A multifunctional nanoplatform integrating multiple therapeutic functions may be an effective strategy to realize satisfactory therapeutic efficacy in the treatment of tumors. However, there is still a certain challenge in integrating multiple therapeutic agents into a single formulation using a simple method due to variations in their properties. In this work, multifunctional CuS-ICG@PDA-FA nanoparticles (CIPF NPs) with excellent ability to produce reactive oxygen species and photothermal conversion performance are fabricated by a simple and gentle method. Hollow mesoporous copper sulfide nanoparticles (HMCuS NPs) not only have excellent loading and photothermal conversion performance but also can cause a highly efficient Fenton-like reaction for chemodynamic therapy (CDT). The loaded photosensitizer indocyanine green (ICG) imparts excellent photodynamic properties to the NPs, which in turn enhances the stability of ICG. The polydopamine (PDA) coating improves the stability and biocompatibility of the NPs and creates the conditions for surface modification of folic acid. The FA-coated NPs show precise targeting of tumor cells. The results of the cellular uptake assay demonstrate that CIPF NPs enter tumor cells through an endocytic pathway. Lysosome colocalization and escape experiments prove that CIPF NPs possess good lysosomal escape ability under irradiation of NIR. Both in vitro and in vivo antitumor studies of CIPF NPs reveal excellent efficacy in photothermal/photodynamic/chemodynamic therapy. The construction of high-performance CIPF NPs offers valuable insights into the design of a multifunctional copper sulfide-based nanoplatform for combined cancer treatment and precise theranostics.

The low cure rate and high mortality associated with cancer pose significant threats to human health. Photodynamic and photothermal therapies have emerged as promising treatment strategies for various types of cancers. In this study, we successfully synthesized a novel type of carbon dot (CD) using 1,2,4-aminobenzene and ethylenediamine as precursors. Surprisingly, these CDs exhibited outstanding nucleolus-targeting capabilities coupled with a remarkable photothermal effect. Through the integration of these nucleolus-targeting CDs with indocyanine green (ICG) and folic acid (FA), we created CDs-ICG-FA nanocomplexes suitable for combined photodynamic and photothermal therapy. In vitro experiments demonstrated that CDs-ICG-FA maintained a robust photothermal ability, achieving a conversion efficiency of up to 34.3%. Furthermore, CDs-ICG-FA generated abundant reactive oxygen species, effectively inducing cancer cell death and demonstrating its potential for photodynamic therapy. In MCF-7 cancer cells, CDs-ICG-FA exhibited a pronounced synergistic photothermal/photodynamic anticancer effect. Subsequent in vivo experiments in mice revealed that CDs-ICG-FA could selectively accumulate at tumor sites, significantly inhibiting tumor growth upon exposure to an 808 nm laser. These findings suggest that the developed nucleolus-targeting CDs-ICG-FA hold promising potential for cancer targeting and the application of combined photothermal/photodynamic therapy.

Precise oncotherapy requires effective cancer treatments that are guided by clinical imaging techniques. One of the most representative cases is multi-imaging-guided phototherapy. This study presents a novel multifunctional theranostic agent of SnxWO3 tungsten bronze, which is an excellent light absorber in the near infrared (NIR) range. Theoretical calculations based on density functional theory confirm that the insertion of donor Sn atoms into orthorhombic WO3 gives rise to the broadband visible-NIR absorption. Accordingly, both the photothermal effect and reactive oxygen species (ROS) production could be realized under NIR light irradiation by SnxWO3 tungsten bronze nanocrystals, thereby triggering the potent in vivo photothermal and photodynamic synergistic therapy. Meanwhile, modified SnxWO3 tungsten bronze has the functions of photoacoustic imaging (PAI), X-ray computed tomography (CT) imaging and near-infrared fluorescence (NIRF) imaging for tumor detection as well. Finally, for investigating the antitumor mechanism of in vivo solid tumors, clinical imaging modalities of B-mode ultrasonography (US) and magnetic resonance imaging (MRI) are employed to monitor the tumor evolution process after the photo-treatment, verifying a typically liquefactive necrosis process. These results indicate that the SnxWO3 tungsten bronze nanostructure is a promising theranostic agent for imaging-guided cancer therapy.

The unique optical properties of squaraine dyes make them promising for cancer phototheranostics, but the reported squaraines for in vivo treatments mainly rely on their photothermal effect, where monotherapy cannot achieve the desired therapeutic effect. Here we generated a type of squaraine capable of killing tumors through both photothermal and photodynamic effects. We optimized squaraine structure with selenium modulation and formulated it into nanoparticles that showed strong absorption of infrared light, negligible fluorescence, good photothermal conversion (66.6 %), and strong photodynamic effects even after several irradiation cycles. In addition, the nanoparticles could be tracked through their strong optoacoustic signal. In mice, the nanoparticles effectively accumulated in tumors and eliminated them upon irradiation, without causing adverse effects. Our work demonstrates the potential of selenium modulation of squaraine for precise cancer diagnosis and treatment through synergistic photothermal and photodynamic effects.

Due to the limited concentration of hydrogen peroxide (H 2 O 2 ) in the tumor microenvironment, peroxidase (POD)-mimicking nanozyme-mediated catalytic therapy usually cannot completely eliminate tumor issues. In this study, we report an H 2 O 2 -responsive nanozyme constructed by loading 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) into POD-like Fe-porphyrin covalent organic frameworks (COFs) (Fe-DhaTph). The resulting ABTS@Fe-DhaTph can function as a multifunctional therapeutic nanoagent for combination therapy of catalytic therapy, photothermal therapy (PTT), and photodynamic therapy (PDT). In the tumor microenvironment, ABTS@Fe-DhaTph can catalyze the decomposition of endogenous H 2 O 2 to produce highly cytotoxic hydroxyl radicals (·OH) for in situ tumor catalytic therapy. Meanwhile, the loaded ABTS is oxidized by H 2 O 2 with the assistance of ABTS@Fe-DhaTph, and the resulting oxABTS, with strong near-infrared absorbance, can enable the H 2 O 2 -dependent PTT. Besides Fe-DhaTph-induced PDT, the generated oxABTS with glutathione (GSH)-depletion ability can synergistically enhance the generation of reactive oxygen species, thereby improving catalytic therapy and PDT. To the best of our knowledge, ABTS@Fe-DhaTph is the first COF-based nanozyme to serve as a cascade-amplified synergistic therapeutic nanoagent for cancer treatment.

BODIPY dyes have recently been used for photothermal and photodynamic therapy of tumors. However, complex multi-material systems, multiple excitation wavelengths and the unclear relationship between BODIPY structures and their PTT/PDT efficiency are still major issues. In our study, nine novel BODIPY near-infrared dyes were designed and successfully synthesized and then, the relationships between BODIPY structures and their PTT/PDT efficiency were investigated in detail. The results showed that modifications at position 3,5 of the BODIPY core with conjugated structures have better effects on photothermal and photodynamic efficiency than the modifications at position 2,6 with halogen atoms. Density functional theory (DFT) calculations showed that this is mainly due to the extension of the conjugated chain and the photoinduced electron transfer (PET) effect. By encapsulating BDPX-M with amphiphilic DSPE-PEG2000-RGD and lecithin, the obtained NPs not only show good water solubility and biological stability, but also could act as superior agents for photothermal and photodynamic synergistic therapy of tumors. Finally, we obtained BODIPY NPs that exhibited excellent photothermal and photodynamic effects at the same time under single irradiation with an 808 nm laser (photothermal conversion efficiency: 42.76%, A/A0: ∼0.05). In conclusion, this work provides a direction to design and construct phototherapeutic nanoparticles based on BODIPY dyes for tumor treatment.

Gold nanoparticles (Au NPs) used as photosensitizers for photothermal therapy (PTT) have attracted extensive attention. However, limited tissue penetration of light seriously hinders the practical application, causing Au NPs to be combined with multiple theranostic agents to improve the treatment effect. Herein, we fabricate one potent gold nanostars (GNS) that can unify the dual nanozyme activities and photothermal property, achieving effective synergistic cascade catalytic and photothermal therapy. Notably, GNS is prepared by a green and facile one-step synthesis method using biodegradable poly(γ-glutamic acid) as a medium. The single GNS exhibits inherent dual nanozyme activities, glucose oxidase- and peroxidase-like activities, which not only convert glucose of tumor tissue into hydrogen peroxide (H2O2) but also catalyze H2O2 to produce the amount of toxic hydroxyl radicals (•OH), leading to an amplified cascade catalytic therapy. Moreover, the cascade catalytic activity of GNS is enhanced 1.4-fold in the photothermal effect. Finally, the GNS can achieve photoacoustic imaging-guided efficient synergistic cascade catalytic and photothermal therapy with a high antitumor rate of 97.0%. This simple but multifunctional GNS could provide a new strategy facilitating the practical treatment application of single Au NPs.

The use of functional nanomaterials to realize tumor therapeutic therapy has received considerable attention. However, a single type of nanomaterial usually can only achieve one specific function, and may cause biosafety problems due to poor metabolism. In this work, we have designed a kind of nanocomposite film based on Bi 2 Se 3 nanosheets and carbon nanotubes, which can achieve efficient photothermal conversion and reactive oxygen species generation under near-infrared laser irradiation. Moreover, the developed films showed good biocompatibility in vivo. The multifunctional films showed remarkable therapeutic effects under 808 nm excitation with 0.3 W/cm 2 , which avoiding the introduction of nano-reagents into the mice and excessive laser power damage. Our work will provide reference for the use of nanomaterials with heterojunctions in synergistic therapy strategies of photothermal and photodynamic therapy.

Sono-photodynamic therapy (SPDT) integrates the advantages of sonodynamic therapy (SDT) and photodynamic therapy (PDT), showing great potential for the minimally invasive treatment of malignant tumors. Nevertheless, the effectiveness of sensitizers in PDT is often limited by insufficient light penetration, restricting the synergistic therapeutic effects of SPDT. Herein, we present a controllable strategy using in situ ultrasound-triggered bioluminescence for enhanced SPDT. This method utilizes a mechanosensitive bioluminescent protein composed of the mechanosensitive channel of large conductance (MscL) and the aequorin (AEQ) photoprotein. Upon ultrasound stimulation, MscL responds to mechanical force by inducing Ca2+ influx, which subsequently activates AEQ to oxidize a luminogenic substrate and generate bioluminescence. By leveraging ultrasound and its associated bioluminescence, the natural sono-photosensitizer, Chlorin e6 (Ce6), exhibits efficient sono-/photocytotoxicity, induces immunogenic cell death, and synergistically enhances the antitumor immune response. This ultrasound-triggered SPDT not only suppresses primary tumor growth and metastasis but also induces a long-term immune memory to resist tumor rechallenge. The integration of SPDT with immune activation forms a multimodal “sono–photo–immuno” platform for in situ breast tumor treatment and simultaneously prevents tumor recurrence after tumor resection. These findings showcase our platform as a versatile tool for the development of more effective cancer therapies.

Photodynamic therapy (PDT) and photothermal therapy (PTT) have been explored widely for application in cancer treatment. In this work, we describe the synthesis of CaTiO3:Yb,Er (CTO) nanofibers co-conjugated with Rose Bengal (RB) and gold nanorods (AuNRs), which offer the potential for combined upconversion photoluminescence (UCPL) and enhanced, synergistic PDT and PTT. Based on this delivery platform, RB and AuNRs served as the PDT and PTT agents, respectively. RB and AuNRs have strong and well-matched absorption with the green and red emissions of UCPL CTO nanofibers respectively, hence a single 980 nm continuous wave laser with deep tissue penetration can be employed to allow PDT and PTT to occur simultaneously. The nanocomposite can effectively convert the near-infrared (NIR) radiation from the laser into a combination of targeted hyperthermia and generation of reactive oxygen species (ROS). In comparison with PDT or PTT alone, the combined PDT/PTT treatment showed significantly enhanced suppression of the viability of Hep G2 cells in vitro, demonstrating its potential for use in oncology.

Photothermal therapy (PTT) based on a second near-infrared (NIR-II, 1000–1700 nm) light source holds great promise in the field of tumor treatment. However, hyperthermia-induced heat shock protein (HSP) expression and limited therapeutic efficacy remain the major challenges for PTT. Herein, a NIR-II laser-responsive nanocapsule PC/AC@GR@HA (abbreviated as P/A@G@H) was designed for carbon oxide (CO)-assisted tumor PTT combined with thermodynamic therapy (TDT). P/A@G@H was constructed by loading the CO storage polymer mPEG(CO) (PC) along with the free radical initiator 4,4′-azobis(4-cyanovaleric acid) (AC) into a gold hollow nanorod (GR), followed by the surface coating of hyaluronic acid (HA). After accumulating in tumor tissue through the active targeting of HA, P/A@G@H can quickly cause local hyperthermia upon 1064 nm laser irradiation and lead to cell damage by GR-mediated PTT. Moreover, the loaded PC can react with in situ H2O2 to produce CO, which inhibits the PTT-induced HSP90 expression. This can effectively weaken the heat resistance of tumor cells and enhance the PTT effect. Additionally, as a thermally decomposable radical initiator, AC can turn to toxic free radicals to ablate tumor cells via TDT under hyperthermia. Our research demonstrates that the synergistic strategy combining CO-assistance and TDT can effectively augment the ultimate therapeutic outcome of PTT, and the integrated nanocapsule P/A@G@H could be a powerful weapon for tumor suppression based on NIR-II light.

Chemodynamic therapy and sonodynamic therapy are two promising tumour therapeutic strategies. However, lack of highly effective sonosensitizers and control over chemodynamic therapy limit their application. Here we synthesize silver-doped zinc selenide quantum dots with atomically dispersed superficial Fe and show that they act as efficient sonosensitizers, catalysers and immunoreagents. Surface modification with an in situ self-assembly peptide drives accumulation in tumours. Superficial Fe III remains stable and converts to Fe II only under ultrasonic processing, reverting to Fe III upon ultrasound cessation. Under ultrasound stimulation, superficial Fe undergoes valence change with concomitant amelioration of the hypoxic tumour microenvironment and production of sonodynamic therapy-beneficial hydroxyl radicals. Furthermore, silver doping suppressed nonradiative recombination of excitons, leading to improved production of singlet oxygen. Meanwhile, selenium promotes robust systemic immune responses for the inhibition of tumour metastases. This nano-platform allows control of valence switching of atomically dispersed catalysts, representing an effective tool for chemodynamic/sonodynamic/immunotherapy.
