Background: Photodynamic therapy (PDT) is widely recognized as a promising way to cure cancer. However, the limited tumor homing property of currently available drug delivery systems (DDSs) is the bottleneck for the delivery of photodynamic agents.Purpose: In our study, we decorated silica nanoparticles (SLN) with cell membrane (CM) derived from SGC7901 cells to construct carrier (CM/SLN) which was able to to specifically target the homogenous SGC7901 cells.Materials and methods: Furthermore, the decent drug loading capability of CM/SLN was adopted to load photodynamic agent chlorins e6 (Ce6) to finally construct aDDS suitable for tumor-targeted PDT of gastric cancer. Results: The experimental results suggested that CM/SLN/Ce6 was nano-sized particles with good dispersion and stability in physiological conditions. Moreover, due to the modification of CM,CM/SLN/Ce6 could specifically target the homogenous SGC7901 cells both in vitro and in vivo. Most importantly, further in vivo results demonstrated that the CM/SLN/Ce6 showed a better anticancer outcome compared to SLN/Ce6.Conclusion: CM/SLN/Ce6 might be a promising platform for effective tumor targeted PDT of gastric cancer.

Sonodynamic therapy (SDT), an emerging cancer treatment with significant potential, offers the advantages of non-invasiveness and deep tissue penetrability. The method involves activating sonosensitizers with ultrasound to generate reactive oxygen species (ROS) capable of eradicating cancer cells, addressing the challenge faced by photodynamic therapy (PDT) where conventional light sources struggle to penetrate deep tissues, impacting treatment efficacy. This study addresses prevalent challenges in numerous nanodiagnostic and therapeutic agents, such as intricate synthesis, poor repeatability, low stability, and high cost, by introducing a streamlined one-step assembly method for nanoparticle preparation. Specifically, the sonosensitizer Chlorin e6 (Ce6) and the chemotherapy drug erlotinib are effortlessly combined and self-assembled under sonication, yielding carrier-free nanoparticles (EC-NPs) for non-small cell lung cancer (NSCLC) treatment. The resulting EC-NPs exhibit optimal drug loading capacity, a simplified preparation process, and robust stability both in vitro and in vivo, owing to their carrier-free characteristics. Under the synergistic treatment of sonodynamic therapy and chemotherapy, EC-NPs induce an excess of reactive oxygen in tumor tissue, prompting apoptosis of cancer cells and reducing their proliferative capacity. Both in vitro and in vivo experiments demonstrate superior therapeutic effects of EC-NPs under ultrasound conditions compared to free Ce6. In summary, our research findings highlight that the innovatively designed carrier-free sonosensitizer EC-NPs present a therapeutic option with commendable efficacy and minimal side effects.

Green foods are usually harvested and stored before ripening, but excessive ethylene generated during storage can lead to the loss of vegetables and fruits. Photocatalytic oxidation provides a promising method to remove ethylene and extend the shelf life of green foods. In this study, a mesoporous nanosilica-loaded Ce6 composite nanomaterial (Ce6@SiO2) was designed and synthesized to adsorb ethylene into its pores and then degrade it under visible light illumination. The photosensitizer Ce6 in the mesopore produced large amounts of reactive oxygen species that degraded ethylene. The specific surface area was greatly increased by coating the material on the surface of a sodium alginate (SA) hydrogel membrane. This improved its degradation efficiency and facilitated the adsorption and degradation of ethylene during fruit storage. The ethylene removal capacity was studied by measuring the ethylene concentration and increases in the humidity in Tedlar gas bags. Practical applications were tested by observing color changes and surface decay of tomatoes stored with and without SA/Ce6@SiO2. The results showed the system may be used in practical commercial preservation systems.

In the field of cancer therapy, photodynamic therapy (PDT) is an emerging treatment modality that utilized light, photosensitizers (PSs), and oxygen to generate reactive oxygen species (ROS) to destroy tumor sites selectively. Conventional PSs experience issues such as poor water solubility and low bioavailability. The widely developed PS delivery nanosystems can address these problems, but most of them usually depend on relatively complex components and time-consuming synthesis procedures. Therefore, PS delivery vehicles with more easily obtained components and simpler preparation methods need to be explored. In this study, amphiphilic and cationic single-isomer cyclodextrin (CD-IL) was designed, which could co-assemble with chlorin e6 (Ce6, a commercially available PS) via multiple non-covalent interactions. The obtained nanovesicles (CD-CE) could not only overcome the inherent drawbacks of free Ce6, but also exhibit improved PDT efficacy in a series of cell-related experiments, including favorable stability, preferable cellular uptake, and enhanced phototoxicity. Thus, CD-CE prepared by the co-assembly of simple building blocks could be a facile option for PDT clinical applications.

Emerging multimodal therapy strategies based on sonodynamic therapy (SDT) are foreseen as foundational in forthcoming tumor theranostics. Nevertheless, the therapeutic effects of SDT are limited by the hypoxic environment of the tumor microenvironment (TME). Based on this, nano-enzymes are envisioned to decompose excess hydrogen peroxide in the tumor microenvironment to generate oxygen. Here, PEG-PLGA-coated CeO 2 nanoparticles loaded with the sonosensitizer chlorin e6 (CC@PP) were prepared for cancer therapy based on a nanoprecipitation method. CC@PP possesses highly efficient catalase-like (CAT) properties, which are able to decompose overexpressed H 2 O 2 into O 2 and enhance the effect of O 2 -dependent sonodynamic therapy. In addition, it can act as a peroxidase (POD) at lower pH for chemodynamic therapy. Under the guidance of the US, CC@PP can perform synergistic sonodynamic and chemodynamic cancer therapy and inhibit tumor growth efficiently, CC@PP is biocompatible, has good in vivo fluorescence imaging effect, and has integrated diagnostic and therapeutic properties, which can be effectively aggregated at the tumor area. In this study, the effect of sonodynamic therapy was enhanced by improving the hypoxic environment in the TME, and multimodal synergistic enhancement of cancer therapy was realized.

A multimodal nanocarrier based on mesoporous silica nanoparticles (MSNs) is developed to co-delivery photosensitizer chlorin e6 (Ce6) and chemotherapeutic agent doxorubicin (Dox) for cancer combination therapy. Ce6 was covalently conjugated with mesoporous silica nanoparticles, which could increase the loading efficiency, and allowed for photodynamic therapy. Doxorubicin was loaded into the pores of mesoporous silica nanoparticles to afford the dual drug delivery system Dox@MSNs-Ce6. These hybrid nanoparticles have an average diameter of about 100 nm and slightly negative charge of about −17 mV. The Dox@MSNs-Ce6 nanoparticles could efficiently enter into cancer cells. The cellular reactive oxygen species level in treated cells increased about 17 times, upon 660 nm light irradiation (10 mW/cm2, 2 min). More importantly, Dox@MSNs-Ce6 exhibited excellent synergistic effect through combining chemotherapy and photodynamic therapy against A549 lung cancer cells. Our work provides an effective strategy for anticancer drug delivery and combination therapy.

Introduction: The effective treatment of glioblastoma still remains a great challenge. We herein report the development of chlorin e6 (Ce6)-conjugated iron oxide (Fe 3 O 4 -Ce6) nanoparticles for ablation of glioblastoma cells via combining photothermal therapy (PTT) with photodynamic therapy (PDT). Methods: Ce6 was conjugated to the synthesized Fe 3 O 4 nanoparticles to form Fe 3 O 4 -Ce6 nanoparticles displaying the optical property of Ce6. Results and discussion: Under 808 nm laser irradiation, Fe 3 O 4 -Ce6 nanoparticles generated heat and the temperature increase did not have obvious changes after five cycles of laser irradiation, suggesting their good photothermal effect and photothermal stability. In addition, 660 nm laser irradiation of Fe 3 O 4 -Ce6 nanoparticles produced singlet oxygen ( 1 O 2 ) to mediate PDT. The Fe 3 O 4 -Ce6 nanoparticles without laser irradiation showed a low cytotoxicity, but they would obviously kill C6 cancer cells after laser irradiation via the combinational effect of PTT and PDT. Fe 3 O 4 -Ce6 nanoparticles thus could be used as a nanotherapeutic agent for combinational ablation of glioblastoma cells.

Aim: Hollow mesoporous copper sulfide nanocapsules conjugated with poly(ethylene glycol) (PEG), doxorubicin and chlorin e6 (HPDC) were synthesized for fluorescence imaging and multimodal tumor therapy. Materials & methods: HPDC were synthesized by encapsulating chlorin e6 and doxorubicin into PEGylated nanocapsules via a simple precipitation method. The photothermal/photodynamic effects, drug release, cellular uptake, imaging capacities and antitumor effects of the HPDCs were evaluated. Results: This smart nanoplatform is stimulus-responsive toward an acidic microenvironment and near infrared laser irradiation. Moreover, fluorescence imaging-guided and combined photothermal/photodynamic/chemotherapies of tumors were promoted under laser activation and led to efficient tumor ablation, as evidenced by exploring animal models in vivo. Conclusion: HPDCs are expected to serve as potent and reliable nanoagents for achieving superior therapeutic outcomes in cancer management.

A unique matrix metalloproteinase 2-targeted photosensitizer delivery platform was developed in this study for tumor-targeting imaging and photodynamic therapy. The model photosensitizer therapeutic agent chlorin e6 (Ce6) was first covalently conjugated with matrix metalloproteinase 2-cleavable polypeptide and then modified with polyethylene glycol via a redox-responsive cleavable disulfide linker. The resultant matrix metalloproteinase 2-cleavable polypeptide modified PEGylated Ce6 (PEG-SS-Ce6-MMP2) nanoparticles, which formed via self-assembly, were observed to be monodisperse and significantly stable in aqueous solution. In addition, owing to their cellular redox-responsiveness at the cleavable disulfide linker, the PEG-SS-Ce6-MMP2 nanoparticles were able to release Ce6 rapidly. Despite displaying enhanced intracellular internalization, the synthesized PEG-SS-Ce6-MMP2 nanoparticles did not compromise their phototoxic effects toward A549 cancer cells when compared with free Ce6 and PEGylated Ce6 nanoparticles. In vivo experiments further revealed that, in contrast with the free Ce6 or with the PEGylated Ce6 nanoparticles, the PEG-SS-Ce6-MMP2 nanoparticles showed a remarkable increase in tumor-targeting ability and a significantly improved photodynamic therapeutic efficiency in A549 tumor-bearing mice. These results suggest that the PEG-SS-Ce6-MMP2 nanoparticles hold great potential for tumor-targeting imaging and photodynamic therapy.

Photodynamic therapy (PDT) is a promising cancer therapy modality due to its intrinsically negligible side effects and treatment resistance. However, the development of the high-efficiency PDT still remains a challenge. Herein, a nanodrug platform PEG-Ce6-PEI@PB combined tumor acidity-induced polyethyleneimine (PEI) cytotoxicity with an oxygen self-supply property is developed for dual-enhanced PDT. The obtained PEG-Ce6-PEI@PB presents suppressed PEI cytotoxicity and chlorin e6 (Ce6) phototoxicity during the bloodstream before becoming active in tumor tissues/cells. The acidic tumor microenvironment can shed PEG coating to rebound PEI positive charges, facilitating tumor cell uptake and reverting the PEI cytotoxicity to enhance following PDT. Moreover, Prussian blue (PB) nanozymes with catalase-like activity can convert endogenous hydrogen peroxide into oxygen to relieve tumor hypoxia, which is attributed to the photosensitizer Ce6 producing more cytotoxic reactive oxygen species upon laser irradiation to further strengthen PDT. Moreover, PEG-Ce6-PEI@PB exhibits good biocompatibility and long blood circulation. More importantly, PEG-Ce6-PEI@PB-treated breast cancer cells and tumor-bearing mice present effective therapeutic efficacy upon laser irradiation, verifying the synergistic antitumor effects of PEI cytotoxicity and oxygen self-supplying PDT.
