ordered structure, with the decreased random coil content. The changes in tertiary structure of PI-DX conjugates were reflected by the results of intrinsic fluorescence and surface hydrophobicity. Moreover, PI-DX conjugates showed better solubility, emulsifying properties, thermal stability and antioxidant activities. These results provided a theoretical basis for the development of PI-based MRPs with desirable characteristics. Keywords:plant protein;Maillard reaction;conjugate;structural property;functional property

Effects of Drying Methods on the Physicochemical and Functional Properties of Cinnamomum camphora Seed Kernel Protein Isolate

Mengqiang Ye,Zhixin Wang,Xianghui Yan,Zheling Zeng,Ting Peng,Jiaheng Xia,Junxin Zhao,Weiyun Wang,Deming Gong,Ping Yu

Cinnamomum camphoraseed kernel protein isolate (CPI) has attracted increasing attention due to its sustainability and potential applications. This study aimed to investigate the effects of freeze-drying (FD), vacuum-drying (VD), and spray-drying (SD) on the physicochemical and functional properties of CPI. The morphology observation results showed that the SD-CPI, SD-CPI, and VD-CPI were spherical, lamellar, and massive, respectively. Compared to FD and SD, VD had more impact on the color, surface hydrophobicity, intermolecular disulfide bonds, intrinsic fluorescence, and thermal stability of CPI. Fourier transform infrared spectroscopy (FTIR) analyses showed that among three CPI samples, VD-CPI had the highest content of β-sheet but the lowest contents of α-helix and β-turn. At different pH values, the solubility, emulsification, and foaming properties of VD-CPI were inferior to those of FD-CPI and SD-CPI. These results provide useful information on the changes in the physicochemical and functional properties of CPI subjected to different drying methods, and offer theoretical guidance for the production and use of CPI in the food industry.

Seasonal emission of monoterpenes from four chemotypes of Cinnamomum camphora

Zhengfeng Tian,Qingyun Luo,Zhaojiang Zuo

Plant terpenes are the main functional agents in forest bathing and have great value in several industries. To uncover the seasonal variation of Cinnamomum camphora monoterpene emission and the difference among the chemotypes for promoting the development of forest bathing and exploitation of monoterpenes using the species, the emission of monoterpenes and related gene expression in the camphor chemotype, eucalyptol chemotype, linalool chemotype and borneol chemotype of C. camphora were analyzed in May (spring), July (summer) and November (autumn), and the stored monoterpenes and storage structure were investigated. Eleocytes were the storage structure of monoterpenes in C. camphora , which widely existed in the leaf palisade tissue and spongy tissue, as well as around the glands and vascular bundles. The 4 chemotypes of C. camphora released different monoterpene components which mainly derived from the immediate emission after synthesis. Camphor, eucalyptol, linalool and endo-borneol showed the uppermost emission rate and were the typical monoterpenes in the corresponding chemotype, respectively, due to the high expression of the genes encoding bornyl diphosphate synthase ( E5.5.1.8 ), linalool synthase ( TSP14 ) and (+)-neomenthol dehydrogenase ( E1.1.1.208 ) for the biosynthesis of camphor, linalool and endo-borneol, respectively, and only expression of the gene encoding 1,8-cineole synthase ( TSP-cin ) for eucalyptol biosynthesis. Among the 4 chemotypes, the linalool chemotype showed the highest emission rate of monoterpenes in the same month (without significant difference with the eucalyptol chemotype in November), due to the high expression of the genes ( dxs , dxr , ispF , gcpE , and GGPS ) in methylerythritol-4-phosphate (MEP) pathway, α-terpineol synthase gene ( E4.2.3.111 ) and TSP14 . Among the 3 months, temperature was the remarkably varied meteorological condition. The highest monoterpene emission rate from the 4 chemotypes of C. camphora was detected in July, which should result from the up-regulated expression of related genes in monoterpene biosynthesis and improved vaporization and diffusion of stored monoterpenes under high temperature, with the former providing a major contribution.

Adjusting function of camphor on primary metabolism in Cinnamomum camphora stressed by high temperature

Yuyan Cai,Haozhe Xu,Chenyi Xu,Zhaojiang Zuo

Cinnamomum camphora has great economic value for its wide utilization in traditional medicine and furniture material, and releases lots of monoterpenes to tolerate high temperature. To uncover the adjusting function of monoterpenes on primary metabolism and promoting their utilization as anti-high temperature agents, the photosynthetic capacities , primary metabolite levels, cell ultrastructure and associated gene expression were surveyed in C. camphora when it was blocked monoterpene biosynthesis with fosmidomycin (Fos) and fumigated with camphor (a typical monoterpene in the plant) under high temperature (Fos+38 °C+camphor). Compared with the control (28 °C), high temperature at 38 °C decreased the starch content and starch grain size, and increased the fructose, glucose, sucrose and soluble sugar content. Meanwhile, high temperature also raised the lipid content, with the increase of lipid droplet size and numbers. These variations were further intensified in Fos+ 38 °C treatment. Compared with Fos+ 38 °C treatment, Fos+ 38 °C+camphor treatment improved the starch accumulation by promoting 4 gene expression in starch biosynthesis, and lowered the sugar content by suppressing 3 gene expression in pentose phosphate pathway and promoting 15 gene expression in glycolysis and tricarboxylic acid cycle . Meanwhile, Fos+ 38 °C+camphor treatment also lowered the lipid content, which may be caused by the down-regulation of 2 genes in fatty acid formation and up-regulation of 4 genes in fatty acid decomposition. Although Fos+ 38 °C+camphor treatment improved the photosynthetic capacities in contrast to Fos+ 38 °C treatment, it cannot explain the variations of these primary metabolite levels. Therefore, camphor should adjust related gene expression to maintain the primary metabolism in C. camphora tolerating high temperature.
