Transplantation of mesenchymal stem cells (MSCs) holds promise to repair severe traumatic injuries. However, current transplantation practices limit the potential of this technique, either by losing the viable MSCs or reducing the performance of resident MSCs. Herein, we design a “bead-jet” printer, specialized for high-throughput intra-operative formulation and printing of MSCs-laden Matrigel beads. We show that high-density encapsulation of MSCs in Matrigel beads is able to augment MSC function, increasing MSC proliferation, migration, and extracellular vesicle production, compared with low-density bead or high-density bulk encapsulation of the equivalent number of MSCs. We find that the high-density MSCs-laden beads in sparse patterns demonstrate significantly improved therapeutic performance, by regenerating skeletal muscles approaching native-like cell density with reduced fibrosis, and regenerating skin with hair follicle growth and increased dermis thickness. MSC proliferation within 1-week post-transplantation and differentiation at 3 − 4 weeks post-transplantation are suggested to contribute therapy augmentation. We expect this “bead-jet” printing system to strengthen the potential of MSC transplantation.

Multicellular spheroids can mimic the in vivo microenvironment and maintain the unique functions of tissues, which has attracted great attention in tissue engineering. However, the traditional culture microenvironment with structural deficiencies complicates the culture and collection process and tends to lose the function of multicellular spheroids with the increase of cell passage. In order to construct efficient and functional multicellular spheroids, in this study, a chitosan/polyvinyl alcohol nanofiber sponge which has an open-cell cellular structure is obtained. The hair follicle (HF) regeneration model was employed to evaluate HF-inducing ability of dermal papilla (DP) multicellular spheroids which formed on the cellular structure nanofiber sponge. Through structural fine-tuning, the nanofiber sponge has appropriate elasticity for the creation of a three-dimensional dynamic microenvironment to regulate cellular behavior. The cellular structure nanofiber sponge tilts the balance of cell–substratum and cell–cell interactions to a state which is more conducive to the formation of controllable multicellular spheroids in a short time. More importantly, it improves the secretory activity of high-passaged dermal papilla cells and restores their intrinsic properties. Experiments using BALB/c nude mice show that cultured DP multicellular spheroids could effectively enhance HF-inducing ability. This novel system provides a simple and efficient strategy for multicellular spheroid formation and HF regeneration.

Traditional wound dressings lack the activity to promote hair follicle regeneration during the treatment of deep burn skin injuries . In this study, a composite electrospun wound dressing (SZS + PG) containing strontium zinc silicon bioceramics (Sr 2 ZnSi 2 O 7 , SZS) was designed, which efficiently released bioactive Sr 2+ , Zn 2+ and SiO 3 2− ions. Owing to the synergistic effect of these three ions, SZS + PG not only significantly activated hair follicle stem cells around the burn wound, but also significantly promoted the regeneration of capillaries in the damaged area, thereby effectively promoting the hair follicle regeneration in burn wounds. This study confirmed that the combination of different functional ions is an effective way to facilitate burn wound healing with high qualitative hair follicle regeneration, which provided a new consideration for the development of novel wound dressings for burn wound treatment.

The pathogenesis of androgenetic alopecia (AGA) is driven by heightened androgen receptor signaling, alongside contributing factors of oxidative stress, follicular inflammation, and impaired perifollicular angiogenesis. Existing treatments such as minoxidil and finasteride suffer from limited efficacy, systemic side effects, and high relapse rates. In this study, we developed a rosemary oil (RO)-based microemulsion (QAR ME) for the co-delivery of quercetin (QT) and adenosine (AD) to synergistically target multiple pathological pathways of AGA. Here, RO can not only serve as a component of the oil phase but also function as a functional excipient with inherent antioxidant activity. The optimized QAR ME formulation demonstrated high stability, a small uniform particle size (24.98 ± 0.41 nm). It not only improved the solubility of QT but also enhanced the skin penetration of each active component. In vitro and in vivo evaluations revealed that QAR ME effectively attenuated oxidative stress, suppressed inflammatory responses, promoted angiogenesis, and activated the Wnt/β-catenin pathway, leading to enhanced hair follicle regeneration. It is worth noting that during the 28-day continuous treatment, QAR ME did not show any adverse reactions, which indicates that this therapy has the potential for long-term treatment of AGA. These findings indicate that QAR ME is a promising topical nanotherapy for the safe and effective treatment of AGA.

Infective diabetic wounds can significantly delay healing and pose severe infection risks. The process of infective diabetic wounds healing (DWH) are not only re-epithelialization and angiogenesis but also anti-bacterial, hair follicles regeneration and nerve growth. In this study, a stable dynamic hydrogel system was formulated by incorporating dynamic borate easter bond and carbon–carbon double bond, which based on tannic acid (TA) − acrylic phenyl boric acid (APBA) and acrylamide (AM). Subsequently, a hydrogel (ATAN-Met) was obtained by modifying the antibacterial ammonium cationic (N + ) and loading metformin (Met). This system demonstrated impressive wet adhesion, self-healing, wearable and peelable properties, responding to glucose/reactive oxygen species (ROS) for drug release. In vitro and vivo experiments illustrated that ATAN-Met recruited mesenchymal stem cell (MSC), facilitating interaction between MSC and neural cells, regulating macrophage polarization, and modulating the diabetic wound microenvironment while exhibiting anti-bacterial property, ultimately leading to re-epithelialization and neurovascular-hair follicles regeneration. RNA and metabolomic sequencing indicated that ATAN-Met activate the Hippo pathway, inhibit the TNF pathway, and regulate wound metabolism to improve the diabetic wound microenvironment. These results suggest that the microenvironment-responsive ATAN-Met hydrogel shows considerable potential for the whole course management of infective DWH.

The structural properties of biomaterials play crucial roles in guiding cell behavior and influencing immune responses against the material. We fabricated electrospun membranes with three types of surface topography (random, aligned, and latticed), introduced them to dorsal skin excisional wounds in mice and rats, and evaluated their effects on wound healing and immunomodulatory properties. An overview of different immune cells in the microenvironment with the help of single-cell RNA sequencing revealed diverse cellular heterogeneity in vivo. The time course of immune response was advanced toward an adaptive immunity–dominant stage by the aligned scaffold. In mice without mature T lymphocytes, lack of wound-induced hair neogenesis indicated a regulatory role of T cells on hair follicle regeneration. The microenvironment around scaffolds involved an intricate interplay of immune and cutaneous cells.

Intermittent fasting has gained global popularity for its potential health benefits, although its impact on somatic stem cells and tissue biology remains elusive. Here, we report that commonly used intermittent fasting regimens inhibit hair follicle regeneration by selectively inducing apoptosis in activated hair follicle stem cells (HFSCs). This effect is independent of calorie reduction, circadian rhythm alterations, or the mTORC1 cellular nutrient-sensing mechanism. Instead, fasting activates crosstalk between adrenal glands and dermal adipocytes in the skin, triggering the rapid release of free fatty acids into the niche, which in turn disrupts the normal metabolism of HFSCs and elevates their cellular reactive oxygen species levels, causing oxidative damage and apoptosis. A randomized clinical trial (NCT05800730) indicates that intermittent fasting inhibits human hair growth. Our study uncovers an inhibitory effect of intermittent fasting on tissue regeneration and identifies interorgan communication that eliminates activated HFSCs and halts tissue regeneration during periods of unstable nutrient supply.

Joint skin wounds are difficult to treat because of the frequent large motion of these active wounds; thus, dressings capable of simultaneous real-time monitoring of joint motion and promoting joint wound healing are highly needed. Herein, inspired by the aligned wrinkling microstructure and sensing functions of normal joint skins, we combine the electrospinning technique with a water-induced self-assembly approach to prepare bioinspired conductive dressings with aligned wrinkles for achieving the above-mentioned bifunctions. The results indicate that both the wavelength and height of the bioinspired aligned wrinkles can be facilely tuned by adjusting the thickness ratio of the two layers and the loading amount of conductive microparticles. Owing to the unique aligned wrinkling structure and good conductivity, the bioinspired dressing can monitor the donor student's diverse joint motions. Interestingly, the dressing can also accurately monitor different types of mouse neck motion, including up/down and left/right movements. The in vivo wound repairing results confirm that the bioinspired dressing can accelerate the healing of active wounds on the mouse neck by promoting collagen deposition, hair follicle regeneration, and epithelialization. The bioinspired dressing with the integration of real-time motion monitoring features and wound repairing functions will open a new avenue to improve the management of joint wounds.

Stem cell therapies have made great progress in the treatment of diabetic wounds during recent decades, while their short in vivo residence, alloimmune reactions, undesired behaviors, and dramatic losses of cell functions still hinder the translation of them into clinic. Here, inspired by the natural components of stem cell niches, we presented novel microfluidic hydrogel microcarriers with extracellular matrix (ECM)-like composition and adipose-derived stem cells (ADSCs) encapsulation for diabetic wound healing. As the hydrogel was synthesized by conjugating hyaluronic acid methacryloyl (HAMA) onto the Fibronectin (FN) molecule chain (FN-HAMA), the laden ADSCs in the microcarriers showed improved bioactivities and pro-regenerative capabilities. Based on these features, we have demonstrated that these ADSCs microcarriers exhibited significant promotion of neovascularization, follicular rejuvenation, and collagen deposition in a mouse diabetic wound model. These results indicated that the stem cell niche-inspired FN-HAMA microcarriers with ADSCs encapsulation have great clinical potential for diabetic wound treatment.

Background: The timely management of large-scale wounds and the regeneration of skin appendages constitute major clinical issues. The production of high-precision and customizable artificial skin via 3D bioprinting offers a feasible means to surmount the predicament, within which the selection of bioactive materials and seed cells is critical. This study is aimed at employing skin stem cells and multicomponent hydrogels to prefabricate artificial skin through 3D bioprinting, which enables the regeneration of skin and its appendages. Methods and Results: We employed gelatin methacrylate (GelMA) and hyaluronic acid methacrylate (HAMA) as bioactive materials, in conjunction with epidermal stem cells (Epi-SCs) and skin-derived precursors (SKPs), to fabricate artificial skin utilizing 3D bioprinting. The photosensitive multicomponent hydrogel, comprising 5% GelMA and 0.5% HAMA, demonstrated excellent printability, suitable solubility and swelling rates, as well as stable mechanical properties. Moreover, this hydrogel exhibited exceptional biocompatibility, effectively facilitating the proliferation of SKPs while maintaining the cellular characteristics of both SKPs and Epi-SCs. The transplantation of this artificial skin into cutaneous wounds in nude mice led to complete wound healing and functional tissue regeneration. The regenerated tissue comprised epidermis, dermis, hair follicles, blood vessels, and sebaceous glands, closely resembling native skin. Remarkably, the artificial skin demonstrated sustained tissue regeneration capacity even after 12 h of in vitro culture, facilitating comprehensive functional skin regeneration. Conclusions: Our research presented a skin repair strategy for prefabricated cell-loaded artificial skin, thereby successfully facilitating the regeneration of the epidermis, dermis, hair follicles, blood vessels, and sebaceous glands within the wound.

Wound repair is challenging for traditional wound dressings. New bioactive dressings need to be developed urgently. Herein, we reported a highly bioactive silk protein wound dressing (SPD) with natural silk fiber-sericin hydrogel interpenetrating double network structure, which combines the dual characteristics of natural silk and sericin hydrogel. Silk fiber scaffolds were secreted directly from silkworms bred by regulating their spinning behaviors. Sericin in SPD is obtained by dissolving silkworm cocoons at high temperature and high pressure, while it remains intact activities to self-assemble a hydrogel. To explore the effect of SPD, we first systematically evaluated its physicochemical properties and biological activities in vitro. The SPD exhibits high porosity, prominent mechanical strength, pH-responsive degradability, and excellent anti-oxidation and cell compatibility. Besides, SPD can load and maintain long-term drug release. Based on the satisfactory performance of SPD in vitro, effective in vivo treatment was achieved in a mouse full-thickness wound model, as demonstrated by a significantly accelerated wound healing process, promote the regeneration of hair follicles and sebaceous glands, increased expression of vascular endothelial growth factor, and reduced inflammation. Further, resveratrol was loaded into SPD to enhance the effects of anti-oxidation and anti-inflammation for wound healing. Our investigation shows that SPD with excellent physicochemical and biological properties applied in a murine full-thickness skin wound model resulted in remarkable and efficient acceleration of healing process, which may inspire the design of new, effective, and safer medical materials for tissue regeneration.

Severe burn injuries pose a critical challenge due to the destruction of neurovascular networks, which are indispensable for skin regeneration and often lead to incomplete healing and functional deficits. This study introduces an organic–inorganic nanocomposite hydrogel that orchestrates neurovascular network reconstruction to achieve full-thickness skin regeneration in burn wounds. Composed of gelatin-based matrices embedding mineralized metal-phenolic nanoparticles (MMF) and stem cell-derived exosomes (EXO), this “dual-engine” system synergizes MMF’s ion-releasing (Zn2+, SiO32–, tannic acid) and EXO’s bioactive signaling to overcome regeneration barriers. MMF stabilizes vascular architectures while exerting antioxidant and antibacterial effects, whereas EXO enhances endothelial cell migration and tube formation. In a murine deep second-degree burn model, the nanocomposite hydrogel could accelerate the wound closure rate (reaching 96.9% within 15 days, four times faster than controls), enhance collagen deposition, promote angiogenesis and innervation (increase vascular density by over 2-fold and nerve fiber regeneration by 3.5-fold), facilitate scarless wound healing (with epidermis thickness comparable to that of native skin), and stimulate skin appendage regeneration (the density of hair follicles/glands was approximately 7.8 times that of controls). In summary, our work offers an attractive approach to designing biomaterials that promote neurovascular regeneration and wound healing in burn injuries.
