Hemostatic powders are widely used for managing bleeding from wounds with various irregular shapes. However, their limited liquid absorption capacity and difficulty in removal after application remain significant clinical challenges. Herein, we introduce a multifunctional hemostatic powder composed of dually crosslinked poly([2-(methacryloyloxy)ethyl]trimethylammonium chloride- co -acrylic acid) (pMATC- co -AA) and N -[(2-hydroxy-3-trimethylammonium)propyl] chitosan chloride (HTCC), which are integrated via electrostatic interactions and hydrogen bonding. This hemostatic powder exhibits superb liquid-absorbing capacity (94.5 times its own weight in water and 9.1 times in blood), sufficient adhesive properties (5.9 kPa on pig skin), and on-demand removability. It significantly reduces the number of viable Escherichia coli and Staphylococcus aureus by 4.61 and 4.04 orders of magnitude, respectively, thus providing an effective microbiological barrier for the wound. Furthermore, a series of in vivo and in vitro experiments confirm the powder's excellent hemostatic properties. The hydrogel formed on the wound after hemostasis can be removed by saline rinsing on demand, due to its superb liquid-absorbing capacity. Notably, the powder demonstrates good in vivo biocompatibility, with minimal risk of impeding wound healing, and it facilitates the healing process during the later stages. In sum, the hemostatic powder offers a promising solution for trauma bleeding control and acute wound treatment.

Hemostatic powder is widely employed for emergency bleeding control due to its ability to conform to irregularly shaped wounds, ease of use, and stable storage. However, current powders exhibit limited tissue adhesion and insufficient support for thrombus formation, making them easily washed away by blood. In this study, a hybrid powder (QAL) was produced by mixing quaternized chitosan (QCS) powder, catechol-modified alginate (Cat-SA) powder, and laponite (Lap) powder. Upon addition of QAL, the blood quickly transformed to a robust and adhesive blood gel. The adhesion strength of the blood gel was up to 31.33 ± 1.56 kPa. When compared with Celox, QAL showed superior performance in promoting hemostasis. Additionally, QAL exhibited effectiveness in eliminating bacteria while also demonstrating outstanding biocompatibility with cells and blood. These favorable properties, including strong coagulation, adhesion to wet tissue, antibacterial activity, biosafety, ease of use, and stable storage, make QAL a promising emergency hemostatic agent.

Hemostatic powder, which can absorb large amounts of water and tends to produce repeated hydration with tissue, has been clinically proven as an ideal engineering material for treating wounds and tissues. We herein designed a polypeptide-based hemostatic powder. A water-soluble polypeptide, γ-polyglutamic acid (γ-PGA), was mixed with the polyethyleneimine (PEI), N-hydroxysuccinimide, and 1-(3-dimethylaminopropyl)-3-ethylcarbodiimide. The solution of these polymers was lyophilized to harvest the γ-PGA/PEI powder (PP hemostatic powder). When deposited on a bleeding wound, the PP hemostatic powder can quickly absorb a large amount of blood and interstitial fluid, concentrate coagulation factors, coagulate blood cells, and eventually form a stable mechanical hydrogel. The wound bleeding time of the PP hemostatic powder group was 1.8 ± 0.4 min, significantly lower than that of the commercial chitosan hemostatic powder group (2.8 ± 0.4 min). The PP hemostatic powder was endowed with antioxidant capacity by introducing protocatechuic aldehyde, which can effectively inhibit inflammation and promote wound healing. Therefore, via preparation through a facile lyophilization method, the PP hemostatic powder is expected to find a wide application prospect as a qualified hemostatic powder. Keywords:hemostatic powder;hydrogel;γ-polyglutamic acid;polyethyleneimine;wound hemostasis

Uncontrollable bleeding and excessive blood loss are the primary causes of death on the battlefield and in accidents. However, commercial hemostatic materials possess individual drawbacks for hemostasis. Here, we present a rapid hemostatic powder (SACC-GO) with high biosafety, which is fabricated by a facile ball milling method using inexpensive raw materials (sodium alginate (SA), calcium chloride, cerium nitrate and graphene oxide (GO)). The SACC-GO could immediately become gel when meeting blood, whose strength was enhanced due to specific surface area of GO and high coordination capacity of Ce 3+ ions. In addition, the SACC-GO had a high water absorption rate (~2000 %) allow it to concentrate blood and release Ca 2+ ions to improve blood coagulation. The in vitro and in vivo results, such as hemostasis in tail amputation, femoral hemostasis and hepatic hemostasis, exhibited excellent hemostasis effects by reducing hemostasis time and blood loss (with the majority of reductions exceeding 50 %). The SACC-GO also enhanced wound healing due to Ce ions scavenging reactive oxygen species (ROS). Thus, the SACC-GO hemostatic powder had great potential for rapid hemostasis.

Hemostatic powders are widely used in incompressible or irregularly shaped bleeding wounds, but traditional hemostatic powders exhibit low adhesion, unsatisfactory hemostatic effect, limited infection control, and are not suitable for clinical or emergency situations. This study developed a novel self-gelling hemostatic powder (QTPM) consisting of quaternized cellulose (QC)/ tannic acid (TA)/ polyethylene glycol (PEG)/ montmorillonite (MMT). QTPM could absorb interfacial liquid hydrating to a stable hydrogel which form a switchable adhesion to tissues. Moreover, QTPM exhibits excellent antibacterial property by the synergistic effect of QC and TA. Furthermore, QTPM directly activate intrinsic and extrinsic coagulation hemostatic pathways to enhance hemostasis, and it concentrate coagulation factors. In vivo hemostasis study results show that QTPM significantly accelerated hemostasis and reduced blood loss compared with the blank group (>75 % reduction in hemostatic time, >85 % reduction in blood loss) in liver bleeding model (hemostasis time of 71.67 ± 7.09 s, blood loss of 19.23 ± 2.60 mg) and tail amputation model (hemostasis time of 91.03 ± 12.05 s, blood loss of 15.24 ± 1.77 mg). Therefore, the advantages of QTPM including rapid and effective hemostasis, easy usage, easy storability and adaptability make it a potential biomaterial for rapid hemostasis direction in the clinical setting.

Although hemostatic powders have excellent adaptability for irregular and inaccessible wounds, their hemostasis for continuous bleeding or bleeding wounds of non-compressible organs remains a critical challenge. Herein, a series of benzeneboronic acid-modified sodium alginate/catechol-modified quaternized chitosan (SA-BA/QCS-C, SBQCC) powders is developed by borate ester crosslinking for non-compressible hemorrhage control. SBQCC powders possess remarkable tissue adhesion, rapid self-gelation, good cytocompatibility and antibacterial activity against S. aureus and E. coil . The blood coagulation assays show that SBQCC powders display excellent blood clotting ability due to the synergistic effect of SA-BA and QCS-C. The SBQCC2 powder with the SA-BA to QCS-C mass ratio of 5 to 3 has the greatest effect on the blood-clotting rate. Upon depositing SBQCC2 powder to bleeding wounds of rabbit liver, the powder can absorb a large amount of blood and form a stable hydrogel physical barrier at the bleeding wounds in situ to achieve non-pressing rapid hemostasis. The SBQCC2 powder also has good biocompatibility and can be degraded in vivo. Altogether, the SBQCC powders can be a promising candidate for rapid hemostasis, and these findings may provide a new perspective for improving the hemostatic efficiency of the hemostatic powder in biomedical fields.

Uncontrolled hemorrhage remains a critical global health challenge, particularly in cases of non-compressible bleeding. This study presents a sequentially dual-crosslinked hemostatic powder that combines rapid hydration-activated electrostatic gelation with photo-triggered covalent reinforcement. The powder consists of negatively charged double bond-functionalized polyacrylic acid (PAA-db) and positively charged polyethyleneimine (PEI), enabling ultrafast fluid absorption and self-gelation upon contact with biological fluids. Subsequent light irradiation induces covalent crosslinking, enhancing the mechanical property. The powder-derived dual-crosslinked PAA-db/PEI (dPAA-db/PEI) hydrogel enables rapid and robust tissue adhesion. In the severe non-compressible hemorrhage models, the hemostatic powder outperforms clinical hemostatic agents (fibrin glue, CELOX powder), in terms of minimizing hemostatic time and blood loss. This hemostatic powder offers a promising solution for managing severe non-compressible hemorrhage, demonstrating rapid sealing, robust mechanical performance, and excellent biocompatibility.

Hemostatic powders with flexible shape are widely used for the noncompressible and inaccessible hemorrhage wounds. However, current hemostatic powders display poor wet tissue adhesion and fragile mechanical strength of the powder-supported blood clots , leading to compromised hemostasis efficacy. Herein, a bi-component of carboxymethyl chitosan (CMCS) and aldehyde-modified hyaluronic acid grafted with catechol groups (COHA) was designed. Upon absorption of blood, the bi-component powders (CMCS-COHA) spontaneously self-crosslinks into an adhesive hydrogel within 10 s, tightly adhering to wound tissue to form a pressure-resistant physical barrier. During gelation , the hydrogel matrix captures and locks the blood cells/platelets to generate a robust thrombus in the bleeding sites. Compared with traditional hemostatic powder Celox™, CMCS-COHA displays superior blood coagulation and hemostatic performance. More importantly, CMCS-COHA has inherent cytocompatibility and hemocompatibility. These prominent advantages in rapid and effective hemostasis, adaptability to fit irregulate defective wound, easy preservation, facile usage, and bio-safety, make CMCS-COHA a promising hemostatic in emergency situations.

Rapid and effective hemostasis on irregular-shaped non-compressible bleeding wounds is still one of the key challenges in clinical practice. Herein, a novel multi-functional microgel assembly powder (MAP) composed of oxidized dextran/methacrylate gelatin (ODex/GelMA) microgel powder and long-chain alkyl quaternized chitosan (LQCS) crosslinker powder is reported for acute hemostasis, antibacterial, and wound healing. When MAP is applied onto bleeding wounds, porous ODex/GelMA microgel powder immediately absorbs massive liquid in blood whilst LQCS induces in situ co-assembly of erythrocytes and microgels. In other words, MAP combines the merits of hemostatic powders and hemostatic hydrogels, that is, the excellent hemostasis performance of MAP is originated from not only the high fluid uptake ratio/rate that causes the aggregation of coagulation factors and erythrocytes, but also the formation of erythrocytes-involved hydrogel-like microgel assembly that dramatically improves the mechanical strength, tissue adhesion and wound sealing performance in blood compared with those in PBS. In addition, MAP also exhibits excellent antibacterial ability, and promotes the wound healing in bacterial-infected full-thickness skin wounds. The fantastic features of MAP including low cost, easy synthesis and usage, excellent hemostasis on irregular-shaped, non-compressible bleeding wounds, outstanding antibacterial and wound healing, good cytocompatibility, make it a promising hemostatic material and wound dressing.

Uncontrolled hemorrhage, especially in non-compressible and deep wounds, remains a critical issue in emergency and surgical care. Existing hemostatic powders often lack rapid gelation, mechanical robustness, and adequate adherence, increasing the risk of rebleeding under high-pressure blood flow. To address these limitations, PQPP, a novel self-gelling hemostatic material composed of polyacrylamide/quaternized chitosan coacervates and polydopamine nanoparticles is developed. PQPP can rapidly absorb blood within 2 s, undergoes in situ gelation, and forms a robust adhesive hydrogel to effectively seal wounds. Its efficacy stems from the electrostatic adsorption and catechol functional groups of polydopamine nanoparticles. Importantly, PQPP exhibits high burst pressure resistance, excellent blood cell aggregation capability, outstanding biocompatibility, and antibacterial properties. Comprehensive in vitro and in vivo studies, including cytotoxicity and blood compatibility tests, as well as trials in mouse liver, heart, and vascular injury models, demonstrate PQPP's superior hemostatic performance under high-pressure conditions without causing inflammation. With its rapid gelation, robust adhesion, and mechanical integrity, PQPP represents a promising hemostatic material for immediate wound management in surgical and emergency applications.

Hemostatic materials combining rapid bleeding control with regenerative potential remain a major clinical need. Here, we report a bioinspired hemostatic powder (GSHD) engineered through molecular conjugation of thiolated gelatin and dopamine-modified hyaluronic acid. Upon contact with biofluids, GSHD rapidly gels via dynamic thiol-catechol cross-linking, achieving ultrafast hydration (<5 s) and forming a robust hydrogel. This hydrogel exhibits high blood absorption (569.2 ± 46.3%), strong tissue adhesion (28.3 ± 3 kPa), and burst pressure resistance (216.2 ± 10.5 mmHg), enabling effective sealing under high-flow conditions. In vivo tests in rat liver, cardiac, and femoral artery models show rapid hemostasis (<30 s) with minimal blood loss. Additionally, GSHD promotes M2 macrophage polarization, enhances angiogenesis, and supports organized collagen deposition, facilitating tissue regeneration. By integrating rapid sealing, antioxidant activity, and biomimetic extracellular matrix cues, GSHD represents a paradigm shift in hemostatic design, bridging emergency care with functional tissue repair.
