Rice root Fe plaque enhances oxidation of microbially available organic carbon via Fe(III) reduction-coupled microbial respiration

Jinzhi Yao,Shuping Qin,Ting Liu,Tim J. Clough,Nicole Wrage-Mönnig,Jiafa Luo,Chunsheng Hu,Tida Ge,Shungui Zhou

Poorly crystalline iron (Fe) oxides are commonly deposited on the surface of rice roots, forming an Fe plaque. This Fe plaque is an important area for Fe redox reactions because the poorly crystalline Fe is available for microbial transformations. However, it remains unclear what the mechanisms are that cause the root Fe plaque to affect CO 2 emissions from paddy soils . Thus, this study investigated the effects of Fe plaque on paddy soil CO 2 emissions and the associated mechanisms. Paddy soil with rice plant roots containing an Fe plaque-coating had 1.5-fold larger CO 2 emissions and higher expression levels of genes involved in soil carbon (C) degradation than Fe plaque-free roots, indicating that Fe plaque stimulated paddy soil derived CO 2 emissions. Fe plaque-coated rice roots, immersed in a 13 C-labeled glucose solution, emitted more CO 2 , with a higher abundance of 13 CO 2 , than the Fe plaque-free roots. These differences were not observed when the surfaces of the rice roots were sterilized. These results indicate that the Fe plaque-stimulated CO 2 emissions were due to soil microbial respiration , not autotrophic root respiration . Chelating the dissolved Fe on the root surface eliminated Fe plaque stimulation of CO 2 emissions, while Fe(III) supplementation correlated with enhanced CO 2 emissions. The stoichiometric mole ratio of the enhanced CO 2 emissions to the Fe(II) generated was comparable to the theoretical ratio of Fe(III) when used as an electron acceptor for organic C decomposition. These results showed that the Fe plaque enhancement of CO 2 emissions was coupled to Fe(III) reduction. Limiting the stimulating effects of Fe plaque on CO 2 emissions may be a potentially useful approach for mitigating organic C loss from paddy soils.

NIR Fluorescent Probe for In Situ Bioimaging of Endogenous H2S in Rice Roots under Al3+ and Flooding Stresses

Jian Wang,Hui Xie,Haoyang Li,Rong Wang,Bo Zhang,Tianrui Ren,Jianli Hua,Nan Chen

Hydrogen sulfide (H2S) is one of the typical reactive sulfur species, which exhibits an important role in regulating both physiological and pathological processes. Recent studies indicate that H2S also serves as a key signaling molecule in a broad range of regulatory processes in plants. However, in situ imaging and detection of the levels of H2S in plant tissues remains a challenge. In this work, a NIR fluorescent probe (HBTP-H2S) was synthesized to achieve H2S imaging in living plant tissues. HBTP-H2S exhibited high sensitivity toward H2S with a large Stokes shift (250 nm). HBTP-H2S could be applied to HeLa cells to detect the fluctuation of endogenous H2S levels in response to physiological stimulations. Importantly, HBTP-H2S was utilized for direct H2S imaging of rice roots and revealed the upregulation of H2S signaling in response to aluminum ions and flooding stresses. Our work thus provides a new tool to investigate H2S-involved signal interaction and protective resistance of crops under environmental stresses.

NaCl Facilitates Cell Wall Phosphorus Reutilization in Abscisic Acid Dependent Manner in Phosphorus Deficient Rice Root

Yang Xiaozheng,Liu Yusong,Huang Jing,Tao Ye,Wang Yifeng,Shen Renfang,Zhu Xiaofang

Phosphorus (P) starvation in rice facilitates the reutilization of root cell wall P by enhancing the pectin content. NaCl modulates pectin content, however, it is still unknown whether NaCl is also involved in the process of pectin regulated cell wall P remobilization in rice under P starved conditions. In this study, we found that 10 mmol/L NaCl increased the shoot and root biomasses under P deficiency to a remarkable extent, in company with the elevated shoot and root soluble P contents in rice. Further analysis indicated that exogenous NaCl enhanced the root cell wall P mobilization by increasing the pectin methylesterase activity and uronic acid content in pectin suggesting the involvement of NaCl in the process of cell wall P reutilization in P starved rice roots. Additionally, exogenous NaCl up-regulated the expression of P transporter OsPT6, which was induced by P deficiency, suggesting that NaCl also facilitated the P translocation prominently from root to shoot in P starved rice. Moreover, exogenous abscisic acid (ABA) can reverse the NaCl-mediated mitigation under P deficiency, indicating the involvement of ABA in the NaCl regulated root cell wall P reutilization. Taken together, our results demonstrated that NaCl can activate the reutilization of root cell wall P in P starved rice, which is dependent on the ABA accumulation pathway.

Rice root exudates enhance desorption and bioavailability of phthalic acid esters (PAEs) in soil associating with cultivar variation in PAE accumulation

Pei-Pei Du,Yu-Hong Huang,Huixiong Lü,Lei Xiang,Yan-Wen Li,Hui Li,Ce-Hui Mo,Quan-Ying Cai,Qing X. Li

Phthalic acid esters (PAEs) is a class of prevalent pollutants in agricultural soil , threating food safety through crop uptake and accumulation of PAEs. Accumulation of PAEs varies largely among crop species and cultivars. Nevertheless, how root exudates affect PAE bioavailability , dissipation, uptake and accumulation is still not well understood. In the present study, desorption and pot experiments were designed to investigate how root exudates from high-( Peizataifeng ) and low-( Fengyousimiao ) PAE accumulating rice cultivars affect soil PAE bioavailability, dissipation, and accumulation variation. Rice root exudates including low molecular weight organic acids (LMWOAs) of Peizataifeng and Fengyousimiao could enhance desorption of two typical PAE compounds, di-n-butyl phthalate (DBP) and di-(2-ethylhexyl) phthalate (DEHP), from aged soil to their available fractions by increasing soil dissolved organic carbon (DOC), thus improving their bioavailability in soil. Peizataifeng produced twice higher amounts of oxalic acid , critic acid and malonic acid in root exudates, and exhibited stronger effects on enhancing desorption and bioavailability of DBP and DEHP than Fengyousimiao . Higher (by about 50%) total organic carbon contents of root exudates from Peizataifeng led to higher (by 10–30%) soil microbial biomass carbon and nitrogen than Fengyousimiao , and thus promoted more PAE dissipation from soil than Fengyousimiao . Nevertheless, higher (by 20–50%) soil DOC and significantly higher PAE bioavailability in the soils planted Peizataifeng resulted in greater (by 53–93%) PAE accumulation in roots and shoots of Peizataifeng than Fengyousimiao , confirming by higher (by 1.82–3.48 folds) shoot and root bioconcentration factors of Peizataifeng than Fengyousimiao . This study reveals that the difference in root exudate extent and LMWOAs between Peizataifeng and Fengyousimiao differentiates PAE accumulation.

Effects of rice root exudates on aggregation, dissolution and bioaccumulation of differently-charged Ag nanoparticles

Jiajia Yang,Hongyu Duan,Xiya Wang,Huan Zhang,Zhifeng Zhang

The biological toxicity and eco-environmental risk of metal nanoparticles (MNPs) is closely related to their stability. The stability of MNPs not only depends on their own properties but also on the effects of biological and environmental factors. To better understand the interaction between biological factors and MNPs in aquatic environments, the effects of total rice root exudates (T-RRE) on the aggregation, dissolution and bioaccumulation of Ag nanoparticles (AgNPs) with different surface charges were investigated in detail. Results indicated that T-RRE can induce the aggregation and sedimentation, and hinder the dissolution of polyethyleneimine-coated AgNPs (AgNPs@PEI) with positive surface charges as well as reducing the bioaccumulation of Ag in rice roots. T-RRE had no obvious effect on the dispersion stability of AgNPs@Cit (negatively charged citrate-coated AgNPs) and AgNPs@PVP (near electrically neutral polyvinylpyrrolidone-coated AgNPs), although T-RRE could induce the dissolution of AgNPs@Cit and AgNPs@PVP. In the molecular fractions of T-RRE, high-molecular-weight root exudates (H-RRE) play a key role in inducing the aggregation of AgNPs@PEI and hindering the bioaccumulation of Ag in rice roots. Compared with H-RRE, low-molecular-weight root exudates (L-RRE) can promote the dissolution of AgNPs@Cit and AgNPs@PVP, but it can obviously promote silver accumulation in rice roots. The difference in charge intensity between L-RRE and T-RRE plays a key role in inducing the aggregation and dissolution of AgNPs with different charges. These findings provide a foundation for investigation of the interactions between rice root exudates and nanoparticles with different surface charges in complex environmental systems.
