As plastic pollution continues to increase and plastic waste is shredded to form smaller plastic particles, there is growing concern about the potential impact of nanoplastics (NPs) on freshwater ecosystems. In this work, the effects of three surface-modified NPs, including polystyrene (PS), PS-NH 2 , and PS-COOH, on the growth, photosynthetic activity, oxidative damage, and microcystins (MCs) production/release of Microcystis aeruginosa ( M. aeruginosa ) were investigated. Results indicated that all three NPs significantly inhibited the growth of M. aeruginosa after a 96 h exposure, and the growth inhibition followed the order of PS-NH 2 > PS > PS-COOH ( p < 0.05). Meanwhile, all three NPs at the concentration of 100 mg/L significantly increased the content of intra-MCs (115 %, 147 %, and 121 % higher than the control, respectively) and extra-MCs (142 %, 175 %, and 151 % higher than the control, respectively) after a 96 h exposure ( p < 0.05). Moreover, our findings also suggested that the potential mechanisms of surface-modified PS NPs on M. aeruginosa growth and MCs production/release were associated with physical constraints, photosynthetic activity obstruct, and oxidative damage. Our findings provided direct evidence for different kinds of surface modifications of PS NPs on freshwater algae and improve the understanding of the potential risk of NPs in aquatic ecosystems.

Gallic acid (GA) is an allelochemical that has been utilized in high concentrations for the management of harmful algal blooms (HABs). However, there is limited knowledge regarding its impact on the growth of M. aeruginosa as the GA concentration transitions from high to low during the HABs control process. This study has revealed that as the GA concentration decreases (from 10 mg/L to 0.001 μg/L), a dose-response relationship becomes apparent in the growth of M. aeruginosa and microcystin production, characterized by high-dose inhibition and low-dose stimulation. Notably, at the concentration of 0.1 μg/L GA, the most significant growth-promoting effect on both growth and MCs synthesis was observed. The growth rate and maximum cell density were increased by 1.09 and 1.16 times, respectively, compared to those of the control group. Additionally, the contents of MCs synthesis saw a remarkable increase, up by 1.85 times. Furthermore, lower GA concentrations stimulated the viability of cyanobacterial cells, resulting in substantially higher levels of reactive oxygen species (ROS) and chlorophyll-a (Chl a ) compared to other concentrations. Most importantly, the expression of genes governing MCs synthesis was significantly upregulated, which appears to be the primary driver behind the significantly higher MCs levels compared to other conditions. The ecological risk quotient (RQ) value of 0.1 μg/L GA was the highest of all experimental groups, which was approximately 30 times higher than that of the control, indicating moderate risk. Therefore, it is essential to pay attention to the effect of M. aeruginosa growth, metabolism and water ecological risk under the process of reducing GA concentration after dosing during the HABs control process.

Quaternary ammonium compounds (QACs) are commonly used in a variety of consumer and commercial products, typically as a component of disinfectants. During the COVID-19 pandemic, QACs became one of the primary agents utilized to inactivate the SARS-CoV-2 virus on surfaces. However, the ecotoxicological effects of QACs upon aquatic organisms have not been fully assessed. In this study, we examined the effects of a widely used QAC (benzalkonium chloride-C 14 , BAC-14) on two toxigenic Microcystis strains and one non-toxigenic freshwater Microcystis strain and carried out an analysis focused on primary, adaptive and compensatory stress responses at apical (growth and photosynthesis) and metabolic levels. This analysis revealed that the two toxic Microcystis strains were more tolerant than the non-toxic strain, with 96 hr-EC 50 values of 0.70, 0.76, and 0.38 mg/L BAC-14 for toxigenic M. aeruginosa FACHB-905, toxigenic M. aeruginosa FACHB-469, and non-toxigenic M. wesenbergii FACHB-908, respectively. The photosynthetic activities of the Microcystis , assessed via F v /F m values, were significantly suppressed under 0.4 mg/L BAC-14. Furthermore, this analysis revealed that BAC-14 altered 14, 12, and 8 metabolic pathways in M. aeruginosa FACHB-905, M. aeruginosa FACHB-469, and M. wesenbergii FACHB-908, respectively. It is noteworthy that BAC-14 enhanced the level of extracellular microcystin production in the toxigenic Microcystis strains , although cell growth was not significantly affected. Collectively, these data show that BAC-14 disrupted the physiological and metabolic status of Microcystis cells and stimulated the production and release of microcystin, which could result in damage to aquatic systems.

Bifidobacterium breve (YH68) is widely used in the fields of food fermentation and biomedicine. In this study, we explored the antibacterial activity of the cell free culture supernatant (CFCS) of YH68 against Clostridioides difficile ATCC 9689 (CD) by measuring multiple indexes, including the growth, spores production, toxin A/B production, and the expression levels of the tcdA and tcdB genes of CD. In addition, we examined the changes in major cellular functional groups, structures, permeability, integrity, and the proton motive force (PMF) of the cytoplasmic membrane. The results showed that double-dilution ratio of YH68-CFCS (3 × 109 CFU/mL) was the MIC value. The cell density, spores production, and the toxin production of CD treated with YH68-CFCS were lower than that of the control (p < 0.05). In addition, the gene expression levels of tcdA and tcdB in CD treated with YH68-CFCS were significant downregulated (p < 0.05). Marked differences were observed in the cell membrane and cell wall by a FT-IR spectroscopy and SEM. Analysis of the cell membrane permeability and integrity of the CD cells revealed that YH68-CFCS induced the leakage of a large amount of intracellular K+, inorganic phosphate, ATP, nucleic acids and proteinaceous substances. Furthermore, PMF analysis indicated that there was a significant change in Δψ and ΔpH. These findings demonstrated that the antibacterial activity of YH68-CFCS against CD involved the inhibition of growth, spore production, toxin production, and virulence genes expression; a consumption of PMF in the cytoplasmic membrane, the formation of pore in the cell membrane, together with the enhanced cell membrane permeability; and, eventually, cell completely disintegration.
