Seed priming with brassinosteroids alleviates aluminum toxicity in rice via improving antioxidant defense system and suppressing aluminum uptake

Basit Farwa,Liu Jiaxin,An Jianyu,Chen Min,He Can,Zhu Xiaobo,Li Zhan,Hu Jin,Guan Yajing

Brassinosteroids (BRs) are growth-promoting hormones that exhibit high biological activities across various plant species. BRs shield plants against various abiotic stresses. In the present study, the effect of BRs against aluminum (Al) toxicity was investigated through seed priming with 24-epibrassinolide (0.01 μM) in two different rice cultivars. BRs application was found effective in confronting plants from Al toxicity (400 μM). The rice seeds primed with BRs showed enhancement in seed germination energy, germination percentage, root and shoot length, as well as fresh and dry weight under Al-absence and Al-stressed conditions as compared to water-priming. Especially under Al stress, BRs priming promoted the growth of rice seedlings more obviously. Al toxicity significantly increased the Al contents in seedling root and shoot, as well as the MDA concentration, H 2 O 2 production, and the activities of antioxidative enzymes including ascorbate peroxidase, catalase, and peroxidase. Meanwhile, the photosynthetic pigments of seedling reduced under Al stress. When compared to sensitive cultivar (CY-927), these modifications were more obvious in the tolerant variety (YLY-689). Surprisingly, BRs were able to alleviate the Al injury by lowering MDA and H 2 O 2 level and increasing antioxidant activities and photosynthetic pigments under Al stress. The results on antioxidant activities were further validated by gene expression study of SOD-Cu-Zn , SOD-Fe 2 , CATa , CATb , APX02 , and APX08 . It suggested that BRs were responsible for the mitigation of Al stress in rice seedlings by inducing antioxidant activities with an effective response to other seed growth parameters and reduced Al uptake under induced metal stress.

24-Epibrassinolide enhances 5-ALA-induced anthocyanin and flavonol accumulation in calli of ‘Fuji’ apple flesh

Zheng Jie,An Yuyan,Wang Liangju

Color is a key factor for fruit commercial value. 5-Aminolevulinic acid (5-ALA), as an eco-friendly plant growth regulator, shows an attractively promotive effect on plant secondary metabolism, especially for fruit coloration. Brassinosteroids (BRs) can also improve plant flavonoid biosynthesis. No information is now available on the relationship between 5-ALA and BR. Here, we found that 1.5 mg L −1 24-epibrassinolide (24-EBL) promoted 50 mg L −1 5-ALA-induced anthocyanin accumulation, while, brassinazole (Brz) significantly inhibited the 5-ALA-induced flavonoid accumulation. HPLC analysis further showed that the inductive effects of 5-ALA on the accumulation of cyanidin-3-galactoside, quercetin-3-galactoside, quercetin and kaempferol were elevated by 24-EBL, but impaired by Brz. These results suggest that brassinolide biosynthesis might involve in 5-ALA-induced flavonoid accumulation. Gene expression analysis showed that 5-ALA and 5-ALA + 24-EBL induced the expression of regulatory genes MdMYB10, MdMYB9, MdbHLH3 and MdbHLH33 . These two treatments also up-regulated the structural gene expressions of anthocyanin biosynthesis and transportation, including MdCHS, MdF3′H, MdDFR, MdANS, MdUFGT, MdGST and MdMATE , as well as flavonol biosynthetic gene MdFLS . But Brz decreased 5-ALA-induced up-regulation of these genes. In addition, 5-ALA also induced the expression of MdBRI1, MdBAK1 and MdBZR1 , which are involved in brassinolide signal transduction. These results indicate that 24-EBL enhances 5-ALA-promoted expression of genes related to flavonoid biosynthesis and brassinolide signal transduction, while Brz exhibits the opposite effects. Taken together, we propose that 24-EBL is involved in 5-ALA-induced anthocyanin and flavonol accumulation in calli of apples. Our results provide new insights into 5-ALA-induced fruit coloration.

Seed Priming with Brassinosteroids Alleviates Chromium Stress in Rice Cultivars via Improving ROS Metabolism and Antioxidant Defense Response at Biochemical and Molecular Levels

Farwa Basit,Min Chen,Temoor Ahmed,Muhammad Shahid,Muhammad Noman,Jiaxin Liu,Jianyu An,Abeer Hashem,Al-Bandari Fahad Al-Arjani,Abdulaziz A. Alqarawi,Mashail Fahad S. Alsayed,Elsayed Fathi Abd_Allah,Jin Hu,Yajing Guan

This research was performed to explore the vital role of seed priming with a 0.01 µM concentration of brassinosteroids (EBL) to alleviate the adverse effects of Cr (100 µM) in two different rice cultivars. Seed priming with EBL significantly enhanced the germination attributes (germination percentage, germination energy, germination index, and vigor index, etc.), photosynthetic rate as well as plant growth (shoot and root length including the fresh and dry weight) under Cr toxicity as compared to the plants primed with water. Cr toxicity induced antioxidant enzyme activities (SOD, POD, CAT, and APX) and ROS level (MDA and H2O2contents) in both rice cultivars; however, a larger increment was observed in YLY-689 (tolerant) than CY-927 (sensitive) cultivar. EBL application stimulatingly increased antioxidant enzyme activities to scavenge ROS production under Cr stress. The gene expression of SOD and POD in EBL-primed rice plants followed a similar increasing trend as observed in the case of enzymatic activities of SOD and POD compared to water-primed rice plants. Simultaneously, Cr uptake was observed to be significantly higher in the water-primed control compared to plants primed with EBL. Moreover, Cr uptake was significant in YLY-689 compared to CY-927. In ultra-structure studies, it was observed that EBL priming relieved the rice plants from sub-cellular damage. Conclusively, our research indicated that seed priming with EBL could be adopted as a promising strategy to enhance rice growth by copping the venomous effect of Cr.Keywords:antioxidants;brassinosteroids;chromium;heavy metals;rice

Identification and validation of a major gene for kernel length at the P1 locus in Triticum polonicum

Songyue Chai,Qin Yao,Rui Liu,Wenhui Xiang,Xue Xiao,Xing Fan,Jian Zeng,Lina Sha,Houyang Kang,Haiqin Zhang,Dan Long,Dandan Wu,Yonghong Zhou,Yi Wang

Kernel size, one of the traits that determine wheat yield, is controlled by multiple quantitative trait loci. Polish wheat ( Triticum polonicum ) has elongated and plump kernel and is a valuable material for breeding high-yielding wheat cultivars. However, genes or loci determining kernel length (KL) in Polish wheat are unknown. We identified and validated a major KL gene, KL-PW , at the P1 locus in Polish wheat. KL-PW is VRT-A2 , which encodes a MIKC-type MADS-box protein (MADS55). An insertion/deletion mutation in intron 1 of VRT-A2 PW led to an alternatively spliced transcript, VRT-A2 PW2 . Quantitative PCR analysis showed that VRT-A2 PW was more highly expressed in developing seeds than was VRT-A2 Ailanmai . Brassinosteroid (BR) sensitivity experiment and the expression of BR-related genes indicated that VRT-A2 PW functions as a positive regulator of BR responses. VRT-A2 PW significantly increased KL of wheat. These findings not only reveal the molecular basis of KL-PW in controlling KL, but also provide a valuable genetic resource for increasing kernel size in wheat.

Determination and analysis of solubility of brassinolide in different solvent systems at different temperatures (T = 278.15–323.15 K)

Zilong Cui,Zhiruo Zhao,Liang Yao,Yonghong Hu

Brassinolide is a new environmental protection plant growth regulator. With the aim of knocking down selfcost and scale up agriculture market-place emulation, come what will manufacture method is adopted, the final step crystallization is significant for getting exclusive substances. The purpose of this study is to provide some authentic and general energetics data for the actual industrial production of brassinolide. In the research, the solid–liquid equilibrium of brassinolide in twelve pure solvents (tetrahydrofuran, dimethyl sulfoxide, ethanol, acetone, methanol, trichloromethane, acetonitrile, 1-butanol, isopropanol, xylenes, n-hexane and n-heptane) and three binary solvent mixtures (trichloromethane + tetrahydrofuran, acetonitrile + tetrahydrofuran and n-hexane + tetrahydrofuran) were tested by the circumstance of atmosphere pressure at range temperatures between 278.15 K and 323.15 K. Four models like the Buchowski–Ksiazaczak λh model, the modified Apelblat model, the Jouyban-Acree-van't-Hoff model and the Redich-Kister (CNIBS/R-K) model were employed to describe the change trend of solubility. On the groundwork of a series of data analysis, tetrahydrofuran is a better organic solvent and n-hexane can be applied as an effective anti-solvent when separating out brassinolides. These solubility data are very important for optimizing the extraction process and recrystallization of brassinolide.

Synergistic interaction between brassinosteroid and jasmonate pathways in rice response to cadmium toxicity

Zaoli Zhang,Tingting Zhang,Long Lu,Shunjiao Qiu,Zecong Huang,Yuan Wang,Xinyu Chen,Long Li,Yanyan Sun,Rongjun Zhang,Pinghua Chen,Yuanyuan Song,Rensen Zeng

Brassinosteroids (BRs) and jasmonic acid (JA) are known to be involved in regulating plant responses to cadmium (Cd) stress. However, their specific roles and interaction in this process remain unclear. In this study, we discovered that exogenous BR alleviated Cd-mediated growth inhibition of rice seedlings. Enhanced Cd tolerance was also observed in m107 , a BR-overproduction mutant. Phenotypic analysis of genetic materials involved in BR signaling confirmed the positive role of BR in regulating rice response to Cd toxicity. OsDLT, a key component in the BR signaling pathway, was found to be crucial for BR-mediated Cd tolerance. Further analysis demonstrated that activation of the BR pathway reduced the accumulation of Cd and reactive oxygen species (ROS) by modulating the expression of genes associated with Cd transport and ROS scavenging. Interestingly, transcriptome analysis indicated that the JA pathway was enriched in OsDLT-regulated differently expressed genes (DEGs). Gene expression and hormone assays showed that BR promoted the expression of JA pathway genes and JA levels in plants. Moreover, BR-induced tolerance was compromised in the JA signaling-deficient mutant osmyc2 , suggesting that BR-mediated Cd resistance depends on the activation of the JA signaling pathway. Overall, our study revealed the synergistic interaction between BR and JA pathways in rice response to Cd stress, providing insights into the complex hormonal interplay in plant tolerance to heavy metals.

Brassinosteroid-Mediated Resistance to Cobalt-Induced Toxicity by Regulating Hormonal Balance, Cellular Metabolism, and Antioxidant Defense in Maize

Abdul Salam,Jinzhe Chang,Liupeng Yang,Muhammad Zeeshan,Anas Iqbal,Ali Raza Khan,Muhammad Siddique Afridi,Zaid Ulhassan,Wardah Azhar,Zhixiang Zhang,Peiwen Zhang

Brassinosteroids (BRs) play an essential role in regulating plant metabolic pathways that influence growth, development, and stress responses. However, their role in alleviating cobalt (Co) stress has not been extensively studied. This research aimed to assess the impact of exogenous BRs (0.1 µM) on maize subjected to Co stress (300 µM) in a hydroponic experiment. The results indicated that BR supplementation significantly decreased the accumulation of H2O2by 17.79 and 16.66%, O2•−by 28.5 and 21.48%, and MDA by 37.5 and 37.9% in shoot and root, respectively, as compared to Co stress alone. Additionally, BRs enhanced endogenous levels of BRs (31.16%) and growth hormones (IAA 50.8%, JA 57.8%, GA 52.5%), and reduced Co contents by 26.3% in roots and 36.1% in shoots. BRs enhanced antioxidant enzyme activity both in the shoot and root, leading to reduced ROS levels as confirmed by laser scanning confocal microscopy. Furthermore, BRs increased phenols, flavonoids, and soluble sugars, and elevated total protein content. Observations from transmission electron microscopy indicated reduced ultrastructural damage in plants treated with BRs under Co stress. Taken together, this study highlights the role of BRs in alleviating Co stress in maize, demonstrating their efficiency in enhancing stress tolerance by modulating hormone levels and key metabolic processes.

Abscisic acid and brassinolide combined application synergistically enhances drought tolerance and photosynthesis of tall fescue under water stress

Zhifei Chen,Zhi Wang,Yungui Yang,Min Li,Bingcheng Xu

Tall fescue ( Festuca arundinacea Schreb.) is a widely used cool-season turfgrass, and its growth is mainly limited by water deficit. Abscisic acid (ABA) and brassinolide (BR) are two important stress hormones regulating plant physiological processes and growth under water deficit. To investigate effects of exogenous ABA and BR on physiological and photosynthetic performance of tall fescue under water stress conditions, ABA (10 and 20   mg   L −1 ) and BR (0.4 and 0.8   mg   L −1 ) were applied individually and in combination (0.4   mg   L −1 BR and 10   mg   L −1 ABA) under three soil water regimes [75   ±   5% FC (HW), 50   ±   5% FC (MW) and 25   ±   5% FC (LW)] in the greenhouse. Results revealed that ABA and BR application markedly decreased the relative conductivity and malondialdehyde, notably increased leaf relative water content, antioxidant enzyme activity and proline content under water stress. ABA plus BR application was equally effective in improving the activities of antioxidant enzyme as ABA at 20   mg   L −1 . ABA application reduced stomatal conductance and decreased both transpiration and net photosynthetic rate ( P n ), while BR application significantly increased P n and water use efficiency (WUE) by enhancing chlorophyll content. The ABA and BR combination application showed higher P n and WUE as well as BR single application. It indicated that ABA and BR combination application increased photosynthetic capacity, and reduced the effect on photosynthetic loss caused by ABA under water stress. All these confirmed that ABA plus BR application exhibited a synergistic interaction on enhancing drought tolerance and photosynthesis of tall fescue under water stress.

Transcriptome Analysis Reveals an Essential Role of Exogenous Brassinolide on the Alkaloid Biosynthesis Pathway in Pinellia Ternata

Chenchen Guo,Ying Chen,Dengyun Wu,Yu Du,Mengyue Wang,Cunqi Liu,Jianzhou Chu,Xiaoqin Yao

Pinellia ternata(Thunb.) Druce is a traditional medicinal plant containing a variety of alkaloids, which are important active ingredients. Brassinolide (BR) is a plant hormone that regulates plant response to environmental stress and promotes the accumulation of secondary metabolites in plants. However, the regulatory mechanism of BR-induced alkaloid accumulation inP. ternatais not clear. In this study, we investigated the effects of BR and BR biosynthesis inhibitor (propiconazole, Pcz) treatments on alkaloid biosynthesis in the bulbil ofP. ternata. The results showed that total alkaloid content and bulbil yield was enhanced by 90.87% and 29.67% under BR treatment, respectively, compared to the control. We identified 818 (476 up-regulated and 342 down-regulated) and 697 (389 up-regulated and 308 down-regulated) DEGs in the BR-treated and Pcz-treated groups, respectively. Through this annotated data and the Kyoto encyclopedia of genes and genomes (KEGG), the expression patterns of unigenes involved in the ephedrine alkaloid, tropane, piperidine, pyridine alkaloid, indole alkaloid, and isoquinoline alkaloid biosynthesis were observed under BR and Pcz treatments. We identified 11, 8, 2, and 13 unigenes in the ephedrine alkaloid, tropane, piperidine, and pyridine alkaloid, indole alkaloid, and isoquinoline alkaloid biosynthesis, respectively. The expression levels of these unigenes were increased by BR treatment and were decreased by Pcz treatment, compared to the control. The results provided molecular insight into the study of the molecular mechanism of BR-promoted alkaloid biosynthesis. Keywords:alkaloid;brassinolide;ephedrine;secondary metabolism;RNA-seq

Exogenous 24-epibrassinolide activates detoxification enzymes to promote degradation of boscalid in cherry tomatoes

Yajie Yang,Linchun Mao,Weiliang Guan,Xiaobo Wei,Yelin Shao,Zisheng Luo,Xingyu Lin,Li Li

BACKGROUND Boscalid is often used to extend the storage time of postharvest cherry tomato. Pesticide residue has become an issue of food safety. This study sought to investigate the spatial distribution of boscalid residue in cherry tomato fruits and to determine the effect of 24-epibrassinolide (EBR) in promoting boscalid degradation. RESULTS Boscalid could quickly penetrate into cherry tomatoes, but mainly remained in the peel. The migration of boscalid from the peel into the core was a time-consuming and complex process during storage. After 72 h, boscalid residues in the pulp and the core began to accumulate gradually. The exogenous application of EBR activated peroxidase, glutathione reductase and glutathione S -transferase, and effectively promoted the degradation of boscalid by a maximum decrease of 44.8% in peel, 54.0% in pulp and 71.2% in core. CONCLUSION As one of the common pesticides, boscalid had a strong ability to enter the cherry tomato and thus become a potential risk for public consumption. Therefore, rational use of pesticides is recommended. The results of this study indicate that the possible risk of boscalid residue could be alleviated by EBR pretreatment through activating detoxification enzymes. © 2020 Society of Chemical Industry

24-Epibrassinolide Enhances Resistance Against Colletotrichum fructicola by Promoting Lignin Biosynthesis in Camellia sinensis L.

Zhang Lan,Zhang Zheng,Ahammed Golam Jalal,Wang Xiaorong,Fang Huihan,Yan Peng,Zhang Li-Ping,Ge Shibei,Fu Jianyu,Han Wen-Yan,Li Xin

Brassinosteroids (BRs) are steroid-type phytohormones that not only control growth and development but also responses to biotic and abiotic stressors in plants. However, little is known about the function of BRs in the protection of tea against the fungal pathogen Colletotrichum fructicola causing anthracnose disease. Here, we revealed that 24-epibrassinolide (EBR) could improve tea resistance against C. fructicola , which was linked to the EBR-induced accumulation of lignin in tea leaves. Time-course of the relative expression of ITS/Cs18SrDNA showed that its expression increased after the inoculation with C. fructicola ; however, EBR pretreatment significantly decreased the ITS/Cs18SrDNA expression at 3, 24 and 96 h postinoculation. Following an initial increase in 3 h postinoculation, lignin content significantly decreased from 6 to 24 h postinoculation with C. fructicola . In contrast to inoculation with only C. fructicola , exogenous EBR pretreatment significantly increased lignin contents throughout the postinoculation period. According to the qRT-PCR assay, EBR and C. fructicola differentially regulated the expression of 14 genes involved in the phenylpropanoid and lignin-specific pathways. Moreover, the activity of peroxidase (POD), a central enzyme in the final phase of lignin biosynthesis increased significantly at 3 h postinoculation with EBR treatment. Gene expression analysis revealed that EBR induced CsMYB4-like expression possibly to activate the transcription of CsPOD5-like gene responsible for lignin biosynthesis in response to C. fructicola . These findings suggest that EBR plays a positive role in disease resistance by inducing lignin synthesis, which may have potential application for the prevention of C. fructicola in tea plants.

Coumarin Promotes Hypocotyl Elongation by Increasing the Synthesis of Brassinosteroids in Plants

Siqi Liu,Aolin Ma,Jie Li,Zhixuan Du,Longfei Zhu,Guanping Feng

Coumarins are natural products commonly found in plants and are typical allelopathic substances that strongly affect the growth of plants after being exudated from the root and help plants absorb Fe in cases of iron deficiency. Although coumarins have been found to have multiple effects, this understanding is still relatively limited. Here, we show that coumarin significantly promotes the elongation of the hypocotyl by enhancing cell elongation. Further research has found that coumarin increases the content of BR in plants by enhancing the expression of brassinosteroid (BR) synthesis genes. The effect of coumarin on promoting hypocotyl elongation is completely blocked by the mutation of the BR synthesis geneDEETIOLATED 2(DET2) or the co-addition of the BR synthesis inhibitor brassinazole (BRZ). Genetic analysis using Arabidopsis mutants showed that coumarin promoting hypocotyl elongation depends on the signaling pathway of the BRs. Overall, coumarin promotes elongation of the hypocotyl by increasing the synthesis of BRs in plants. These results provide us with new insights into the role of coumarins and offer strong theoretical support for the mechanisms of interactions between plants.
