Histopathological analysis of Artemia franciscana nauplii under different forms of mercury stress

Yanlin Wei,Wei Liu,Fengling Zhang,Yang Chen,Dongxin Wei,Hangyu Lin,Tao He

Inorganic mercury (Hg) and methylmercury (MeHg) have emerged as global pollutants owing to their long-term environmental stability and bioaccumulation. These heavy metals enter aquatic systems via industrial emissions, coal combustion, and natural processes, posing a serious threat to ecosystems and human health. This study assesses the impact of Hg2+ and MeHg on the growth and development of brine shrimp (Artemia franciscana) nauplii by analyzing the histopathological effects on their tissues. Brine shrimp nauplii from the Bohai Bay in China were selected as the study subjects and exposed to 1 μmol/L solutions of HgCl₂ and MeHgCl. Tissue sections were continuously taken at different immersion times. After staining with the standard hematoxylin-eosin (HE) method, the tissue morphology of brine shrimp nauplii under different forms of Hg stress was observed under a light microscope. The results showed that MeHg exhibited significantly greater toxicity to brine shrimp nauplii than Hg2+. Under the same exposure time, the MeHg group exhibited more pronounced epithelial cell damage, nuclear material disorder, and nucleoplasm diffusion outside the nucleus than the HgCl₂ group. This finding provides an important theoretical support for further research into the toxicological mechanisms of MeHg and Hg2+, and highlights that the toxic effects of methylmercury on aquatic organisms.

Inhibition of Tyrosinase by Mercury Chloride: Spectroscopic and Docking Studies

Chen Jianmin,Ye Yaling,Ran Mengnan,Li Qinglian,Ruan Zhipeng,Jin Nan

Inorganic mercury compounds have been used in skin-lightening products since ancient times. Although a previous study demonstrated that mercury impeded the transfer of Cu2+ to the apotyrosinase, the effect of mercury on tyrosinase is still unclear. In the present study, the mechanism of mercury chloride (HgCl2) induced inactivation of tyrosinase was investigated for the first time. The IC50 values were 29.97 and 77.93 μmol/L for monophenolase and diphenolase, respectively. A kinetic analysis revealed that HgCl2 inhibited tyrosinase activity in an irreversible non-competitive manner. The strong intrinsic fluorescence quenching suggested that the formation of the HgCl2-tyrosinase complex induced conformational changes of the enzyme, and HgCl2 had only one single binding site or a single class of binding site on tyrosinase. The molecular docking and further experiments demonstrated that HgCl2 bound to the amino residuals (His) in the catalytic center of tyrosinase. To our knowledge, these findings presented in this paper were the first evidence of the direct interactions between HgCl2 and tyrosinase, which provided a deep understanding of the inhibition mechanism of mercury on tyrosinase.

Mechanistic insights into HgCl2 adsorption over zinc sulfide: The role of inherent sulfur and newly-formed zinc chloride

Mingguang Zhang,Jianping Yang,Lijian Leng,Hongxiao Zu,Wei Zheng,Jiefeng Chen,Hailin Zheng,Wenqi Qu,Zequn Yang,Hailong Li

Divalent mercury (Hg 2 + , mainly as HgCl 2 ) dominates the mercury species discharged from cement kiln flue gas , however, achieving effective control remains challenging. Herein, zinc sulfide (ZnS) was applied for HgCl 2 removal from cement kiln flue gas , showing superior adsorption performance among metal sulfides. ZnS achieved an unprecedented adsorption capacity of 426.1 mg ·g −1 at an optimal temperature of 160 °C, nearly 300 times higher than its Hg 0 adsorption capacity. The typical flue gas components (O 2 , H 2 O, NO and SO 2 ) displayed insignificant adverse effects on HgCl 2 adsorption. Such intriguing observation was ascribed to the inherent sulfur (S 2 2- and S 2- ) and newly-formed zinc chloride (ZnCl 2 ). The S 2 2- and S 2- sites facilitated the conversion of HgCl 2 into β-HgS and α-HgS, respectively. The HgCl 2 dissociation resulted in chlorination of ZnS to form ZnCl 2 which accelerated the initial HgCl 2 adsorption process . The moderate binding energy of HgCl 2 on the Zn sites of ZnCl 2 facilitated the migration of intermediate adsorption products to the S sites of ZnS to form HgS. This work not only provided a promising alternative to commercial activated carbons (ACs) for HgCl 2 removal from cement kiln flue gas, but also proposed new insights to guide the design of HgCl 2 adsorbents. Synopsis A high-efficient HgCl 2 removal strategy for cement kiln flue gas was developed, and the role of inherent sulfur and newly-formed ZnCl 2 in HgCl 2 adsorption over ZnS was proposed for the first time.

miR-15b-5p promotes HgCl2-induced chicken embryo kidney cells ferroptosis by targeting β-TrCP-mediated ATF4 ubiquitin degradation

Hong-Yu Fu,Yue Li,Han Cui,Jiu-Zhi Li,Wan-Xue Xu,Xi Wang,Rui-Feng Fan

Mercuric chloride (HgCl 2 ), a widespread environmental pollutant, induces ferroptosis in chicken embryonic kidney (CEK) cells. Whereas activating transcription factor 4 (ATF4), a critical mediator of oxidative homeostasis, plays a dual role in ferroptosis, but its precise mechanisms in HgCl 2 -induced ferroptosis remain elusive. This study aims to investigate the function and molecular mechanism of ATF4 in HgCl 2 -induced ferroptosis. Our results revealed that ATF4 was downregulated during HgCl 2 -induced ferroptosis in CEK cells. Surprisingly, HgCl 2 exposure has no significant impact on ATF4 mRNA level. Further investigation indicated that HgCl 2 enhanced the expression of the E3 ligase beta-transducin repeat-containing protein (β-TrCP) and increased ATF4 ubiquitination. Subsequent findings identified that miR-15b-5p as an upstream modulator of β-TrCP, with miR-15b-5p downregulation observed in HgCl 2 -exposed CEK cells. Importantly, miR-15b-5p mimics suppressed β-TrCP expression and reversed HgCl 2 -induced cellular ferroptosis. Mechanistically, HgCl 2 inhibited miR-15b-5p, and promoted β-TrCP-mediated ubiquitin degradation of ATF4, thereby inhibited the expression of antioxidant-related target genes and promoted ferroptosis. In conclusion, our study highlighted the crucial role of the miR-15b-5p/β-TrCP/ATF4 axis in HgCl 2 -induced nephrotoxicity, offering a new therapeutic target for understanding the mechanism of HgCl 2 nephrotoxicity.

Noninvasive Dual-Modality Photoacoustic-Ultrasonic Imaging to Detect Mammalian Embryo Abnormalities after Prenatal Exposure to Methylmercury Chloride (MMC): A Mouse Study

Qi Qiu,Yali Huang,Bei Zhang,Doudou Huang,Xin Chen,Zhongxiong Fan,Jinpei Lin,Wensheng Yang,Kai Wang,Ning Qu,Juan Li,Zhihong Li,Jingyu Huang,Shenrui Li,Jiaxing Zhang,Gang Liu,Gang Rui,Xiaoyuan Chen,Qingliang Zhao

Background: Severe environmental pollution and contaminants left in the environment due to the abuse of chemicals, such as methylmercury, are associated with an increasing number of embryonic disorders. Ultrasound imaging has been widely used to investigate embryonic development malformation and dysorganoplasia in both research and clinics. However, this technique is limited by its low contrast and lacking functional parameters such as the ability to measure blood oxygen saturation ( SaO 2 ) and hemoglobin content (HbT) in tissues, measures that could be early vital indicators for embryonic development abnormality. Herein, we proposed combining two highly complementary techniques into a photoacoustic-ultrasound (PA-US) dual-modality imaging approach to noninvasively detect early mouse embryo abnormalities caused by methylmercury chloride (MMC) in real time. Objectives: This study aimed to assess the use of PA-US dual-modality imaging for noninvasive detection of embryonic toxicity at different stages of growth following prenatal MMC exposure. Additionally, we compared the PA-US imagining results to traditional histological methods to determine whether this noninvasive method could detect early developmental defects in utero . Methods: Different dosages of MMC were administrated to pregnant mice by gavage to establish models of different levels of embryonic malformation. Ultrasound, photoacoustic signal intensity (PSI), blood oxygen saturation ( SaO 2 ), and hemoglobin content (HbT) were quantified in all experimental groups. Furthermore, the embryos were sectioned and examined for pathological changes. Results: Using PA-US imaging, we detected differences in PSI, SaO 2 , HbT, and heart volume at embryonic day (E)14.5 and E11.5 for low and high dosages of MMC, respectively. More important, our results showed that differences between control and treated embryos identified by in utero PA-US imaging were consistent with those identified in ex vivo embryos using histological methods. Conclusion: Our results suggest that noninvasive dual-modality PA-US is a promising strategy for detecting developmental toxicology in the uterus. Overall, this study presents a new approach for detecting embryonic toxicities, which could be crucial in clinics when diagnosing aberrant embryonic development. https://doi.org/10.1289/EHP8907

Leaching Behavior and Risk Control of Waste Mercury Chloride Catalyst Through Mechanochemical Reaction Using Sulfur

Yafeng Ren,Jianxin Zhu

A large volume of waste mercury chloride catalyst (HgCl2) is generated by calcium carbide-based polyvinyl chloride production in China, which accounts for 30% of global mercury consumption. However, there are few studies on the leaching behavior and risk control of waste catalyst where mercury is in the form of HgCl2, which is more soluble than metal mercury and poses great threat to human health and environment. This study investigated mechanochemical stabilization and leaching behavior of waste mercury catalyst using sulfur as additive. We found that mercuric sulfide (HgS) formed through mechanical milling, although the reaction between HgCl2 and S is not thermodynamically favored. Moreover, toxicity characteristic leaching procedure (TCLP) results showed mercury leaching values are lower than the TCLP limit value of 0.2 mg/L after mechanochemical treatment. Results of sequential extraction procedure showed that ball mill could transfer more than 99.9% of the Hg in the catalyst to the least mobile fraction.
