Toxicity of Arsenic to Photobacterium phosphoreum, Daphnia magna, and Danio rerio at Different pH Levels

Qun He,Ruijuan Qu,Xinghao Wang,Zhongbo Wei,Ping Sun,Zunyao Wang

Although it has been widely known that arsenic poses great threat to organisms and arsenic species account for the toxic effect of the metalloid, relatively little attention has been given to the arsenic toxicity to different organisms at different pH values. Three kinds of aquatic organisms including Photobacterium phosphoreum , Daphnia magna , and Danio rerio were used to study the effects of pH on the toxicity of arsenic. Toxicity test results were analyzed by calculating EC 50 values and for arsenic species at five different pH values (5.0, 6.0, 7.0, 8.0, and 9.0). As a result, As(III) is more toxic to the tested organisms than As(V), and P. phosphoreum was more sensitive to the toxicity of arsenic than D. magna and D. rerio . The toxicity of As(III) species is different for different organisms: dihydrogen arsenite, H 2 AsO 3 – , is more toxic to P. phosphoreum and D. rerio than arsenous acid, H 3 AsO 3 , while H 3 AsO 3 is more toxic to D. magna . As for As(V), arsenate, HAsO 4 2− , is the most toxic to P. phosphoreum , whereas dihydrogen arsenate, H 2 AsO 4 – , may be responsible for the toxicity to D. magna and D. rerio . As(III) and As(V) are more toxic to P. phosphoreum in an alkaline environment, while for D. magna in an acidic environment and As(III) is more toxic to D. rerio in an alkaline environment and As(V) in an acidic environment.

Using DeepLabCut for markerless cardiac physiology and toxicity estimation in water fleas (Daphnia magna)

Ferry Saputra,Michael Edbert Suryanto,Gilbert Audira,Cao Thang Luong,Chih-Hsin Hung,Marri Jmelou Roldan,Ross D. Vasquez,Chung-Der Hsiao

Daphnia magna is one species of water flea that has been used for a long time for ecotoxicity studies. In addition, Daphnia has a myogenic heart that is very useful for cardiotoxicity studies. Previous attempts to calculate the cardiac parameter endpoints in Daphnia suffer from the drawback of tedious operation and high variation due to manual counting errors. Even the previous method that utilized deep learning to help the process suffer from either overestimation of parameters or the need for specialized equipment to perform the analysis. In this study, we utilized DeepLabCut software previously used for animal pose tracking and demonstrated that ResNet_152 was the best fit for training the network. The trained network also showed comparable results with ImageJ and Kymograph, which was mostly done manually. In addition to that, several macro scripts in either Excel or Python format were developed to help summarize the data for faster analysis. The trained network was then challenged to analyze the potential cardiotoxicity of imidacloprid and pendimethalin in D. magna , and it showed that both pesticides cause alteration in their cardiac performance. Overall, this method provides a simple and automatic method to analyze the cardiac performance of Daphnia by utilizing DeepLabCut . The method proposed in this paper can contribute greatly to scientists conducting fast and accurate cardiotoxicity measurements when using Daphnia as a model.

Multiple Screening of Pesticides Toxicity in Zebrafish and Daphnia Based on Locomotor Activity Alterations

Akhlaq Hussain,Gilbert Audira,Nemi Malhotra,Boontida Uapipatanakul,Jung-Ren Chen,Yu-Heng Lai,Jong-Chin Huang,Kelvin H.-C. Chen,Hong-Thih Lai,Chung-Der Hsiao

Pesticides are widely used to eradicate insects, weed species, and fungi in agriculture. The half-lives of some pesticides are relatively long and may have the dire potential to induce adverse effects when released into the soil, terrestrial and aquatic systems. To assess the potential adverse effects of pesticide pollution in the aquatic environment, zebrafish (Danio rerio) andDaphnia magnaare two excellent animal models because of their transparent bodies, relatively short development processes, and well-established genetic information. Moreover, they are also suitable for performing high-throughput toxicity assays. In this study, we used both zebrafish larvae and water flea daphnia neonates as a model system to explore and compare the potential toxicity by monitoring locomotor activity. Tested animals were exposed to 12 various types of pesticides (three fungicides and 9 insecticides) for 24 h and their corresponding locomotor activities, in terms of distance traveled, burst movement, and rotation were quantified. By adapting principal component analysis (PCA) and hierarchical clustering analysis, we were able to minimize data complexity and compare pesticide toxicity based on locomotor activity for zebrafish and daphnia. Results showed distinct locomotor activity alteration patterns between zebrafish and daphnia towards pesticide exposure. The majority of pesticides tested in this study induced locomotor hypo-activity in daphnia neonates but triggered locomotor hyper-activity in zebrafish larvae. According to our PCA and clustering results, the toxicity for 12 pesticides was grouped into two major groups based on all locomotor activity endpoints collected from both zebrafish and daphnia. In conclusion, all pesticides resulted in swimming alterations in both animal models by either producing hypo-activity, hyperactivity, or other changes in swimming patterns. In addition, zebrafish and daphnia displayed distinct sensitivity and response against different pesticides, and the combinational analysis approach by using a phenomic approach to combine data collected from zebrafish and daphnia provided better resolution for toxicological assessment. Keywords:zebrafish;daphnia;pesticide;locomotion;behavior;phenomics

Transgenerational Effects of Atrazine on Daphnia magna Based on Life-History Traits and Population Dynamics

Wang Jun,Li Meng,Gu Lei,Sun Yunfei,Zhu Xuexia,Yang Zhou

Atrazine is a predominant herbicide globally, and its residues are commonly found in natural water bodies due to its extensive use. Atrazine is known for its detrimental effects on the reproductive abilities of aquatic plants and animals. Our study explored the impact of maternal exposure to atrazine on the survival and performance of offspring using the water flea Daphnia magna as a model organism. We investigated how the life-history traits and population dynamics of maternal D. magna and their offspring are influenced by atrazine. We hypothesized that exposure to atrazine in mothers would negatively affect the survival and performance of their offspring. The findings revealed that short-term exposure to atrazine adversely affects fitness-related traits, manifesting as decreased survivorship and reduced fecundity. Offspring from atrazine-exposed mothers displayed a limited recovery in fitness; no mortality was observed during the 14-day experiment, yet fecundity remained significantly reduced. At the population level, maternal exposure to atrazine led to a decreased population carrying capacity. The notable maternal effects on offspring reproduction underscore the long-term toxicity of atrazine to D. magna populations, suggesting prolonged consequences even after the cessation of atrazine pollution. This study underscores the transgenerational effects of atrazine, highlighting its persistent threat to D. magna populations.

The detrimental effects and mechanisms of Orlistat in disrupting energy homeostasis and reproduction in Daphnia magna

Zhihua Ni,Xinling Tian,Wenbo Zhao,Wenkai Hu,Jinghua Lv,Xiaoli Sun,Yajie Zhang,Yiwen Zhang,Yuming Zhang,Baoku Li,Fengsong Liu

Orlistat (ORL) has been employed as an anti-obesity pharmaceutical for several decades. Given its low absorption rate, the majority of administered ORL is excreted into the environment with feces. It is crucial to collect scientific information regarding the possible ecological risks associated with ORL. Here, the effects of ORL on Daphnia magna were evaluated using a 21-day chronic test at concentrations of 1, 10, 100, and 1000 μg/L. We found the inhibition of feeding and swimming activities in the 100 and 1000 μg/L ORL exposed D. magna , respectively. Their digestive enzyme activities and metabolites were reduced even at 1 μg/L ORL exposure. It is noteworthy that exposure to 100 μg/L ORL induced a decrease in the reproductive capacity of D. magna , although no discernible genotoxicity was observed. To identify the toxicological mechanisms of ORL, a metabolic analysis was conducted on D. magna exposed to 1000 μg/L ORL. A comprehensive reduction in carbohydrates, lipids, and amino acids was observed, which resulted in a blockage of metabolic flux towards the TCA cycle, as evidenced by mitochondrial dysfunction. These findings substantiate the detrimental impact of ORL on D. magna and provide insights into the underlying molecular mechanisms from a metabolic perspective.

Investigation on toxicity and mechanism to Daphnia magna for 14 disinfection by-products: Enzyme activity and molecular docking

Jin J. Li,Min J. Dai,Jun Z. Xue

Exposure to disinfection by-products (DBPs) has been found to induce a range of toxic effects in aquatic organism. Previous studies have consistently demonstrated that a majority of DBPs have the ability to induce in vivo toxicity in aquatic organisms. However, the impact of DBPs on the metabolic processes of Daphnia magna ( D. magna ) and the underlying molecular toxicity mechanisms are still not well understood. Therefore, we investigated the effects of 14 DBPs on two oxidative stress enzymes and malondialdehyde (MDA) levels in D. magna . Additionally, we employed molecular docking to simulate the toxicity of DBPs to D. magna at the molecular level. This comprehensive analysis allowed us to gain further insights into the toxicity of DBPs on D. magna . The results showed that among the aliphatic DBPs, the more bromine substituents, the lower the toxicity effect, and it's opposite in the aromatic DBPs. In the detection of oxidative stress level, catalase (CAT) enzyme and superoxide dismutase (SOD) enzyme in D. magna under compound stress showed a low increase and decrease with the increase of concentration. The level of MDA showed a positive correlation with the concentration. In the last, molecular docking simulations have shown promise in predicting the toxicity of DBPs and providing insights into their toxic effects to a certain extent, and the docking situation of P53 is slightly different. Hence, it is imperative to further regulate the presence of aromatic DBPs due to their pronounced toxic effects on D. magna , and these simulations can be complemented with actual experiments to enhance our understanding of the toxicity mechanisms of DBPs.

Evaluation of the Acute Effects and Oxidative Stress Responses of Phenicol Antibiotics and Suspended Particles in Daphnia magna

Yuxuan Zhang,Peiyong Guo,Yanmei Wu,Meixian Wang,Jun Deng,Haitao Su,Yinshi Sun

Suspended particles (SP) exist widely in various water systems and are able to adsorb other pollutants in water, producing ecotoxic effects on aquatic nontarget species. Until now, however, few studies have focused on the effects of SP on antibiotics. Therefore, the present study investigated the effects of the mixtures of SP and phenicol antibiotics (chloramphenicol [CAP], thiamphenicol [TAP]) on acute toxicity and oxidative stress responses in Daphnia magna . The results indicated that the acute toxicity of phenicol antibiotics in D. magna was increased when combined with SP. Besides, the immobilization of daphnids caused by phenicol drugs in the presence of 10 mg/L of SP was more intense than that with 200 mg/L of SP. Furthermore, the impact of SP with diverse concentrations on the activity of catalase and the level of reduced glutathione in D. magna was different. Notably, almost all CAP + TAP + SP treatments markedly increased malondialdehyde content in D. magna , causing potential cellular oxidative damage in D. magna . In summary, the present study provides insights into the toxic effects of phenicol antibiotic and SP mixtures on aquatic organisms. Environ Toxicol Chem 2021;40:2463–2473. © 2021 SETAC

Cardiovascular Performance Measurement in Water Fleas by Utilizing High-Speed Videography and ImageJ Software and Its Application for Pesticide Toxicity Assessment

Fiorency Santoso,Viacheslav V. Krylov,Agnes L. Castillo,Ferry Saputra,Hong-Ming Chen,Hong-Thih Lai,Chung-Der Hsiao

Simple SummaryWith the advantages of easy culture, body transparency, and high sensitivity to chemical pollution, water fleas have been recognized as a good model for ecotoxicity studies. In this paper, we established ImageJ-based methods to measure cardiovascular performance by evaluating the heart rate and blood flow velocity in three water fleas for the first time. Among the three water fleas,Daphnia magnawas identified as having the most robust heartbeat and blood flow rate and is therefore suitable for ecotoxicity assessment. Many important parameters like heart rate, blood flow rate, stroke volume, ejection fraction, fractional shortening, cardiac output heartbeat regularity can be extracted from videotaping and mathematical calculation. In utilizing those physiological parameters, the potential impacts of ambient water temperature and pesticide pollution on water fleas can be precisely measured.AbstractWater fleas are a good model for ecotoxicity studies, and were proposed for this purpose by the United States Environmental Protection Agency, due to their easy culture, body transparency, and high sensitivity to chemical pollution. Cardiovascular function parameters are usually used as an indicator of toxicity evaluation. However, due to the nature of the heart and blood flow, and the speed of the heartbeat, it is difficult to perform precise heartbeat and blood flow measurements with a low level of bias. In addition, the other cardiovascular parameters, including stroke volume, cardiac output, fractional shortening, and ejection fraction, have seldom been carefully addressed in previous studies. In this paper, high-speed videography and ImageJ-based methods were adopted to analyze cardiovascular function in water fleas. The heartbeat and blood flow for three water flea species,Daphnia magna,Daphnia silimis, andMoinasp., were captured by high-speed videography and analyzed using open-source ImageJ software. We found the heartbeat is species-dependent but not size-dependent in water fleas. Among the three water fleas tested,D. magnawas identified as having the most robust heartbeat and blood flow rate, and is therefore suitable for the ecotoxicity test. Moreover, by calculating the diameter of the heart, we succeeded in measuring other cardiovascular parameters.D. magnawere challenged with temperature changes and a pesticide (imidacloprid) to analyze variations in its cardiovascular function. We found that the heartbeat ofD. magnawas temperature-dependent, since the heartbeat was increasing with temperature. A similar result was shown in the cardiac output parameter. We also observed that the heartbeat, cardiac output, and heartbeat regularity are significantly reduced when exposed to imidacloprid at a low dose of 1 ppb (parts per billion). The blood flow rate, stroke volume, ejection fraction, and fractional shortening, on the contrary, did not display significant changes. In conclusion, in this study, we report a simple, highly accurate, and cost-effective method to perform physiological and toxicological assessments in water fleas. Keywords:water flea;Daphnia;cardiovascular performance;ImageJ

Exposure Medium: Key in Identifying Free Ag+ as the Exclusive Species of Silver Nanoparticles with Acute Toxicity to Daphnia magna

Shen Mo-Hai,Zhou Xiao-Xia,Yang Xiao-Ya,Chao Jing-Bo,Liu Rui,Liu Jing-Fu

It is still not very clear what roles the various Ag species play in the toxicity of silver nanoparticles (AgNPs). In this study, we found that traditional exposure media result in uncontrollable but consistent physicochemical transformation of AgNPs, causing artifacts in determination of median lethal concentration (LC50) and hindering the identification of Ag species responsible for the acute toxicity of AgNPs to Daphnia magna . This obstacle was overcome by using 8 h exposure in 0.1 mmol L −1 NaNO 3 medium, in which we measured the 8-h LC50 of seven AgNPs with different sizes and coatings and determined the concentrations of various Ag species. The LC50 as free Ag + of the seven AgNPs (0.37–0.44 μg L −1 ) agreed very well with that of AgNO 3 (0.40 μg L −1 ) and showed the lowest value compared to that as total Ag, total Ag + and dissolved Ag, demonstrating free Ag + is exclusively responsible for the acute toxicity of AgNPs to D. magna , while other Ag species in AgNPs have no contribution to the acute toxicity. Our results demonstrated the great importance of developing appropriate exposure media for evaluating risk of nanomaterials.

Residual β-lactam antibiotics and ecotoxicity to Vibrio fischeri, Daphnia magna of pharmaceutical wastewater in the treatment process

Yaru Hu,Dandan Lei,Da Wu,Jing Xia,Wang Zhou,Changzheng Cui

The discharge of pharmaceutical wastewater introduces numerous pollutants into the environment, and their pollution level reduction has aroused extensive concern. This study investigated the variation in residual antibiotics and ecotoxicity to two nutritional-level model organisms in the pharmaceutical wastewater treatment process (PWTP). The wastewater in the equalization tank contained massive organic matters (2.9–18.7 times higher than the permissible values in GB21903-2008) and antibiotics (310.88 μg/L), posing extremely toxic effects to Vibrio fischeri ( V. fischeri ) and Daphnia magna ( D. magna ) . The biological anaerobic/aerobic treatment units contributed the most to the reduction of antibiotics and the ecotoxicity to both organisms, with the removal rates of 72% and > 90%, respectively. The ecotoxicity of pharmaceutical wastewater was strongly and positively correlated with the residual antibiotics, amoxicillin, cephalexin, ammonia nitrogen, and total phosphorus ( P  < 0.05). However, the detected amounts of amoxicillin and cephalexin were approximately 10 5 times lower than the predicted no-effect concentrations of amoxicillin and cephalexin to V. fischeri and D. magna in freshwater, which implied the joint ecotoxicity posed by multicomponent mixtures, such as the residual antibiotics and organic toxic substances, rather than the specific residual antibiotics. This study provides a better understanding of the variations and residual levels of pollutants in PWTPs , including their ecotoxicity risk to the aquatic environment, highlighting the need to optimize pharmaceutical wastewater treatment technologies .

Evaluation of the subtle effects and oxidative stress response of chloramphenicol, thiamphenicol, and florfenicol in Daphnia magna

Zhang Yuxuan,Guo Peiyong,Wu Yanmei,Zhang Xiaoyan,Wang Meixian,Yang Simin,Sun Yinshi,Deng Jun,Su Haitao

Phenicol antibiotics, such as chloramphenicol, thiamphenicol, and florfenicol, are commonly used in the veterinary and aquaculture fields to treat diseases and have frequently been detected in aquatic environments. Nevertheless, there is limited information regarding the effects of phenicol antibiotics on aquatic nontarget species. Thus, the present study aims to investigate the long-term (21-d) influence on the reproduction and growth of and the acute (24-h) oxidative response and tissue damage in the crustacean Daphnia magna after exposure to phenicol drugs, including their environmental concentrations. The results indicate that D. magna exposed to florfenicol are likely to cause more adverse effects than those exposed to chloramphenicol and thiamphenicol over long-term (21-d) exposures. Furthermore, changes in biochemical biomarkers such as malondialdehyde (MDA), catalase (CAT), and reduced glutathione (GSH) induced by individual and mixtures of phenicol antibiotics were also observed. Low concentrations of chloramphenicol, thiamphenicol + florfenicol, and chloramphenicol + thiamphenicol significantly increased the MDA levels of D. magna after 24-h exposures, causing cellular oxidative damage in the animals. In addition, discrepancies between CAT activities and GSH levels were observed, underscoring the need to evaluate multiple indicators of oxidative stress in toxicological studies using D. magna as a model. The toxicity of cadmium to three aquatic organisms (Photobacterium phosphoreum, Daphnia magna and Carassius auratus) under different pH levels

R.-J. Qu,X.-H. Wang,M.-B. Feng,Y. Li,H.-X. Liu,L.-S. Wang,Z.-Y. Wang

This study investigated the effect of pH on cadmium toxicity to three aquatic organisms: Photobacterium phosphoreum , Daphnia magna and Carassius auratus . The acute toxicity of Cd 2+ to P. phosphoreum and D. magna at five pH values (5.0, 6.0, 7.0, 8.0, and 9.0) was assessed by calculating EC 50 values. We determined that Cd 2+ was least toxic under acidic conditions, and D. magna was more sensitive to the toxicity of Cd than P. phosphoreum . To evaluate Cd 2+ -induced hepatic oxidative stress in C. auratus at three pH levels (5.0, 7.25, 9.0), the activity of antioxidant enzymes (superoxide dismutase, catalase and glutathione peroxidase), the level of glutathione and the malondialdehyde content in the liver were measured. Oxidative damage was observed after 7   d Cd exposure at pH 9.0. An important finding of the current research was that Cd 2+ was generally more toxic to the three test organisms in alkaline environments than in acidic environments.
