Knockdown of cytochrome P450 1 A (cyp1a) gene suppresses growth and oxygen tolerance in zebrafish

Min Li,Dongjie Wang,Xiaoping Huang,Shulan Wang,Zhenhan Chen,Muhammad Junaid,Shaolin Xie

Cyp1a (cytochrome P450 1 A) is critical for metabolizing endogenous substances and environmental chemicals. In this study, a zebrafish strain KI ( cyp1a:mcherry ), exhibiting low cyp1a gene expression, was compared with wild-type zebrafish (WT) to investigate the effects of cyp1a on growth and hypoxia tolerance. The results demonstrated that low cyp1a expression significantly inhibited zebrafish growth and reduced hypoxia tolerance. Specifically, KI zebrafish exhibited slower growth rates and higher sensitivity to low oxygen conditions compared to WT. These physiological phenotypes directly link low cyp1a expression to impaired growth and reduced environmental adaptation. Transcriptomic analysis revealed potential mechanisms underlying these effects, including up-regulation of digestive system-related genes (e.g., cpa1 , cpb1 ) and dysregulation of pathways involved in detoxification, stress response, and steroid biosynthesis. These findings highlight the importance of maintaining normal cyp1a expression for healthy growth and environmental adaptation in zebrafish.

Zebrafish ELL-associated factors Eaf1/2 modulate erythropoiesis via regulating gata1a expression and WNT signaling to facilitate hypoxia tolerance

Liu WenYe,Lin ShuHui,Li LingYa,Tai ZhiPeng,Liu Jing-Xia

EAF1 and EAF2, the eleven-nineteen lysine-rich leukemia (ELL)-associated factors which can assemble to the super elongation complex (AFF1/4, AF9/ENL, ELL, and P-TEFb), are reported to participate in RNA polymerase II to actively regulate a variety of biological processes, including leukemia and embryogenesis, but whether and how EAF1/2 function in hematopoietic system related hypoxia tolerance during embryogenesis remains unclear. Here, we unveiled that deletion of EAF1/2 (eaf1−/− and eaf2−/−) caused reduction in hypoxia tolerance in zebrafish, leading to reduced erythropoiesis during hematopoietic processes. Meanwhile, eaf1−/− and eaf2−/− mutants showed significant reduction in the expression of key transcriptional regulators scl, lmo2, and gata1a in erythropoiesis at both 24 h post fertilization (hpf) and 72 hpf, with gata1a downregulated while scl and lmo2 upregulated at 14 hpf. Mechanistically, eaf1−/− and eaf2−/− mutants exhibited significant changes in the expression of epigenetic modified histones, with a significant increase in the binding enrichment of modified histone H3K27me3 in gata1a promoter rather than scl and lmo2 promoters. Additionally, eaf1−/− and eaf2−/− mutants exhibited a dynamic expression of canonical WNT/β-catenin signaling during erythropoiesis, with significant reduction in p-β-Catenin level and in the binding enrichment of both scl and lmo2 promoters with the WNT transcriptional factor TCF4 at 24 hpf. These findings demonstrate an important role of Eaf1/2 in erythropoiesis in zebrafish and may have shed some light on regeneration medicine for anemia and related diseases and on molecular basis for fish economic or productive traits, such as growth, disease resistance, hypoxia tolerance, and so on.

Enzyme-assisted Photoinitiated Polymerization-induced Self-assembly in Continuous Flow Reactors with Oxygen Tolerance

Cai Wei-Bin,Liu Dong-Dong,Chen Ying,Zhang Li,Tan Jian-Bo

Polymerization-induced self-assembly (PISA) is an emerging method for the preparation of block copolymer nano-objects at high concentrations. However, most PISA formulations have oxygen inhibition problems and inert atmospheres ( e.g. argon, nitrogen) are usually required. Moreover, the large-scale preparation of block copolymer nano-objects at room temperature is challenging. Herein, we report an enzyme-assisted photoinitiated polymerization-induced self-assembly (photo-PISA) in continuous flow reactors with oxygen tolerance. The addition of glucose oxidase (GOx) and glucose into the reaction mixture can consume oxygen efficiently and constantly, allow the flow photo-PISA to be performed under open-air conditions. Polymerization kinetics indicated that only a small amount of GOx (0.5 μmol/L) was needed to achieve the oxygen tolerance. Block copolymer nano-objects with different morphologies can be prepared by varying reaction conditions including the degree of polymerization (DP) of core-forming block, monomer concentration, reaction temperature, and solvent composition. We expect this study will provide a facile platform for the large-scale production of block copolymer nano-objects with different morphologies at room temperature.

An oxygen-tolerant photo-induced metal-free reversible addition-fragmentation chain transfer polymerization

Lei Yu,Yanlong Wei,Yuanyuan Tu,Shudong Lin,Zhenzhu Huang,Jiwen Hu,Yue Chen,Hang Qiao,Wei Zou

We report herein a visible light-induced metal-free living polymerization with high oxygen tolerance that can be performed in aqueous media. In contrast with ordinary living/controlled radical polymerizations, oxygen can be present throughout the entire reaction process. This reaction can be photo-induced and proceeds at room temperature. First, we have successfully synthesized a well-defined polymer in an ambient atmosphere by the photo-induced radical polymerization method, using acrylic acid as a monomer and fluorescein as a photocatalyst. However, the subsequent chain extension reaction did not occur, possibly due to oxidation of the chain transfer agent (CTA). Despite this, we found that the addition of vitamin C (ascorbic acid) imparted the process with oxygen tolerance. We conducted a systematic study to optimize the best concentrations of the key reagents including the monomer, CTA, fluorescein, and vitamin C. Through these optimizations we were able to synthesize in the presence of oxygen a series of well-defined poly(acrylic acid)s (PAAs) with dispersities ( Ð ) below 1.3 and molecular weights that closely matched the theoretical values. The kinetic study showed that the molecular weight of the produced PAA increased linearly with the conversion of the monomer, and chain extension reaction also yielded a block polymer with a higher molecular weight than that of the previous polymer. Therefore, we developed a novel photo-induced living polymerization method that can be conducted both in the absence of oxygen and in the presence of air. © 2018 Wiley Periodicals, Inc. J. Polym. Photoenzymatic RAFT Emulsion Polymerization with Oxygen Tolerance

Li Ruo-Yu,An Ze-Sheng

Photoenzymatic reversible addition-fragmenatation chain transfer (RAFT) emulsion polymerization, surfactant-free or ab initio, of various monomers is reported with oxygen tolerance. In surfactant-free emulsion polymerizatoin, poly( N, N -dimethylacrylamide)s were used as stabilizer blocks for emulsion polymerization of methyl acrylate, n-butyl acrylate and styrene, producing well-defined amphiphilic block copolymers, including those with an ultrahigh molecular weight, at quantitative conversions. The controlled character of surfactant-free emulsion polymerization was confirmed by kinetic studies, chain extension studies and GPC analyses. Temporal control was demonstrated by light ON/OFF experiments. In ab initio emulsion polymerization of methyl acrylate and methyl methacrylate, low-dispersity hydrophobic polymers were synthesized with predictable molecular weights. This study extends the monomer scope suitable for photoenzymatic RAFT polymerization from hydrophilic to hydrophobic monomers and demonstrates that oxygen-tolerance can be equally achieved for emulsion polymerization with excellent RAFT control.

Floatable artificial leaf to couple oxygen-tolerant CO2 conversion with water purification

Zhang Zhiyong,Wang Yang,Xie Yangen,Tsukamoto Toru,Zhao Qi,Huang Qing,Huang Xingmiao,Zhang Boyang,Song Wenjing,Chen Chuncheng,Sheng Hua,Zhao Jincai

To enable open environment application of artificial photosynthesis, the direct utilization of environmental CO 2 via an oxygen-tolerant reductive procedure is necessary. Herein, we introduce an in situ growth strategy for fabricating two-dimensional heterojunctions between indium porphyrin metal-organic framework (In-MOF) and single-layer graphene oxide (GO). Upon illumination, the In-MOF/GO heterostructure facilitates a tandem CO 2 capture and photocatalytic reduction on its hydroxylated In-node, prioritizing the reduction of dilute CO 2 even in the presence of air-level O 2 . The In-MOF/GO heterostructure photocatalyst is integrated with a porous polytetrafluoroethylene (PTFE) membrane to construct a floatable artificial leaf. Through a triphase photocatalytic reaction, the floatable artificial leaf can remove aqueous contaminants from real water while efficiently reducing CO 2 at low concentrations (10%, approximately the CO 2 concentration in combustion flue gases) upon air-level O 2 . This study provides a scalable approach for the construction of photocatalytic devices for CO 2 conversion in open environments.
