A highly water-soluble hydrogen-bond-induced emission carbon dots for ratiometric fluorescent detection of water content in organic solvents

Yanfei Jia,Bing Han,Xiao-Ting Liu,Yuhao Liu,Yuanqiang Sun,Chao Lu

Water in organic solvents is a prevalent impurity that significantly influences chemical reactions and industrial processes. Carbon dots (CDs) gained attention as promising tools for chemosensing due to their advantageous characteristics, including easy synthesis, cost-effectiveness, and excellent adjustability and stability. However, limited solubility in water and turn off fluorescence response mode hinder the practical utilization of CDs for water sensing. To tackle such dilemma, a highly water-soluble CDs with distinctive hydrogen-bond-induced emission (HBIE) was rationally designed through introducing sulfone group into the widely employed p -phenylenediamine precursor. The inclusion of sulfone group imparts the CDs with notable water solubility, as well as distinctive ratiometric fluorescent response towards water content, exhibiting high sensitivity and selectivity. Upon exposure to water, the emission color of CDs undergoes a real-time transition from blue to yellow-green, which can be readily discerned by naked eyes. The CDs have been successfully applied in detecting water in commercially available alcohol. This study presents a straightforward yet efficacious approach for rationally design of CDs with unique HBIE characteristics and ratiometric fluorescent response, offering great potential for practical chemosensing applications.

Hydrogen-Bond-Induced Emission of Carbon Dots for Wash-Free Nucleus Imaging

Haifang Liu,Jie Yang,Zhaohui Li,Lehui Xiao,Aaron Albert Aryee,Yuanqiang Sun,Ran Yang,Hongmin Meng,Lingbo Qu,Yuehe Lin,Xiaobing Zhang

Carbon dots (CDs) are emerging as powerful tools for biosensing and bioimaging because of their intrinsic properties such as abundant precursors, facile synthesis, high biocompatibility, low cost, and particularly robust tunability and stability. In this work, a new type of CDs was prepared from m-phenylenediamine and folic acid by hydrothermal method. Interestingly, the as-prepared CDs show blue emission in non-hydrogen-bonding solution, whereas robust green emission in hydrogen-bonding solution. Based on this phenomenon, a novel fluorescence sensing mechanism named as hydrogen-bonding-induced emission (HBIE) was proposed. The HBIE-CDs have large Stokes shift (141 nm) in water, good biocompatibility, and ultrasmall size, which facilitates their translocation into living cells. Very importantly, the as-prepared HBIE-CDs show strong affinity toward nucleic acid without interference from other biological species. After binding with DNA/RNA through hydrogen bond, as high as 6-fold green fluorescence enhancement of HBIE-CDs was observed. Since the nucleus is rich in DNA/RNA, these HBIE-CDs were successfully used for rapid and, especially, wash-free subcellular in situ imaging of the nucleus in living cells in a fluorescence turn on mode, which has a great practicability to be used for nucleus imaging in bioanalytical studies and clinical applications.

Hydrogen bond-induced bright enhancement of fluorescent silica cross-linked micellar nanoparticles

Fangyuan Gai,Mengmeng Fan,Xudong Yang,Ce Hao,Yuhui Ao,Chunying Duan,Yunling Liu,Qisheng Huo

This work demonstrated the synthesis and design of ultra-bright and ultra-small fluorescent nanoparticles , which were prepared by encapsulating 4-(diphenylamino)benzaldehyde (DPB) in silica cross-linked micellar nanoparticles (SCMNPs). The DPB-doped SCMNPs (DPB-SCMNPs) exhibited ultra-bright fluorescence in an aqueous medium that was 22 times brighter than that of free DPB molecules in an organic solvent. For the first time, density functional theory (DFT) and time-dependent density functional theory (TDDFT) calculations were used to confirm that the enhanced brightness of the DPB-SCMNPs was due to a hydrogen bond-induced mechanism. In addition, the 3D fluorescence spectra and the Commission Internationale de L'Eclairage (CIE) diagram were employed to determine the optical properties and emission colour of the DPB-SCMNPs. Moreover, the DPB-SCMNPs were water-soluble, monodisperse and ultra-small (∼12 nm) and should be robust and stable in aqueous media and biological systems.

A ratiometric fluorescent dark box and smartphone integrated portable sensing platform based on hydrogen bonding induction for on-site determination of enrofloxacin

Qi Pu,Chumeng Wang,Xinyue Yin,Nengsheng Ye,Lu Zhang,Yuhong Xiang

Enrofloxacin (ENR) residues in animal-derived food and water threaten human health. Simple, low-cost and on-site detection methods are urgently needed. Blue emitting carbon quantum dots (CQDs) and orange rhodamine B (RhB) were used as recognition and reference signals, respectively, to construct a ratiometric fluorescence sensor. After the addition of ENR, the color of the sensor changed from orange to blue because hydrogen bonding induced a considerable increase in CQDs fluorescence. Based on this mechanism, a simple and low cost on-site portable sensing platform was constructed, which integrated a stable UV light strip and a smartphone with voice-controlled phototaking function and an RGB app. The t -test results of spiked ENR recoveries for diluted milk, honey and drinking water revealed no significant differences between the ratiometric fluorescent sensor and portable sensing platform. Thus, this portable sensing platform provides a novel strategy for on-site quantification of quinolone antibiotics in foodstuffs and environmental water.
