llicit drug detection is an important part of drug problem management, and existing detection strategies suffer from inefficiency and narrow scope of application, which cannot fully meet the needs of practical work. Here, a multi-color fluorescent carbon dots was synthesized in one step and used as probes for the fluorescence sensing detection of three habitual illicit drugs , heroin, ketamine and methamphetamine. The fluorescence response results were recorded in the form of image fixation and spectral detection, which contained the subtle fingerprint information of these illicit drugs. Further, a multi-modal detector based on multi-task deep learning was constructed to achieve qualitative and semi-quantitative illicit drug detection, with accuracies of 98.4 % and 84.4 % for image and 99.9 % and 99.6 % for spectral, respectively, and a broad detection range from 1 ng/mL to 1 mg/mL. The process showed that deep learning significantly enhanced the detection sensitivity and range of fluorescent probes , and the dual-modal image-spectrum recognition met the practical needs of illicit drug on-site fast detection and accurate analysis, providing a new technical solution for illicit drug detection.

Portable mass spectrometers have played an important role in the rapid field detection of drugs; however, interference from matrix components, such as salts, solvents, and other metabolites, often leads to high rates of false positives and false negatives, making it challenging to meet the requirements for rapid field detection. To mitigate these challenges, a solvent-assisted thermal desorption ionization source has been developed, which enables rapid screening, improved accuracy and quantitative MS analysis. This ionization source is designed to allow the introduction of sample solution into the thermal desorption chamber with coaxial solvent and gas flows. The sample is introduced into a closed high-temperature thermal desorption chamber by the coaxial gas flow, where it is rapidly vaporized, enabling a concentrated ionization and avoiding chemical interferences from the environment. The use of both a solvent flow and a guiding gas flow in the new ion source design effectively reduces the sample deposition on the inner surface of the glass liner in the thermal desorption chamber and minimize cross contamination and the interferences from salts in the sample, a significant 1.8–5 fold enhancement in the signal intensity of the drug was observed, and accommodates a large sample volume injection for a significantly increased sensitivity, repeatability, and accuracy of MS detection. By integrating this novel solvent-assisted thermal desorption ionization source with a portable mass spectrometer and employing a selected ion monitoring (SIM) mode, the linear dynamic ranges of detection for ketamine, cocaine, and morphine in the hair matrix have shown to be 1–20 ng/mg, with R 2 > 0.99. The recovery rates for the three tested illicit drugs are all within the range of 92.28 %–104.66 %. Both intra-day and inter-day precision remain to be below 10 %, indicating high measurement accuracy and stability. The limit of detection (LOD) for ketamine, cocaine, and morphine at 0.2 ng/mg has also been achieved, which is significantly better than those by using conventional methodologies. The technique developed through this study provides a reliable tool for in situ rapid analysis of illicit drugs in hair.

Fluorescent-colorimetric nanoparticles (FCNPs) attract considerable interest as an emerging dual-signal reporter for on-site qualitative/quantitative point-of-care testing. However, the suboptimal signaling components and self-assembled structure lacking physical isolation in traditional FCNPs result in low fluorescence brightness, poor stability, and strong internal filtration effect (IFE), which severely limits their wide application in lateral flow assay (LFA). Here, ordered self-assembly for hydrophobic upconversion nanoparticles (UCNPs) is developed using 3D porous space magnetic dendritic mesoporous silica (MS), stepwise surface silanization, and polydopamine (PDA) flexible scaffold modification to fabricate MS@UCNPs@PDA (MSUD). With rational design, MSUD improves stability and luminescence intensity (131 times higher than quantum dot-based fluorophores), and also eliminates IFE and fluorescence background interference on LFA strips. The detection limits of MSUD-labeled LFA for qualitative and quantitative detection of methamphetamine by naked eye-based colorimetric and smartphone-based fluorescence strategy are 1.047 × 10 4 pg mL −1 and 47.25 pg mL −1 , ≈10- and 2116- times lower than that of gold nanoparticles-LFA, respectively. The practicality of the MSUD-based LFA is validated in 83 urine/hair forensic samples, with the quantitative determination results in good agreement with the liquid chromatography-mass spectrometer data. This work presents an innovative strategy for constructing FCNPs, facilitating their progressive development and widespread applications.

Illicit drug abuse has brought big challenges to public health, because it has caused severe damage to the human central nervous system. The development of a rapid, quantified, and on-site detection of illicit drugs is of key importance for the control of drug abuse. However, it remains challenging due to biological sample interference and difficulties in achieving specific recognition of the drug molecule. Here, we report a molecule-responsive two-component surface-enhanced Raman scattering (SERS) platform for the capture and detection of illicit drugs. Under optimized conditions, the system demonstrates a linear response to cocaine concentrations ranging from 1 ppb to 10 ppm (R2 > 0.99), with an ultralow detection limit of 1.5 ppb. Successful application in spiked bovine serum validates the method’s robustness against complex biological matrices. This work establishes a novel paradigm for detecting trace-level illicit drugs with significant forensic potential. The design principle might be applied to other chemical detection.

In forensic drug analysis, detecting low concentrations of illicit drugs in samples presents a significant challenge. This paper introduces a novel approach for developing molecularly imprinted quantum dot materials (Co-AD QDs-MIPs) capable of efficiently detecting methamphetamine (METH) in artificial urine without the need for extraction or pretreatment. Using free radical polymerization, molecularly imprinted materials were coated onto the surface of quantum dots, forming a Co-AD QDs-MIPs fluorescence nanoprobe. The resulting Co-AD QDs-MIPs demonstrated enhanced specificity for METH detection and showed strong fluorescence quenching. Under optimal conditions, the method enabled selective and quantitative detection of METH. A good linear correlation was observed between the fluorescence quenching effect and increasing METH concentration in the range of 67–469 nM, with a detection limit of 23 nM. The Co-AD QDs-MIPs nanoprobe successfully detected METH in simulated human urine samples, with a recovery rate of 96.2% to 102.1%. Therefore, it is a simple, cost-effective, and selective nanoprobe for detecting low concentrations of METH.

The efficient detection of illicit drugs such as cocaine continues to be important for the fight against drug trafficking. Herein, we report a one-step method for rapid and specific cocaine detection. The method is based on our finding that small-molecule Thioflavin T (ThT) can act as a fluorescence indicator, which can be bonded with the anti-cocaine aptamer (MNS-4.1) to generate an enhanced fluorescence signal. More interestingly, upon cocaine binding, the intercalated ThT can be replaced, causing a drastic fluorescence reduction. We further optimized the sequence of MNS-4.1 and a new anti-cocaine aptamer (coc.ap2-GC) was obtained. This aptamer showed a higher affinity to both ligands, which increased the ThT binding fluorescence intensity and showed the highest quenching efficiency. Based on the fluorescence change induced by competitive binding, cocaine detection could be accomplished by a “mix-and-detect” strategy within seconds. Such a label-free method exhibits high sensitivity to cocaine with a low detection limit of 250 nM. Moreover, the practical sample analysis (2.5% human urine and saliva) also exhibits good precision and high sensitivity.

The sandwich-type lateral flow assays relying on dual aptamers with high sensitivity and specificity have been broadly explored. However, it is unlikely to match a pair of specific aptamers that can bind a small molecular target (e.g., cocaine) simultaneously due to the steric hindrance. In response, we herein introduced the strategy of “one divides into two” into the construction of sandwich-type lateral flow strip assay (LFSA). Specifically, we split a single cocaine-recognizing aptamer into two segments, either of which was conjugated with gold nanoparticles (AuNPs) or labeled with biotin, serving as signal probe and capture probe, respectively. Upon the presence of the target molecule, a ternary sandwich complex comprised of the two halves of the aptamer and the target formed. Such sandwich-type LFSA exhibited an excellent nonlinear logarithmic response in the range from 10 μmol/L to 5 mmol/L with R 2 = 0.9994. The sensitive on-site detection of cocaine in artificial biological samples including urine and sweat was achieved within 15 min, with the visual limit of detection as low as 50 μmol/L for urine and 200 μmol/L for sweat, and the recovery rates of 83.6–107.4%.
