Enzyme-Triggered Size-Switchable Nanosystem for Deep Tumor Penetration and Hydrogen Therapy

Yongju He,Xiangjie Tian,Xingyu Fan,Xiyu Gong,Songwen Tan,Anqiang Pan,Shuquan Liang,Hui Xu,Fangfang Zhou

The poor penetration of nanocarriers within tumor dense extracellular matrices (ECM) greatly restricts the access of anticancer drugs to the deep tumor cells, resulting in low therapeutic efficacy. Moreover, the high toxicity of the traditional chemotherapeutics inevitably causes undesirable side effects. Herein, taking the advantages of biosafe H2 and small-sized nanoparticles in diffusion within tumor ECM, we develop a matrix metalloprotease 2 (MMP-2) responsive size-switchable nanoparticle (UAMSN@Gel-PEG) that is composed of ultrasmall amino-modified mesoporous silica nanoparticles (UAMSN) wrapped within a PEG-conjugated gelatin to deliver H2 to the deep part of tumors for effective gas therapy. Ammonia borane (AB) is chosen as the H2 prodrug that can be effectively loaded into UAMSN by hydrogen-bonding adsorption. Gelatin is used as the substrate of MMP-2 to trigger size change and block AB inside UAMSN during blood circulation. PEG is introduced to further increase the particle size and endow the nanoparticle with long blood circulation to achieve effective tumor accumulation via the EPR effect. After accumulation into the tumor site, MMP-2 promptly digests gelatin to expose UAMSN loading AB for deep tumor penetration. Upon stimulation by the acidic tumor microenvironment, AB decomposes into H2 for further intratumor diffusion to achieve effective hydrogen therapy. Consequently, such a simultaneous deep tumor penetration of nanocarriers and H2 results in an evident suppression on tumor growth in a 4T1 tumor-bearing model without any obvious toxicity on normal tissues. Our synthetic nanosystem provides a promising strategy for the development of nanomedicines with enhanced tumor permeability and good biosafety for efficient tumor treatment.

Palladium Nanocrystals-Engineered Metal–Organic Frameworks for Enhanced Tumor Inhibition by Synergistic Hydrogen/Photodynamic Therapy

Jiajie Chen,Shiyang Lin,Doudou Zhao,Lei Guan,Yaping Hu,Yitong Wang,Kaili Lin,Yufang Zhu

Hydrogen therapy, as a star therapeutic modality, has recently acquired much attention in the field of anticancer medicine. Evidence suggests that hydrogen can selectively reduce intratumoral overexpressed hydroxyl radicals (•OH) to break the redox homoeostasis and thereby result in redox stress and cell damage. As a reactive oxygen species-related noninvasive modality, photodynamic therapy (PDT) has been approved for varied tumor treatments clinically. For implementing tumor therapy with enhanced anticancer efficacy and attenuated side effects, here a biocompatible palladium nanocrystals-integrated nanoscale porphyrinic metal–organic framework (NPMOF) is designed to develop a novel combined therapy modality, that is, synergistic hydrogen/photodynamic therapy. The NPMOF is employed simultaneously as the photosensitizer for PDT and as the nanocarrier to support palladium nanocrystals, which is further used as the hydrogen vehicle. The final hydrogen-containing nanosystems exhibit a persistent reductive hydrogen release behavior and considerable light-activated singlet oxygen ( 1 O 2 ) generation without mutual interference, contributing to the adequate disturbance of tumor microenvironment redox steady-state for synergistically inducing tumor cell death. Ultimately, by coupling of tumor-selective hydrogen therapy and PDT, the designed nanosystems realize the augmented therapeutic outcome with minimal side effects, providing a safe and efficient tumor treatment for future clinical translation.

A PdMo bimetallene with precise wavelength adjustment and catalysis for synergistic photothermal ablation and hydrogen therapy of cancer at different depths

Lei Bai,Wenhui Yi,Yanru Wang,Yilong Tian,Baona Zhou,Tao Yi,Ping Zhang,Xin Cheng,Jinhai Si,Xun Hou,Jin Hou

By delivering the idea of green and safe hydrogen energy and novel photothermal therapy to the biomedical field, engineering of therapeutic nanomaterials for treatment of major diseases (such as cancer) holds great significance. In this work, a novel PdMo bimetallene was synthesized by a solvothermal reduction method, and it was explored and applied in the field of anti-tumor therapy for the first time. The absorption peak of the PdMo bimetallene can be precisely adjusted in the NIR biological window (700–1350 nm) only by changing the synthesis time. At the same time, it also shows strong light absorption and high photothermal conversion efficiency. Specifically, the photothermal conversion efficiencies at 808 nm, 980 nm and 1064 nm are 43.1%, 51.7% and 69.15%, respectively. Surprisingly, a PdMo bimetallene is an efficient catalyst, which can effectively promote hydrogen production from the hydrolysis of ammonia borane (AB) under acidic and photothermal conditions. Benefitting from these excellent properties, a multifunctional composite nano therapeutic agent (PdMo@AB@HA) was developed via layer-by-layer surface modification with AB and hyaluronic acid (HA). In this way, the synergistic PTT/hydrogen therapy of PdMo@AB@HA composite nanosheets in the NIR-I and NIR-II windows (808 nm, 980 nm, and 1064 nm) on mouse tumor xenografts of different depths was realized. Furthermore, the controlled release of hydrogen, targeted endocytosis, efficient eradication of tumors of different depths and high biosafety were systematically proved in vitro and in vivo. This work not only provides a novel and efficient theranostic nanoplatform for efficient cancer theranostics, but also provides a new strategy for the development of safe and efficient new anti-tumor therapies.

Qing Wang,Daixing Zhang,Yining Qi,Changbao Huang,Dejun Ding,Chuanliang Liu

Introduction: The growing threat of bacterial infections poses a critical challenge to public health, underscoring the urgent need for innovative antibacterial agents and therapeutic strategies. In response, we have developed a multifunctional nanoplatform based on palladium-hydride metal–organic frameworks (P(H)ZPAg) for synergistic hydrogen and photothermal antibacterial therapy. Methods: This nanoplatform integrates palladium hydride (PdH) encapsulated within a zeolitic imidazolate framework (ZIF-8), surface modification with polydopamine (PDA), and in situ generation of silver nanoparticles (Ag NPs) to achieve enhanced antibacterial efficacy. Comprehensive characterization was performed to assess hydrogen release kinetics, photothermal performance, and silver-mediated bactericidal activity. The therapeutic potential of P(H)ZPAg was further evaluated in vivo using a Staphylococcus aureus-infected rat wound model. Results: The P(H)ZPAg nanoplatform demonstrated a successful combination of hydrogen release, photothermal conversion, and silver ion-based antibacterial mechanisms. In vitro assays revealed potent synergistic antibacterial effects against both Escherichia coli and Staphylococcus aureus. In vivo studies showed that treatment with P(H)ZPAg nanoparticles significantly enhanced wound healing and bacterial clearance compared to control groups.Discussion: These findings highlight the potential of combining hydrogen therapy, photothermal therapy, and silver ion release within a single nanoplatform to markedly improve antibacterial outcomes. This study presents a promising strategy for the development of multifunctional nanotherapeutics, offering a novel and effective approach for managing topical bacterial infections and promoting wound healing.

Silicone dressing combined with topical oxygen therapy alleviates incontinence‐associated dermatitis via NF‐κB p65/STAT1 signaling pathway

Guiyu an Chen,Yingxun Chen,Yan Zhang,Shufeng Zheng,Louying Zhu,Mingxing Ding

Background Incontinence‐associated dermatitis (IAD) is a tough problem in clinical settings, not only increasing the risk of complications like catheter‐related urinary tract infections and pressure ulcers in elderly and critically ill patients, but also prolonging hospital stays, raising hospital costs, and possibly leading to medical disputes. This study is aimed to evaluate the therapeutic effect of silicone dressing combined with topical oxygen therapy on IAD in a rat model. Methods An IAD rat model induced by synthetic urine with trypsin was established. Hematoxylin & eosin staining was carried out to examine skin histology. Using immunofluorescence, the microvessel density in the affected skin tissues was determined. ELISA was performed to measure the concentrations of inflammatory cytokines and angiogenic factors in serum. The mRNA expression of EGF, PDGF, and VEGF was detected via qRT‐PCR. Western blotting was employed to determine NF‐κB p65/STAT1 pathway‐related protein levels. Results Compared to single therapy, silicone dressing combined with topical oxygen therapy could significantly reduce the severity of IAD, improve skin histology, inhibit inflammation, and promote angiogenesis in IAD rat models. Additionally, the results showed that relatively speaking, the combined therapy suppressed the NF‐κB p65/STAT1 signaling pathway more effectively. Conclusion These findings indicated that silicone dressing combined with topical oxygen therapy can alleviate IAD through promoting wound healing and inhibiting inflammation via NF‐κB p65/STAT1 signaling pathway in a rat model, which provided a theoretical basis for the prevention and treatment of IAD in clinic.

Cold beer-inspired multifunctional nanozyme for ischemic stroke with rapid thrombus clearance and long-lasting hydrogen therapy

Jinlu Wei,Zhibin Zhou,Xinyue Pu,Xuefei Wu,Yuyang Zhang,Tingting Zhong,Wenjing Huang,Zhiwei Zhong,Xiaolei Wang

Ischemic stroke is a thrombus-inflammatory disease caused by cerebral vascular blockage. The narrow treatment window presents challenges for timely intervention and neurological recovery. Inspired by cold beer, we developed a multifunctional nanozyme (CREKA-MOF@Pd/H 2 -uPA, denoted as CMPd(H)U). After injection, CMPd(H)U efficiently targeted the thrombus and combined with gas therapy to expedite thrombolysis and restore blood flow. Additionally, CMPd(H)U released hydrogen (H 2 ) to eliminate reactive oxygen species (ROS), thereby mitigating reperfusion injury. Furthermore, CMPd(H)U demonstrated anti-inflammatory and neuroprotective properties by modulating microglial polarization towards the M2 phenotype and inhibiting neuronal pyroptosis. Animal behavioral experiments confirmed the effectiveness of the system in promoting neuromotor function recovery post-stroke.

Self-Assembled Nanoparticles of Silicon (IV)–NO Donor Phthalocyanine Conjugate for Tumor Photodynamic Therapy in Red Light

Kadireya Aikelamu,Jingya Bai,Qian Zhang,Jiamin Huang,Mei Wang,Chunhong Zhong

The combination of photodynamic therapy (PDT) and pneumatotherapy is emerging as one of the most effective strategies for increasing cancer treatment efficacy while minimizing side effects. Photodynamic forces affect nitric oxide (NO) levels as activated photosensitizers produce NO, and NO levels in the tumor and microenvironment directly impact tumor cell responsiveness to PDT. In this paper, 3-benzenesulfonyl-4-(1-hydroxy ether)-1,2,5-oxadiazole-2-oxide NO donor–silicon phthalocyanine coupling (SiPc–NO) was designed and prepared into self-assembled nanoparticles (SiPc–NO@NPs) by precipitation method. By further introducing arginyl-glycyl-aspartic acid (RGD) on the surface of nanoparticles, NO-photosensitizer delivery systems (SiPc–NO@RGD NPs) with photo-responsive and tumor-targeting properties were finally prepared and preliminarily evaluated in terms of their formulation properties, NO release, and photosensitizing effects. Furthermore, high reactive oxygen species (ROS) generation efficiency and high PDT efficiency in two breast cancer cell lines (human MCF-7 and mouse 4T1) under irradiation were also demonstrated. The novel SiPc–NO@RGD NPs show great potential for application in NO delivery and two-photon bioimaging-guided photodynamic tumor therapy.

Modulating the Electronic Structure of MnNi2S3 Nanoelectrodes to Activate Pyroptosis for Electrocatalytic Hydrogen-Immunotherapy

Jingrui Li,Gang Wang,Zhaoyu Wen,Shumin Sun,Zhihui Han,Yuqi Yang,Jie Wu,Zifan Pei,Luyao Liu,Youdong Chen,Liang Cheng

Hydrogen (H 2 ) therapy has demonstrated antitumor effect, but the therapeutic efficacy is restricted by the low solubility and nontarget delivery of H 2 . Electrolysis of H 2 O by electrocatalysts sustainably releases enormous amounts of H 2 and inspires the precise delivery of H 2 for tumor therapy. Herein, manganese-doped Ni 2 S 3 nanoelectrodes (MnNi 2 S 3 NEs) are designed for the electrocatalytic delivery of H 2 and the activation of antitumor immunity to effectively potentiate H 2 -immunotherapy. Ni atoms featuring empty 3d orbitals reduce the initial energy barrier of the hydrogen evolution reaction (HER) by promoting the adsorption of H 2 O. Moreover, Mn atoms with different electronegativity modulate the electronic structure of Ni atoms and facilitate the desorption of the generated H 2 , thus enhancing the HER activity of the MnNi 2 S 3 NEs. Based on the high HER activity, controllable delivery of H 2 for electrocatalytic hydrogen therapy (EHT) is achieved in a voltage-dependent manner. Mechanistically, MnNi 2 S 3 NE-mediated EHT induces mitochondrial dysfunction and oxidative stress, which subsequently activates pyroptosis through the typical ROS/caspase-1/GSDMD signaling pathway. Furthermore, MnNi 2 S 3 NE-mediated EHT enhances the infiltration of CD8 + T lymphocytes into tumors and reverses the immunosuppressive microenvironment. This work demonstrates an electrocatalyst with high HER activity for synergistic gas-immunotherapy, which may spark electrocatalyst-based tumor therapy strategies.

Hydrogen Therapy and Lipopolysaccharide Trapping with MgB2@Inulin Gel for Reversing Colitis and Its Associated Psychiatric Complications via the Gut–Brain Axis

Yaoyu Hu,Pengfei Zhu,Junwei Deng,Yi Yu,Qian Chen,Haitao Wu,Zhengbao Zha,Hua Wang,Yan Ma

Individuals diagnosed with inflammatory bowel disease (IBD) frequently encounter dysbiosis of the intestinal microbiota and are predisposed to psychiatric comorbidities, such as depression and anxiety. The development of oral therapeutics that concurrently attenuate intestinal inflammation, modulate gut microbiota, and ameliorate psychiatric symptoms is imperative for effective IBD management. This study introduced an orally administered Inulin gel incorporating magnesium diboride nanosheets (MgB2@Inulin gel). The hydrolysis of MgB2 nanosheets could generate reductive hydrogen gas (H2), which is critical for scavenging excess reactive oxygen/nitrogen species (RONS), and boron ions that neutralize harmful lipopolysaccharides (LPS). Prebiotic Inulin could effectively protect MgB2 nanosheets from gastric acid degradation, ensuring their persistence in the intestinal tract, where it modulated the gut microbiota and preserved intestinal barrier integrity. Notably, MgB2@Inulin gel markedly alleviated anxiety and depressive behaviors in the model of colitis by regulating the gut–brain axis. Thus, this work underscores the pivotal role of the gut–brain axis in the therapeutic approach to IBD and its associated psychiatric manifestations, proposing a straightforward, safe, and efficacious oral hydrogen treatment strategy for IBD and concurrent mental health disorders.

Calcium Peroxide-Based Hydrogel Patch with Sustainable Oxygenation for Diabetic Wound Healing

Na Li,Xiaohuan Lu,Yueying Yang,Shan Ning,Ye Tian,Mengyuan Zhou,Zheng Wang,Lin Wang,Jianfeng Zang

Nonhealing diabetic wounds are predominantly attributed to the inhibition of angiogenesis, re-epithelialization, and extracellular matrix (ECM) synthesis caused by hypoxia. Although oxygen therapy has demonstrated efficacy in promoting healing, its therapeutic impact remains suboptimal due to unsustainable oxygenation. Here, this work proposes an oxygen-releasing hydrogel patch embedded with polyethylene glycol-modified calcium peroxide microparticles, which sustainably releases oxygen for 7 days without requiring any supplementary conditions. The released oxygen effectively promotes cell migration and angiogenesis under hypoxic conditions as validated in vitro. The in vivo tests in diabetic mice models show that the sustainably released oxygen significantly facilitates the synthesis of ECM, induces angiogenesis, and decreases the expression of inflammatory cytokines, achieving a diabetic wound healing rate of 84.2% on day 7, outperforming the existing oxygen-releasing approaches. Moreover, the proposed hydrogel patch is designed with porous, soft, antibacterial, biodegradable, and storage stability for 15 days. The proposed hydrogel patch is expected to be promising in clinics treating diabetic wounds.
