Arbuscular Mycorrhiza Reduced Nitrogen Loss via Runoff, Leaching, and Emission of N2O and NH3 from Microcosms of Paddy Fields

Zhang Shujuan,Yun Wenfei,Xia Yu,Wu Sikai,You Zhaoyang,Matthias C. Rillig

This study aims to investigate the role of arbuscular mycorrhizal (AM) symbiosis in reducing N loss from paddy fields, using two rice lines: a mycorrhiza-defective rice line (non-mycorrhizal) and its mycorrhizal progenitor. Two rice lines were grown in the presence of an AM fungal isolate. In this study, N loss of runoff, leaching, N 2 O emission, and NH 3 volatilization were measured, and in addition, N uptake of rice, soil aggregates, and plant available N concentration of soil. The results obtained suggest that N loss via runoff, leaching, NH 3 volatilization, and N 2 O emission of mycorrhizal rice was 11%, 8%, 6%, and 1%, lower than that of non-mycorrhizal rice, respectively. Meanwhile, mycorrhizal rice has higher biomass and plant N uptake. Our study shows that the AM symbiosis contributes to the sustainability of rice production by reducing N loss, enhancing soil aggregation and increasing plant N uptake.

Mitigating nitrogen loss in paddy field microcosms through indigenous arbuscular mycorrhizal fungi assemblage

Shujuan Zhang,Xinlong Chen,Yuexiao Dong,Jingfan Qi,Zhaoyang You

Whether farmers should consider the role of arbuscular mycorrhizal fungi (AMF) in agriculture is a hotly debated topic. We aimed to investigate the role of indigenous AMF in reducing nitrogen (N) loss from paddy fields via runoff, leaching, NH 3 volatilization, and N 2 O emission. We conducted a pot experiment employing a mycorrhiza-defective rice mutant (non-mycorrhizal) as the control, grown in soil containing indigenous AMF. The corresponding AMF treatment used the progenitor of this mutant with the same soil. The plants were fertilized with nitrogen, phosphorus and potassium 6 weeks after sowing. The root colonization was 23% in mycorrhizal rice, and no typical AMF structures were observed in the roots of non-mycorrhizal rice. Our findings indicated that the mycorrhizal system exhibited lower N concentrations of runoff and leachate further compounded by reduced fluxes of N 2 O and NH 3 . This led to 14% decrease (mycorrhizal rice 111 kg N ha −1 ; the non-mycorrhizal rice: 129 kg N ha −1 ) in cumulative N loss within 3 days post-fertilization. While this AMF effect was consistent across the four tested N loss pathways, differences were observed between NH 4 + -N and NO 3 − -N in the runoff pathway. Notably, our results revealed no evidence of trade-offs in AMF effect on N loss among the tested pathways. Additionally, mycorrhizal rice had larger shoots and roots than their non-mycorrhizal counterparts. Our study underscores the potential benefits of indigenous AMF in paddy fields for mitigating water pollution and reducing greenhouse gas emission.

From waste to value: Multi-omics reveal pomegranate peel addition improves corn silage antioxidant activity and reduces methane and nitrogen losses

Xia Zhang,Huixian Zhang,Dongcai Wang,Yuanqing Zhang

Fermentation technology presents promising opportunities for food waste valorization. Pomegranate peel ( PP ), a food by-product, has potential applications in fermented feed. This study examined the effects of a 6% dry PP additive on the ensiling characteristics, antioxidant activity, metabolites, bacterial community, and in vitro ruminal fermentation, methane (CH 4 ) emission of corn silage ensiled for 7 days and 60 days using microbiome and metabolome analyses. PP-treated silage inhibited ( P  < 0.05) protein degradation by reducing ammonia nitrogen and non-protein nitrogen concentrations during ensiling. The PP additive increased ( P  < 0.05) water-soluble carbohydrate content and reduced ethanol production in corn silage. Lactiplantibacillus dominated PP-treated silage at the initial ensiling stage, while Levilactobacillus prevailed at the final stage . Notably, the PP additive exhibited strong antioxidant activity by modulating antioxidant enzymes and flavonoid biosynthesis mediated by key metabolites (epigallocatechin and catechin). Correlation analysis identified Lactiplantibacillus , Citrobacter , Phytobacter and Burkholderia as key microbes in the production of antioxidant metabolites and enzymes in PP-treated silage. Additionally, PP supplementation reduced ( P < 0.05) in vitro ruminal CH 4 and nitrogen losses, while decreasing dry matter (DM) digestibility in corn silage. In summary, PP-treated corn silage enhanced antioxidant properties and reduced the nitrogen losses and in vitro ruminal CH 4 emissions, but lowered DM digestibility. Thus, PP can be recommended as a silage additive, though the dry PP level should be lower than that used in this study.

Suitability of Mycorrhiza-Defective Rice and Its Progenitor for Studies on the Control of Nitrogen Loss in Paddy Fields via Arbuscular Mycorrhiza

Zhang Shujuan,You Zhaoyang,Guo Xinyue,Yun Wenfei,Xia Yu,Rillig Matthias C.

Employing mycorrhiza-defective mutants and their progenitors does not require inoculation or elimination of the resident microbial community in the experimental study of mycorrhizal soil ecology. We aimed to examine the suitability of mycorrhiza-defective rice (non-mycorrhizal, Oryza sativa L., cv. Nipponbare) and its progenitor (mycorrhizal) to evaluate nitrogen (N) loss control from paddy fields via arbuscular mycorrhizal (AM) fungi. We grew the two rice lines in soils with the full community of AM fungi and investigated root AM colonization. In the absence of AM fungi, we estimated rice N content, soil N concentration and microbial community on the basis of phospholipid fatty acids; we also quantified N loss via NH3 volatilization, N2O emission, runoff and leaching. In the presence of AM fungi, we did not find any evidence of AM colonization for non-mycorrhizal rice while mycorrhizal rice was colonized and percentage of root colonization was 17–24%. In the absence of AM fungi, the two rice lines had similar N content, soil N concentration and microbial community. Importantly, there was no significant difference in N loss via all the four pathways between mycorrhizal and non-mycorrhizal systems. This mycorrhizal/non-mycorrhizal rice pair is suitable for further research on the role of AM fungi in the control of soil N loss in paddy fields.

Microplastics enhance nitrogen loss from a black paddy soil by shifting nitrate reduction from DNRA to denitrification and Anammox

Xiaofang Ma,Jun Shan,Yanchao Chai,Zhijun Wei,Chenglin Li,Ke Jin,Han Zhou,Xiaoyuan Yan,Rong Ji

Microplastics (MPs) are frequently detected emerging pollutants in soil that can endanger farmland ecosystems; however, little is known about their impacts on dissimilatory nitrate reduction processes in paddy soil . Here, using the 15 N-tracer and microbial molecular techniques, we investigated the effects of MPs (200–400 μm) made of polystyrene (PS), polyvinyl chloride (PVC), and polyethylene (PE) on denitrification, anaerobic ammonium oxidation (Anammox), and dissimilatory nitrate reduction to ammonium (DNRA) and the associated microbial community in a black paddy soil. All MPs increased the Anammox rate by 6.6 %–745 % and decreased the DNRA rate by 15.1 %–74.2 %, while MPs of PS and PE significantly increased the denitrification rate by 79.3 %–102.3 % and 34.8 %–62.1 %, respectively. The MPs promoted the partitioning of NO 3 − towards denitrification and Anammox while inhibiting DNRA, as suggested by the decreased relative contributions of DNRA from 24.1 % to 5.4 %–14.2 % following MPs amendment. This was attributed to the increased denitrification gene abundance and the enriched specific denitrifier taxa, as well as the decreased DNRA gene abundance. Our findings suggest that the stimulated denitrification and Anammox by MPs, accompanied by the suppression of DNRA, may lead to substantial nitrogen loss in paddy fields , underscoring the need to further evaluate the environmental behaviors of MPs in agricultural ecosystems.
