Evaluation of the interaction of chlorhexidine and MDP and its effects on the durability of dentin bonding

Jiadi Shen,Haifeng Xie,Qi Wang,Xinyi Wu,Jiaxue Yang,Chen Chen

Objective This study aimed to evaluate the potential interaction of chlorhexidine (CHX) and 10-methacryloyloxydecyl dihydrogen phosphate (MDP) and its effects on the durability of dentin bonding. Methods Two commercial adhesives were tested: a MDP-free adhesive (Single Bond 2, SB2) and a MDP-containing adhesive (Single Bond Universal, SBU). Teeth were randomly assigned to six groups and tested for micro-tensile bond strength (μTBS): Ctr, direct bonding with SB2; CHX, CHX conditioning and SB2; MDP, MDP conditioning and SB2; CHX   +   MDP, combined CHX and MDP conditioning and SB2; SBU, direct bonding with SBU; CHX   +   SBU, CHX conditioning and SBU. The potential interaction of CHX and MDP was assessed by measuring nanoleakage, in situ zymography, and chemoanalytic characterization via Fourier transform infrared spectroscopy (FTIR), X-ray photoelectron spectroscopy (XPS) and nuclear magnetic resonance (NMR). Specimens for μTBS and nanoleakage tests were first subjected to water storage for 24   h or 6 months. Results The initial μTBS values of the Ctr and CHX groups were significantly lower than those of the other four groups (P   <   0.05). Water storage for 6 months significantly weakened all groups (P   <   0.05), with the Ctr group showing the lowest μTBS. This group also showed more obvious nanoleakage than the other five groups. In situ zymography revealed that the Ctr group showed the strongest fluorescence and that the CHX   +   MDP group showed greater fluorescence than either CHX or MDP group. FTIR, XPS, and NMR indicated that MDP can interact with hydroxyapatite. NMR detected no Ca 2+ salt peak for MDP when it was combined with CHX. Significance The application of either CHX or MDP alone can improve dentin bond durability. However, CHX may interfere with the formation of MDP–Ca salts.

pH-Responsive polymeric nanocarriers for efficient killing of cariogenic bacteria in biofilms

Zhouxiang Zhao,Chunmei Ding,Yaqin Wang,Hong Tan,Jianshu Li

Traditional antibacterial treatments, such as chlorhexidine (CHX), destroy cariogenic biofilms. However, they exert negative effects in clinical applications, for example, teeth staining, taste disturbance and harm to oral tissue after a long-term exposure. Therefore, biocompatible strategies for efficient antibacterial drug delivery are in high demand. In this study, aimed at dental caries therapy enhancement, we designed a pH-responsive nanocarrier system, capable of releasing CHX in an acidic environment within cariogenic biofilms. Cationic poly(ethylene glycol)-block-poly(2-(((2-aminoethyl)carbamoyl)oxy)ethyl methacrylate) (PEG-b-PAECOEMA) was synthesized first. Modification of PAECOEMA by citraconic anhydride (CA) forms negatively charged PEG-b-PAECOEMA/CA, which could assemble into core–shell polyionic complex micelles (PICMs) when mixed with cationic CHX via electrostatic interactions. PICMs are stable in healthy neutral oral microenvironments with CHX encapsulated in the core and PEG shell exposed. Once in acidic milieu within caries-producing biofilms, they rapidly disassemble and release CHX cargo owing to degradation of citraconic amide groups. Molecular structures of the above copolymers were confirmed using 1H NMR and gel permeation chromatography (GPC) analysis. The pH-dependent degradation rates of citraconic amide in PEG-b-PAECOEMA/CA copolymer were measured by fluorescamine method. Atomic force microscopy (AFM) studies confirmed successful assembly of well-defined spherical PICMs in aqueous solution. The disassembly of PICMs in acidic microenvironment was observed using dynamic light scattering (DLS). PICMs showed an obvious pH-dependent drug release profile when the pH changed from 7.4 to 5.5. More importantly, the micellar system could reduce drug toxicity of CHX and exhibited outstanding antibacterial capability in the biofilm of Streptococcus mutans. Micelles constructed from pH-sensitive PEG-b-PAECOEMA/CA are highly promising for dental caries therapy and provide guidelines for drug-delivery system design in other acidic pathologic systems.

Preparation of keratin/chlorhexidine complex nanoparticles for long-term and dual stimuli-responsive release

Xuelian Zhi,Yanfang Wang,Pengfei Li,Jiang Yuan,Jian Shen

Nanoscale polyion complex formation via the electrostatic complexation of a polyelectrolyte and a charged drug is the most convenient method for building a drug delivery system that simultaneously realizes the carrier preparation and drug embedding. Herein, we prepared keratin/chlorhexidine complex nanoparticles (KCNPs) based on a drug-induced ionic gelation technique without using a crosslinker, organic solvent or surfactant. These KCNPs exhibited good stability even after having been long-standing for 14 days. The KCNPs were characterized using FTIR, DLS, SEM and TEM. It was found that these nanoparticles had a spherical morphology with a diameter of about 180 nm, and a negatively charged surface with a zeta potential of about −39.1 mV. The cell toxicity of the KCNPs at different dosage levels was evaluated using the MTT assay method, indicating their slight cytotoxicity at lower dosages. The antibacterial activity against E. coli and S. aureus was determined using the zone of inhibition method. It seemed that the KCNPs had better antibacterial activity against S. aureus than against E. coli. Drug delivery profiles showed that the chlorhexidine (CHX) loaded nanoparticles exhibited both pH- and glutathione-responsive character. These keratin-based complex nanoparticles can be regarded as a valuable stimuli-responsive strategy for the delivery of anticancer agents.

Mussel-inspired monomer – A new selective protease inhibitor against dentine collagen degradation

Kang Li,Fung Man Ngo,Angela Yat Laam Yau,Winnie Wai Ling Tam,Edmund Chun Ming Tse,James Kit Hon Tsoi,Cynthia Kar Yung Yiu

Objectives To evaluate the inhibitory effect of a novel mussel-inspired monomer ( N -(3,4-dihydroxyphenethyl)methacrylamide (DMA) on the soluble and matrix-bound proteases . Methods The inhibitory effect of DMA (0, 1, 5, and 10 mM) and 1 mM chlorhexidine (CHX) dissolved in 50% ethanol/water on soluble recombinant human matrix metalloproteinases (rhMMP-2, −8, and −9), as well as cysteine cathepsins (B and K) were evaluated using both fluorometric assay kits and molecular docking . The effect of CHX and DMA on matrix-bound proteases was examined by in situ zymography , and the fluorescence intensity and relative area were calculated by Image J software. All data obtained were analyzed by one-way ANOVA followed by Tukey test (α = 0.05). Results The anti-proteolytic ability of DMA increased in a dose-dependent manner except that of rhMMP-9. Inhibitory effect of 1 mM DMA against rhMMP-2, − 8, − 9, as well as cathepsin B and K was all significantly lower than 1 mM CHX ( p  < 0.05). The molecular docking analysis was in good agreement with the experimental results, that the binding energy of DMA was lower than CHX for all proteases. In situ zymography revealed that all DMA- and CHX-treated groups significantly inactivated the matrix-bound proteases, with a dramatic reduction of the fluorescence intensity and relative area compared with the control group ( p  < 0.05). Significance Under the prerequisite condition that the overall inhibitory performance on matrix-bound proteases was comparable by DMA and CHX, the more selective property of DMA could avoid inducing potential negative effects by suppressing MMP-9 when applied in dental treatment compared with CHX.

A liquid crystalline precursor incorporating chlorhexidine acetate and silver nanoparticles for root canal disinfection

Tengyi Zheng,Xintian Huang,Jintian Chen,Disang Feng,Liling Mei,Ying Huang,Guilan Quan,Chune Zhu,Vikramjeet Singh,Hao Ran,Xin Pan,Chuan-Yu Wu,Chuanbin Wu

Lyotropic liquid crystals (LLC) have received increasing attention as a drug delivery system. In this study, a novel intra-canal disinfectant based on the glycerol monooleate (GMO) LLC precursor incorporation with chlorhexidine (CHX) and silver nanoparticles (Ag-NPs) was designed and evaluated. The LLC precursor with excellent fluidity was able to penetrate deeply into the complex tiny collateral branch root canals. The transformation of cubic LLC in root canals upon coming into contact with water provided long-lasting disinfection against multidrug-resistant bacteria to avoid the endodontic reinfection and follow-up visits. The GMO–ethanol–water (48% : 12% : 40%, w/w) formulation containing 0.5% CHX and 0.02% Ag-NPs was selected for further studies. The low viscosity of the precursor presented excellent injectability and flowabilities. From the in vitro release test, the release behaviours were found to be influenced by CHX and Ag-NP contents, allowing the optimized precursor to obtain a 28-day release profile. The CHX-Ag-NP containing LLC precursor exhibited an excellent and sustained sterilization effect on Enterococcus faecalis for more than one month with a bacterial inactivation rate of ≥98.5%, which was far more than the minimum clinical requirement (7 days). Furthermore, no in vitro toxicity was observed in the cytotoxicity evaluation. The CHX-Ag-NP containing LLC precursor was proved to be a promising intra-canal disinfectant in our study.

Could chloroxylenol be used as WBE biomarker in gravity sewers? Fates, behaviors and feasible conditions

Xinhui Xia,Huizhi Mu,Yujia Du,Shuocheng Shao,Yaqun Li,Dan Li,Qingliang Zhao,Liangliang Wei

Understanding the in-sewer stability of chemical biomarkers is crucial for effective wastewater-based epidemiology (WBE) studying. Sewer conditions, including environmental and biological factors, significantly influence biomarker transformations. This study investigated the stability of chloroxylenol (PCMX) under different levels of pH, temperature, shear force, and ventilation status, and then clarified the fate and behavior of PCMX in gravity sewers (GS). Results indicated the stability of PCMX obviously increased with higher pH and shear force, and lower temperature in both well- and partially-ventilated GS reactors. In poorly-ventilated GS reactors, the highest degradation rates occurred under normal conditions (pH = 7.0, T = 20 °C, shear = 1.15 N/m 2 ). Biological activity (MPR>SPR) and dissolved oxygen (DO) primarily drove PCMX transformation, with minimal effects from pH, temperature, and shear force. A positive correlation existed between PCMX transformation and DO, and a negative correlation existed between PCMX transformation and biological activity. Mass balance analysis indicated that adsorption and bioaccumulation dominated PCMX transformation in GS, while biotransformation occurred with the increasing of DO and prolongation of HRT. Additionally, the suitability of PCMX as a WBE biomarker under different GS conditions was assessed. PCMX was viable as a biomarker in partially-ventilated GS under pH 8 or shears force of 0.48 N/m 2 conditions, and in poorly-ventilated GS under pH 6 or shears force of 0.48 N/m 2 conditions. This study enhances understanding of factors affecting PCMX stability and supports its application as a WBE biomarker in community health assessments.

Engineering Antibacterial, Biocompatible, Anti-Oxidant Titanium-Based Implants Using Polyphenols-Chlorhexidine Networks for Bone Regeneration

Yankai Wang,Biao Hong,Baozang Yin,Yuan Li,Liuwei Huang,Shuang Tan,Tiancheng Gao,Manfred F. Maitz,Junsheng Zhang,Shunli Zheng,Can Huang,Hua Qiu,Jialong Chen,Xiangyang Li

Implant-associated infections leading to osteolysis and implant loosening are an ongoing clinical challenge. Various strategies have been proposed to equip bone implants with antibacterial properties to prevent infection. However, the cytotoxicity associated with antimicrobial effects adversely impacts the osseointegration. Herein, a facile and safe strategy is proposed to endow bone implants with infection prevention, good cytocompatibility, inflammatory-responsive antimicrobial properties, thus promoting bone healing. The coating is fabricated on the implant through both covalent and non-covalent bonds of polyphenols with chlorhexidine (CHX). The covalent bonds guarantee long-term stability, while non-covalent bonds facilitate early release of CHX. Furthermore, the inclusion of polyphenols reduces the electrophilicity of CHX, inhibits reactive oxygen species generated by CHX, and minimizes interference with the mitochondrial electron transport chain, thereby reducing cellular toxicity. Consequently, the coating effectively fortified the bone implant, successfully impeding bacterial invasion within 7 days in Sprague-Dawley rats and suppressing inflammation as well as bone resorption caused by bacteria during a 60-day femoral implantation, thus facilitated osseointegration on the implant. The study investigated the cytotoxicity associated with mitochondrial interference induced by CHX and proposed a strategy to enhance its cellular compatibility, thereby providing a novel approach for fabricating biocompatible antibacterial bone implants.

Degradation and mechanism analysis of chloroxylenol in aqueous solution by gas–liquid discharge plasma combined with ozonation

Keke Ma,Lu Zhou,Yu Bai,Yiying Xin,Mingru Chen,Heping Li,Chengyu Bao,Yuexi Zhou

Gas–liquid discharge non-thermal plasma (NTP) coupled with an ozonation reactor was used to investigate the removal of a broad-spectrum antibacterial agent, chloroxylenol (PCMX), from aqueous solution. Under the same experimental conditions (discharge power of 50.25 W, the initial concentration of PCMX of 60 mg L−1, oxygen flow of 1.0 L min−1 and PCMX solution flow of 150 mL min−1), the PCMX degradation rates in the ozonation-only, NTP-only and NTP/O3 systems were 29.25%, 67.04% and 79.43%, respectively. Correspondingly, the energy efficiency has also been greatly improved, and increased to 0.45, 1.03 and 1.21 g kW−1 h−1. In addition, the effects of the initial concentration of PCMX, initial pH, the flow rate of oxygen, the addition of H2O2 and the addition of a radical scavenger on the degradation rate of PCMX were investigated in the NTP/O3 system. The degradation rate in acidic solutions was higher than that in alkaline solutions. During the removal process of PCMX, the rate of degradation was strongly increased with the addition of H2O2 and acutely decreased with the addition of the radical scavenger. Compared with deionized water the degradation rates of PCMX in secondary effluent were inhibited. Four main intermediates of PCMX degradation by the NTP/O3 system were identified by gas chromatography-mass spectrometry (GC-MS) and a possible degradation pathway of PCMX was proposed. The changes in toxicity of the PCMX solution during the NTP/O3 system oxidation process were also evaluated using bioluminescent bacteria and Quantitative Structure Activity Relationship (QSAR) models with the help of the ECOSAR software.

Calcium-silicate mesoporous nanoparticles loaded with chlorhexidine for both anti- Enterococcus faecalis and mineralization properties

Fan Wei,Li Yanyun,Sun Qing,Ma Tengjiao,Fan Bing

Background In infected periapical tissues, Enterococcus faecalis is one of the most common dominant bacteria. Chlorhexidine has been proved to show strong antibacterial ability against E. faecalis but is ineffective in promoting mineralization for tissues around root apex. Mesoporous calcium-silicate nanoparticles are newly synthesized biomaterials with excellent ability to promote mineralization and carry-release bioactive molecules in a controlled manner. In this study, mesoporous calcium-silicate nanoparticles were functionalized with chlorhexidine and their releasing profile, antibacterial ability, effect on cell proliferation and in vitro mineralization property were evaluated. Results The chlorhexidine was successfully incorporated into mesoporous calcium-silicate nanoparticles by a mixing-coupling method. The new material could release chlorhexidine as well as Ca2+ and SiO3 2− in a sustained manner with an alkaline pH value under different conditions. The antimicrobial ability against planktonic E. faecalis was dramatically improved after chlorhexidine incorporation. The nanoparticles with chlorhexidine showed no negative effect on cell proliferation with low concentrations. On dentin slices, the new synthesized material demonstrated a similar inhibitory effect on E. faecalis as the chlorhexidine. After being immersed in SBF for 9 days, numerous apatite crystals could be observed on surfaces of the material tablets. Conclusion sMesoporous calcium-silicate nanoparticles loaded with chlorhexidine exhibited release of ions and chlorhexidine, low cytotoxicity, excellent antibacterial ability and in vitro mineralization. This material could be developed into a new effective intra-canal medication in dentistry or a new bone defect filling material for infected bone defects.

Construction of organic compatible kaolinite antibacterial material via a dry process and its enhanced antibacterial activity

Xiaoyu Zhang,Shengping Li,Ning Zhao,Yueqing Deng,Zengjie Zuo,Chunquan Li,Shuilin Zheng,Zhiming Sun

In today’s world of antibiotic abuse and bacterial resistance, it is urgent to propose a non-antibiotic solution against bacterial wound infections. Hence, the present work prepared kaolinite (KAO)-based organic antibacterial material (CA/KAO) modified with chlorhexidine acetate (CA) via a dry process. And the potential of CA/KAO in the doping preparation of antibacterial wound dressings was evaluated with poly(vinyl alcohol) (PVA) as an organic matrix. The results demonstrated that a synergistic promotion system between CA and kaolinite was formed. Kaolinite optimized the thermal stability of CA and controlled it to achieve a demand-oriented release mode of “burst + slow” release. CA was mainly attached to the surface of kaolinite by hydrogen bonding to impart its antibacterial properties and enhanced organic compatibility. For Staphylococcus aureus ( S. aureus ), which is prone to induce wound infections, CA/KAO showed a low-level MIC of 11.72 mg/L and achieved a 98.37% antibacterial rate by contact inactivation and released CA sterilization. CA/KAO was homogeneously dispersed in PVA film and achieved inhibition zones of 15.393 mm and 12.663 mm against S. aureus and Escherichia coli (E. coli) , respectively, at 30 wt% doping. It is expected that CA/KAO would be an option for the mass production of antibacterial wound dressings.

Stability Evaluation of Lyotropic Liquid Crystalline Precursor for the Co-delivery of Chlorhexidine and Silver Nanoparticles

Fu Fangqin,Li Xin,Zheng Tengyi,Xia Xiao,Du Minqun,Huang Zhengwei,Huang Ying,Pan Xin,Wu Chuanbin

Sealing the therapeutic agents in the root canal is considered to be an essential step in root canal therapy. The lyotropic liquid crystalline precursor (LLCP) incorporated with chlorhexidine (CHX) and silver nanoparticles (Ag-NPs) has been confirmed as a promising candidate for root canal therapy in the previous study. Importantly, the stability of the LLCP system was a significant determinant for its therapeutic effect and further application. The objective of this study was to comprehensively investigate the stability of the LLCP incorporated with CHX and Ag-NPs. The oil-water partition coefficient of CHX and Ag-NPs was measured. The water absorption and the physical stability of drug-loaded LLCP solution were studied. Stability under high temperature, high humidity, and strong light irradiation was also investigated. The results demonstrated that CHX and Ag-NPs could be entrapped in the water channel of LLCP, indicating the low tendency of drugs leakage. The drug-loaded LLCP was a pseudoplastic fluid and it showed an excellent physical stability with a sedimentation rate of 0.981 and a settling time of 26~28 h. The payload of LLCP was confirmed to weaken the water absorption behavior, which facilitated its transformation to cubic liquid crystal. The stress testing under high temperature, high humidity, and strong light irradiation also manifested that the LLCP was stable when stored under moisture-proof condition. In conclusion, the developed LLCP incorporated with CHX and Ag-NPs was highly stable during storage and qualified for further application.

Dual-Function Hydrogel Coating on Silicone Urinary Catheters with Durable Antibacterial Property and Lubricity

Shuai Gao,Wei Zeng,Zheng Liu,Fanjun Zhang,Yunfeng Zhang,Xi Liu,Dimeng Wu,Yunbing Wang

Silicone urinary catheters are broadly employed in medical practice. However, they are susceptible to inducing catheter-associated urinary tract infections (CAUTIs) due to bacterial adherence to the catheter’s surface, and they exhibit a high friction coefficient, which can greatly affect their effectiveness and functionality. Thus, the development of a silicone urinary catheter with antibacterial properties and lubricity is in strong demand. We hereby developed a poly(vinyl acetate) carrier coating to load chlorhexidine acetate and applied a hydrogel coating primarily composed of polyvinylpyrrolidone (PVP) and poly(ethylene glycol) diacrylate (PEGDA), which was then coated onto the silicone urinary catheters and cured through a thermal curing process and could provide lubricity. Subsequently, we analyzed its surface characteristics and assessed the antibacterial property, lubricity, cytotoxicity, and potential for vaginal irritation. The findings from the Fourier transform infrared spectrometer (FTIR), scanning electron microscope (SEM), water contact angle (WCA), inhibition zone measurements, and friction coefficient analysis confirmed the successful modification of the silicone urinary catheter. Additionally, the outcomes from the cytotoxicity and vaginal irritation assessments demonstrated that the dual-function hydrogel coating-coated silicone urinary catheters exhibit outstanding biocompatibility. This study illustrates that the prepared silicone urinary catheters possess durable antibacterial properties and lubricity, which thus gives them broad clinical application prospects.
