Combination of Derivatization–HPLC–MS and Enzymatic Hydrolysis–Edman Degradation for Amino Acid Sequence and Configuration of Polymyxin B Components

Zhang Hanzhi,Li Feng,Dun Junling,Sun Ning,Liu Hao,Chen Guiliang

An analysis strategy that can simultaneously determine the amino acid site information and configuration for polymyxin (PM) components is reported in this study. The main components of PMB1, PMB2, and PMB1-I (the Leucine at position 7 changed to be Isoleucine) were purified from the mixture of PMB. The PMB component was hydrolyzed by hydrochloric acid to be a mixture of amino acids, which were derivatized with Marfey’s reagent of N α -(5-fluoro-2,4-dinitrophenyl)- l -alanine amide ( l -FDAA), and then, the configurations of amino acids were easily determined by HPLC–MS. However, the aforementioned method could not offer the information of amino acid at each site, a specific enzymatic hydrolysis method combined Edman degradation was developed. The fatty acyl-diaminobutyric acid (position 1) of PMB component was removed by ficin, and the PMB components were transformed to be PMB nonapeptide. The amino acids at each site of the nonapeptide were determined by N-terminal sequencing. Combining the results of the above methods, the amino acid information and configuration of each site in PM components can be determined, especially the l -Leucine and l -Isoleucine in PMB1 and PMB1-I. The workflow was useful for other PM analogues to identify the sequence and configuration for quality control.

Self-Assembly of Peptide Chiral Nanostructures with Sequence-Encoded Enantioseparation Capability

Yuqi Fan,Qiguo Xing,Jiaxing Zhang,Yuefei Wang,Yaoyu Liang,Wei Qi,Rongxin Su,Zhimin He

Biopolymers such as polysaccharides and proteins have been widely used for the chiral separation of various components due to the intrinsic chirality of the polymers. Amyloid-like short peptides can also self-assemble into diverse chiral supramolecular nanostructures or polymers with precisely tailored architectures driving by noncovalent interactions. However, the use of such supramolecular nanostructures for the resolution and separation of chiral components remains largely unexplored. Here, we report that the self-assembled peptide supramolecular nanostructures can be used for the highly efficient chiral separation of various enantiomers. By rationally designing the constituent amino acid sequence of the peptides and the self-assembling environment, we can fabricate supramolecular polymers with distinct surface charges and architectures, including nanohelices, nanoribbons, nanosheets, nanofibrils, and nanospheres. The various supramolecular nanostructures were then used to resolve the racemic mixtures of α-methylbenzylamine, 2-phenylpropionic acid, and 1-phenylethanol. The results indicated that the self-assembled peptide polymers showed excellent enantioselective separation efficiency for different chiral molecules. The enantioselective separation efficiency of the peptide nanostructures can be tailored by changing their surface charges, morphology, and the constituent amino acid sequences of the peptides.

Characterization of Peanut Protein Hydrolysate and Structural Identification of Umami-Enhancing Peptides

Lixia Zhang,Xiaojing Sun,Xin Lu,Songli Wei,Qiang Sun,Lu Jin,Guohui Song,Jing You,Fei Li

Umami peptides are naturally found in various foods and have been proven to be essential components contributing to food taste. Defatted peanut powder hydrolysate produced by a multiprotease (Flavorzyme, Alcalase, and Protamex) was found to elicit an umami taste and umami-enhancing effect. The taste profiles, hydrolysis efficiency, amino acids, molecular weight distribution, Fourier transform infrared spectroscopy (FT-IR), and separation fractions obtained by ultrafiltration were evaluated. The results showed that peanut protein was extensively hydrolyzed to give mainly (up to 96.84%) free amino acids and peptides with low molecular weights (<1000 Da). Furthermore, β-sheets were the major secondary structure. Fractions of 1–3000 Da and <1000 Da prominently contributed to the umami taste and umami enhancement. To obtain umami-enhancing peptides, these two fractions were further purified by gel filtration chromatography, followed by sensory evaluation. These peptides were identified as ADSYRLP, DPLKY, EAFRVL, EFHNR, and SDLYVR by ultra-performance liquid chromatography (UPLC), and had estimated thresholds of 0.107, 0.164, 0.134, 0.148, and 0.132 mmol/L, respectively. According to the results of this work, defatted peanut powder hydrolysate had an umami taste and umami-enhancing effect, and is a potential excellent umami peptide precursor material for the food industry. Keywords:peanut protein hydrolysate;umami-enhancing peptide;characterization;isolation and purification;amino acid sequence

Peptide capped Pd nanoparticles for oxygen electroreduction: Strong surface effects

Hongyu Yang,Zhenghua Tang,Wei Yan,Likai Wang,Qiannan Wang,Yongqing Zhang,Zhen Liu,Shaowei Chen

Peptide-capped nanoparticles represent a unique type of nanomaterials with emergent optical and electrochemical properties . Herein, a series of peptide capped palladium nanoparticles have been prepared and employed as highly efficient catalysts for oxygen electroreduction. The peptide sequence was tuned by substituting amino acid residues at specialized positions, and strong surface effects were observed between the peptide sequence and the electrocatalytic activity. The findings corresponded well with the previously reported C C coupling catalytic reactions, indicating that the residue-specific binding effects but not the overall binding strength governed the electrocatalytic activity. The results may shed light on the rational design of bio-inspired nanomaterials with optimized electrocatalytic properties.

Effect of sturgeon protein in promoting the adhesion of Lactobacillus plantarum and Lactobacillus rhamnosus

Erdong Yuan,Miao Zhou,Zhengyang Liang,William Kwame Amakye,Chuanli Hou,Jiaoyan Ren

Dietary protein could serve as primary nitrogen source for intestinal flora to enhance the intestinal environment by influencing probiotic adhesion. Our work aims to demonstrate a framework for probiotic activity guided isolation of potential effective fractions from the sturgeon protein. We confirmed the isolated fractions of sturgeon protein as having probiotic adhesion activity against Lactobacillus plantarum 45 (LP45) and Lactobacillus rhamnosus GG (LGG). Three protein fractions (0%, 40%, and 60% NaCl elution fractions) were obtained through anion exchange resin column. The molecular weights of the three fractions were identified by polyacrylamide gel electrophoresis in the ranges of 20.1–66.4 kDa (Fr1), 14.3–44.3 kDa (Fr2) and 14.3–97.2 kDa (Fr3), respectively. A detailed study was performed using an in vitro adhesion model to obtain the bioactive protein. In this model, probiotics were incubated on isolated intestinal segments of mice, and the adhesion effect of probiotics was evaluated through polysaccharide analysis, biofilm quantification, viable count measurement and electron microscopy . Notably, Fr2 was identified as the most effective fractions, significantly increasing the intestinal adhesion rates of LP45 and LGG by 144.8% and 53.7%, respectively. Ultimately, the primary structure of the protein 14.3 kDa in Fr2 was identified by mass spectrometry . Partial peptide amino acid sequences of the protein were confirmed to be W-W-Y-C-F (m/z = 804), K-N-H-D-I/L-Q-A-D-D-I/L-T-C (m/z = 1510) and Q-M-V-H-K-I/L-E-D-E-E-I/L-C-T-S-A-D-K-D (m/z = 2300). In conclusion, we found evidence that purified fractions of sturgeon protein facilitates the adhesion of intestinal probiotics and can be used as an active protein beneficial for intestinal health.
