Citrinin is a common mycotoxin produced by different fungi. Although considerable endeavors have been made to increase citrinin output to meet the requirements of research and industrial needs, few methods are effective, owing to potential safety concerns and technical limitations. In this study, a blue fungus that produces a yellow pigment was isolated from Ziziphus jujuba . The strain was then identified as Penicillium citrinum and named HR-087. Preparative liquid chromatography was used to extract the yellow pigment. Both LC–MS and NMR assays showed that it was citrinin. To enhance the citrinin production of this strain, the culture medium was optimized through orthogonal experiments, as well as batch and fed-batch process. The final titer and productivity of citrinin reached 9.62 g/L and 0.1 g/(L × h), respectively. The high titer achieved indicates the possibility of large-scale production of high purity citrinin for low-cost supply for both academic and diagnostic analysis standard.

Zearalenone (ZEN), a common mycotoxin, has significant threat to human and animal health, so its rapid and sensitive detection in foods is quite important. In this work, a magnetic molecular imprinting electrochemiluminescence (ECL) sensor was proposed for its detection, which was constructed by loading carbon dots (CDs) with Fe 3 O 4 magnetic nanoparticles (MNPs) and then coating them with a molecular imprinted polymer (MIP). The CDs were prepared using waste coffee grounds, enabling low-cost and large-scale production. After polyethyleneimine modification, the CDs could generate strong ECL signal in the presence of tri-n-propylamine. Through siloxane cross-linking, the CDs were anchored onto the MNPs surface, and the MIP was simultaneously formed. The resulting molecularly imprinted nanoparticle exhibited low cytotoxicity under tested conditions and was able to rapidly enrich and separate ZEN. The obtained sensor demonstrated high stability, sensitivity, reproducibility, selectivity, and anti-interference capability. Under optimized conditions, the ECL intensity was linear to the logarithm of ZEN concentration in the range of 1.0 × 10 −3 –3.2 × 10 2 ng mL. When applied to detect ZEN in grain samples (including corn, wheat flour, and rice), the spiked recoveries ranged from 90.1 % to 109 %, with relative standard deviation below 7.2 %. This work provides a novel approach for the low-cost and rapid detection of mycotoxins.

To enable sensitive quantitative detection of zearalenone (ZEN) in cereal matrices, this study develops a molecularly imprinted electrochemical sensor. A three-dimensional flower-shaped carbon composite material (CoNiZn@C), synthesized from a ternary CoNiZn metal–organic framework, was utilized as a highly conductive substrate featuring excellent electrical conductivity and abundant binding sites. Density functional theory calculations were conducted to elucidate the intermolecular interactions and cooperative recognition mechanisms between zearalenone and the selected monomers. Experimentally, the AuNPs-based thioaniline units (PATP@AuNPs) and pyrrole were employed as bifunctional monomers. Specifically, PATP@AuNPs can be stably immobilized on the substrate surface to enhance interfacial electron transfer efficiency, while simultaneous in situ copolymerization of the two monomers generates numerous non-covalent binding cavities capable of specifically recognizing ZEN. Under optimized experimental conditions, the sensor exhibited a linear detection range from 10 nM to 1000 µM and a detection limit as low as 1.6 nM. This detection approach is straightforward, cost-effective, and demonstrates excellent specificity and sensitivity, indicating substantial potential for rapid screening and quantitative analysis of mycotoxins in agricultural products. Herein, a novel ratiometric molecularly imprinted electrochemical sensor (MIP-RECS) based on Ag nanoparticles (AgNPs) and ionic liquid (IL) co-modified Ti 2 NTx (AgNPs@Ti 2 NT x @IL) was developed for the determination of patulin (PAT). Firstly, the AgNPs@Ti 2 NT x @IL nanocomposite, was prepared by an in situ reduction and self-assembled method, and then, was cast on the glassy carbon electrode (GCE) to build an electrochemical interface with excellent conductivity and high surface area. After that, the MIP was deposited on the nanocomposite modified GCE by the electro-polymerization technique, with thionine (Thi) and PAT as functional monomer and template molecules. In the sensing platform, the oxidation peaks generated by Thi and AgNPs were used as reference and probe signal, respectively. The introduction of reference effectively eliminates the influence of the matrix effect. At the same time, the oxidation signal of the Ag probe is greatly improved by the high conductivity and large surface area of the AgNPs@Ti 2 NT x @IL nanocomposite. Therefore, the MIP-RECS presented a wide linear range, a very low low limit of detection (LOD), together with high selectivity and excellent repeatability. Additionally, the linearity ranges and LODs of the sensor can be controlled by the amount of Ag, highlighting its robustness in different types of real samples. Furthermore, this sensing model can be used to sensitively detect other residues with poor redox activity, such as imidacloprid and catechol, highlighting its versatility and universality.

This study developed an electrochemical sensor incorporating a dual recognition strategy that integrates molecularly imprinted polymer (MIP) and aptamers for the ultra-sensitive detection of zearalenone (ZEN) in grains. Exploiting the high specific surface area of the Cu-Zr bimetallic metal-organic framework-818 (MOF-818), aptamers were covalently immobilized on the olefin-functionalized MOF-818 surface via thioenol reaction. Employing 3-thiopheneacetic acid functionalized gold nanoparticles (3-TAA@AuNPs) as functional monomers, nanoscale molecularly imprinted cavities were electro-polymerized onto the modified electrode surface in the presence of the ZEN template. This synergistic recognition system integrates the aptamer's specific binding capability with the spatial complementarity of MIP, displaying an excellent linear response over a concentration range of 10 −13 to 10 −5 g/L and a detection limit as low as 2.8 × 10 −14 g/L. Analysis of real samples exhibited ZEN recoveries ranging from 96.5 % to 105.0 % (RSD <5.2 %) in corn and wheat matrices, confirming its practical applicability for rapid screening of fungal toxins in agricultural commodities.

In recent years, food-borne pathogen Pseudomonas cocovenenans have caused many food safety incidents. Therefore, the detection of toxoflavin produced by the bacteria has aroused widespread attention. In this study, two fluorescent sensors were prepared using nitrogen-doped carbon dots (NCDs) materials as precursors: (1) Luminol-functionalized NCDs were synthesized by NHS-EDC reaction, and L-NCDs fluorescence sensor was constructed for the detection of toxoflavins in actual food samples. The linear range of the L-NCDs fluorescence sensor is 0.1–50 μM, the detection limit is 0.05 μM and the excitation wavelength is 375 nm. (2) The NCDs-CMC fluorescent film sensor was constructed by incorporating NCDs into sodium carboxymethyl cellulose. It was excited at a wavelength of 385 nm for visual detection of toxoflavin. The film sensor has a linear range of 1–60 μM and a detection limit of 0.45 μM. The results showed that the method has good sensitivity and reliability. This study extends the application of NCDs-based fluorescence sensing to detect toxoflavin. It enables simple, rapid, low-cost sensitive detection and visual monitoring of toxoflavin. Additionally, it offers a fast and convenient new technology for preventing and controlling Pseudomonas cocovenenans contamination in food.

The relationship between human health and patulin (PAT) in the diet is a complex and intertwined one. The development of a sensing approach for the field detection of patulin is crucial, as the current approach lacks real-time detection capabilities and is costly in terms of material and technology. This paper presents a portable ratiometric fluorescence sensor that can be used to rapidly, accurately, and efficiently detect patulin in food items at the point of origin. The sensor employs a combination of sulfhydryl functionalized gold nanoclusters (MUA-AuNCs) and blue emission carbon dots (B-CDs), which have been engineered to serve as highly effective “on-off” nanoprobes. The modified sulfhydryl ( SH) groups present on the gold clusters serve as specific recognition sites for patulin binding. The probes exhibit a discernible shift in hue, from orange-red to blue. The sensitivity detection limit (LOD) for patulin was found to be 0.019 μM, with a substantial linear correlation observed in the range of 0–2.2 μM. The objective of the combined chromatographic test strip and color recognition platform was to facilitate the sensitive, accurate, and real-time detection of patulin in foodstuffs, which is of paramount importance for the prevention of early disease. To facilitate rapid and straightforward preliminary testing of food security, it is anticipated that the integrated chromatographic strip ratiometric fluorescence sensing platform will be developed into portable home detection equipment.

BACKGROUND Bioherbicides are becoming more attractive as safe weed control tools towards sustainable agriculture. Natural products constitute an important source chemicals and chemical leads for discovery and development of novel pesticide target sites. Citrinin is a bioactive compound produced by fungi of the genera Penicillium and Aspergillus . However, its physiological-biochemical mechanism as a phytotoxin remains unclear. RESULTS Citrinin causes visible leaf lesions on Ageratina adenophora similar to those produced by the commercial herbicide bromoxynil. Phytotoxicity bioassay tests using 24 plant species confirmed that citrinin has a broad activity spectrum and therefore has potential as a bioherbicide. Based on chlorophyll fluorescence studies, citrinin mainly blocks PSII electron flow beyond plastoquinone Q A at the acceptor side, resulting in the inactivation of PSII reaction centers. Furthermore, molecular modeling of citrinin docking to the A. adenophora D1 protein suggests that it binds to the plastoquinone Q B site by a hydrogen bond between the O1 hydroxy oxygen atom of citrinin and the histidine 215 of the D1 protein, the same way as classical phenolic PSII herbicides do. Finally, 32 new citrinin derivatives were designed and sorted according to free energies on the basis of the molecular model of an interaction between the citrinin molecule and the D1 protein. Five of the modeled compounds had much higher ligand binding affinity within the D1 protein compared with lead compound citrinin. CONCLUSION Citrinin is a novel natural PSII inhibitor that has the potential to be developed into a bioherbicide or utilized as a lead compound for discovery of new derivatives with high herbicidal potency. © 2023 Society of Chemical Industry.

A fast and sensitive method involving ultra-performance liquid chromatography–tandem mass spectrometry (UPLC–MS/MS) was introduced to detect citrinin in dried orange peel. A series of extraction, purification and chromatographic conditions was also systematically examined. With the proposed method, the obtained calibration graph was linear, with an R of 0.9996 within a concentration range of 0.5–10 ng/mL. The estimated limits of detection and quantification were 0.05 and 0.17 ng/mL, respectively. Under the selected conditions, the relative recoveries in different citrus products spiked with 1–10 ng/mL citrinin were 89.4–98.7% with RSDs of <2.5%. Compared with previously reported analytical methods, the newly developed UPLC–MS/MS method showed excellent sensitivity and good precision in detecting citrinin. The results indicated that it is a reliable and effective technique for the detection of trace citrinin in dried orange peel.

A novel dual-dummy templates imprinted electrochemical sensor has been fabricated for the detection of patulin. Herein, 2-oxindole (2-oxin) and 6-hydroxynicotinic acid (6-HNA) as the dummy templates, 4-aminothiophenol as functional monomer, and ionic liquid (IL) as electropolymerization electrolyte are employed to prepare molecularly imprinted polymer (MIP) film. 2-Oxin and 6-HNA have multiple groups and the obtained MIP possesses different types of imprinted sites, thereby achieving a better recognition capacity than that of single-dummy imprinted film. ILs can regulate the density of molecularly imprinted film and facilitate effective molecular recognition. The composite of PtPd decorated N-doped porous carbon has good conductivity and large surface area, and can amplify the signal. With the aid of electrochemical probe [Fe(CN) 6 ] 3-/4- (0.16 V vs. SCE) patulin can be detected. Under the optimal conditions, this sensor shows a detection range from 0.01 to 10 μg L −1 , with a detection limit of 7.5 × 10 −3 μg L −1 ( S / N = 3). Two spiked juice samples were analyzed by this method, and the recovery ranges from 94 to 99.8% with RSD values of 2.4–4.6% ( n = 3), indicating that this method can be applied for the detection of patulin in real samples. AimsRed yeast rice (RYR), produced by inoculating Monascus strains to steamed rice, contains many kinds of physiologically bioactive compounds, among which monacolin K can be used as an antihypercholesterolaemic agent. However, RYR can be polluted by the mycotoxin citrinin, which has nephrotoxic and hepatotoxic activities. To avoid the risk of citrinin contamination in Monascus fermented products, it is important to screen for Monascus strains that produce no or low citrinin. Methods and ResultsFive autochthonous Monascus strains with high‐yield monacolin K and undetectable citrinin were obtained using high‐performance liquid chromatography (HPLC). All five strains were identified as Monascus ruber based on Genealogical Concordance Phylogenetic Species Recognition criteria. Polymerase chain reaction revealed that citrinin polyketide synthase (pksCT) gene was found in these strains, but transcriptional regulator (ctnA) was not found.ConclusionsFive strains are potential strains for producing high‐quality RYR. The distribution of the pksCT gene was not restricted to Monascus purpureus and Monascus sanguineus, and M. ruber strains were diverse in pksCT and ctnA genes.Significance and Impact of the StudyThe integration of citrinin HPLC analysis and pksCT and ctnA genes amplification could provide a complementary approach in valuable Monascus strains screening.
