The Recognition of Sweat Latent Fingerprints with Green-Emitting Carbon Dots

Dan Zhao,Wenting Ma,Xincai Xiao

The recognition of fingerprints has played an extremely important role in criminal investigations, due to its uniqueness. This paper reports on the recognition of sweat latent fingerprints using green-emitting, environment-friendly carbon dots prepared with DL-malic acid and ethylenediamine, and the exploration of impacting factors in the development process of fingerprints. The experiments showed that better fingerprint images could be obtained when the latent fingerprints are developed in green-emitting carbon dots with pH 9 for 30 min, at room temperature. The reported method was also effective for latent fingerprints on a variety of substrates, as well as for those water-immersed ones, where the developed fingerprint remained stable after long-term preservation. Furthermore, the fluorescent three-dimensional fingerprint image could provide direct and simple evidence on pressing habits. The objective of this paper was to present this method. The method may help to narrow the range of suspects during criminal investigations and in forensic science. Keywords:green;carbon dots;sweat latent fingerprints;forensic science

Validation of the Investigator 24plex QS Kit: A 6-dye Multiplex PCR Assay for Forensic Application in the Chinese Han Population

Tao Ruiyang,Chen Chong,Sheng Xiang,Xia Ruocheng,Zhang Xiaochun,Zhang Jingyi,Yang Zihao,Zhang Suhua,Li Chengtao

The Investigator 24plex QS Kit (QIAGEN, Hilden, Germany) is a 6-dye fluorescent chemistry short tandem repeat (STR) polymerase chain reaction (PCR) amplification system that simultaneously amplifies 20 of the expanded Combined DNA Index System (CODIS) core STR loci, SE33, DYS391, and the standard sex-determining locus, amelogenin, as well as two special internal performance quality sensor controls (QS1 and QS2), which are included in the primer mix to check the PCR performance. This study was designed to be a pilot evaluation of this STR-PCR kit in a Chinese Han population regarding the PCR conditions, sensitivity, precision, accuracy, repeatability, reproducibility, and concordance; tolerance to PCR inhibitors; applicability to real “forensic-type” samples; species specificity; mixture, balance and stutter analyses, and utility in a population investigation. The exhaustive validation studies demonstrated that the Investigator 24plex QS system is accurate, sensitive and robust for STR genotyping. In addition, these genetic markers in the population data in our study indicated that they can also be useful for forensic identification and paternity testing in the Chinese Han population.

Bacteria-targeting BSA-stabilized SiC nanoparticles as a fluorescent nanoprobe for forensic identification of saliva

Li Xingmei,Ding Yanjun,Ling Jiang,Yao Wenyan,Zha Lagabaiyla,Li Na,Chang Yunfeng,Wang Yong,Cai Jifeng

Forensic saliva identification represents an increasingly useful auxiliary means of crime investigations, particularly in sex crimes. Salivary bacteria detection techniques have been shown to be viable methods for identifying the presence of saliva. A one-pot method is described for the fabrication of bovine serum albumin-stabilized SiC nanoparticles (SiC@BSA NPs). The SiC@BSA NPs were conjugated to antibacterial peptide GH12 to allow for fluorometric detection and imaging of bacteria in saliva. More specifically, the nanoprobe, with fluorescence excitation/emission maxima at 320/410 nm, was used to detect the oral bacteria S. salivarius levels. The detection limit is 25 cfu·mL −1 , and the assay can be performed within 40 min. The nanoprobe was also used to detect bacteria in forensic body fluids including blood, urine, and semen. In all cases, positive results were obtained with (mixed) samples containing saliva, while other saliva samples without saliva showed negative results. Fluorescent images of S. salivarius cells were obtained by implementing a high-content image analysis system. These results suggest that this new nanoprobe can be applied to screen for forensic saliva stains. Schematic representation of the preparation of SiC@BSA-GH12 nanoprobe for fluorometric detection and imaging of S. salivarius in saliva.

Temporal evolution stages classification and aging time prediction of gel-pen ink using GC-IMS combined with machine learning for forensic science applications

Wenhui Lu,Jing Chen,Liangliang Zhang,Zixuan Nie

Determining the temporal evolution of inks remains a critical challenge in forensic document analysis. The temporal evolution stages classification and aging time prediction of gel-pen ink were investigated by integrating gas chromatography-ion mobility spectrometry (GC-IMS) with machine learning algorithms. Ink-specific volatile markers were correlated with aging mechanisms through kinetic modeling and heatmap analysis. Three distinct temporal evolution stages were categorized into the rapid evaporation stage, slow-release stage, and chemical stabilization stage through multivariate analysis of volatiles. Furthermore, six tree-based machine learning algorithms were systematically evaluated, with the Categorical Boosting (CatBoost) model achieving superior performance (accuracy=100 %) in classifying five detailed aging stages of gel-pen ink. The decision tree regression model demonstrated high temporal prediction accuracy (test R²=0.954) through interpretable feature engineering. A stepwise strategy combining classification and regression models was proposed, enabling simultaneous ink characterization and age estimation. This methodology is expected to provide a validated approach for classifying temporal evolution stages and predicting aging time, significantly improving the efficiency of forensic analysis in judicial investigations.

Development and validation of a novel SiFaSTRTM 23-plex system

Hancheng Zheng,Ruiyang Tao,Jingyi Zhang,Jiashuo Zhang,Shouyu Wang,Zihao Yang,Qiannan Xu,Yuzhen Gao,Suhua Zhang,Chengtao Li

Human identification and paternity testing are usually based on the study of STRs depending on their particular characteristics in the forensic investigation. In this paper, we developed a sensitive genotyping system, SiFaSTR ™ 23-plex, which is able to characterize 18 expanded Combined DNA Index System STRs (D3S1358, D5S818, D2S1338, TPOX, CSF1PO, TH01, vWA, D7S820, D21S11, D10S1248, D8S1179, D1S1656, D18S51, D12S391, D19S433, D16S539, D13S317, and FGA), three highly polymorphic STRs among Chinese people (Penta D, Penta E, and D6S1043), one Y-chromosome Indel and amelogenin using a multiplex PCR; the PCR amplified products were analyzed using the Applied Biosystems 3500 Genetic Analyzer. Full genotyping profiles were obtained using only 31.25 pg of control DNA; various case-type specimens, as well as ten-year-old samples were also successfully genotyped. Additionally, in the validation studies, this multiplex was demonstrated to be human-specific and compatible with the interference of multiple PCR inhibitors. The system also enabled the detection of mixtures, and complete profiles could be obtained at the mixed ratios of 1:1, 1:3, and 3:1. The development and validation study here illustrated that the SiFaSTR ™ 23-plex system is accurate, powerful, and more sensitive than many other systems. What's more, the population data in our study not only illustrated that this 23-plex typing system was straightforward and efficient but also expanded the Chinese STR database, which could facilitate the appropriate application of the 23 genetic markers in forensic genetics, especially in the Chinese population.

Fast Imaging of Eccrine Latent Fingerprints with Nontoxic Mn-Doped ZnS QDs

Chaoying Xu,Ronghui Zhou,Wenwei He,Lan Wu,Peng Wu,Xiandeng Hou

Fingerprints are unique characteristics of an individual, and their imaging and recognition is a top-priority task in forensic science. Fast LFP (latent fingerprint) acquirement can greatly help policemen in screening the potential criminal scenes and capturing fingerprint clues. Of the two major latent fingerprints (LFP), eccrine is expected to be more representative than sebaceous in LFP identification. Here we explored the heavy metal-free Mn-doped ZnS quantum dots (QDs) as a new imaging moiety for eccrine LFPs. To study the effects of different ligands on the LFP image quality, we prepared Mn-doped ZnS QDs with various surface-capping ligands using QDs synthesized in high-temperature organic media as starting material. The orange fluorescence emission from Mn-doped ZnS QDs clearly revealed the optical images of eccrine LFPs. Interestingly, N-acetyl-cysteine-capped Mn-doped ZnS QDs could stain the eccrine LFPs in as fast as 5 s. Meanwhile, the levels 2 and 3 substructures of the fingerprints could also be simultaneously and clearly identified. While in the absence of QDs or without rubbing and stamping the finger onto foil, no fluorescent fingerprint images could be visualized. Besides fresh fingerprint, aged (5, 10, and 50 days), incomplete eccrine LFPs could also be successfully stained with N-acetyl-cysteine-capped Mn-doped ZnS QDs, demonstrating the analytical potential of this method in real world applications. The method was also robust for imaging of eccrine LFPs on a series of nonporous surfaces, such as aluminum foil, compact discs, glass, and black plastic bags.

Simultaneous determination of ten neonicotinoid insecticides and a metabolite in human whole blood by QuEChERS coupled with UPLC-Q Exactive orbitrap high-resolution mass spectrometry

Jingwen Zhang,Junning Liu,Yuheng Wang,Yong Wang,Ruiqin Yang,Xinyang Zhou

Since neonicotinoid insecticides are now the most extensively used insecticides worldwide, there are increasing cases of neonicotinoid poisoning. A rapid and sensitive method was developed for the determination of ten neonicotinoid insecticides and a metabolite 6-chloronicotinic acid in human whole blood. The types and amounts of extraction solvent, salting-out agent, and adsorbent in the QuEChERS method were optimized by comparing the absolute recoveries of 11 analytes. The separation was performed on an Agilent EC18 column with the gradient elution with 0.1% formic acid in water and acetonitrile as the mobile phase. The quantification was achieved by Q Exactive orbitrap high-resolution mass spectrometry under parallel reaction monitoring scan mode. The 11 analytes showed good linearity with R 2  ≥ 0.9950, LODs ranging from 0.01 μg/L to 0.30 μg/L, and LOQs from 0.05 μg/L to 1.00 μg/L. The recoveries ranged from 78.3% to 119.9% at low, medium, and high spiked concentrations of blank blood, with matrix effects ranging from 80.9% to 117.8%, inter-day RSDs from 0.7% to 6.7%, and intra-day RSDs from 2.7% to 9.8%. The method was furthermore applied to a real case of neonicotinoid insecticide poisoning to demonstrate its feasibility. The proposed method is suitable for the rapid screening of neonicotinoid insecticides in poisoned human blood in the field of forensic science, as well as monitoring of neonicotinoid insecticide residues in humans in the field of environmental safety, compensating for a lack of studies on neonicotinoid insecticide determination in biological samples.

Rapid Sample Pretreatment Facilitating SERS Detection of Trace Weak Organic Acids/Bases in Complex Matrices

Jing Xu,Shu Zhang,Si-heng Luo,Chen-ru Xiong,Minghuai Zhu,Jing Chang,Bo Zou,Bin Ren,Zhong-qun Tian,Guo-kun Liu

Fast and efficient sample pretreatment is the prerequisite for realizing surface-enhanced Raman spectroscopy (SERS) detection of trace targets in complex matrices, which is still a big issue for the practical application of SERS. Recently, we have proposed a highly performed liquid–liquid extraction (LLE)–back extraction (BE) for weak acids/bases extraction in drinking water and beverage samples. However, the performance efficiency decreased drastically on facing matrices like food and biological blood. Based on the total interaction energies among target, interferent, and extractant molecules, solid-phase extraction (SPE) with a higher selectivity was introduced in advance of LLE–BE, which enabled the sensitive (μg L–1 level) and rapid (within 10 min) SERS detection of both koumine (a weak base) and celastrol (a weak acid) in different food and biological samples. Further, the high SERS sensitivity was determined unmanned by Vis-CAD (a machine learning algorithm), instead of the highly demanded expert recognition. The generality of SPE–LLE–BE for various weak acids/bases (2 < pKa < 12), accompanied by the high efficiency, easy operation, and low cost, offers SERS as a powerful on-site and efficient inspection tool in food safety and forensics.

Luminescent nanofibers for human skin textures photocopying

Tian Tian,Han Huixuan,Lin Xinyi,Kang Hui,Yang Meifang,Chen Zihao,Wu Chao,Jiang Yuansheng,Li Wenguang,Chang Xueqing,Xu Qin,Zhang Yizhou,Chen Yu-Xin,Ouyang Gangfeng,Pang Huan

The domain of forensic science, dermatology, and regenerative medicine critically relies on the precise replication of human skin details. Nevertheless, conducting on-site analysis poses challenges due to the stringent requirements for stability, accuracy, and the use of safe imaging materials. Current skin imaging methodologies are hindered by the inherent limitations of their hardware components, particularly when it comes to capturing the intricate, micrometer-scale textures of human skin. To address these challenges, we develop a low-cost (<$800), portable nanofiber-based imaging technique (NFIT) using CsPbBr 3 @HPβCD luminescent nanofibers. NFIT achieves in-situ, multi-regional imaging with ultrahigh-resolution (1450 dpi) and micron-scale similarity (93.24 ± 4.6%), capturing intricate details from sweat pores to large skin areas. Its non-contact design eliminates chemical pre/post-treatments, ensuring safety, hygiene and ease of use. NFIT demonstrates robustness and reliability as it maintains clear imaging under extreme temperature (−50 °C to +50 °C) and over extended periods (Level 3 ≥ 81 days, Level 2 ≥ 108 days). An algorithm was developed to support 3D skin texture model reconstruction, offering a transformative solution for forensic evidence analysis, dermatological assessments, and personalized medicine.

Development of a Novel Fluorescence Nanoprobe Based on Molecularly Imprinted Polymer on Cobalt-Carbon Quantum Dots for Rapid Detection of Methamphetamine

Yin Qinhong,Yang Lihua,Liu Mei,Zhu Yanqin

In forensic drug analysis, detecting low concentrations of illicit drugs in samples presents a significant challenge. This paper introduces a novel approach for developing molecularly imprinted quantum dot materials (Co-AD QDs-MIPs) capable of efficiently detecting methamphetamine (METH) in artificial urine without the need for extraction or pretreatment. Using free radical polymerization, molecularly imprinted materials were coated onto the surface of quantum dots, forming a Co-AD QDs-MIPs fluorescence nanoprobe. The resulting Co-AD QDs-MIPs demonstrated enhanced specificity for METH detection and showed strong fluorescence quenching. Under optimal conditions, the method enabled selective and quantitative detection of METH. A good linear correlation was observed between the fluorescence quenching effect and increasing METH concentration in the range of 67–469 nM, with a detection limit of 23 nM. The Co-AD QDs-MIPs nanoprobe successfully detected METH in simulated human urine samples, with a recovery rate of 96.2% to 102.1%. Therefore, it is a simple, cost-effective, and selective nanoprobe for detecting low concentrations of METH.
