Paper-based analytical device for high-throughput monitoring tetracycline residue in milk

Huimin Lu,Miaosi Li,Azadeh Nilghaz,Lizi Li,Guangxue Chen,Yanbin Jiang,Junfei Tian

A low-cost and portable paper-based analytical device has been developed for high throughput and on-site monitoring TC residue in milk through visualized colorimetric reaction. The filtration and concentration effect induced by the porous nature of paper contribute to strengthen the color intensity, leading to quantitative and sensitive detection of tetracycline reaching 1 ppm detection limit, with the linear range of 1–100 ppm both in water and milk samples. The applicability was demonstrated by detection of TC in 18 different types of real milk samples with good recovery ranging from 88% to 113%. Furthermore, the dynamic degradation behavior of tetracycline was monitored through the device. To the best of our knowledge, this is the first report of colorimetric detection of tetracycline in milk using the paper-based device. This simple, fast, cost-effective (~$0.50 per device) and equipment-free paper-based platform provides a promising tool for future application in food and environmental safety.

Multiple Loci DNA Methylation Profiling for Rapid Colorectal Cancer Diagnosis Based on a Smartphone-Integrated Microfluidic Biosensing Platform

Shixin Cai,Ke Shi,Aohua Ji,Yilin Long,Xiaoli Wang,Yuting Zhang,Nandi Zhou

Methylation level of circulating tumor DNA (ctDNA) is widely recognized as a pivotal biomarker in precision oncology for tumor characterization and risk assessment. However, current point-of-care testing platforms for methylation analysis are constrained in achieving sufficient detection specificity due to their limitations in single-locus detection and their dependence on complex instrumentation. In this study, to overcome the above limitations, a smartphone-integrated, microfluidic chip technology-based biosensing platform was engineered for simultaneous analysis of multiple methylation loci in ctDNA. In this platform, whole blood samples from suspected individuals underwent GlaI methylation-sensitive enzyme digestion and target-specific primer hybridization, followed by microfluidic chip loading for on-chip colorimetric reactions. Methylation levels at specific loci were quantified through chamber-specific absorbance measurements by using the developed portable biosensing platform. The biosensor achieved simultaneous detection of three specific methylation loci (CpG sites) on the Septin 9 gene, which are associated with colorectal cancer. The detection limits (LODs) for the individual methylation loci are 0.25, 0.4, and 0.525 fM, respectively. This platform demonstrates high accuracy in detecting methylation signatures within blood samples from colorectal cancer patients, showing a significant correlation with conventional diagnostic methods. With its low cost and portable features, the developed biosensing platform may provide an economical solution for early-stage screening of cancer for the healthcare system in remote and under-resourced regions.

Evaporation-induced ion enrichment for high-performance glucose detection in a microfluidic thread/paper-based analytical system

Xiong Jing,Wu Liang,Xiao Gang,Ma Yan,Ran Ruilong,Sun Wei,Wang Wei,Mao Cuiping,Qiao Yan,Lu Zhisong

A microfluidic thread/paper-based analytical device (µTPAD) is of great significance in biomedical analysis and point-of-care diagnostics. Nevertheless, the research has not been done on the ion distribution on the thread and how it affects sensing so far. Inspired by the sweat stains on the clothes, a high-performance µTPAD based on the evaporation-induced ion enrichment is presented for glucose monitoring in salty solutions. In comparison to the multi-ply cotton thread, the single-ply cotton thread exhibits the most significant ion-enrichment capability at the rear segment, which could be attributed to the balance between the water evaporation and the solution replenishment in the thread. The salts accumulated at the rear segment could increase the ionic strength of the solution passed through the thread, promoting the colorimetric reaction on the paper-based sensing unit. The single-ply thread with strong ion-enrichment capability was integrated with a paper-based glucose sensor to prepare a µTPAD for highly sensitive glucose detection in both PBS and artificial sweat (-0.673µM −1 in PBS and − 0.253 µM −1 in artificial sweat). Moreover, the system could detect glucose in human sweat samples with high accuracy and precision. This work may provide a strategy to prepare a highly sensitive µTPAD for precise analysis of substances in bio-fluids, particularly human sweat. Graphical Abstract

Enzyme-Free Colorimetric Immunoassay for Protein Biomarker Enabled by Loading and Disassembly Behaviors of Polydopamine Nanoparticles

Li Li,Zhenyuan Xing,Qiaorong Tang,Lei Yang,Li Dai,Huan Wang,Tao Yan,Weiying Xu,Hongmin Ma,Qin Wei

In this work, a mussel-inspired polydopamine (PDA) nanoparticle with biocompatible and reactive surface characteristics was desirably incorporated with high loading capacity and pH-induced disassembly-releasing behavior toward Fe(III) ions to develop an enzyme-free colorimetric immunoassay. The catechol functional network of PDA can act as a scaffold to coordinate with large amounts of Fe(III) ions to form the PDA-Fe(III) NPs whose coordination state can be tailored by changing the pH values. In detail, PDA-Fe(III) NPs can be maintained in a disassembled tri-coordinate state under alkaline conditions while the highly loaded Fe(III) ions can be easily released under acid conditions to react with ferrocyanide for the in situ generation of Prussian blue (PB) NPs. The detection sensitivity of prostate-specific antigen (PSA) was significantly improved, owing to the high peroxidase-like activity of PB NPs that triggered excellent catalytic effect by the colorimetric reaction. In addition, favorable linearity was found in the range of 0.0005–20 ng mL–1 with a low detection limit of 0.84 pg mL–1 (S/N = 3). Furthermore, excellent selectivity and reproducibility were exhibited by the developed enzyme-free colorimetric immunoassay. It is believed that this proposed PDA-Fe(III) NP-based enzyme-free colorimetric system will offer a facile and reliable tool for the sensitive detection of PSA and other cancer biomarkers in human serum.
