Tangeretin, a polymethoxyflavone abundant in citrus fruits, is acclaimed for its diverse biological benefits to human health and is a significant component of traditional Chinese medicine. This study reports the development of an indirect competitive enzyme-linked immunosorbent assay (icELISA) with monoclonal antibody for quantifying tangeretin in various citrus and herbal medicines. The optimized icELISA demonstrated a half-maximal inhibitory concentration (IC 50 ) of 5.57 ng/mL, encompassing a dynamic range of 1.54–25.38 ng/mL, and showcased average recoveries ranging from 81.65 % to 105.75 % in citrus peel and pulp samples. Extensive analysis of six citrus cultivar samples and four herbal samples was conducted using the developed assay, with results corroborated by Ultra–Performance Liquid Chromatography Tandem Mass Spectrometry (UPLC–MS/MS). These findings confirm the assay’s efficacy and reliability for tangeretin detection, offering a practical method for evaluating this bioactive compound in food.

In this work, water-resistant poly (vinyl alcohol) (PVA)/poly (acrylic acid) (PAA) electrospun fibers encapsulating tangeretin (Tan) were fabricated by emulsion-electrospinning. To optimize the electrospinning condition, the size and morphology of electrospun fibers were characterized by dynamic light scattering (DLS), optical light microscope, fluorescence microscopy, and scanning electron microscopy (SEM), respectively. The optimized initial concentration of PVA/PAA was 10% (w/w) with a mass ratio of 3:7. The time and temperature of optimized thermal crosslinking treatment were 2 h and 145 °C, respectively. The results of XRD and SEM showed that the Tan was successfully incorporated into smooth PVA/PAA electrospun fibers and those fibers possessed nano-diameter size and high porosity. The encapsulation of Tan had no significant impact on the antioxidant activity of PVA/PAA/Tan crosslinking electrospun fibers. The in vitro release test showed the PVA/PAA/Tan crosslinking electrospun fibers achieved durable release profiles and lower burst release rates than that from the pure Tan emulsion. Based on these results, it is concluded that PVA/PAA/Tan crosslinking electrospun fibers prepared by emulsion-electrospinning serve as a promising technique in the fields of water-insoluble drug delivery and slow-release.

This study aimed to reveal the interaction and inhibitory mechanisms of tangeretin (TAN), nobiletin (NBT), and their acidic hydroxylated forms, 5-demethyltangeretin (5-DT) and 5-demethylnobiletin (5-DN) on porcine pancreatic lipase (PPL) using spectroscopic techniques and molecular dynamics (MD) simulation. PPL inhibition assay showed that the inhibitory activity of NBT (IC 50 value of 3.60 ± 0.19 μM) was superior to those of three polymethoxylated flavones (PMFs), indicating it may be related to the methoxy groups at the 3′-position in its molecular structure. Inhibition kinetic analyses demonstrated that the inhibition types of the 4 PMFs were consistent with the mixed inhibition model, which agreed well with the results from the ultraviolet-visible (UV–Vis) spectroscopy, Circular dichroism (CD), fluorescence spectroscopy, molecular docking , and MD simulation that PMFs could bind to the PPL catalytic site and non-catalytic site, affecting the normal spatial conformation of PPL and weakening its ability to decompose the substrate. All these findings suggest that PMFs are a kind of natural lipase inhibitors, and NBT has the potential as a lipase inhibition precursor because of its unique flavone skeleton structure.

White spot syndrome virus (WSSV) is a serious pathogen threatening global crustacean aquaculture with no commercially available drugs. Herbal medicines widely used in antiviral research offer a rich reserve for drug discovery. Here, we investigated the inhibitory activity of 13 herbal medicines against WSSV in crayfish Procambarus clarkii and discovered that naringenin (NAR) has potent anti-WSSV activity. In the preliminary screening, the extracts of Typha angustifolia displayed the highest inhibitory activity on WSSV replication (84.62%, 100 mg/kg). Further, NAR, the main active compound of T. angustifolia, showed a much higher inhibition rate (92.85%, 50 mg/kg). NAR repressed WSSV proliferation followed a dose-dependent manner and significantly improved the survival of WSSV-challenged crayfish. Moreover, pre- or post-treatment of NAR displayed a comparable inhibition on the viral loads. NAR decreased the transcriptional levels of vital genes in viral life cycle, particularly for the immediately early-stage gene ie1 . Further results showed that NAR could decrease the STAT gene expression to block ie1 transcription. Besides, NAR modulated immune-related gene Hsp70 , antioxidant ( cMnSOD, mMnSOD, CAT, GST ), anti-inflammatory ( COX-1 , COX-2 ) and pro-apoptosis-related factors (Bax and BI-1) to inhibit WSSV replication. Overall, these results suggest that NAR may have the potential to be developed as preventive or therapeutic agent against WSSV.

Tangeretin, a flavonoid from Citri Reticulatae Pericarpium, is known for its neuroprotective effects, but the mechanisms are not fully understood. Alzheimer's disease, a leading neurodegenerative disorder, characterized by amyloid-beta (Aβ) accumulation, represents a significant therapeutic challenge. This study investigates the protective effects of tangeretin against Aβ1-42-induced neurotoxicity using HT22 cells and zebrafish larvae as experimental models. Tangeretin mitigated Aβ 1-42 -induced cytotoxicity, as evidenced by enhanced cell viability and reduced apoptosis. Tangeretin treatment mitigated Aβ 1-42 -induced cytotoxicity in HT22 cells, as evidenced by enhanced cell viability and reduced apoptosis. Mechanistically, tangeretin ameliorated mitochondrial dysfunction by reducing mitochondrial fragmentation, decreasing donut-shaped mitochondria, restoring mitochondrial membrane potential, and attenuating reactive oxygen species (ROS) production. Moreover, tangeretin modulated mitochondria-lysosome interactions by promoting mitophagy and normalizing the prolonged mitochondria-lysosome contact induced by Aβ 1-42 . In zebrafish larvae, Aβ 1-42 exposure resulted in developmental malformations, including pericardial and yolk sac edema, elevated ROS levels, increased apoptosis, and impaired neurodevelopment. Tangeretin effectively counteracted these deficits, as revealed by live imaging, supporting its neuroprotective role observed in cellular models. Collectively, our study suggests that tangeretin may serve as a promising protective agent against Aβ 1-42 -induced neurotoxicity.

Grapefruit ( Citrus × paradisi Macf.) peel, a by-product of the citrus-processing industry, possesses an important economic value due to the richness of bioactive compounds. In this study, boron-linked covalent organic frameworks integrated with molecularly imprinted polymers (CMIPs) were developed via a facile one-pot bulk polymerization approach for the selective extraction of naringenin from grapefruit peel extract. The obtained CMIPs possessed a three-dimensional network structure with uniform pore size distribution, large surface areas (476 m 2 /g), and high crystallinity. Benefiting from the hybrid functional monomer APTES-MAA, the acylamino group can coordinate with the boronate ligands of the boroxine-based framework to form B-N bands, facilitating the integration of imprinted cavities with the aromatic skeleton. The composite materials exhibited a high adsorption capacity of 153.65 mg/g, and a short adsorption equilibrium time of 30 min for naringenin, together with favorable selectivity towards other flavonoid analogues. Additionally, the CMIPs captured the template molecules through π–π* interaction and hydrogen bonding, as verified by FT-IR and XPS. Furthermore, they had good performance when employed to enrich naringenin in grapefruit peels extract compared with the common adsorbent materials including AB-8, D101, cationic exchange resin, and active carbon. This research highlights the potential of CMIPs composite materials as a promising alternative adsorbent for naringenin extraction from grapefruit peel.

Summary The oral bioavailability of tangeretin is limited by its low water solubility and high crystallisation. In the present study, tangeretin was entrapped in whey protein concentrate (WPC)-stabilised emulsions to enhance its bioaccessibility. Hydroxypropyl methylcellulose (HPMC) was added to modify the interface of the emulsion droplets, and Ca 2+ -crosslinking was used to induce the gel formation. Spray drying of the emulsions or gels enabled to produce microcapsules. The physicochemical characteristics and in vitro digestion behavior of the microcapsules were investigated. Results indicated that HPMC modification and Ca 2+ -crosslinking increased the average particle size, altered the microstructure, ameliorated the quality of the WPC-based microcapsules and improved the encapsulation efficiency of tangeretin. Furthermore, the in vitro studies showed that HPMC modified and Ca 2+ -added microcapsules could control the release of free fatty acid. HPMC greatly increased the bioaccessibility of tangeretin from around 28% to 80%. Overall, these results confirmed that HPMC-modified WPC microcapsules could be a promising carrier to protect hydrophobic functional ingredients and improve their oral bioavailability.

A dual-drug delivery system (DDDS) based on oxidized alginate (OxAlg) and carboxymethyl chitosan (CMCS) is developed, which can be used for dual-responsive delivery of methotrexate (MTX) and naringin (Nar) for efficient treatment of osteosarcoma. Nar is first loaded onto graphene oxide (GO) through π-π stacking and hydrogen bonding, and then the Nar loaded GO (GO/Nar) is co-encapsulated with MTX by the hydrogel of OxAlg and CMCS generated through Schiff base reaction. The acylhydrazone (–N = C–) bonds between OxAlg and CMCS are prone to be hydrolyzed in an acidic medium, speeding the delivery of the two drugs; meanwhile, GO in this DDDS is an outstanding photothermal agent, leading to near-infrared light (NIR)-responsive delivery of the two drugs. The cumulative release of MTX and Nar can reach 91.09% and 85.69%, respectively, at pH 5.0 with NIR irradiation. The produced hyperthermia under NIR irradiation can result in the ablation of the osteosarcoma cells, and thus the therapeutic efficacy of the DDDS against osteosarcoma is significantly improved via the chemo-photothermal synergistic therapy mode, which can be further confirmed by the cytotoxicity test.

Bone tissue engineering is a promising approach for tackling clinical challenges. Osteoprogenitor cells, osteogenic factors, and osteoinductive/osteoconductive scaffolds are employed in bone tissue engineering. However, scaffold materials remain limited due to their source, low biocompatibility, and so on. In this study, a composite hydrogel scaffold composed of hydroxyapatite (HA) and sodium alginate (SA) was manufactured using three-dimensional printing. Naringin (NG) and calcitonin-gene-related peptide (CGRP) were used as osteogenic factors in the fabrication of drug-loaded scaffolds. Investigation using animal experiments, as well as scanning electron microscopy, cell counting kit-8 testing, alkaline phosphatase staining, and alizarin red-D staining of bone marrow mesenchymal stem cell culture showed that the three scaffolds displayed similar physicochemical properties and that the HA/SA/NG and HA/SA/CGRP scaffolds displayed better osteogenesis than that of the HA/SA scaffold. Thus, the HA/SA scaffold could be a biocompatible material with potential applications in bone regeneration. Meanwhile, NG and CGRP doping could result in better and more positive proliferation and differentiation.
