Omadacycline Efficacy against Streptococcus Agalactiae Isolated in China: Correlation between Resistance and Virulence Gene and Biofilm Formation

Guiqiu Li,Ying Wei,Yan Guo,Hui Gong,Jie Lian,Guangjian Xu,Bing Bai,Zhijian Yu,Qiwen Deng

This study aimed to evaluate the activity, resistance, clonality of MIC distribution, and the correlation between virulence and resistance genes and biofilm formation of omadacycline (OMC) in clinics for Streptococcus agalactiae isolates from China. 162 isolates were collected retrospectively in China. The S. agalactiae were collected from the body’s cervical secretions, wound secretions, ear swabs, secretions, semen, venous blood, cerebrospinal fluid, pee, etc. The MIC of OMC against S. agalactiae was determined by broth microdilution. The inhibition zone diameters of OMC and other common antibiotics were measured using filter paper. D-test was performed to determine the phenotype of cross resistance between erythromycin and clindamycin. In Multilocus sequence typing (MLST), some commonly-detected resistance genes and virulence gene of these S. agalactiae isolates were investigated using polymerase chain reaction (PCR). Biofilms were detected by crystal violet staining. Our data demonstrated the correalation of the biofilm formation and OMA antimicrobial susceptibility of S.agalactiae clinical isolates with the carrier of virulence gene scpB. Conclusively, OMC exhibits the robust antimcirobial activity against clinical S. agalactiae isolates from China compared with DOX or MIN, and the carrier of the virulence gene scpB might correlate with the biofilm formation in OMC-resistant S. agalactiae.

Identification of Virulence Genes and Antibiotic Resistance in Extraintestinal Pathogenic Escherichia coli Isolated from Broiler Carcasses Using MALDI-TOF MS

Jia-Tong Han,Yu-Xuan Tang,Si-Yi Wu,Yi-Ran Chen,Zhan-Peng Zou,Hang Zeng,Zhongjia Yu

Escherichia colicontamination in poultry is a significant concern due to its potential to cause foodborne illness. The presence of extraintestinal pathogenicE. coli(ExPEC) strains in chicken carcasses can lead to severe human infections. This study investigates the prevalence, virulence, and antibiotic resistance ofE. coliisolates from chicken carcasses processed in both wet market and industrial environments, with a focus on the detection capabilities of MALDI-TOF MS. A total of 119E. coliisolates were obtained. Only a small proportion (5/119) carried enteropathogenic virulence genes. In contrast, 71.42% (85/119) of the isolates harbored multiple extraintestinal virulence genes. Among these, iucCandsitA, which are associated with systemic infections, were present in 68.24% (58/85) and 43.53% (37/85) of the isolates, respectively. Furthermore, 47.06% (56/119) of the isolates carrying at least two extraintestinal virulence genes were classified as ExPEC. Additionally, 94.6% (54/56) of ExPEC isolates were multidrug resistant (MDR), showing resistance to over three antibiotic classes, raising concerns about the spread of antibiotic resistance. MALDI-TOF MS profiling revealed significant heterogeneity among the ExPEC isolates, with no distinct clustering patterns based on processing environment or sampling site. These findings underscore the public health risks posed by ExPEC in poultry and emphasize the need for improved surveillance, stringent hygiene practices, and responsible antibiotic use in poultry production.

Inhibition effect of aerobic respiratory Lactococcus lactis on Salmonella enterica subsp. enterica serovar Typhimurium SL 1344 and its intestinal colonization ability

Yulong Zhang,Suo Zheng,Jingru Chen,Zhiliang Cheng,Fei Liu,Yuehua Jiao

Lactococcus lactis undergoing aerobic respiration can consume oxygen and form an anaerobic environment. Salmonella , as a facultative anaerobic foodborne pathogen, multiply better under aerobic conditions. The purpose of this study was to explore aerobic respiration of Lactococcus lactis subsp. lactis ( L. lactis ) KLDS 4.0325 inhibitory effect on Salmonella enterica subsp. enterica serovar Typhimurium ( S. Typhimurium) SL 1344 in vitro and its colonization ability in vivo . Results indicate that L. lactis KLDS 4.0325 could inhibit the growth of S. Typhimurium SL1344 by producing bacteriocin. And L. lactis KLDS 4.0325 undergoing aerobic respiration could enhance this inhibitory effect, while reducing the expression levels of virulence genes sipB , sipC , and sopE2 in S. Typhimurium SL1344. L. lactis KLDS 4.0325 could be successfully labeled by CFDA-SE fluorescent probe, and the labeling rate was more than 97%. Murine experiments showed that L. lactis KLDS 4.0325 could adhere and colonize in the cecum and colon. Thus, L. lactis KLDS 4.0325 is a promising probiotic strain for the potential inhibition of Salmonella infection via its oxygen-scavenging in host gut.

Probiotic of Lactiplantibacillus plantarum NWAFU-BIO-BS29 Isolated from Chinese Traditional Fermented Milk and Its Potential Therapeutic Applications Based on Gut Microbiota Regulation

Mohamedelfatieh Ismael,Yaxin Gu,Yanlong Cui,Tao Wang,Fangfang Yue,Yanting Qin,Xin Lü

Lactic acid bacteria are one of the bioresources that can promote the host’s health and have potential therapeutic applications. This study aimed to evaluate the probiotic properties of novelLactiplantibacillus plantarumNWAFU-BISO-BS29 isolated in vitro from traditional Chinese fermented milk, assess its safety, and study its interaction with the gut microbiota using a BALB/c mouse model. The findings reveal that this strain had a high tolerance to gastric acidity (64.4%) and bile salts (19.83–87.92%) with remarkable auto-aggregation and co-aggregation abilities (33.01–83.96%), respectively. Furthermore, it lowered the cholesterol levels in dead cells (44.02%) and live cells (34.95%) and produced short-chain fatty acids (SCFAs). Likewise, it showed good antioxidant properties and strong antipathogen activity againstEscherichia coliandStaphylococcus aureuswith inhibition zones at 21 and 25 mm, respectively. The safety assessment results indicate that all of the virulence factor genes were not detected in the whole DNA; additionally, no hemolysis or resistance to antibiotics commonly used in food and feed was observed. Interestingly, the 16S rRNA gene sequencing of the mouse gut microbiota showed a marked alteration in the microbial composition of the administrated group, with a noticeable increase in Firmicutes, Patescibacteria, Campylobacterota, Deferribacterota, Proteobacteria, and Cyanobacteria at the phylum level. The modulation of gut microbial diversity significantly improved the production of SCFCs due to the abundance of lactobacillus genera, which was consistent with the functional gene predictive analysis and is believed to have health-promoting properties. Based on these results, our novel strain is considered a safe and good probiotic and could hold high potential to be used as a starter culture or to safely supplement functional foods as a probiotic and may provide new insights into therapeutic interventions. Keywords:probiotic;Lactiplantibacillus plantarum;virulence genes;antioxidants;hemolytic activity;cholesterol-lowering;gut microbiota

Mangrove plastisphere as a hotspot for high-risk antibiotic resistance genes and pathogens

Huan-Qin Li,Wen-Lei Wang,Ying-Jia Shen,Jian-Qiang Su

Microplastics (MPs) are critical vectors for the dissemination of antibiotic resistance genes (ARGs); however, the prevalence and ecological risks of high-risk ARGs in mangrove ecosystems—globally vital yet understudied coastal habitats—remain poorly understood. To address this gap, this study investigated polyethylene, polystyrene, and polyvinyl chloride incubated in mangrove sediments for one month, focusing on high-risk ARGs, virulence gene (VGs), and pathogenic antibiotic-resistant bacteria within the mangrove plastisphere. High-throughput PCR and metagenomic analyses revealed that high-risk ARGs, VGs, and mobile genetic elements (MGEs) were significantly enriched on MPs compared to surrounding sediments. Pathogenic bacteria and MGEs were also more abundant in the plastisphere, highlighting its role as a hotspot for ARG dispersal. Metagenome-assembled genome analysis identified Pseudomonas and Bacillus as key hosts for ARGs, MGEs, and VGs, particularly multidrug resistance genes, integrase genes, and adherence factors. Notably, polystyrene harbored the highest abundance of pathogenic bacteria carrying ARGs, MGEs, and VGs, and mangrove root exudates were found to amplify horizontal gene transfer on MPs, uncovering a previously overlooked mechanism driving antibiotic resistance in coastal ecosystems. These findings not only elucidate how MPs accelerate the spread of ARGs, but also underscore the urgent need for targeted mitigation strategies to address the adverse impacts microplastic pollution on human, animal, and environmental health.

Characterization of Clinically Relevant Strains of Extended-Spectrum β-Lactamase-Producing Klebsiella pneumoniae Occurring in Environmental Sources in a Rural Area of China by Using Whole-Genome Sequencing

Chi Xiaohui,Berglund Björn,Zou Huiyun,Zheng Beiwen,Börjesson Stefan,Ji Xiang,Ottoson Jakob,Lundborg Cecilia Stålsby,Li Xuewen,Nilsson Lennart E.

Klebsiella pneumoniae is a gram-negative, opportunistic pathogen, and a common cause of healthcare-associated infections such as pneumonia, septicemia, and urinary tract infection. The purpose of this study was to survey the occurrence of and characterize K. pneumoniae in different environmental sources in a rural area of Shandong province, China. Two hundred and thirty-one samples from different environmental sources in 12 villages were screened for extended-spectrum β-lactamase-(ESBL)-producing K. pneumoniae, and 14 (6%) samples were positive. All isolates were multidrug-resistant and a few of them belonged to clinically relevant strains which are known to cause hospital outbreaks worldwide. Serotypes, virulence genes, serum survival, and phagocytosis survival were analyzed and the results showed the presence of virulence factors associated with highly virulent clones and a high degree of phagocytosis survivability, indicating the potential virulence of these isolates. These results emphasize the need for further studies designed to elucidate the role of the environment in transmission and dissemination of ESBL-producing K. pneumoniae and the potential risk posed to human and environmental health.

Novel SCCmec type XII methicillin-resistant Staphylococcus aureus isolates identified from a swine production and processing chain

Wenyuan Zhou,Xinhui Li,Lei Shi,Hua H. Wang,He Yan

Methicillin-resistant Staphylococcus aureus (MRSA) has been a major public health concern. In this study, a total of 1485 samples from three swine farms, one slaughterhouse and one indoor market in Xiamen, China were collected in 2015, and the prevalence and profiles of MRSA were assessed. All the MRSA isolates were characterized by molecular typing, antibiotic susceptibility, coagulation activity, as well as PCR screening for 38 antibiotic resistance genes, two mobile genetic elements ( lsa (E)-containing multiresistance gene cluster and Tn 558 ), and 36 virulence genes. During the study, 54 of 1485 (3.6%) samples from the swine production, processing and retail chain were found positive for MRSA. A relatively rare SCC mec XII genotype was prevalent in swine farm (84.6%, 11/13) and slaughterhouse isolates (80.6%, 25/31), but absent in the market isolates (0%, 0/10). Notably, all staphylococcal cassette chromosome mec (SCC mec ) type XII MRSA isolates were resistant to at least 6 classes of antibiotics, carried two mobile genetic elements ( lsa (E)-containing multiresistance gene cluster and Tn 558 ) and harbored multiple virulence genes. These multidrug resistant MRSA isolates could also coagulate both bovine and caprine plasma. Our results on the SCC mec XII MRSA isolates, particularly their profiles of related genotypes, antibiotic resistance and virulence determinants, illustrated the evolvement of livestock-associated (LA)-MRSA in the swine production environment and spread along the processing chain. The dominance of the SCC mec XII in MRSA isolates found in this study, differed from previous reports from China, indicated potential contribution associated with the production process.

Hemolysin function of Listeria is related to biofilm formation: transcriptomics analysis

Li Ruidan,Liang Qian,Tian Sicheng,Zhang Yunwen,Liu Sijing,Ou Qian,Chen Zhaobin,Wang Chuan

Listeriolysin O (LLO) is the main virulence protein of Listeria monocytogenes (LM), that helps LM escape lysosomes. We previously found that the cellular immune response elicited by L.ivanovii (LI) is weaker than that elicited by LM. We speculated that this may be related to the function of ivanolysin O (ILO). Here, we constructed hemolysin gene deletion strain, LIΔ ilo , and a modified strain, LIΔ ilo :: hly , in which ilo was replaced by hly . Prokaryotic transcriptome sequencing was performed on LI, LIΔ ilo , and LIΔ ilo :: hly . Transcriptome differences between the three strains were compared, and genes and pathways with significant differences between the three strains were analyzed. Prokaryotic transcriptome sequencing results revealed the relationship of ilo to the ribosome, quorum sensing, and phosphotransferase system (PTS) pathways, etc. LIΔ ilo exhibited attenuated biofilm formation ability compared to LI. Biofilm formation was significantly recovered or even increased after replenishing hly . After knocking out ilo , the relative expression levels of some virulence genes, including sigB , prfA , actA , smcL , and virR , were up-regulated compared to LI. After replenishing hly , these genes were down-regulated compared to LIΔ ilo. The trend and degree of such variation were not completely consistent when cultured in media containing only monosaccharides or disaccharides. The results confirmed that hemolysin is related to some important biological properties of Listeria , including biofilm formation and virulence gene expression levels. This is the first comprehensive study on ILO function at the transcriptomic level and the first evidence of a relationship between Listeria hemolysin and biofilm formation.

Punicalagin inhibits biofilm formation and virulence gene expression of Vibrio parahaemolyticus

Hongli Liu,Wenxiu Zhu,Yu Cao,Jingzhu Gao,Tong Jin,Ningbo Qin,Xiaodong Xia

Vibrio parahaemolyticus is a major food-borne pathogen and the leading cause of seafood-associated enteric infections worldwide. In addition, imprudent use of antibiotics has contributed to increasing antibiotic resistance in V. parahaemolyticus. Therefore, there is an urgent need to develop novel control strategies to reduce V. parahaemolyticus contamination and infection. V. parahaemolyticus could form biofilm on different surfaces and contains several virulence factors, which together contribute to its persistence and pathogenicity. In this study, anti-biofilm and potential anti-virulence effects of punicalagin against V. parahaemolyticus were investigated. Punicalagin at sub-inhibitory concentrations (3.125–12.5 μg/mL) exerted significant anti-biofilm activity (20.9–74.4% reduction). It decreased bacterial motility, and attenuated metabolic activity of cells inside biofilm. Meanwhile, punicalagin inhibited EPS production and caused reduction of exopolysaccharides in biofilms, and decreased Raman peak intensities of different biofilm components. Moreover, punicalagin significantly inhibited V. parahaemolyticus adhesion to Caco-2 cells (23.8–68.1% reduction) and downregulated expression of genes involved in V. parahaemolyticus biofilm formation and virulence. These findings suggest that punicalagin could be potentially developed as an alternative strategy to control V. parahaemolyticus biofilms in food industry and mitigate diseases caused by this pathogen.

Genomic characteristics of a novel strain Lactiplantibacillus plantarum X7021 isolated from the brine of stinky tofu for the application in food fermentation

Guang Liu,Yafan Liu,Kum-Song Ro,Lei Du,Ya-Jie Tang,Li Zhao,Jingli Xie,Dongzhi Wei

Lactiplantibacillus plantarum X7021 is a novel strain originated from the brine of stinky tofu. In order to investigate the application potential of the strain in food industry, the genomic characteristics as well as some metabolic features were studied using comparison genomics and transcriptional assay. The complete genome of X7021includes a 3,407,054 bp chromosome with 3259 protein coding sequences, and 6 plasmids. L. plantarum strains, WCFS1, JDM1, and ST-III were selected for comparative genomic analysis. Results revealed that X7021 is safe for the application in foods with less antibiotic resistance and virulence genes. The strain has 25 complete phosphotransferase system transporters and strong proteolytic system for its adaptability to various foodstuffs. The strain containes a complete purine metabolic pathway incorporating 4 permeases, resulting in the reductions of 38.9% guanine, 26.1% hypoxanthine and 70.2% xanthine during the soymilk fermentation. Soy isoflavone glycosides could be converted to corresponding aglycones by two β-glucosidases. The strain could grow in 700 mg/L nitrite and entirely degrade 100 mg/L nitrite during 24 h based on highly active nitrite transporter and nitrite reductase. Accordingly, the results suggest that L. plantarum X7021 is a prospect starter candidate for the fermentation of plant foodstuffs.
