Driven by economic interests, the abuse of antibiotics has become a significant concern for humans worldwide. As one of the most commonly used antibiotics, oxytetracycline (OTC) residue in animal-derived foods occurs occasionally, which has caused danger to humanity. However, there is still no simple and efficient solution to detect OTC residue. Here, an easily-operated colorimetric strategy for OTC detection was developed based on nickel ions (Ni2+) immobilized carboxymethylcellulose/polyacrylonitrile nanofibrous membranes (Ni@CMC/PAN NFMs). Owing to numerous O- and N-containing groups OTC has a strong tendency to complex with Ni2+on the strips, inducing a color change from light green to yellow visible to the naked eye. The NFMs structural features, CMC functionalization process, and Ni2+immobilization amount was carefully regulated to assure OTC detection whilst maintaining the inherent characteristics of NFMs. With the benefits of the large specific surface area (SSA) and small pore size of NFMs, the strips not only exhibited a rapid response (2 min), and low detection limit (5 nM) but also performed with good reversibility and selectivity concerning OTC detection over other antibiotics. The successful development of such enchanting nanofibrous materials may provide a new comprehension into the design and improvement of colorimetric strips. Keywords:nanofibrous membranes;nickel ions;colorimetric strips;oxytetracycline detection

The development of nanozymes with minimized color interference and target-specific recognition capability represents a critical advancement in colorimetric test strip. Here, white nanozymes are facilely synthesized by coordination polymerization. The optimization of surfactant modification endowed these nanozymes with enhanced alkaline phosphatase (ALP)-like activity by a larger specific surface area, and electrostatic interactions. Notably, the ALP-like activity of the nanozyme can be specifically inhibited by emamectin benzoate (EMB) rather than other pesticides. A reversible uncompetitive inhibition mechanism is systematically investigated at both enzymological and molecular levels, establishing a framework for studying nanozyme-small molecule interactions. Further, an EMB test strip is developed. Crucially, the whiteness of nanozyme effectively minimized nanozyme-induced background interference and shows a whitening effect on the paper substrate of test strip, significantly enhancing the visibility of colorimetric signal. Due to EMB-specific recognition, high ALP-like activity, and the whiteness of nanozyme, the developed nanozyme-based test strip demonstrated excellent selectivity, high sensitivity, and reliable performance for the real-time detection in real samples. This work not only demonstrates the seamless integration of white nanozymes with colorimetric test strips, but also provides a promising solution for developing enzyme-free, visual, and rapid detection platforms by screening highly specific nanozyme recognition elements for commercial pesticides.

Nanoparticle-natural enzyme complexes are receiving increasing attention as the promising signal reporters for colorimetric lateral flow immunoassay (LFIA). Nonetheless, it remains a challenge to develop the nanocomplexes with high loading efficiency, catalytic efficiency, and colorimetric signal brightness. Herein, inspired by pomegranate structure, we reported the synthesis of a colorimetric catalytic nanocomplex ((HRP@ZIF-8)3@PDA@HRP), using dopamine flexible scaffold-coated multishell porous zeolitic imidazolate framework-8 (ZIF-8) as a hierarchical scaffold to encapsulate horseradish peroxidase (HRP), and described its potential to promote an ultrasensitive colorimetric LFIA of cardiac troponin I (cTnI). (HRP@ZIF-8)3@PDA@HRP exhibited ultrahigh HRP loading efficiency and catalytic activity due to the epitaxial shell-by-shell overgrowth of porous ZIF-8 scaffold, which provided more cavities for enzyme immobilization and a diffusion path for the catalytic substrate. Furthermore, the polydopamine (PDA) layer on the (HRP@ZIF-8)3 surface both enhanced the colorimetric signal brightness and acted as a flexible scaffold to immobilize HRP, further increasing the amount of enzyme. Following integration with LFIA, the developed platform achieved an ultrasensitive colorimetric test strip assay for cTnI with pre- and postcatalytic naked-eye detection sensitivities of 0.5 ng mL–1 and 0.01 ng mL–1, respectively, which were 4/2- and 200/100-fold higher than gold nanoparticles (AuNPs)/PDA-based LFIA and comparable to chemiluminescence immunoassay. Further, the quantitative testing results of the developed colorimetric LFIA on 57 clinical serum samples agreed well with the clinical data. This work provides ideas for the design of natural enzymes-based colorimetric catalytic nanocomplex to encourage applications for the development of ultrasensitive LFIA for early diseases diagnosis.
