Most injectable hydrogels are prone to irreversible deformation or damage under sustained shear forces and exhibit low mechanical properties, which limit their practical applications in biological engineering. In this work, the first hydrogen-bonding reinforced injectable thermoresponsive hydrogel system has been fabricated through an in situ hydrazide–aldehyde cross-linking reaction between hydrazide-functionalized poly(N-isopropylacrylamide) (PNIPAM) and dialdehyde dextrin. The mechanical strength and stability of the hydrogel were reinforced by the introduction of intermolecular hydrogen-bonding from poly(N-acryloyl glycinamide) (PNAGA), which possesses great biocompatibility and similarity to PNIPAM. The internal morphology, dynamic moduli, de-cross-linking properties, and temperature responsiveness of the hydrogels were systematically investigated. Upon hydrogen-bonding reinforcement, the hydrogel elasticity, reflected by the maximum storage modulus, dramatically increased from 90.2 to 517.0 kPa, which is about a 570% increase compared with unreinforced hydrogels. In addition, the mechanical properties of the hydrogel were reinforced as the de-cross-linking could be inhibited by PNAGA-based hydrogen-bonding. The temperature release behavior of the hydrogel was assessed with a model drug (propranolol hydrochloride, PHCl) and could be precisely controlled by adjusting the environmental temperature between 25 and 37 °C. Therefore, PNAGA reinforced thermoresponsive hydrogels are promising functional biological materials for use in tissue-repair engineering and controlled-release drug delivery.

Oxidative stress arises from an imbalance between excessive production of reactive oxygen species (ROS) and the body’s antioxidant defenses. In neurodegenerative diseases, this imbalance leads to ROS accumulation, causing neuronal dysfunction and cell death. Traditional drug therapies often fail to address the dynamic nature of neuroinflammation, limiting their therapeutic efficacy. To overcome this challenge, we have developed an innovative ROS-responsive injectable hydrogel. This hydrogel is designed to detect oxidative stress sensitively and release glutathione in a controlled manner, thereby modulating inflammation and restoring the damaged immune microenvironment to facilitate tissue repair. The hydrogel was synthesized by crosslinking polyvinyl alcohol (PVA) with sodium alginate modified with 3-aminophenylboronic acid (Alg-PBA). We investigated the hydrogel’s formation mechanism and analyzed how component variations affect its morphological and rheological properties. Our findings demonstrate that an optimal Alg-PBA to PVA weight ratio of 2:1 yields a hydrogel with superior mechanical strength. Glutathione (GSH) release studies confirmed the hydrogel’s pronounced ROS-responsive drug release behavior. Furthermore, biocompatibility assessments revealed that the hydrogel loaded with 100 μg/mL GSH exhibited excellent compatibility and significantly inhibited neuronal apoptosis under oxygen-glucose deprivation (OGD) conditions. This work presents a promising strategy for treating inflammation-related diseases and provides valuable insights for designing next-generation hydrogels that adapt to injury-responsive microenvironments.

An injectable hydrogel loading α-N-heterocyclic thiosemicarbazones with antibacterial activity was designed to attain therapy against wounds with high efficiency. In the present study, the injectable FPZ-H2L hydrogel was synthesized by loading 2,6-diacetylpyridine bis(N4-methylthiosemicarbazone) (H2L) on transition-metal ions (Zn2+)-cross-linked-folic acid (FA)-co-polydopamine (PDA) hydrogel (FPZ hydrogel). The injectable hydrogel (FPZ-H2L) cross-linked by coordination and hydrogen bonds displayed an excellent self-healing performance and completely covered the irregular wound. Compared with the FPZ hydrogel, the FPZ-H2L hydrogel has stronger antibacterial efficacy on typical Gram-positive/negative bacteria and drug-resistant bacteria. The antibacterial mechanisms of the hydrogel were related to biofilm ablation, the disruption of the cell membrane integrity, leakage of nucleic acids and proteins, and the production of oxidative stress response, thus causing bacterial death. Cell Counting Kit-8 (CCK-8) test shows that FPZ-H2L hydrogel has excellent cell compatibility, and gel drug loading technology greatly overcomes the problem of high cytotoxicity of H2L. The wound healing experiments indicated that the hydrogel enhanced the wound healing efficacy of mice. These results prove that the mixed hydrogel has broadened the field of vision for the design of antibacterial wound dressings.

As the forefront of the digestive system, a mouth in a high moist microenvironment is easy to breed bacteria and the inevitable injury from oral surgeries will cause a series of physiological disorders. The injectable hydrogels are promising biomaterials to solve this challenge, but the limited mechanical performance and weak mechanical adjustment are insufficient to fit in the whole healing process. Herein, we propose and design a new spatiotemporal self-strengthening injectable hydrogel via manipulating the time-dependent cross-linking effects after the Schiff base reaction. The resultant hydrogels possess various promising properties and functions, including rapid initial gelation efficiency (∼30 s), spatiotemporal adjustable mechanical strength , great antifouling property and biocompatibility, leading to effective promote oral tissue regeneration and bone repair. We believe both our concept and spatiotemporal self-strengthening injectable hydrogels will provide new insight into the hydrogel-based wound dressing and broaden the application of the bioinspired hydrogels in biomedical fields .

Despite the excessive of antibiotics causes the emergence of multidrug-resistant (MDR) bacteria, antibiotic therapy remains the most effective and fastest-acting strategy currently available. Water-soluble metal–organic cages (MOCs) are ideal platforms for enhancing antibiotic efficacy through host–guest interactions and mediating the transformation of infected microenvironments due to their defined sizes, shapes, and high modularization of ligands. Here, we have developed a novel water-soluble MOCs crosslinked injectable hydrogel antibacterial platform (GG-ENR@Fe Gel) with multi-mode treatment strategies. In this platform, guar gum (GG), as a carrier, exhibits excellent moisturizing property, biosafety and rapid degradation ability, making it suitable as a medical dressing. Water-soluble MOCs (Fe-1•SO 4 ) was employed in the hydrogel to enhance the solubility and antibacterial efficacy of enrofloxacin (ENR) through host–guest interaction, further triggering additional crosslinking among the GG polymers. Furthermore, after effective adhesion to the infected wound, the Fe-1•SO 4 can generate singlet oxygen ( 1 O 2 ) by PDT and rapidly produce massive hydroxyl radicals (•OH) through Fenton-like reaction in the infected microenvironment. This study demonstrates that the water-soluble MOCs is a versatile and synergistic therapeutic platform that can provide promising strategies for treatment of bacterial infections.

Wound dressings with excellent adhesiveness, antibacterial, self-healing, hemostasis properties, and therapeutic effects have great significance for the treatment of acute trauma. So far, numerous mussel-inspired catechol-based wet adhesives have been reported, opening a pathway for the treatment of acute trauma. However, catechol-based hydrogels are easily oxidized, which limits their applications. Here, the design of a polyphosphazene and non-catechol based antibacterial injectable hydrogel is reported as a multifunctional first aid bandage. Inspired by barnacle cement proteins, a series of dynamic phenylborate ester based adhesive hydrogels are prepared by combining the cation– π structure modified polyphosphazene with polyvinyl alcohol. The inherent antibacterial property (4 h antibacterial rate 99.6 ± 0.2%), anti-mechanical damage, and hemostatic behavior are investigated to confirm multi-functions of wound dressings. In water, the hydrogels firmly adhere to tissue surfaces through cation– π and π – π interactions as well as hydrogen bonding (adhesion strength = 45 kPa). Moreover, in vivo experiments indicate the hydrogels can shorten the bleeding time and reduce the amount of bleeding by 88%, and significantly accelerate the wound healing rate. These hydrogels have a promising application in the treatment of acute trauma, which is in urgent need of anti-infection and hemostasis.

Hydrogel stands out as one of the most attractive wound dressings due to its excellent moisturizing properties and capacity to absorb wound exudates. However, conventional hydrogel dressings often lack responsiveness to the microenvironment, merely acting as protective barriers for the wound. Consequently, they exhibit limited effectiveness in preventing infection and facilitating wound repair. To address these problems, we have developed a multifunctional injectable hydrogel, CF/MS@HG, based on peroxidase-like nanozymes, aiming at rapidly healing bacterial-infected wounds. The hydrogel is mainly composed of oxidized sodium alginate, aminated gelatin, and polylysine, encapsulating MIL-101(CuFe) NPs (CF) and manganese selenide nanoparticles (MnSe 2 NPs, or MS NPs). After injection, the complex rapidly gelatinizes at the infected wound site through a Schiff base reaction. In vitro experiments have demonstrated the hydrogel’s strong adhesion and self-healing capabilities. Moreover, CF exhibiting peroxidase (POD)-like activity, catalyzes in situ hydrogen peroxide (H 2 O 2 ) to generate highly toxic hydroxyl radicals (·OH) within the wound microenvironment, inducing oxidative damage to bacteria. Meanwhile, MS decomposes into H 2 Se in the slightly acidic wound microenvironment, disrupting bacterial metabolism and inhibiting proliferation. The addition of polylysine further enhances the hydrogel’s antibacterial properties. In vivo experiments have shown that the hydrogel exhibits excellent biological safety and significantly promotes wound healing. This multifunctional smart hydrogel holds great promise for the treatment of bacterial-infected wounds.

The design of injectable hydrogels with excellent biocompatibility, medical imaging, and antitumor activity to improve the therapeutic efficiency on tumor tissues and to reduce toxic side effects is very important in clinical practice. A novel medically imageable pH-, temperature- and NIR- intelligent cellulose nanofibril (TOCNF)-based injectable hydrogel was designed using pH-, and temperature-responsive TOCNF as the carrier, citric-acid-stabilized Prussian blue nanoparticles (PBNPs) as the contrast agent and photothermal switch, and doxorubicin (DOX) as the chemotherapeutics agent. The hydrogel possessed a nanocage structure and a 3D network structure, which allowed a high loading capacity of PBNPs (2 mg/g) and DOX (415 mg/g). The PBNPs stabilized on TOCNF through electrostatic interactions and hydrogen bond could aid medical magnetic resonance imaging (MRI) and photoacoustic imaging (PAI). Interestingly, the temperature, pH, and NIR tri-stimuli-responsive intelligent switches, inspired by the mimosa leaf, contributed to the controllable sustained DOX release property of the hydrogel. Therefore, in vivo bimodal MRI/PAI and tumor therapy results showed that the previously prepared hydrogel could be ideal for the rapid diagnosis and chemo-photothermal therapy in the clinical treatment of tumors.

Developing highly efficient and biocompatible biolubricants for arthritis treatment is extraordinarily demanded. Herein, inspired by the efficient lubrication of synovial joints, a paradigm that combines natural polysaccharide (chitosan) with zwitterionic poly[2-(methacryloyloxy) ethyl phosphorylcholine] (PMPC), to design a series of brush-like Chitosan- g -PMPC copolymers with highly efficient biological lubrication and good biocompatibility is presented. The Chitosan- g -PMPC copolymers are prepared via facile one-step graft polymerization in aqueous medium without using any toxic catalysts and organic solvents. The as-prepared Chitosan- g -PMPC copolymers exhibit very low coefficient of friction ( μ < 0.01) on Ti 6 Al 4 V alloy substrate in both pure water and biological fluids. The superior lubrication is attributed primarily to the hydrated feature of PMPC side chains, interface adsorption of copolymer as well as to the hydrodynamic effect. In vivo experiments confirm that Chitosan- g -PMPC can alleviate the swelling symptom of arthritis and protect the bone and cartilage from destruction. Due to their facile preparation, distinctive lubrication properties, and good biocompatibility, Chitosan- g -PMPC copolymers represent a new type of biomimetic lubricants derived from natural biopolymer for promising arthritis treatment and artificial joint lubrication.

Overproduction of free radicals in vivo leads to oxidative stress, which substantially slows the healing of wound. Here, a series of injectable antioxidant hydrogels were designed by protocatechuic acid (PA)-mediated carboxylated chitosan (CCS) composed by conjugating oxidized hyaluronic acid (OHA). These biocompatible OHA/CCS-PA composite hydrogels presented stable chemical structure , physical properties, and effective antibacterial properties against common pathogens exceeding 99%. In addition, OHA/CCS-PA hydrogels demonstrated satisfactory antioxidant capacity to scavenge over 85% of overproduced free radicals, which could reduce the damage of oxidative stress to cellular activity and diminish the expression of reactive oxygen species in vitro by the long-lasting release of phenolic hydroxyl element. More importantly, the results of in vivo animal tests confirmed that OHA/CCS-PA hydrogels could significantly promote wound healing with higher collagen disposition (86.29%) in comparison with commercial Tegaderm™ film and the control hydrogel with physical mixing of PA. These findings indicated that OHA/CCS-PA hydrogel had great potential for wound healing.

In situ injectable hydrogels, which have great potential in tissue engineering, are characterized by simple preparation, minimal invasiveness and adaptation to complex shapes. However, injectable hydrogels have higher requirements for biocompatibility and safety due to their use in vivo implantation. Therefore, in this study, a human-like collagen (HLC)-based in situ gel-forming injectable HLC-HPA hydrogel was synthesized by combining the amino group of HLC with the carboxyl group of HPA activated using 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC) and N-hydroxysuccinimide (NHS), followed by crosslinking by horseradish peroxidase (HRP) and H 2 O 2 , and used as scaffold material for tissue engineering. The hydrogel stiffness, gel time and biodegradation rate could be easily and independently adjusted by varying the H 2 O 2 and HRP concentrations. Scanning electron microscope (SEM) clarified the homogeneous porous and interconnected internal structures of the hydrogel. In vitro cell viability and in vivo degradation experiments confirmed that the HLC-HPA hydrogel had good biodegradability and excellent biocompatibility. Interestingly, in cultured macrophages, the HLC-HPA hydrogel showed anti-inflammatory activity by reducing the amount of the pro-inflammatory cytokines tumor necrosis factor-α (TNF-α) and interleukin- 6 (IL-6) and increasing the secretion of the anti-inflammatory cytokine interleukin-10 (IL-10) induced by lipopolysaccharide (LPS). Meanwhile, in animal experiments, HLC-HPA hydrogels exhibit excellent biocompatibility and have properties of hemostasis and reduction of inflammatory response. Therefore, the HLC-HPA hydrogel prepared in this study has great potential for development for use in the biomedical field.
