Yarn-woven triboelectric nanogenerators (TENGs) have greatly advanced wearable sensor technology, but their limited sensitivity and stability hinder broad adoption. To address these limitations, Poly(VDF-TrFE) and P(olyadiohexylenediamine (PA66)-based nanofibers coaxial yarns (NCYs) combining coaxial conjugated electrospinning and online conductive adhesive coating are developed. The integration of these NCYs led to enhanced TENGs (NCY-TENGs), notable for their flexibility, stretchability, and improved sensitivity, which is ideal for capturing body motion signals. One significant application of this technology is the fabrication of smart insoles from NCY-TENG plain-woven fabrics. These insoles are highly sensitive and possess antibacterial, breathable, and washable properties, making them ideal for real-time gait monitoring in patients with diabetic foot conditions. The NCY-TENGs and their derivatives show immense potential for a variety of wearable electronic devices, representing a considerable advancement in the field of wearable sensors.

Incorporating biomass (e.g., bacterial cellulose, BC) pressure-sensing structures into plantar monitoring technologies could effectively defend users against insecure wear and inaccurate detection, while their capability in sensing pressure continuously when subjected to large pressure is hindered by their natural two-dimensional structure. Here, we present a microbial engineering strategy for fabricating hierarchical fully biomass pressure sensors (HFBPSs) with sufficient sensing capabilities and good structural stability for plantar pressure detection. Particularly, the incorporation of the biobased material loofah enables HFBPS to adopt a hierarchical structure from micro- and nanostructured sensing networks, allowing for dynamic changes in the series-parallel configuration of the circuit under large pressure. This innovative structure design breaks the bottleneck of low operating range encountered by traditional rigid two-dimensional BC pressure sensors, ensuring the HFBPS with ultrahigh sensitivity (2.82 kPa–1) and wide detection range (0–600 kPa). In addition to stably monitoring gait motions, the HFBPS integrated into smart insoles can recognize nonstandard Tai Chi movements. Our HFBPS with exceptional pressure-sensing performance provides significant impact on the future development of smart insoles for plantar monitoring.

Objective: The limited ability to regenerate axons after spinal cord injury (SCI) is influenced by factors such as astrocyte activation, reactive proliferation, and glial scar formation. The TGF-β/Smad (transforming growth factor-β/mothers against decapentaplegic homolog) pathway, associated with astrocytic scarring, plays a crucial role in recovery post-injury. This study aims to investigate how icariin (ICA) interacts with reactive astrocytes in the treatment of spinal cord injury. Methods: A rat SCI model was constructed, and the recovery of motor function was observed after treatment with ICA.HE staining, LFB staining, immunofluorescence staining, and Western blotting were employed to assess ICA's ability to inhibit astrocyte proliferation in rats following spinal cord injury by modulating YAP, as well as to evaluate the reparative effects of ICA on the injured spinal cord tissue. Primary astrocytes were isolated and cultured. Immunoprecipitation-Western Blot (IP-WB) ubiquitination and cytoplasm-nuclear separation were employed to assess PPM1B ubiquitination and nuclear translocation. Results: The CatWalk XT gait analysis, BBB (Basso, Beattie, and Bresnahan) score, electrophysiological measurements, HE staining, and LFB staining collectively demonstrated that ICA promotes motor function and tissue recovery following spinal cord injury in rats. Immunofluorescence staining and Western Blot analyses revealed that ICA inhibits astrocyte proliferation in rats post-spinal cord injury by suppressing YAP activity. Furthermore, the activation of YAP by XMU-MP-1 was shown to compromise the efficacy of ICA in these rats after spinal cord injury. Additional immunofluorescence staining and Western Blot experiments confirmed that ICA inhibits TGFβ1-induced astrocyte activation through the regulation of YAP. The knockdown of PPM1B (protein phosphatase, Mg2+/Mn2+-dependent 1B) in astrocytes was found to inhibit TGFβ signaling. Additionally, YAP was shown to regulate PPM1B ubiquitination and nuclear translocation through immunoprecipitation-Western blot analysis, along with the segregation of cytoplasm and nucleus. Conclusion: Icariin promotes functional recovery in rats after spinal cord injury by inhibiting YAP and regulating PPM1B ubiquitination to inhibiting the activation of reactive astrocytes.

Background: Oxidative stress and apoptosis of neurons significantly contribute to the pathophysiological cascade of spinal cord injury (SCI). However, the role of hypoxic-preconditioned mesenchymal stem cell-derived small extracellular vesicles (H-sEVs) in promoting SCI repair remains unclear. Hence, the present study aims to investigate the regulatory effects of H-sEVs on neuronal oxidative stress and apoptotic responses following SCI.Methods: The administration of H-sEVs of SCI rats was assessed using behavioral evaluations such as Basso-Beattie-Bresnahan (BBB) scores, neuroelectrophysiological monitoring, and Catwalk gait analysis. Indices of oxidative stress (including superoxide dismutase [SOD], total antioxidant capacity [T-AOC], and malondialdehyde [MDA]) were measured. Neuronal survival was evaluated through Nissl staining, while the expression level of sirtuin 1 (SIRT1) was examined using immunohistochemical staining. Additionally, histological evaluation of lesion size was performed using hematoxylin-eosin (HE) staining. Tunel cell apoptosis staining and analysis of apoptosis-associated proteins (B-cell lymphoma-2 [Bcl2] and BCL2-Associated X [Bax]) were conducted through immunofluorescence staining and western blot, respectively. Furthermore, the model of oxidative stress was established using PC12 cells, and apoptosis levels were assessed via flow cytometry and western blot analysis. Importantly, to ascertain the critical role of SIRT1, we performed SIRT1 knockout experiments in PC12 cells using lentivirus transfection, followed by western blot. Results: Using those behavioral evaluations, we observed significant functional improvement after H-sEVs treatment. Nissl staining revealed that H-sEVs treatment promoted neuronal survival. Moreover, we found that H-sEVs effectively reduced oxidative stress levels after SCI. HE staining demonstrated that H-sEVs could reduce lesion area. Immunohistochemical analysis revealed that H-sEVs enhanced SIRT1 expression. Furthermore, Tunel cell apoptosis staining and western blot analysis of apoptosis-related proteins confirmed the anti-apoptotic effects of H-sEVs. The PC12 cells were used to further substantiate the neuroprotective properties of H-sEVs by significantly inhibiting neuronal death and attenuating oxidative stress. Remarkably, SIRT1 knockout in PC12 cells reversed the antioxidant stress effects induced by H-sEVs treatment. Additionally, we elucidated the involvement of the downstream Nrf2/HO-1 signaling pathway. Conclusion: Our study provides valuable insights into the effects of H-sEVs on neuronal oxidative stress and apoptosis after SCI. These findings underscore the potential clinical significance of H-sEVs-based therapies for SCI.

Human gait is an important indicator for diagnosis of various movement-related disorders. Three-dimensional (3D) plantar force monitoring enables real-time recording of contact forces in both the normal and tangential directions in each step, providing useful clues for identification of irregular gait patterns or gait-related health issues. Most flexible sensors for gait monitoring focus on contact forces in the normal direction, being limited to detect forces in the tangential direction. Herein we have developed a porous elastomer with a cavity array (PECA) to measure 3D contact forces for gait analysis. The sensor consists of two fabric electrodes and a dielectric layer of porous Ecoflex elastomer containing a 5 × 5 cavity array, forming four parallel-plate capacitor units in a soft construction. Therefore, a 3D force in any arbitrary direction generates four unique capacitive electrical signals. Importantly, the PECA includes a well-designed combination of micrometer-scale pores and millimeter-sized cavities, dramatically improving the detection sensitivity and linear elasticity range. We have demonstrated high sensitivities of 0.41 N −1 and 0.23 N −1 in the normal and tangential directions, respectively. The flexible PECA-based sensors have been tested to sensitively distinguish various standing postures and different stride lengths for gait analysis, promising reliable translation into areas such as healthcare monitoring, sports training, and rehabilitation engineering.

Continuous and reliable monitoring of gait is crucial for health monitoring, such as postoperative recovery of bone joint surgery and early diagnosis of disease. However, existing gait analysis systems often suffer from large volumes and the requirement of special space for setting motion capture systems, limiting their application in daily life. Here, we develop an intelligent gait monitoring and analysis prediction system based on flexible piezoelectric sensors and deep learning neural networks with high sensitivity (241.29 mV/N), quick response (66 ms loading, 87 ms recovery), and excellent stability (R2 = 0.9946). The theoretical simulations and experiments confirm that the sensor provides exceptional signal feedback, which can easily acquire accurate gait data when fitted to shoe soles. By integrating high-quality gait data with a custom-built deep learning model, the system can detect and infer human motion states in real time (the recognition accuracy reaches 94.7%). To further validate the sensor’s application in real life, we constructed a flexible wearable recognition system with human–computer interaction interface and a simple operation process for long-term and continuous tracking of athletes’ gait, potentially aiding personalized health management, early detection of disease, and remote medical care.
