Combining the internal force-driven chemodynamic therapy (CDT) and the external energy-triggered photodynamic therapy (PDT) holds great promise to achieve an advanced anticancer effect based on reactive oxygen species (ROS). However, the insufficient oxy-substrates supply in tumor microenvironment, like hydrogen peroxide (H 2 O 2 ) and oxygen (O 2 ), is the Achilles heel that greatly restricts the efficacy of this ROS-based treatment. Herein, the construction of a c opper p e roxide-based tumor pH-re s ponsive a utocatalytic nano r eactor (CESAR), via an albumin-mediated biomimetic mineralization strategy is described. The decoration of human serum albumin endows the nanoreactor good hydrophilicity and biocompatibility, which is highly desired for the metal-based materials. Upon exposure to acidic tumor microenvironment, CESAR presents a pH-triggered disintegration with Cu 2+ , H 2 O 2 and O 2 generated instantly. The generated H 2 O 2 complements the hyperoxide deficiency and initiates a localized Fenton-like reaction with the assistance of Cu 2+ for highly toxic hydroxyl radicals (•OH) production for improving CDT. The evolved O 2 gas enables hypoxia relief for enhanced Ce6-mediated PDT. This H 2 O 2 /O 2 self-supplying strategy significantly amplifies the tumor oxidative damage and gains an optimal treatment outcome, which offers a new paradigm for optimizing the tumor therapeutic options limited by oxide or hyperoxide deficiency, not only for CDT/PDT, but also other oxy-substrates involved strategies. Statement of significance The shortage of oxy-substrates in the tumor microenvironment remains a great challenge for ROS-based cancer therapy. Herein, we introduce human serum albumin as a scaffold to stabilize copper peroxide nanomaterials for constant production of H 2 O 2 and O 2 to enhance chemodynamic/photodynamic therapy. The tumor pH-triggered H 2 O 2 /O 2 production and Cu 2+ release are confirmed, assuring the strategy of a highly precise, effective way to destroy tumor without any side effects. This work lends new and exciting insights into the engineering design of autocatalytic oxy-substrates self-supply nanoreactor for overcoming the bottlenecks, like the oxy-substrates deficiency of CDT/PDT and the poor stability of metal peroxides, to achieve highly effective chemodynamic/photodynamic therapy.

Diabetic wounds present multiple functional impairments, including neurovascular dysregulation, oxidative imbalance, and immune dysfunction, making wound healing particularly challenging, while traditional therapeutical strategies fail to address these complex issues effectively. Herein, we propose a strategy utilizing dual-layer microneedles to deliver therapeutic gases by modulating neurovascular coupling and immune functions for diabetic wound treatment. The microneedle can respond to reactive oxygen species (ROS) in the diabetic microenvironment and subsequently generate oxygen (O 2 ) and nitric oxide (NO). These gases comprehensively promote neuro-vascular regeneration, reduce oxidative stress levels, and attenuate inflammation. In vivo studies demonstrate that the microneedle can accelerate diabetic wound healing by modulating neurovascular regeneration and inflammatory processes. Transcriptomic analyses further validate the involvement of related advantageous signaling pathways. The potential mechanism involves the activation of the PI3K-AKT-mTOR pathway to facilitate autophagy, ultimately accelerating the healing process. Thus, our multifunctional dual-layer microneedles provide an effective strategy for treating diabetic wounds.

Periodontitis, a chronic inflammatory condition associated with bacterial infection, is the leading cause of tooth loss in adults. Pathogenic bacteria that persist in deep periodontal pockets with hyperglycemic levels following traditional manual debridement can contribute to a persistent local inflammatory microenvironment, posing a significant challenge and highlighting the critical need for improved therapeutic approaches. In addition, the anaerobic environment during these treatments is beneficial to the colonization of anaerobes and may amplify inflammatory responses. Herein, we designed a glucose oxidase (GOx)-coating MXene-SnS 2 bio-heterojunctions (MS@G bio-HJs) for multi-modal and effective treatment of anti-bacteria and anti-inflammatory. Upon the hyperglycemic nidus in periodontal pockets, GOx catalyzes the conversion of glucose into hydrogen peroxide (H 2 O 2 ). Subsequently, the breakdown of H 2 O 2 into hydroxyl radicals through Sn 2+ /Sn 4+ -involved Fenton-like reactions enables in situ liberation of oxygen (O 2 ), contributing to relieve local hypoxia. The proposed bio-HJs presents a satisfactory bactericidal capability with anti-bacterial rates of more than 99 % upon near-infrared irradiation, in virtue of multi-modal therapies including photodynamic, photothermal and chemodynamic treatments. Furthermore, in vivo assays utilizing infectious rat models demonstrated that the proposed bio-HJs exhibits high biocompatibility and capacity to alleviate hypoxia and reduce pathogenic virulence factors, potentially contributing to periodontal tissue regeneration. These findings suggest that the versatile bio-HJs could be a promising therapeutic candidate for treating infectious periodontal diseases.

Bacterial infection is a growing public health concern and causes a huge medical and financial burden. It is of significance to efficiently construct multifunctional platforms for bacterial point-of-care testing (POCT) and elimination. Herein, near-infrared (NIR) light-responded vancomycin-doped prussian blue nanoparticles (PB-VAN NPs ) with high efficient photothermal conversion was synthesized for binding, dual-mode portable detection, and elimination of bacteria. The PB-VAN NPs can bind to the surface of Gram-positive bacteria such as Staphylococcus aureus ( S. aureus ), forming complex of PB-VAN NPs / S. aureus . After being centrifugated, the suspension solution of PB-VAN NPs can stimulate perfluorohexane (PFH) to rapidly release oxygen (O 2 ) under NIR irradiation. Thus, the bacteria can be sensitively detected with portable pressure meter as signal reader, reporting a limit of detection (LOD) of 1.0 CFU mL -1 . On the other side, the sediment of PB-VAN NPs / S. aureus can be detected via thermal camera, reporting a LOD of 1.0 CFU mL -1 . Interestingly, the bacteria can be effectively inactivated with the local temperature elevation during temperature-based detection. The antibacterial efficiency reaches as high as 99.8%. The developed multifunctional nanoplatform not only provides a straightforward “mix-then-test” way for portable detection of bacteria with high sensitivity, also realizes high efficiency elimination of bacteria simultaneously. The developed strategy was further applied for promoting wound healing of bacteria-infected mice.

A multi-functional nanocatalytic system based on combined therapies has attracted considerable research attention in recent years due to its potential in the treatment of cancer. Herein, ZnO 2 @ [email protected] nanoparticles (NPs) based on hydroxyl radical (•OH) mediated chemodynamic therapy (CDT) and glucose-exhausting starvation therapy (ST) were constructed. Specifically, in the acidic tumor microenvironment (TME), the pH responsive decomposition of the shell ZIF-67 triggered the release of the Fenton-like catalyst Co 2+ , after which the exposed zinc peroxide (ZnO 2 ) reacted with H 2 O (H + ) to generate O 2 and hydrogen peroxide (H 2 O 2 ). The generated O 2 could alleviate hypoxia in the TEM and interact with ultra-small Au NPs originally coated on ZnO 2 to catalyze intracellular glucose and to produce another source of H 2 O 2 . While the glucose consumption caused the starvation of tumor cells, the generated H 2 O 2 from dual sources reacted with the catalyst Co 2+ to generate highly toxic •OH for CDT. Systematic in vitro and in vivo experiments were carried out to evaluate this nanocatalytic system, and the results showed an enhanced efficacy of this cancer therapy.

Reduced drug uptake and elevated drug efflux are two major mechanisms in cancer multidrug resistance (MDR). In the present study, a new multistage O 2 -producing liposome with NAG/R8-dual-ligand and stimuli-responsive dePEGylation was developed to address the abovementioned issues simultaneously. The designed C-NAG-R8-PTXL/MnO 2 -lip could also achieve magnetic resonance imaging (MRI)-guided synergistic chemodynamic/chemotherapy (CDT/CT). In vitro and in vivo studies showed that C-NAG-R8-PTXL/MnO 2 -lip enhanced circulation time by PEG and targeted the tumor site. After tumor accumulation, endogenous l -cysteine was administered, and the PEG-attached disulfide bond was broken, resulting in the dissociation of PEG shells. The previously hidden positively charged R8 by different lengths of PEG chains was exposed and mediated efficient internalization. In addition, the oxygen (O 2 ) generated by C-NAG-R8-PTXL/MnO 2 -lip relieved the hypoxic environment within the tumor, thus reducing the efflux of chemotherapeutic drug. O 2 was able to burst liposomes and triggered the release of PTXL . The toxic hydroxyl radical (·OH), which was produced by H 2 O 2 and Mn 2+ , strengthened CDT/CT. C-NAG-R8-PTXL/MnO 2 -lip was also used as MRI contrast agent , which blazed the trail to rationally design theranostic agents for tumor imaging.

Invasive bacterial infection interferes the normal orchestrated course of healing cascades during cutaneous regeneration. Hypoxia in the deep-seated infection microenvironment (IME) considerably precludes photonic disinfection efficacy, especially antibacterial photodynamic therapy (aPDT). Herein we develop IME-unlocked oxygen-evolving bio-catalytic fabrics composed of electrospun poly(ε-caprolactone) scaffolds, MXene/SnS 2 bio-heterojunctions (MX/SnS bio-HJs), and lactate oxidase (LOx) for deep-seated bacteria-infected cutaneous regeneration. LOx can deplete lactic acid produced by bacteria at infected site and yield hydrogen peroxide (H 2 O 2 ). The MX/SnS bio-HJs not only catalyze H 2 O 2 to produce hydroxyl radicals (·OH) through Fenton-like reaction, but also generate oxygen (O 2 ) to fortify aPDT under Near-infrared (NIR) light irradiation, consequently, expeditious disinfection is achieved. Moreover, in vivo results reveal that the bio-catalytic fabrics exert benign cutaneous regeneration ability by slaughtering bacteria, promoting hemostasis and collagen deposition. This work provides a tactic to instill fabrics with an IME-unlocked catalytic sterilization capability through endogenous O 2 evolution for remedying infected wounds.
