Natural deep eutectic solvent-ultrasound for the extraction of flavonoids from Fructus aurantii: Theoretical screening, experimental and mechanism

Hua Du,Wenhui Wu,Yaodeng Wang,Ping Tang,Yi Wu,Jing Qiu,Chong Xu,Lingzi Li,Min Yang

To reduce the environmental pollution caused by organic solvents and improve the extraction efficiency, a green and high-effective natural deep eutectic solvent (NADES) was screened out and prepared for the extraction of flavonoids from Fructus aurantii based on the COMOS-RS and experimental verification. The results showed the results of theoretical screening and experimental verification were consistent, and Ch-Lea was the best extraction solvent, which further proved the COMOS-RS was an efficient tool for the solvent screening. Then, the extraction conditions were investigated. The optimum extraction conditions were as follows: molar ratio (1:2), water content (30%), solid–liquid ratio (1/60 g/g) and extraction time (15 min), yet the extraction temperature and power did not significantly affect the extraction yield. Under the optimal extraction condition, the extraction yield of flavonoids was 122.68 mg/g. Compared with the traditional extraction solvents (methanol and ethanol), Ch-Lea has a higher extraction yield. And the green assessment was conducted based on Eco-Scale methodology, which results indicated the Ch-Lea was more environmentally friendly than methanol and ethanol. Meanwhile, the Ch-Lea has good reusability, and the extraction yields from four reuses were 122.68 mg/g, 125.13 mg/g, 122.56 mg/g and 105.63 mg/g, respectively. Moreover, the extraction mechanism was delved into using molecular dynamics (MD) simulation and reduced density gradient (RDG) analysis. The hydrogen bond interaction, particularly the O-H bonds, between Ch-Lea and the naringin molecule emerged as the primary force driving the extraction process. In summary, Ch-Lea is a green, economical, and highly effective solvent, making it a promising candidate for flavonoid extraction from Fructus aurantii . Its potential as an alternative to traditional extraction solvents further underscores its value.

Integrated transcriptomic and metabolomic analyses reveal key genes controlling flavonoid biosynthesis in Citrus grandis ‘Tomentosa’ fruits

Ruiyi Fan,Congyi Zhu,Diyang Qiu,Genlin Mao,Bernd Mueller-Roeber,Jiwu Zeng

As a well-recognized traditional Chinese medicine (TCM), immature fruits of Citrus grandis ‘Tomentosa’ (CGT) serve to cure chronic cough in humans. Specialized metabolites including flavonoids may have contribute to this curing effect. Knowledge about the molecular mechanisms underlying flavonoid biosynthesis in ‘Tomentosa’ fruits will, therefore, support the breeding of varieties with improved medicinal properties . Hence, we profiled the transcriptomes and metabolites of the fruits of two contrasting C. grandis varieties, namely ‘Zheng-Mao’ (‘ZM’) used in TCM production, and a locally cultivated pomelo, namely ‘Guang-Qing’ (‘GQ’), at four developmental stages. A total of 39 flavonoids, including 14 flavanone/flavone, 5 isoflavonoids , 12 flavonols, and 6 anthocyanins , were identified, and 16 of which were quantitatively determined in the fruits of the two varieties. We found that ‘ZM’ fruits contain more flavonoids than ‘GQ’. Specifically, rhoifolin levels were significantly higher in ‘ZM’ than in ‘GQ’. We annotated 31,510 genes, including 1,387 previously unknown ones, via transcriptome sequencing of ‘ZM’ and ‘GQ.’ A total of 646 genes were found to be differentially expressed between ‘ZM’ and ‘GQ’ throughout at all four fruit developmental stages, indicating that they are robust expression markers for future breeding programs. Weighted gene co-expression network analysis identified 18 modules. Combined transcriptional and metabolic analysis revealed 25 genes related to flavonoid biosynthesis and 16 transcriptional regulators (MYBs, bHLHs, WD40) that may be involved in the flavonoids biosynthesis in C. grandis ‘Tomentosa’ fruits.

Pectin oligosaccharides from Citri Reticulatae Pericarpium ‘Chachi’ promote wound healing in HaCaT keratinocytes by enhancing cell proliferation and migration

Zhongcan Peng,Shurong Tian,Depo Yang,Longping Zhu,Jianing Zhang,Wenfeng Li,Guodong Zheng,Zhimin Zhao

ABSTRACT Citri Reticulatae Pericarpium ‘Chachi’ ( Citrus reticulata ‘Chachi’) is a traditional Chinese medicine with dual medicinal and dietary uses. As its main component, pectin exhibited various biological activities. However, research and development on it, especially pectin oligosaccharides, remain limited. In this study, ‘Chachi’ pectin oligosaccharides (CPOS) was extracted and prepared for the first time, resulting in the purification of two oligosaccharide components, CPOS3 and CPOS4. Structural characterization of the predominant oligosaccharide, CPOS3, revealed it to be composed of a backbone α-D-Gal p A-(1→2)-α-L-Rha p -(1→4)-α-D-3-OAc-Gal p A-(1→4)-α-D-Gal p A-(1→4)-α-D-Gal p A and a side chain [→5)-α-L-Ara f -(1→] 4 attached at the C-4 of Rha p . Subsequent experiments have shown that CPOS3 can significantly promote the proliferation and migration of human immortalized keratinocyte cell. Further investigation revealed that it facilitated cell proliferation by stimulating DNA synthesis in the S phase of the cell cycle. Additionally, CPOS3 exhibited good thermal stability and rheological characteristics. Based on the good wound healing activity and favorable physicochemical properties of CPOS3, its prospects in the fields of wound dressings and tissue repair are promising.

Metabolomic Profiling Reveals the Quality Variations in Citri Reticulatae Pericarpium (Citrus reticulata Blanco cv. Chachiensis) with Different Storage Ages in Response to “Candidatus Liberibacter Asiaticus” Infection

Jiayin Liang,Yuqing Xi,Jiaming Li,Shugui Xu,Yongqin Zheng,Meirong Xu,Zheng Zheng,Xiaoling Deng

Citri ReticulataePericarpium, especially the pericarp ofCitrus reticulataBlanco cv. Chachiensis (PCRC), is an important edible and medicinal ingredient for health and pharmacological properties. Citrus Huanglongbing, a devastating disease that currently threatens the citrus industry worldwide, is caused by a phloem-limited alpha-proteobacterium, “CandidatusLiberibacter asiaticus” (CLas). The industry of cultivar Chachiensis has been suffering from HLB. Although HLB affected the quality of citrus fruit, whether the quality of PCRC was affected by HLB remains unclear. In this study, we compared the metabolite profiles between HLB-affected and healthy PCRC from three sources: fresh, 6-month-old, and 9-year-old PCRC, through the untargeted LC–MS method. Compared to healthy controls, various types of bioactive compounds, mainly flavonoids, terpenoids, alkaloids, coumarins, polysaccharides, and phenolic acids, accumulated in HLB-affected PCRC, especially in the HLB-affected 9-year PCRC. In particular, isorhamnetin, isoliquiritigenin, luteolin 7-O-beta-D-glucoside, limonin, geniposide, pyrimidodiazepine, scoparone, chitobiose, m-coumaric acid, malonate, and pantothenic acid, which contributed to the pharmacological activity and health care effects of PCRC, were highly accumulated in HLB-affected 9-year-old PCRC compared to the healthy control. Multibioassay analyses revealed that HLB-affected 9-year-old PCRC had a higher content of total flavonoids and total polyphenols and exhibited similar antioxidant capacity as compared to healthy controls. The results of this study provided detailed information on the quality of HLB-affected PCRC.

Microbial-Metabolite Interactions Underlying Quality Differences Between Green and Red Citrus Reticulata “Chachi” peel

Yimei Zhao,Xinrui Xiao,Jiadai Tang,Xueqiu Wang,Cheng Wu,Lu Qiao

By using non-targeted metabolomics technology and high-throughput sequencing methods, the differences in metabolites between the green and red Citrus reticulata “Chachi” peels (CRCP), and their associations with microbial communities were systematically analyzed. Combined with physicochemical indicators (browning degree, total flavonoid content, antioxidant capacity), the material basis of their quality and functional components was explored. The results showed that the total flavonoid content (21.89 mg/g) and the antioxidant indicators (ABTS, FRAP) in green CRCP were significantly higher than those in red CRCP ( P < 0.05), while there was no significant difference in browning degree. Metabolomics analysis showed that a total of 1,322 differential metabolites were identified in green and red CRCP. A total of 723 metabolites in green CRCP were significantly up regulated (∼54.70%) ( p < 0.05), including active components such as lariciresinol, ginsenoside Rb1, and salidroside, with potential anti-diabetic, anti-tumor and anti-depression properties, respectively. Correlation analysis of physicochemical indicators and metabolites indicated that antioxidant capacity was positively correlated with most metabolites (83.56%), and the accumulation of core metabolites such as dodecatetraene and genipin was synergistically regulated by FRAP, ABTS, and total flavonoids. The abundance of fungi such as Aspergillus spp. In green CRCP was relatively high, which was positively correlated with the total flavonoid and antioxidant indicators, driving the synthesis of flavonoids. Cladosporium in red CRCP was negatively correlated with metabolites, and bacteria were negatively correlated with antioxidant indicators. This study reveals the advantages of green CRCP in antioxidant activity, and provides a theoretical basis at the metabolite and microbial levels.

Development of a sensitive monoclonal antibody-based indirect competitive enzyme-linked immunosorbent assay for analysing nobiletin in citrus and herb samples

Yongliang Cui,Jing Zhao,Jie Zhou,Guiyu Tan,Qiyang Zhao,Yaohai Zhang,Baomin Wang,Bining Jiao

Nobiletin, a polymethoxyflavone mainly found in citrus fruits, have been reported to exhibit various beneficial biological activities for human health. It is an important bioactive compound in traditional Chinese medicine, Pericarpium Citri Reticulatae and Fructus Aurantii . To detect the contents of nobiletin in citrus and herb samples, we developed an indirect competitive enzyme-linked immunosorbent assay (icELISA) based on monoclonal antibodies. It possessed a median inhibition concentration (IC 50 ) of 2.43 ± 0.19 ng/mL and a working range of 0.52–12.3 ng/mL. The assay exhibited the average recoveries of 72.5–85.3% in citrus peel, pulp and juice samples. Moreover, eleven citrus cultivars samples and four herb samples were also detected by the icELISA. The nobiletin content varied in different citrus cultivars samples and herb samples, which were confirmed by the ultra-performance liquid chromatography tandem mass spectrometry (UPLC-MS). These results indicated that the developed immunoassay was suitable for detecting nobiletin in citrus and herb samples.

Integrated metabolomics and network pharmacology to reveal antioxidant mechanisms and potential pharmacological ingredients of citrus herbs

Xiaomin Cao,Kaixin Shi,Yang Xu,Peipei Zhang,Hongyan Zhang,Siyi Pan

The benefits of citrus herbs are strongly associated with their secondary metabolites. In the study, we conducted widely-targeted metabolomics and ultra-high performance liquid chromatography (UPLC) to compare the variability of ingredients in four citrus herbs. In total, we discovered 1126 secondary metabolites, primarily comprising flavonoids, phenolic acids, lignans and coumarins, and alkaloids. Differential metabolites of citrus herbs were searched by multivariate statistical analysis. Notably, Citri Reticulatae Pericarpium contained higher levels of flavonoids, while Zhique and Huajuhong demonstrated a greater abundance of coumarins. Among the flavonoids determined by UPLC, Guangchenpi demonstrated significantly elevated levels of polymethoxyflavones (tangeretin and nobiletin) compared to other citrus herbs. Additionally, we determined their antioxidant capacity (Chenpi > Guangchenpi > Huajuhong > Zhique) using in vitro assays. Finally, we utilized network pharmacology to explore the antioxidant mechanisms and potential pharmacological ingredients, providing a basis for future preventive and therapeutic applications of these metabolites.
