Pneumatic extrusion-based three-dimensional (3D) printing can be used to fabricate custom-made scaffolds to restore irregular bone defects. During the 3D printing process, therapeutic agents can be added to the scaffolds. This study aimed to develop a polycaprolactone (PCL) scaffold loaded with Ag3PO4 to prevent infections and lidocaine for pain relief by one-step 3D printing. We hypothesized that the drug release could be controlled by varying the filament diameter of the 3D printed scaffolds. To this end, PCL slurry mixed with different amounts of silver phosphate and lidocaine was printed via differently sized nozzles. The obtained cylindric scaffolds displayed a porous interconnected microstructure with high fidelity. The Ag3PO4 and lidocaine were distributed homogeneously. The lidocaine release could be controlled by adjusting the filament diameter while the silver release is correlated with the Ag3PO4 loading amount. The released medium from silver-loaded scaffolds exhibited an obvious inhibition zone against Staphylococcus aureus and Escherichia coli upon loading with 1% Ag3PO4 for up to 6 days and with 3% Ag3PO4 for at least 7 days. Cytotoxicity of all scaffolds was screened by cell assay. In conclusion, the pneumatic extrusion-based 3D printing provides a practical technique to fabricate drug-loaded scaffolds. The Ag3PO4 and lidocaine loaded PCL scaffolds showed the potential for infection prevention and pain relief.

In this study, a multi-component integrated dissolution evaluation system of Yuanhu Zhitong tablets (YZTs) was established based on in vitro and in vivo correlation (IVIVC). The dissolution tests of five quality markers (Q-markers), including tetrahydropalmatine, α-allocryptopine, protopine, corydaline, and byakangelicin, in YZTs were conducted under different dissolution conditions, and pharmacokinetic studies were performed in beagle dogs to construct a correlation model using numerical deconvolution. The data of the five ingredients were integrated in vitro and in vivo according to the biopharmaceutical classification system (BCS) to establish an IVIVC integrating multiple Q-markers. The dissolution media with the best correlation of components were obtained and validated. The results showed that all five components were classified as BCS I compounds, and α-allocryptopine, byakangelicin, tetrahydropalmatine, and corydaline showed good correlation in the paddle method, 75 rpm, with dissolution media of artificial gastric fluid, acetate buffer, acetate buffer and 0.1 M HCl, respectively. Protopine showed good correlation in the paddle method, 100 rpm, with dissolution media of 0.1 M HCl. The integrated BCS I Q-markers showed the best correlation in the medium of acetate buffer. The multi-component integrated dissolution evaluation system established in this experiment accurately predicted the pharmacokinetic data of YZTs by verifying the media, which can be used for the quality control of YZTs. The present study provides an effective and promising strategy for the dissolution evaluation for traditional Chinese medicine preparations.

Chiral compounds commonly exist in traditional Chinese medicine (TCM), but little research on the quality control of TCM has been conducted. In this study, a new strategy is proposed, taking Yuanhuzhitong tablet [YHZT, consisting of Radix Angelicae Dahuricae and Rhizoma Corydalis (Yan Hu Suo, YHS)] for example, which is based on chiral isomer ratio analysis to monitor the production process of Chinese patent medicine companies. In the process of content determination for tetrahydropalmatine (THP) in YHZT from different companies, noticeable differences were observed in their chromatographic behaviors. It is known that THP has two enantiomers, naturally coexisting in YHS as a racemic mixture, so we prepared THP twice and subsequently performed chiral separation analysis using supercritical fluid chromatography. As a result, the peak area ratios of two enantiomers from different companies varied remarkably, demonstrating that some companies did not probably manufacture YHZT products in accordance with the prescription proportion, used inferior or extracted YSH crude materials in the production process, and added raw chemical medicine in the production to reach the standard and lower the costs. In conclusion, the peak area ratio of chiral isomers could be taken as a key quality index.

Graphical A ready-to-use collagen triboelectric nanogenerator (TENG) wound dressing system has been developed. The electric field generated by the TENG promotes the release of the analgesic drug from the collagen patch to achieve rapid pain relief. This system provides stable electrical stimulation output and its low cost and portability also offer an effective strategy for emergency on-site applications. Healing of exposed wounds is a key process in the repair of skin injuries and is crucial to prevent infection, maintain fluid balance, and promote tissue regeneration. Conventional dressings have been widely used; however, they are often inconvenient to use and have limited analgesic effects. In this work, we have developed a ready to use, disposable collagen-triboelectric nanogenerator (Col-TENG) wound protection dressing system. A porous collagen patch acts as a drug reservoir and a contact layer for the skin to protect the wound and remove traumatic contaminants. The frictional electrical field generated by TENG effectively promotes the release of analgesic lidocaine drug from the collagen patch. In vitro and in vivo experiments have shown that compared to conventional drug release systems, this novel friction-facilitated drug release device achieves faster and more drug release, as well as superior analgesia effects, which is promising in application of pain management in wild environments.

BackgroundPrimary dysmenorrhea (PDM) is a prevalent menstrual disorder among women, often underreported and undertreated. Wen-Jing-Zhi-Tong Decoction (WJZTD), a patented Traditional Chinese Medicine (TCM) herbal decoction, has shown efficacy in treating PDM. However, the underlying therapeutic mechanism of WJZTD in PDM treatment remains to be elucidated.ObjectiveThis study aimed to employ integrative pharmacology and experimental validation to investigate the potential therapeutic mechanisms of WJZTD in treating PDM.MethodsThe bioactive compounds of WJZTD were identified by UPLC-Q-Exactive-Orbitrap MS/MS and GC-MS. Putative targets of WJZTD were obtained from Swiss Target Prediction, STITCH, and BATMAN-TCM databases. Known targets of PDM were retrieved from Gene Cards and Drug Bank databases. Protein-to-protein interactions were constructed to screen key targets using the STRING database. Subsequently, GO and KEGG pathway enrichment analyses were performed based on Metascape. Finally, a PDM rat model was established to validate the potential therapeutic mechanisms of WJZTD using Western Blot, PCR, and ELISA.Results390 bioactive compounds in WJZTD were identified through UPLC-Q-Exactive-Orbitrap MS/MS and GC-MS. Network pharmacology revealed 7 key compounds with 20 targets and pathways that are crucial for WJZTD in treating PDM. Behavioral tests confirmed that WJZTD can effectively ameliorate menstrual pain in PDM. WJZTD also inhibited prostaglandin production, thereby relieving uterine smooth muscle contraction. The downregulation of the BDNF/TrkB/ERK/CREB signaling pathway, identified as the key target and pathway through network pharmacology, may be crucial to the anti-nociceptive and anti-inflammatory effects of WJZTD in treating PDM.ConclusionThis study provides the first comprehensive analysis of the key compounds, targets, and pathways of WJZTD, laying a solid foundation for future pharmacological studies on PDM. The anti-nociceptive and anti-inflammatory effect may be attributed to the downregulation of the BDNF/TrkB/ERK/CREB signaling pathway.

In photodynamic therapy (PDT), reactive oxygen species (ROS) are key products that induce cell death, and increasing amount of ROS is a crucial way to enhance PDT efficacy. However, the generated ROS stimulates the transient receptor potential vanilloid 1 channel (TRPV1), which can be activated in the pain pathway and then exacerbate pain. Herein, we utilized arginine−glycine−aspartate (RGD) peptide-modified liposomes for encapsulation Chlorin e6 (Ce6) and capsazepine (Cz), a receptor antagonist of TRPV1, to prepare drug-loaded liposomes, RLCC. Soluble hyaluronic acid microneedle arrays (MNs), which possess sufficient skin penetration capability and excellent biosafety, was applied for in situ delivery of RLCC. With the aid of RGD peptides, the efficiency of intracellular liposomal uptake and the dispersion of drugs in tumor after delivery by MNs were significantly enhanced, showcasing tremendous potential for improving the PDT efficacy. Besides, through the analysis of sciatic nerve signals in mice during PDT, RLCC demonstrated remarkable effectiveness in alleviating pain by significantly reducing nerve impulses. Hence, RLCC demonstrated outstanding effectiveness in PDT and effectively alleviated the associated pain. Overall, this research highlights the potential of utilizing MNs for the in situ delivery of RLCC, facilitating effective PDT while addressing the issue of pain during the treatment.

A cellulose-based hydrogel with synergistic therapeutic effect was prepared by a one-pot method. The hydrogel had hydroxyethyl cellulose (HEC) as a framework and epichlorohydrin (EPI) as a cross-linking agent. Cationic-β-cyclodextrin (C-β-CD) was homogeneously embedded in the grid of the cross-linked structure. Zinc oxide nanoparticles (ZnO NPs) and menthol were used to provide analgesic and bactericidal effects . Menthol was uniformly loaded onto the hydrogel matrix by the addition of C-β-CD and increased the solubility of menthol in water. ZnO NPs were synthesized by in situ hydrothermal treatment . The positively charged group of C-β-CD and the active hydroxyl group of HEC were used to distribute the ZnO NPs uniformly on the hydrogel matrix without significant agglomeration. The hydrogel had a smoothly ordered and compact morphology. It exhibited adequate mechanical, swelling, water retention and oxygen permeability properties in tests. In addition, the hydrogel had an efficient bactericidal effect. Release experiments showed that the encapsulation of menthol by C-β-CD allowed for a slow release of menthol. Biocompatibility tests had shown that the hydrogel was non-toxic to normal human cells.

Licorice is a traditional Chinese medicine and recorded to have pain relief effects in national pharmacopoeia, but the mechanisms behind these effects have not been fully explored. Among the hundreds of compounds in licorice, licochalcone A (LCA) and licochalcone B (LCB) are two important components belonging to the chalcone family. In this study, we compared the analgesic effects of these two licochalcones and the molecular mechanisms. LCA and LCB were applied in cultured dorsal root ganglion (DRG) neurons, and the voltage-gated sodium (NaV) currents and action potentials were recorded. The electrophysiological experiments showed that LCA can inhibit NaV currents and dampen excitabilities of DRG neurons, whereas LCB did not show inhibition effect on NaV currents. Because the NaV1.7 channel can modulate Subthreshold membrane potential oscillations in DRG neuron, which can palliate neuropathic pain, HEK293T cells were transfected with NaV1.7 channel and recorded with whole-cell patch clamp. LCA can also inhibit NaV1.7 channels exogenously expressed in HEK293T cells. We further explored the analgesic effects of LCA and LCB on formalin-induced pain animal models. The animal behavior tests revealed that LCA can inhibit the pain responses during phase 1 and phase 2 of formalin test, and LCB can inhibit the pain responses during phase 2. The differences of the effects on NaV currents between LCA and LCB provide us with the basis for developing NaV channel inhibitors, and the novel findings of analgesic effects indicate that licochalcones can be developed into effective analgesic medicines.

Background Migraine is the third most common disease worldwide, leading to severely decreased quality of life for the patients. In spite of great efforts endeavored in pharmacological and nonpharmacological therapeutic strategies for treating migraine, the outcome is rather disappointing in terms of efficacy. Compelling evidence shows that the expression level of dopamine receptor D2 (DRD2) plays an essential role in progression of migraine. Purpose To explore potential therapeutical possibilities, the attention was paid to Yuanhu Zhitong formula (YHZTF), which is a classical traditional Chinese medicine prescription frequently applied to relieve pain. The aim of this study was to identify the promising compounds derived from YHZTF with anti-migraine effects and investigate the underlying molecular mechanism. Methods The high-resolution mass spectrometry and molecular networking were performed for comprehensive chemical profiling of YHZTF. Network pharmacology was used to generate herbal-component-target-pathway network. Based on the pathway enrichment analysis, the active substances of anti-migraine and the potential molecular mechanism were further determined by performing animal experiments combined with molecular docking strategy. Results In total, 31 substances were identified in YHZTF, including alkaloids such as tetrahydropalmatine and protopine . The analysis of herbal-component-target-pathway network suggests that the alkaloid substances (e.g. tetrahydropalmatine and protopine) from YHZTF target dopamine receptors , thus can be linked to neuroactive ligand-receptor interaction pathways. In a nitroglycerin-induced migraine animal model, pretreatment with tetrahydropalmatine or protopine substantially lessened the aberrant migraine-like symptoms. The results of molecular docking analysis showed that tetrahydropalmatine and protopine had strong affinities to dopamine receptor D2 (DRD2). Using RT-qPCR, the investigators found that DRD2 was significantly down-regulated at the mRNA level in brain tissues of tetrahydropalmatine and protopine-treated group compared to the control group. Conclusion Collectively, the results provide reliable evidence showing that the active substances tetrahydropalmatine and protopine from YHZTF lessens migraine symptoms in an in vivo mouse model suggestively via regulating expression of DRD2. These findings shed light on novel therapeutic strategies and targets to treat migraine using natural products.

Abstract As one of the most common forms of skin injuries, skin burns are often accompanied by edema pain, suppuration of infection, slow tissue regeneration, and severe scar formation, which significantly delay wound healing as well as affect the quality of life. We prepared multifunctional electrospun poly(L-lactide-co-glycolide)/gelatin (P/G)-based dressings to synergistically harness the therapeutic benefits of peppermint essential oil (T), burn ointment (B), and Oregano essential oil (O) (P/G@TBO) for skin regeneration in punch and burn injury models. The P/G@TBO can afford the sustained release of bioactive cues for up to 72 h as well as remarkably promote cell migration ( ca. P/G@TBO, 89% vs. control group, 51%) at 24 h. The P/G@TBO membranes also showed significant angiogenic effect as well as antibacterial and anti-inflammatory properties than that of the control group in vitro. Moreover, P/G@TBO dressings enabled fast wound healing ( ca. P/G@TBO, 100% wound closure vs. control group, 95%) in a full-thickness excisional defect model up to 14 days in rats. Further evaluation of membranes in different animal models, including tail wagging model, facial itch model, and hot burn injury model showed significant pain relieve effect as well as itching and swelling relief functions during earlier stages of wound healing. Membranes were next transplanted into a scalded wound model in rats and an ear punch wound model in rabbits, which manifested antibacterial and anti-inflammatory properties and promoted re-epithelialization to achieve scarless wound healing percentage wound closure at day 28: P/G@TBO, 96% vs. control group 66%. Taken together our approach of simultaneously harnessing T, B, and O to enable multifunctionality to fibrous dressings may hold great promise for burn wound healing applications and other related disciplines. Qufeng Zhitong capsule (QZC) is a well-known Chinese patent medicine that has been widely applied for the clinical treatment of rheumatoid arthritis and other inflammatory diseases. To date, its material basis is still unclear, which has greatly limited its clinical application. In this study, by taking advantage of ultra-high-performance liquid chromatography tandem Q-Exactive Orbitrap high-resolution mass spectrometry, 16 chemical components such as gallic acid, protocatechuic acid, and neochlorogenic acid in QZC were characterized and unambiguously identified based on comparison with the corresponding reference standards. In addition, the correlation between the focused components and their corresponding raw herbs from QZC prescription was investigated. For the first time, the relationship between the components mentioned above and their anti-inflammatory activity was explored via network pharmacology analysis, and a visualized network of “medicinal materials-QZC-compounds-targets-pathways” was established. Based on the brief prediction results of network pharmacological analysis, ultra-performance liquid chromatography coupled with photodiode array detector method was validated in terms of linearity, limit of detection, limit of quantification, precision, repeatability, stability, and recovery test and was successfully employed to determine 16 compounds in 28 batches of QZCs, which confirmed the feasibility and reliability of the established method for the quantitative analysis of 16 compounds in QZC. Considering the content and bioactivity of the tested components, four compounds were recommended as candidate indicators for quality evaluation ultimately. The potential value of this study could not only support a quality evaluation of QZC but also provide a theoretical basis for further in-depth research of QZC in clinical research.
