Epidemiological studies indicate that exposure to persistent organic pollutants is positively associated with the prevalence of obesity. To delineate the potential role of technical-grade chlordane in obesity development, chlordane metabolism and chlordane-induced metabolic changes were investigated in mice fed high-fat diet (HFD) over a 6-week period. Gas chromatography–electron capture detector analysis showed that HFD induced more accumulation of technical chlordane in the liver, muscle and adipose tissue . The enantioselectivities of oxychlordane in selected tissues were also influenced by HFD. 1 H NMR-based liver metabolome indicated that technical chlordane can enhance the metabolic alterations induced by HFD. Compared with the low-fat diet (LFD) group, no differences were observed in the LFD + chlordane group. However, as many as 16 metabolites were significantly different between the HFD group and HFD + chlordane group. Moreover, compared to the LFD + chlordane group, the abundances of 24 metabolites significantly increased or decreased in the HFD + chlordane group. Twenty metabolites were altered in the HFD group compared to the LFD group. Tryptophan profiling suggested that both chlordane and HFD can disturb tryptophan catabolism. These interactions between technical chlordane and HFD suggest that technical chlordane is a candidate obesogen.

Chlordane, a previously extensively utilized insecticidal pesticide, has since been prohibited, however, owing to its limited degradability, it continues to persist significantly in soil and water reservoirs, subsequently accumulating within plant and animal organisms, representing a substantial threat to human health. Despite extensive research conducted over the past few decades to investigate the toxic effects of chlordane, there remains a notable dearth of studies focusing on its impact on sleep activity. Therefore, in this study, the effects of short-term and long-term exposure to chlordane on the activity and sleep of Drosophila were investigated. When exposed to chlordane at a concentration of 1 μM, Drosophila lost body weight, decreased body size and resulted in lipid metabolism disorders. In addition, chlordane exposure altered the arousal and sleep behaviors of Drosophila . Short-term exposure to chlordane resulted in an increase in night-time sleep duration, while long-term exposure to chlordane resulted in an increase in activity and a decrease in sleep, as evidenced by a decrease in the duration of each sleep session and the appearance of sleep fragmentation. Under conditions of long-term chlordane exposure, reactive oxygen species levels were significantly up-regulated in Drosophila . Our results suggest that long-term chlordane exposure triggers oxidative stress damage in Drosophila , leading to sleep disruption. This study offers novel insights into the harmful impacts of environmental pollutants on human sleep patterns and proposes that mitigating the presence of chlordane in the environment could potentially contribute to the reduction of global sleep disorder prevalence.

Although chlordane-related compounds (CHLs) have been regulated, a variety of CHLs are still identified and detected in wild birds and eggs. Embryo is one of fragile periods and is very susceptible to toxic effects of pollutants. In this study, the fate of CHLs during embryo development and degradation of CHLs in neonatal chick were investigated. During embryo development, CHLs were mainly distributed to the liver and muscle, in which trans -nonachlor and an octachlorochlordane (MC5) were hardly metabolized and showed the high persistence, implying a greater risk to birds’ offspring . CHLs with the lower K ow were found to be higher uptake efficiency in embryo, implying contaminants with the lower lipophilicity may contribute to their transport to embryo. Furthermore, the effects of CHLs on the metabolome of neonatal chicks was evaluated. The ether lipid metabolism and glycerophospholipid metabolism were found to be significantly affected, which may disturb the angiogenesis and endothelial cell migration in embryogenesis . Taken together, the lipophilicity of contaminants might be a main factor influencing their transport to embryo, and metabolomics results improve understanding of the effects of CHLs on embryo.

Chlordane-related compounds are ubiquitously detected in the environment and can transfer and accumulate to animals through food chain to cause adverse effects . In this study, the dynamic distribution and the enantiomeric profile of chlordane-related compounds in laying hens over time were investigated. The effect of these compounds on immune-associated cells in the intestinal tract and histopathology in some tissues were also evaluated after long-term exposure. The chlordane-related compounds preferentially accumulated in fat, followed by the intestines, ovum, and egg yolk during long-term exposure. The metabolites heptachlor epoxide and oxychlordane were mainly formed in the liver of hens by epoxidation or hydroxylation . The high accumulation ratios of trans -nonachlor and MC5 were found in ovum and egg yolk after long-term exposure, implying a greater risk to the hens’ offspring. Chlordane-related compounds may cause abnormal lipometabolism and glycometabolism in liver of hens. Additionally, (−)-Cis-chlordane was dominant in all tissues of laying hens and its dominance increased over time. Conversely, (+)-isomer of metabolite oxychlordane was overwhelmingly dominant during the experiment. These findings about enantioselectivity , metabolic processes and toxicological effects are crucial in understanding the exposure risk of chlordane-related compounds.

Nanoplastics are emerging contaminants that may co-exist with organochlorine pesticides and adversely affect invertebrates in the environment. However, the impact of environmental aging on the combined toxicity of nanoplastics and organochlorine pesticides remains unclear. This study investigated the effects of aging on the combined toxicity of polystyrene nanoplastics (PS NPs) and chlordane against Caenorhabditis elegans . The results showed that photo-aging altered the physicochemical properties of PS NPs and promoted the combined toxicity of PS NPs and chlordane to nematodes by reducing survival rate, body length and enhancing germline apoptosis. Additionally, combined exposure of nematodes to aged PS NPs and chlordane significantly increased reactive oxygen species production and intestinal permeability, suggesting that aging enhances combined toxicity through oxidative stress and intestinal damage. Moreover, aging increased chlordane contents in nematodes without promoting PS NPs accumulation, potentially leading to increased combined toxicity of PS NPs and chlordane. Notably, aging significantly increased the accumulation of PS NPs in the posterior intestine of the nematode during co-exposure, which may be responsible for the most sensitive and highest degree of change in germline apoptosis. These observations emphasize the significance of accounting for environmental aging as well as the accumulation and distribution of nanoplastics in organisms when assessing the combined effects of nanoplastics and coexisting pollutants.

The incidence of metabolic diseases is increasing every year, and several studies have highlighted the activity of persistent organic pollutants (POPs) in causing hyperlipidemia and diabetes, and these compounds are considered to be endocrine disrupting chemicals (EDCs). Chlordane is classified as an endocrine disruptor, but the mechanism of how it functions is still unclear. This study investigates the effects of chlordane exposure on Drosophila larvae. Drosophila was cultured in diet containing 0.01 μM, 0.1 μM, 1 μM, 5 μM, and 10 μM chlordane, and the toxicity of chlordane, the growth and development of Drosophila , the homeostasis of glucose and lipid metabolism and insulin signaling pathway, lipid peroxidation-related indicators and Nrf2 signaling pathway were evaluated. We here found that exposure to high concentrations of chlordane decreased the survival rate of Drosophila and that exposure to low concentrations of chlordane caused disruption of glucose and lipid metabolism, increased insulin secretion and impairment of insulin signaling. Notably, it also led to massive ROS production and lipid peroxidation despite of the activation of Nrf2 signaling pathway, an important pathway for maintaining redox homeostasis. Collectively, chlordane causes lipid peroxidation and disrupts redox homeostasis, which may be a potential mechanism leading to impaired insulin signaling and the metabolism of glucose and lipid, ultimately affects Drosophila development.

Hazardous waste dechlorination reduces the potential of creating dioxins during the incineration process. To investigate the salt effect on waste dechlorination, molten hydroxides with a low melting temperature were utilized for the pre-dechlorination and decomposition of chlorine-containing organic wastes (COWs) including trichlorobenzene (TCB), perchloroethylene , hexachlorobenzene and chlordane . The results showed that a eutectic mixture of caustic sodium and potassium hydroxides (41 wt.% NaOH and 59 wt.% KOH) led to a low melting point below 300°C and a relatively high chlorine retention efficiency (CRE) with TCB as a representative COWs. The amounts of hydroxides, reaction time, and temperature all had notable influence on CRE. When the mass ratio of hydroxides to TCB reached 30:1, approximately 98.1% of the TCB was destroyed within 2.5 hr at 300°C with CRE of 71.6%. According to the residue analysis , the shapes of reaction residues were irregular with particles becoming swollen and porous. The benzene ring and C–Cl bonds disappeared, while carboxyl groups formed in the residues. The stripped chlorine was retained and condensed to form chloride salts, and the relative abundance of the chloride ions associated with the mass of TCB in residues increased from 0 to 75.0% within the 2.5 hr reaction time. The observed concentration of dioxins in residues was 5.6 ngTEQ/kg. A reaction pathway and possible additional reactions that occur in this dechlorination system were proposed. Oxidizing agents may attack TCB and facilitate hydrogenation/dechlorination reactions, making this process a promising and environmentally friendly approach for chlorine-containing organic waste treatment .
