Self-Healing Oxidized Dextran/Sodium Alginate Hydrogel Dressing with Hemostatic Activity Speeds Up Wound Healing in Burn Injuries

Jingya Zhao,Huiqing Zhu,Tenglong Xu,Suyue Xu,Yuxin Wang,Yanwei Hou,Meng Zhuo,Kun Du,Yingyu Su,Xiao Han,Yongli Shi,Xueyan Hou

This study introduced a hydrogel dressing, termed SODex-gel, which was constructed by establishing Schiff base and hydrogen bonds with the precursors of oxidized dextran (ODex) and succinic dihydrazide (SD)-modified sodium alginate (SD-mod-SA). Through comprehensive in vitro and in vivo studies, the adhesive properties, self-healing capabilities, hemostatic potential, and wound healing efficacy of the SODex-gel dressing were meticulously evaluated. The 1H NMR, FTIR, and TGA analyses confirmed the fabrication of the SODex-gel dressing and its constituent elements. Scanning electron microscopy (SEM) imaging showcased the uniform pore structures in the SODex-gel dressing. In vitro assessments demonstrated that the SODex-gel dressing was noncytotoxic and exhibits strong adhesion, enabling it to attach to various surfaces. Noteworthy findings from studies of mouse liver incisions and tail amputation models proved the hemostatic ability of the SODex-gel dressing. Moreover, their remarkable wound-healing capabilities were prominently demonstrated through the treatment of a mouse model afflicted with burn skin injuries. Evidence of neovascularization effects was corroborated by the upregulation of CD31 and vascular endothelial growth factor (VEGF) expression in the treated skin samples. Collectively, the experimental data unequivocally established that the SODex-gel dressing is a promising therapeutic approach to accelerate wound recovery, thereby exhibiting substantial potential for clinical applications in treating burn injuries.

Iridoid Glycosides from Phlomis Medicinalis Diels: Optimized Extraction and Hemostasis Evaluation

Sitong Lu,Zhaoyu Zhang,Youling Liang,Qingkun Zhong,Guangfa Li,Zhang Hu

Phlomis medicinalis Diels, an important perennial herbal plant unique to the Qinghai-Tibet Plateau, is often used as Tibetan Materia Medicine Radix Phlomii for the treatment of cold, cough, and convergence trauma. In order to efficiently extract the iridoid glycosides from P. medicinalis , an ultrasound-assisted deep eutectic solvent extraction technique was employed. The main parameters influencing the extraction process were studied through single-factor tests and the extraction was optimized by using response surface methodology. The hemostasis activity of total iridoid glycosides (TIG) from P. medicinalis was evaluated in vitro and in mice. The optimization results revealed that the optimal process parameters were liquid-solid ratio 20 : 1, choline chloride-lactic acid concentration 79 %, and sonication time 34 min, under which a TIG extraction yield of 20.73 % was obtained. Meanwhile, high-performance liquid chromatography-photodiode array/mass spectrometry (HPLC-PDA/MS) was employed to characterize the optimized extract and indicated that TIG from P. medicinalis mainly consisted of sixteen reported iridoid glycosides with a total content of 91.22 %. The experimental results in vivo and in vitro indicated that TIG from P. medicinalis had strong hemostasis activities, which may be achieved by increasing the fibrinogen levels. Therefore, the ultrasound-assisted deep eutectic solvent extraction is an effective method to extract iridoid glycosides from P. medicinalis and they will be promising candidates to be developed for medical hemostasis agents. The Antimicrobial, Hemostatic, And Anti-Adhesion Effects of A Peptide Hydrogel Constructed by The All-D-enantiomer of Antimicrobial Peptide Jelleine-1

Hanru Zhang,Zhiyu Wu,Jingjing Zhou,Zhaopeng Wang,Changyan Yang,Panpan Wang,Muhammad Subaan Fareed,Yuhang He,Jie Su,Ruitao Cha,Kairong Wang

Peptide hydrogels were believed to be potential biomaterials with wide application in the biomedical field because of their good biocompatibility, injectability, and three-dimensional (3D) printability. Most of the previously reported polypeptide hydrogels are composed of L-peptides, while the hydrogels formed by self-assembly of D-peptides are rarely reported. Herein, we reported a peptide hydrogel constructed by D-J-1, which was the all-D-enantiomer of antimicrobial peptide Jelleine-1. Field emission scanning electron microscope (FE-SEM) and rheologic study were performed to characterize the hydrogel. Antimicrobial, hemostatic, and anti-adhesion studies were carried out to evaluate its biofunction. Our results showed that D-J-1 hydrogel was formed by self-assembly and cross-linking driven by hydrogen bonding, hydrophobic interaction, and π-π stacking force of aromatic ring in the structure of D-J-1. It exhibited promising antimicrobial activity, hemostatic activity, and anti-adhesion efficiency in a rat sidewall defect-cecum abrasion model. In addition, it also exhibited good biocompatibility. Notably, D-J-1 hydrogel showed improved in vitro and in vivo stability when compared with its L-enantiomer J-1 hydrogel. Therefore, the present study would provide new insight into the application of D-peptide hydrogel, and provide a new peptide hydrogel with antibacterial, hemostatic, and anti-adhesion efficacy for clinical use. A natural biological adhesive from snail mucus for wound repair

Deng Tuo,Gao Dongxiu,Song Xuemei,Zhou Zhipeng,Zhou Lixiao,Tao Maixian,Jiang Zexiu,Yang Lian,Luo Lan,Zhou Ankun,Hu Lin,Qin Hongbo,Wu Mingyi

The discovery of natural adhesion phenomena and mechanisms has advanced the development of a new generation of tissue adhesives in recent decades. In this study, we develop a natural biological adhesive from snail mucus gel, which consists a network of positively charged protein and polyanionic glycosaminoglycan. The malleable bulk adhesive matrix can adhere to wet tissue through multiple interactions. The biomaterial exhibits excellent haemostatic activity, biocompatibility and biodegradability, and it is effective in accelerating the healing of full-thickness skin wounds in both normal and diabetic male rats. Further mechanistic study shows it effectively promotes the polarization of macrophages towards the anti-inflammatory phenotype, alleviates inflammation in chronic wounds, and significantly improves epithelial regeneration and angiogenesis. Its abundant heparin-like glycosaminoglycan component is the main active ingredient. These findings provide theoretical and material insights into bio-inspired tissue adhesives and bioengineered scaffold designs.

Hydroxypropyl chitosan/ε-poly-l-lysine based injectable and self-healing hydrogels with antimicrobial and hemostatic activity for wound repair

Shuting Peng,Sen Niu,Qin Gao,Ruiyuan Song,Zhengxiao Wang,Ziyun Luo,Xi Zhang,Xiaofei Qin

The biggest obstacle to treating wound healing continues to be the production of simple, inexpensive wound dressings that satisfy the demands associated with full process of repair at the same time. Herein, a series of injectable composite hydrogels were successfully prepared by a one-pot method by utilizing the Schiff base reaction as well as hydrogen bonding forces between hydroxypropyl chitosan (HCS), ε-poly- l -lysine (EPL), and 2,3,4-trihydroxybenzaldehyde (TBA), and multiple cross-links formed by the reversible coordination between iron (III) and pyrogallol moieties. Notably, hydrogel exhibits excellent physicochemical properties, including injectability, self-healing, water retention, and adhesion, which enable to fill irregular wounds for a long period, providing a suitable moist environment for wound healing. Interestingly, the excellent hemostatic properties of the hydrogel can quickly stop bleeding and avoid the serious sequelae of massive blood loss in acute trauma. Moreover, the powerful antimicrobial and antioxidant properties also protect against bacterial infections and reduce inflammation at the wound site, thus promoting healing at all stages of the wound. The study of biohydrogel with multifunctional integration of wound treatment and smart medical treatment is clarified by this line of research.
