A Novel AHAS-Inhibiting Herbicide Candidate for Controlling Leptochloa chinensis: A Devastating Weedy Grass in Rice Fields

Yinglu Chen,Yitao Yan,Jie Chen,Ben Zheng,Youwei Jiang,Zhengzhong Kang,Jun Wu

Given the prevalence of the malignant weed Chinese Sprangletop (Leptochloa chinensis (L.) Nees) in rice fields, the development of novel herbicides against this weed has aroused wide interest. Here, we report a novel diphenyl ether-pyrimidine hybrid, DEP-5, serving as a systematic pre/postemergence herbicide candidate for broad-spectrum weed control in rice fields, specifically for L. chinensis. Notably, DEP-5 exhibits over 80% herbicidal activity against the resistant biotypes even at 37.5 g a.i./ha under greenhouse conditions and has complete control of L. chinensis at 150 g a.i./ha in the rice fields. We uncover that DEP-5 acts as a noncompetitive inhibitor of acetohydroxyacid synthase (AHAS) with an inhibition constant (Ki) of 39.4 μM. We propose that DEP-5 binds to AHAS in two hydrophobic-driven binding modes that differ from commercial AHAS inhibitors. Overall, these findings demonstrate that DEP-5 has great potential to be developed into a herbicide for L. chinensis control and inspire fresh concepts for novel AHAS-inhibiting herbicide design.

The divergent effects of nitrate and ammonium application on mercury methylation, demethylation, and reduction in flooded paddy slurries

Ji Chen,Gongren Hu,Jiang Liu,Alexandre J. Poulain,Qiang Pu,Rong Huang,Bo Meng,Xinbin Feng

The production of methylmercury (MeHg) in flooded paddy fields determines its accumulation in rice grains; this, in turn, results in MeHg exposure risks for not only rice-eating humans but also wildlife. Nitrogen (N) fertilizers have been widely applied in rice cultivation fields to supply essential nutrients. However, the effects of N fertilizer addition on mercury (Hg) transformations are not unclear. This limits our understanding of MeHg formation in rice paddy ecosystems. In this study, we spiked three Hg tracers ( 200 Hg II , Me 198 Hg, and 202 Hg 0 ) in paddy slurries fertilized with urea, ammonium, and nitrate. The influences of N fertilization on Hg methylation, demethylation, and reduction and the underlying mechanisms were elucidated. The results revealed that dissimilatory nitrate reduction was the dominant process in the incubated paddy slurries. Nitrate addition inhibited Hg II reduction, Hg II methylation, and MeHg demethylation. Competition between nitrates and other electron acceptors (e.g., Hg II , sulfate, or carbon dioxide) under dark conditions was the mechanism underlying nitrate-regulated Hg transformation. Ammonium and urea additions promoted Hg II reduction, and anaerobic ammonium oxidation coupled with Hg II reduction (Hgammox) was likely the reason. This work highlighted that nitrate addition not only inhibited Hg II methylation but also reduced the demethylation of MeHg and therefore may generate more accumulation of MeHg in the incubated paddy slurries. Findings from this study link the biogeochemical cycling of N and Hg and provide crucial knowledge for assessing Hg risks in intermittently flooded wetland ecosystems .

Widespread occurrence of cyhalofop-resistant Leptochloa chinensis in eastern China was mainly propelled by target-site mutations

Guoqi Chen,Kai An,Jiahao Xue,Wei Deng,Zeyue Huang,Yang Xu

BACKGROUND Various mechanisms or factors may cause the wide infestation of herbicide-resistant weeds with different contributions, such as target-site resistance (TSR), non-target-site resistance (NTSR), hormesis, and dissemination of herbicide-resistant weeds. Here we tested this hypothesis in eastern China in Leptochloa chinensis , for which cyhalofop resistance is a major challenge to rice planting. RESULTS Our results revealed that 30% of the total 242 populations held high (10 < RI < 100) or very high resistance (RI > 100) to cyhalofop. Resistance increased from north to south, and from transplanted rice fields to direct-seeded rice fields. Six mutant types conferring TSR to cyhalofop were found in 89% sampled populations, with 18% carrying two types, 2% carrying three types, and another 2% carrying four types of mutations. W2027C was the most frequent TSR mutation, present in 47.3% of sampled populations, followed by I2041N (21.8%), W1999C (20%), L1818F (20%), W1999G (5.5%) and C2088R (1.8%). Non-target-site resistance linked with P450 monooxygenases (P450) and glutathione S-transferases (GST) were observed in 7% and 10% cyhalofop-resistant populations sampled. No significant correlation between genetic and geographic distances were observed among cyhalofop-resistant populations. Hormetic effects of cyhalofop to L. chinensis were evident in 13% of overall populations. CONCLUSIONS This study compared the contribution of different mechanisms towards herbicide resistance, revealed herbicide hormesis and revealed the distribution of resistance. Insights from this study could be important for weed management practices in other areas of the world. © 2025 Society of Chemical Industry.

Irrigation-introduced quinolone antibiotics activated the soil-water interface redistribution of per- and polyfluoroalkyl substances in paddy fields

Jianyi Wu,Xinlin Huang,Miao Chen,Lingxuan Li,Wenhui Zhao,Wenqing Tu

Quinolone antibiotic pollution (QAP) introduced by irrigation may cause the release of per- and polyfluoroalkyl substances (PFASs) at the soil-water interface in paddy fields, endangering the growth of rice crops and the security of human food sources. Thus, the comprehensive investigation of the joint and individual effects of quinolone antibiotics with environment-relevant concentrations on the soil PFASs migration were conducted. Results showed that the QAP dose of 1 μg/L had the highest activation for the release of PFASs into the overlying water, resulting in the highest net concentration of 99.32 μg/L. Structural equation modeling revealed that the QAP-mediated microbial adsorption was the hub transition for PFASs migration ( p  < 0.001). The PFASs re-partitioning after QAP was primarily driven by physicochemistry and nutrient cycling variations induced by QAP, which was facilitated by soil microbial urease and acid phosphatase. The soil microbial genes nasC , nasE , phnG , nasD , napD , phnA , nirK , nosZ , and nasB were identified as the critical regulators of PFASs migration under QAP by the aggregate boosted tree modeling. Molecularly, the soil microbial metabolisms of Galactosylglycerol and 3-beta-D-Galactosyl-sn-glycerol were responsible for PFASs release under the QAP level of 1 μg/L ( p  < 0.001). These findings were beneficial in elucidating the ecological risks of bioavailable PFASs in paddy fields subjected to the integrated pollution of quinolone antibiotics.
