Does N deposition mitigate the adverse impacts of drought stress on plant seed germination and seedling growth?

Shu Wang,Mei Wei,Bingde Wu,Huiyuan Cheng,Kun Jiang,Congyan Wang

Large amounts of nitrogen (N) enter ecosystems via N deposition. The increased drought stress has incurred serious threats to plant seed germination and seedling growth (SGe-SGr). It is significant to characterize the combined impacts of N deposition and drought stress on SGe-SGr of plants to enable a further explanation of the response mechanism of SGe-SGr to N deposition and drought stress. This study aims to explore the combined impacts of N deposition with a concentration gradient and drought stress (mimicked using PEG 6000) with different levels on SGe-SGr of wheat. N treatment raises seedling growth of wheat mainly due to the increased levels of available N and the decreased level of peroxidation of the cell cytoplasm membrane of wheat. Drought stress declines SGe-SGr of wheat mostly due to the progressively enlarged osmotic pressure triggered by water deficit and the advanced level of peroxidation of the cell cytoplasm membrane of wheat. N treatment regardless of the concentration dramatically mitigates the adverse impacts of drought stress on seedling growth of wheat. This may be owed to the improved competitive ability for sunlight acquisition and leaf photosynthetic area via the deactivation of nutrition limits (wheat can absorb more N nutrients and improve its drought resistance ability) implemented by N addition. This may also due to the down-regulated level of peroxidation of the cell cytoplasm membrane of wheat under the combined N and drought stress. However, the combined treatments of the high level of N supply and light drought stress decline seed germination of wheat. This may be due to the promoted osmotic pressure contributed by the high level of N supply under water deficit. Thus, N deposition mitigates the adverse impacts of drought stress on seedling growth of wheat but aggravates the adverse impacts of drought stress on seed germination of wheat mostly.

Does Atmospheric Nitrogen Deposition Confer a Competitive Advantage to Invasive Bidens pilosa L. over Native Pterocypsela laciniata (Houtt.) Shih?

Chuang Li,Yue Li,Yingsheng Liu,Shanshan Zhong,Huanshi Zhang,Zhelun Xu,Zhongyi Xu,Daolin Du,Congyan Wang

One of the key reasons for the success of invasive plants is the functional differences between invasive plants and native plants. However, atmospheric nitrogen deposition may disrupt the level of available nitrogen in soil and the functional differences between invasive plants and native plants, which may alter the colonization of invasive plants. Thus, there is a pressing necessity to examine the effects of atmospheric nitrogen deposition containing different nitrogen components on the functional differences between invasive plants and native plants. However, the progress made thus far in this field is not sufficiently detailed. This study aimed to elucidate the effects of artificially simulated nitrogen deposition containing different nitrogen components (i.e., nitrate, ammonium, urea, and mixed nitrogen) on the functional differences between the Asteraceae invasive plantBidens pilosaL. and the Asteraceae native plantPterocypsela laciniata(Houtt.) Shih. The study was conducted over a four-month period using a pot-competitive co-culture experiment. The growth performance ofP. laciniata, in particular with regard to the sunlight capture capacity (55.12% lower), plant supporting capacity (45.92% lower), leaf photosynthetic area (51.24% lower), and plant growth competitiveness (79.92% lower), may be significantly inhibited under co-cultivation condition in comparison to monoculture condition.Bidens pilosaexhibited a more pronounced competitive advantage overP. laciniata, particularly in terms of the sunlight capture capacity (129.43% higher), leaf photosynthetic capacity (40.06% higher), and enzymatic defense capacity under stress to oxidative stress (956.44% higher). The application of artificially simulated nitrogen deposition was found to facilitate the growth performance of monoculturalP. laciniata, particularly in terms of the sunlight capture capacity and leaf photosynthetic area.Bidens pilosaexhibited a more pronounced competitive advantage (the average value of the relative dominance index ofB. pilosais ≈ 0.8995) thanP. laciniataunder artificially simulated nitrogen deposition containing different nitrogen components, especially when treated with ammonium (the relative dominance index ofB. pilosais ≈ 0.9363) and mixed nitrogen (the relative dominance index ofB. pilosais ≈ 0.9328). Consequently, atmospheric nitrogen deposition, especially the increased relative proportion of ammonium in atmospheric nitrogen deposition, may facilitate the colonization ofB. pilosavia a stronger competitive advantage.

Differences in functional traits between invasive and native Amaranthus species under different forms of N deposition

Wang Congyan,Zhou Jiawei,Liu Jun,Jiang Kun

Differences in functional traits between invasive and native plant species are believed to determine the invasion success of the former. Increasing amounts of anthropogenic nitrogen (N) are continually deposited into natural ecosystems, which may change the relative occurrence of the different N deposition forms (such as NH 4 –N, NO 3 –N, and CO(NH 2 ) 2 –N) naturally deposited. Under high N deposition scenarios, some invasive species may grow faster, gaining advantage over native species. In a greenhouse experiment, we grew invasive and native Amaranthus species from seed both alone and in competition under simulated N enriched environments with different forms of N over 3 months. Then, we measured different leaf traits (i.e., plant height, leaf length, leaf width, leaf shape index, specific leaf area (SLA), and leaf chlorophyll and N concentrations). Results showed that the competition intensity between A. retroflexus and A. tricolor decreased under N deposition. This may be due to the large functional divergence between A. retroflexus and A. tricolor under simulated N deposition. Phenotypic plasticity of SLA and leaf chlorophyll concentration of A. retroflexus were significantly lower than in A. tricolor . The lower range of phenotypic plasticity of SLA and leaf chlorophyll concentration of A. retroflexus may indicate a fitness cost for plastic functional traits under adverse environments. The restricted phenotypic plasticity of SLA and leaf chlorophyll concentration of A. retroflexus may also stabilize leaf construction costs and the growth rate. Meanwhile, the two Amaranthus species possessed greater plasticity in leaf N concentration under NO 3 –N fertilization, which enhanced their competitiveness.

Responses of soil N-fixing bacteria communities to Amaranthus retroflexus invasion under different forms of N deposition

Congyan Wang,Jiawei Zhou,Jun Liu,Kun Jiang,Daolin Du

Soil N-fixing bacteria communities (SNB) can increase soil N availability, which can facilitate invasions of many kinds of invasive plant species. Meanwhile, variability in the forms of anthropogenic N deposition can mediate obvious shifts in both soil physicochemical properties and soil microbial communities (especially SNB). This can alter the soil micro-ecological mechanisms of plant invasions via changes in soil characteristics and soil microbial communities, especially SNB. This study uses next-generation high throughput sequencing technology to provide insight into the effects of invasion by Amaranthus retroflexus L. on the structure and diversity of SNB under different forms of simulated N deposition (SND). Soil pH under A. retroflexus invasion under various forms of SND was 8.08% lower than controls under a NH 4 -N treatment ( P   <   0.05), 15.15% lower than controls under a NO 3 -N treatment ( P   <   0.05), and 6.57% lower than controls under CO(NH 2 ) 2 -N and Mix-N treatments ( P   <   0.05). Meanwhile, soil pH was one of the most important environmental variables explaining changes in the structure and diversity of SNB. The total number of species of SNB under A. retroflexus invasion was approximately 13.13% higher than controls for the NH 4 -N treatment ( P   >   0.05), 11.00% higher than controls for the NO 3 -N treatment ( P   >   0.05), 39.42% higher than controls for the CO(NH 2 ) 2 -N treatment ( P   <   0.05), and 27.67 higher than controls for the Mix-N treatment ( P   <   0.05). The positive effects mediated by the combined treatments of A. retroflexus invasion and SND may be ascribed to enhanced soil nutrient content (especially N), which can enhance the abundance of the nifH gene. The effects of A. retroflexus invasion on the total number of species of SNB under organic and mixed-N addition were greater than under inorganic N addition. This may be a consequence of a disturbance of the balance of inorganic to organic N for SND, as a result of the addition of a single form of inorganic N, which can diminish the stimulatory effects of soil nutrient content on SNB under inorganic N. Another potential cause of this pattern may be stronger acidification under inorganic N addition, which can exert more pronounced effects on SNB. LEfSe analysis showed that some species changed significantly under A. retroflexus invasion in the presence of SND (i.e., Rhodobacteraceae under NH 4 -N and Proteobacteria under CO(NH 2 ) 2 -N). Thus, A. retroflexus invasion in the presence of SND caused obvious shifts in certain species of SNB and increased the total number of species of SNB in particular, to facilitate its further invasion.

Nitrogen Deposition Influences the Allelopathic Effect of an Invasive Plant on the Reproduction of a Native Plant: Solidago canadensis versus Pterocypsela laciniata

Congyan Wang,Jun Liu,Hongguang Xiao,Jiawei Zhou,Daolin Du

This study aims to gain insights into the allelopathic effects (by using leaf extracts) of the notorious invasive Solidago canadensis L. on seed germination of the associated Pterocypsela laciniata (Houtt.) C. Shih under different nitrogen (N) forms added: inorganic (NO3--N and NH4 -N), organic (urea-N), and mixed N (a mixture of the three N forms at 1:1:1 ratio). Among the two used concentrations of S. canadensis leaf extracts the higher exhibited inhibitory allelopathic effects on seedling height and biomass, germination potential, germination index, and vigor index of P. laciniata. N demonstrated positive effects on seed germination of P. laciniata. The effects of mixed and organic N on the seedling biomass of P. laciniata were more pronounced than those of inorganic N and control treatment. The vigor index of P. laciniata under mixed N was significantly higher than those under single N form and control treatment. Thus, organic and mixed N showed higher ecological effects on seed germination of P. laciniata than inorganic and single N form. All N forms could alleviate the inhibitory allelopathic effects of S. canadensis on seed germination of P. laciniata. Overall, the inhibitory allelopathic impact of S. canadensis on seed germination of native species may be attenuated under increased and diverse N deposition, thus it could prevent its further invasion.

The driving effects of nitrogen deposition on nitrous oxide and associated gene abundances at two water table levels in an alpine peatland

Wantong Zhang,Xiuping Tao,Zhengyi Hu,Enze Kang,Zhongqing Yan,Xiaodong Zhang,Jinzhi Wang

Alpine peatlands are recognized as a weak or negligible source of nitrous oxide (N 2 O). Anthropogenic activities and climate change resulted in the altered water table (WT) levels and increased nitrogen (N) deposition, which could potentially transition this habitat into a N 2 O emission hotspot. However, the underlying mechanism related with the effects is still uncertain. Hence, we conducted a mesocosm experiment to address the response of growing-season N 2 O emissions and the gene abundances of nitrification (bacterial amoA ) and denitrification ( narG , nirS , norB and nosZ ) to the increased N deposition (20 kg N ha −1  yr −1 ) at two WT levels (WT − 30 , 30 cm below soil surface; WT 10 , 10 cm above soil surface) in the Zoige alpine peatland, Qinghai-Tibetan Plateau. The results showed that the WT did not affect N 2 O emissions, and this was attributed with the limitation of soil NO 3 − . The higher WT level increased denitrification ( narG and nirS gene abundance) resulting in the depletion of soil NO 3 − , but the consequent NO 3 − deficiency further limited denitrification, while the WT did not affect nitrification (bacterial amoA gene abundance). Meanwhile, the N deposition increased N 2 O emissions, regardless of WT levels. This was associated with the N-deposition induced increase in denitrification-related gene abundances of narG , nirS , norB and nosZ at WT − 30 and narG at WT 10 . Additionally, the N 2 O emission factor assigned to N deposition was 1.3 % at WT − 30 and 0.9 % at WT 10 , respectively. Our study provided comprehensive understanding of the mechanisms referring N 2 O emissions in response to the interactions between climate change and human disturbance from this high-altitude peatland.

Does Bidens pilosa L. Affect Carbon and Nitrogen Contents, Enzymatic Activities, and Bacterial Communities in Soil Treated with Different Forms of Nitrogen Deposition?

Yingsheng Liu,Yizhuo Du,Yue Li,Chuang Li,Shanshan Zhong,Zhelun Xu,Congyan Wang,Daolin Du

The deposition of nitrogen in soil may be influenced by the presence of different nitrogen components, which may affect the accessibility of soil nitrogen and invasive plant–soil microbe interactions. This, in turn, may alter the success of invasive plants. This study aimed to clarify the influences of the invasive plantBidens pilosaL. on the physicochemical properties, carbon and nitrogen contents, enzymatic activities, and bacterial communities in soil in comparison to the native plantPterocypsela laciniata(Houtt.) Shih treated with simulated nitrogen deposition at 5 g nitrogen m−2yr−1in four forms (nitrate, ammonium, urea, and mixed nitrogen). MonoculturalB. pilosaresulted in a notable increase in soil pH but a substantial decrease in the moisture, electrical conductivity, ammonium content, and the activities of polyphenol oxidase, β-xylosidase, FDA hydrolase, and sucrase in soil in comparison to the control. Co-cultivatingB. pilosaandP. laciniataresulted in a notable increase in total soil organic carbon content in comparison to the control. MonoculturalB. pilosaresulted in a notable decrease in soil bacterial alpha diversity in comparison to monoculturalP. laciniata. Soil FDA hydrolase activity and soil bacterial alpha diversity, especially the indices of Shannon’s diversity, Simpson’s dominance, and Pielou’s evenness, exhibited a notable decline under co-cultivatedB. pilosaandP. laciniatatreated with nitrate in comparison to those treated with ammonium, urea, and mixed nitrogen.
