Rapid hemostasis, biosafety, and adaptability to different bleeding sites are the focus of research on hemostatic materials. In this work, an efficient and simple preparation of aldehyde starch hemostatic microspheres (ASHMs) crosslinked with L-cystine dimethyl ester was developed. Through a series of physicochemical parameters and in vitro and in vivo hemostatic properties measurements, the results showed that under the optimal crosslinking degree, ASHMs had a regular and uniform spherical shape, and it had a liquid suction capacity of up to 880%. At the same time, the aldehyde groups on its surface can increase the adhesion and aggregation of blood cells, thereby enhancing the hemostatic effect, which can be considered as a synergistic effect. More importantly, ASHMs have good biocompatibility and minimal intradermal irritation. Taken together,ASHMs could be used as potential novel hemostatic materials for surgical treatment.

Chitosan has been made into various hemostats, but their hemostatic efficiency for controlling severe traumatic bleeding is still inadequate. The aim of this work is to make quick hemostats by incorporating mesoporous silica nanoparticles into chitosan. Porous chitosan-silica composite microspheres (CSMS-S) with high hemostatic efficacy were fabricated through a combination of the microemulsion , thermally induced phase separation, and surfactant templating method. A large number of mesoporous silica nanoparticles were formed on and within the CSMS-S microspheres, which had abundant surface and inner macropores. The synergetic two hemostatic mechanisms from chitosan and mesoporous silica nanoparticles let CSMS-S composite microspheres with proper amount of silica displayed better hemostatic potential than the single component porous chitosan microspheres (CSMS). Within a same time interval, the whole blood clotting kinetics showed that CSMS-S could form larger blood clots than CSMS. The hemostatic time of CSMS-S was down to 97 s from 114 s of CSMS in the rat liver laceration model. The cytotoxicity and histological analysis proved that CSMS-S was a safe hemostatic agent without noticeable adverse effects on tissues around the wound. Our results demonstrate that CSMS-K is a promising quick hemostatic agent for traumatic hemorrhaging control.
Keratinocyte growth factor-2 (KGF-2) can regulate the proliferation and differentiation of keratinocyte, which plays a remarkable role in maintaining normal tissue structure and promoting wound healing. As an effective strategy, KGF-2 solution is widely used in the treatment of wounds in clinical applications. However, KGF-2 in solution cannot achieve sustained release, which results in drug loss and unnecessary waste. Polysaccharide hemostasis microspheres (PHMs) are an ideal drug loading platform due to their special “lotus seedpod surface-like” morphology and structure. Herein, to realize the controllable release of KGF-2, PHMs loaded with KGF-2 (KGF-2@PHMs) were prepared. It was found that the bioavailability of KGF-2 was improved greatly. Most importantly, KGF-2@PHMs can reduce inflammation and accelerate the wound healing process due to the controlled release of KGF-2. KGF-2@PHMs might be a potential alternative strategy for wound healing in future clinical applications.
