The determination of dissociation constants for active ingredients from herbal extracts using a liquid–liquid equilibrium method

Xingchu Gong,Shichao Huang,Ruonan Jiao,Jianyang Pan,Yao Li,Haibin Qu

A liquid–liquid equilibrium method to determine dissociation constants was presented in this work using phenolic acids in the water extract of Salvia miltiorrhiza as a sample. The models to quantitatively describe the relationship between aqueous phase pH value and apparent distribution coefficient value were built. Apparent distribution coefficients are determined after liquid–liquid equilibrium experiments. The pK a values of phenolic acids in S. miltiorrhiza extracts were fitted successfully. The dissociation constants of active ingredients that are in a mixture in which most components are unknown can be determined using this method. The distribution coefficients of active ingredient molecules and ions can also be obtained. 1-Octanol is a recommended solvent with low solubility, high boiling point, and extra information of K ow .

Discrimination of aqueous and vinegary extracts of Shixiao San using metabolomics coupled with multivariate data analysis and evaluation of anti-hyperlipidemic effect

Xiaofan Wang,Xu Zhao,Liqiang Gu,Yuanyuan Zhang,Kaishun Bi,Xiaohui Chen

A novel study using LC–MS (Liquid chromatography tandem mass spectrometry) coupled with multivariate data analysis and bioactivity evaluation was established for discrimination of aqueous extract and vinegar extract of Shixiao San. Batches of these two kinds of samples were subjected to analysis, and the datasets of sample codes, t R - m / z pairs and ion intensities were processed with principal component analysis (PCA). The result of score plot showed a clear classification of the aqueous and vinegar groups. And the chemical markers having great contributions to the differentiation were screened out on the loading plot. The identities of the chemical markers were performed by comparing the mass fragments and retention times with those of reference compounds and/or the known compounds published in the literatures. Based on the proposed strategy, quercetin-3-O-neohesperidoside, isorhamnetin-3-O-neohespeeridoside, kaempferol-3-O-neohesperidoside, isorhamnetin-3-O-rutinoside and isorhamnetin-3-O-(2G-α-l-rhamnosyl)-rutinoside were explored as representative markers in distinguishing the vinegar extract from the aqueous extract. The anti-hyperlipidemic activities of two processed extracts of Shixiao San were examined on serum levels of lipids, lipoprotein and blood antioxidant enzymes in a rat hyperlipidemia model, and the vinegary extract, exerting strong lipid-lowering and antioxidative effects, was superior to the aqueous extract. Therefore, boiling with vinegary was predicted as the greatest processing procedure for anti-hyperlipidemic effect of Shixiao San. Furthermore, combining the changes in the metabolic profiling and bioactivity evaluation, the five representative markers may be related to the observed anti-hyperlipidemic effect.

Enhancing the Textural Properties of Tibetan Pig Sausages via Zanthoxylum bungeanum Aqueous Extract: Polyphenol-Mediated Quality Improvements

Jingjing Huang,Haiqiu Wei,Zhang Luo,Liang Li,Zhendong Liu,Ningning Xie

The mechanistic effects ofZanthoxylum bungeanumon the textural properties of Tibetan pig sausages remain inadequately elucidated. We conducted a dose–response analysis usingZ. bungeanumaqueous extraction (ZBAE) containing 36.97 mg GAE/g polyphenols, applied at concentrations from 0.125% to 1.20% in the meat paste. Optimal textural enhancement was achieved at 0.25% ZBAE, as proved by significantly improving water holding capacity (1.77% increase), hardness (139.87% increase), and gel strength (46.04% increase) relative to the control group (p< 0.05). Specifically, this concentration: (i) promoted protein molecular rearrangement of by enhancing hydrophobic interactions (33.33% increase) and hydrogen bonding (287.99% increase); (ii) induced conformational transitions fromα-helix (42.62% decrease) toβ-sheet formations (21.11% increase); and (iii) generated a homogeneous three-dimensional protein network characterized by a fractal dimension of 2.769 ± 0.006 and a porosity of 38.350 ± 0.333%. The addition of natural polyphenols from Z.bungeanummay optimize the textural quality of processed meat products.

Evaluation of antinociceptive and anti-inflammatory effects of aqueous extract of Armadillidium vulgare Latreille

Guo Shen-shen,Ren Meng-yue,Song Shuai,Wei Ping,Luo Jia-bo

Objective To assess the antinociceptive and anti-inflammatory properties of the aqueous extract of Armadillidium vulgare (AV). Methods The antinociceptive effect of AV (400, 600 and 800 mg/kg) was investigated in mice using the acetic acid-induced writhing, formalin-induced nociceptive, and hot plate tests. Phlogogen-induced paw edema using carrageenan, dextran, or compound 48/80 as phlogogen was used as inflammatory models to evaluate AV's anti-inflammatory effect. Additionally, the bioactive substances glucosamine (GLcN) and taurine in AV were determined using high-performance liquid chromatography. Results Oral treatment of the mice with AV (600 and 800 mg/kg) significantly reduced the number of writhes in the acetic acid-induced writhing test ( P <0.01) but not the hot plate test ( P >0.05). All doses tested significantly inhibited paw-withdrawal during the second phase of the formalin-induced nociceptive model ( P <0.01). AV demonstrated a strong anti-inflammatory effect in all those inflammatory models ( P <0.05). Conclusions AV has antinociceptive and anti-inflammatory effects, providing scientific evidence of the efficacy of its traditional use in pain treatment. Furthermore, GLcN and taurine contribute, at least in part, to the anti-inflammatory activity of AV.

Human Gastrointestinal Metabolism of the Cistanches Herba Water Extract in Vitro: Elucidation of the Metabolic Profile Based on Comprehensive Metabolite Identification in Gastric Juice, Intestinal Juice, Human Intestinal Bacteria, and Intestinal Microsomes

Yang Li,Ying Peng,Mengyue Wang,Pengfei Tu,Xiaobo Li

Cistanches Herba is taken orally as a health food supplement and medicinal plant in Asian countries. It consists of the stems of Cistanche deserticola (CD) and Cistanche tubulosa (CT). The gastrointestinal metabolism of the multiple components contained in Cistanches Herba is crucial for the discovery of bioactive constituents. This study aims to elucidate the comprehensive metabolic profile of the Cistanches Herba water extract by simulating human gastrointestinal metabolism in vitro independently and sequentially using four models: gastric juice, intestinal juice, human intestinal bacteria, and human intestinal microsomes. A total of 35 and 18 metabolites were characterized from CD and CT water extracts, respectively. These metabolites were formed through reduction, methylation, dimethylation, deglycosylation, decaffeoyl, derhamnose, dehydrogenation, and glucuronidation. The difference in metabolites of the Cistanches Herba water extract and single compounds and the difference in metabolites of CD and CT water extracts were caused by the oligosaccharides and polysaccharides in Cistanches Herba.

Anti-cyclooxygenase, anti-glycation, and anti-skin aging effect of Dendrobium officinale flowers’ aqueous extract and its phytochemical validation in aging

Zhou Huiji,Zhou Luxian,Li Bo,Yue Rongcai

Introduction: Dendrobium officinale Kimura et Migo (D. officinale) , widely called as “life-saving immortal grass” by Chinese folk, is a scarce and endangered species. The edible stems of D. officinale have been extensively studied for active chemical components and various bioactivities. However, few studies have reported the well-being beneficial effects of D. officinale flowers (DOF). Therefore, the present study aimed to investigate the in vitro biological potency of its aqueous extract and screen its active components. Methods: Antioxidant tests, including 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), the ferric reducing ability of plasma (FRAP), and intracellular reactive oxygen species (ROS) level analyses in primary human epidermal keratinocytes, anti-cyclooxygenase2 (COX-2) assay, anti-glycation assay (both fluorescent AGEs formation in a BSA fructose/glucose system and glycation cell assay), and anti-aging assay (quantification of collagen types I and III, and SA-β-gal staining assay) were conducted to determine the potential biological effects of DOF extracts and its major compounds. Ultra-performance liquid chromatography-electrospray ionisation-quadrupole-time-of-flight-mass spectrometry (UPLC-ESI-QTOF-MS/MS) was performed to investigate the composition of DOF extracts. Online antioxidant post-column bioassay tests were applied to rapidly screen major antioxidants in DOF extracts. Results and discussion: The aqueous extract of D. officinale flowers was found to have potential antioxidant capacity, anti-cyclooxygenase2 (COX-2) effect, anti-glycation potency, and anti-aging effects. A total of 34 compounds were identified using UPLC-ESI-QTOF-MS/MS. Online ABTS radical analysis demonstrated that 1-O-caffeoyl-β-D-glucoside, vicenin-2, luteolin-6-C-β-D-xyloside-8-C-β--D-glucoside, quercetin-3-O-sophoroside, rutin, isoquercitrin, and quercetin 3-O-(6″-O-malonyl)-β-D-glucoside are the major potential antioxidants. In addition, all selected 16 compounds exerted significant ABTS radical scavenging ability and effective AGE suppressive activities. However, only certain compounds, such as rutin and isoquercitrin, displayed selective and significant antioxidant abilities, as shown by DPPH and FRAP, as well as potent COX-2 inhibitory capacity, whereas the remaining compounds displayed relatively weak or no effects. This indicates that specific components contributed to different functionalities. Our findings justified that DOF and its active compound targeted related enzymes and highlighted their potential application in anti-aging.

Insights into dynamic evolution of glucuronofucogalactoglucan from water extract of Agrocybe cylindracea during maturation

Jin-Jin Liu,Shi-Kang Chen,Hui Luo,Yan Wang,Xiao-Xiao Song,Wei-Wei He,Xiao-Jun Huang,Jun-Yi Yin,Shao-Ping Nie

This study explored the physicochemical properties and structural characteristics of Agrocybe cylindracea polysaccharides at four developmental stages, as well as their dynamic evolution during maturation. Results showed that the polysaccharides from A. cylindracea water extract exhibited similar structural characteristics across all four maturity stages, despite a significant reduction in yields. Four water-soluble heteroglycans, including one high molecular weight (ACPM-Et50-I) and three low molecular weight (ACPM-Et50-II, ACPM-Et60, ACPM-Et80), were isolated from A. cylindracea at each maturity stage. ACPM-Et50-I was identified as branched heterogalactans, while ACPM-Et60 and ACPM-Et80 were branched heteroglucans. However, ACPM-Et50-II was characterized as a branched glucuronofucogalactoglucan at the tide-turning stage but a glucuronofucoglucogalactan at the pileus expansion stage due to the increase of its α-(1 → 6)-D-Gal p . In general, although the structural skeletons of most A. cylindracea heteroglycans were similar during maturation as shown by their highly consistent glycosyl linkages, there were still differences in the distribution of some heteroglucans. This work has for the first time reported a glucuronofucogalactoglucan in A. cylindracea and its dynamic evolution during maturation, which may facilitate the potential application of A. cylindracea in food and biomedicine industries.

Pharmacokinetics and tissue distribution of cantharidin after oral administration of aqueous extracts from Mylabris in rats

Cancan Duan,Weina Cheng,Qihong Chen,Xiaofei Li,Jianyong Zhang

A sensitive gas chromatography–mass spectroscopy method was established for the determination of cantharidin (CTD) in rat plasma and liver homogenates. During the experiment, rats were randomly divided into two groups (low, high) and were administered aqueous extract of Mylabris compound for 7 days. Then, plasma and tissue samples were taken at different time points to study the pharmacokinetics and tissue distribution of CTD in rats. The selected reaction monitoring transitions for CTD and clofibrate (internal standard) were m / z 128 → 85 and m / z 169 → 141, respectively. The calibration curve ranged from 10.26 to 3,078 ng/ml for plasma and from 10.26 to 246.24 ng/ml for liver homogenates. The lower limits of quantification were 10.26 ng/ml for both plasma and liver. The intra- and inter-day precision and accuracy were <20% for both plasma and liver homogenates. Extraction recovery ranged from 89.21 to 103.61% for CTD in rat plasma and liver and from 83.79 to 102.74% for IS in rat plasma and liver. Matrix effects ranged from 93.06 to 110.44% for CTD and from 91.65 to 110.80% for IS.

Water Extract of Senecio scandens Buch.-Ham Ameliorates Pruritus by Inhibiting MrgprB2 Receptor

Fan Ye,Yucui Jiang,Jian Zhang,Yingxin Zong,Mei Yu,Cuihua Chen,Chan Zhu,Yan Yang,Keke Jia,Gongxi Chen,Zongxiang Tang

Background Senecio scandens Buch.-Ham (S. scandens) belongs to the Compositae family. As a Traditional Chinese medicine, S. scandens has been used in China to treat conjunctivitis, mastitis and vaginitis, it also has the function of antibacterial and relieving itching. Methods Water extract of S. scandens (WSS) was prepared and its quality was controlled by HPLC. The antipruritic effects of WSS were evaluated by itch behavioral experiments. The oxazolone and compound 48/80 were induced to mice scratch behavior, scratch was recorded 30 min after sensitization. The relationship between the antipruritic mechanism and MrgprB2 on mast cell was studied by using mast cell-deficient Kit (W-sh) “Sash” mice and MrgprB2−/− mice. The mast cells were observed by toluidine blue staining. In vitro, the effects of WSS on MrgprB2 were studied by calcium imaging; The whole-cell patch clamp method recorded the MrgprB2 mediate voltage-dependent currents in mast cells. Results The content of rutin (0.012%) and hyperin (0.014%) in the WSS were determined. WSS could ameliorate the pruritus induced by Oxazolone (inhibition was 41.19%, p = 0.004) and compound 48/80 (inhibition was 50.29%, p = 0.001). Meanwhile, WSS could reduce the number of mast cells in mice skin tissue with allergic contact dermatitis (ACD) (p = 0.002) or compound 48/80 (p = 0.013). In addition, WSS could inhibit the calcium influx (1 mg/mL: p = 0.001, 3 mg/mL: p < 0.0001) and the voltage-dependent currents induced by activation of MrgprB2 on mast cell. WSS also attenuated the calcium influx induced by compound 48/80 in HEK293 cells overexpressing MrgprB2/X2.Conclusion These results showed that WSS could ameliorate pruritus by inhibiting MrgprB2 receptor on mast cells.

Laxative Ingredients of Fresh Hemerocallis citrina Baroni

Yuan Hu,FaYing Jiang,Honglin Jiang,Yishuo Zhu,Lewei Zhao,Fengyi Qin,Xiubin Liu,Jianguo Zeng,Zhixing Qing

Hemerocallis citrina Baroni is a healthy vegetable, and consumption of fresh H. citrina can lead to defecation. However, the laxative ingredients are still not known. In the present study, the laxative effect of fresh H. citrina extracts was primarily evaluated by loperamide-induced constipated mice, and the aqueous extracts (1500 and 3000 mg/kg) showed a significant laxative effect. The aqueous extracts were divided into three parts (0%, 30%, and 50% HCW) by macroreticular resin, and all of them were further evaluated. Based on comprehensive analysis of multiple indices, the constipation-relieving effect of 30% HCW was found to be superior to that of 0% and 50% HCW. A total of 16 compounds were identified from 30% HCW by the ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry (UPLC-Q-TOF-MS) method. Three high-level ingredients including chlorogenic acid (3), 4- O -p-coumaroylquinic acid (6) and rutin (13) were selected to further evaluate their laxative effect. The experimental results demonstrate that 4- O -p-coumaroylquinic acid increases defecation frequency in constipated mice, enhances fecal moisture content, and promotes small intestinal motility in constipated mice. The high-dose 4- O -p-coumaroylquinic acid treatment group exhibited higher mean small intestinal transit rates compared to the high-dose chlorogenic acid and rutin groups. Additionally, this compound effectively alleviated loperamide-induced pathological damage, including thinning of the muscle layer and reduced goblet cell numbers, thereby enhancing small intestinal motility, increasing fecal moisture content, and facilitating defecation. This is the first study to identify 4- O -p-coumaroylquinic acid as a key laxative component in fresh H. citrina Baroni, providing novel insights into its application in functional food or pharmaceutical development.

Dynamics of α-glucan from Agrocybe cylindracea water extract at different developmental stages and its structure characteristics

Jin-Jin Liu,Yu-Ke Hou,Xin Wang,Wei-Wei He,Xiao-Jun Huang,Jun-Yi Yin,Shao-Ping Nie

Polysaccharides are the important bioactive macromolecules in Agrocybe cylindracea , but their changes are as yet elusive during developmental process. This study investigated the dynamic changes of polysaccharides from A. cylindracea fruiting body water extract at four developmental stages and its structure characteristics. Results revealed that the polysaccharides from A. cylindracea water extract significantly increased at the pileus expansion stage and the increased fraction could be α-glucan. The further purification and identification indicated that this α-glucan was a glycogen. It had typical morphology of β particles with a molecular weight of 1375 kDa. Its backbone comprised α-D-(1 → 4)-Glc p and α-D-(1 → 4,6)-Glc p residues at a ratio of 5:1, terminated by α-D-Glc p residue. Rheological behavior suggested that it was a Newtonian fluid at the concentration of 1 %. In addition, despite both the glycogen and natural starch were composed of d -glucose, they exhibited the entirely distinct Maltese cross characteristic and unique crystalline structure. This study is the first to demonstrate the presence of abundant glycogen in the pileus expansion stage of A. cylindracea , which provides new insights on the change patterns of fungal polysaccharides.
